通过细胞学和分子标记技术对切花菊‘长紫’辐照后代进行分析,探讨辐射诱变机理,为菊花辐射育种提供理论依据。结果发现,辐照后代在减数分裂后期Ⅰ和后期Ⅱ出现落后染色体和染色体桥的异常率明显高于对照,且随着剂量升高异常率增加,两... 通过细胞学和分子标记技术对切花菊‘长紫’辐照后代进行分析,探讨辐射诱变机理,为菊花辐射育种提供理论依据。结果发现,辐照后代在减数分裂后期Ⅰ和后期Ⅱ出现落后染色体和染色体桥的异常率明显高于对照,且随着剂量升高异常率增加,两种异常现象发生的最高比率在后期Ⅰ分别为9.0%和11.3%,后期Ⅱ为15.5%和8.6%。用21个ISSR引物扩增得到多态性条带112条,多态性比率为71.3%,表明DNA在不同程度上发生了变异。通过UPGMA法将它们分为5类,聚类结果基本上与花型和花色的变异类型相关。 展开更多

微型凝视高光谱成像仪可以同时获取两个空间维度和一个光谱维度图像,且仅记录整个高光谱图像的外方位元素而不是记录每一帧图像的外方位元素,避免了扫描时的几何不稳定性,有效解决了小型线扫式高光谱成像仪成像几何变形大的问题,适合于... 微型凝视高光谱成像仪可以同时获取两个空间维度和一个光谱维度图像,且仅记录整个高光谱图像的外方位元素而不是记录每一帧图像的外方位元素,避免了扫描时的几何不稳定性,有效解决了小型线扫式高光谱成像仪成像几何变形大的问题,适合于姿态不稳定小型无人机负载平台。目前,研究大多集中于线扫式高光谱成像仪辐射校正方法。凝视型高光谱成像仪应用时间较短,国内外没有较为成熟的数据处理研究,阻碍了无人机微型凝视高光谱成像系统的应用。本文研究了无人机载微型凝视高光谱成像仪辐射响应线性度特性和辐射响应变异性的校正方法,并定量评估该方法的有效性。结果表明,辐射响应变异性校正前,高光谱图像存在明显的渐晕效应和条带现象,校正后,不同波段中像元的辐射响应变异系数显著下降,且渐晕效应和图像条带明显减少。本文提出了基于多目标辐射定标方法,并通过比较定标后的高光谱图像光谱与地面光谱仪实测地物光谱来验证辐射定标的精度。结果表明,多目标辐射定标方法的定标结果表现出更好的效果,特别对于近红外波段,与光谱仪实测的地物反射率差异较小。 展开更多

[Method] Ripe pollen grains of upland cotton were irradiated by 60Co-γ Ray with doses of 5, 10, 15, and 20 Gy, respectively, to learn radiation effect and select appropriate dose. [Result] Most properties of M1 obviously showed variation when dose was over 10 Gy; vitality, growth, and fertility were greatly inhibited when dose was 15 Gy which was almost semi-lethal concentration, and variation species were richest at the same time, which provided materi- als for practical breeding. [Conclusion] 60Co-γ Ray of 15 Gy is more suitable for mutagenesis research on ripe pollen grains of upland cotton. 展开更多

A novel unselective regrowth buried heterostructure long-wavelength superluminescent diode (SLD) with a graded composition bulk InGaAs active region is developed by metalorganic vapor phase epitaxy (MOVPE). At a 150mA injection current, the full width at half maximum of the emission spectrum of the SLD is about 72nm, ranging from 1602 to 1674nm. The emission spectrum is smooth and flat. The ripple of the spectrum is less than 0.3dB at any wavelength from 1550 to 1700nm. An output power of 4.3mW is obtained at a 200mA injection current under continuous-wave operation at room temperature. This device is suitable for the applications of light sources for gas detectors and L-band optical fiber communications. 展开更多

A novel embedded sensor network records changes in key climatic-environmental variables over a range of altitude in the BaekduDaegan Mountain (BDM) of Gangwon Province in Korea, a protected mountain region with unique biodiversity undergoing climate change research. The investigated area is subdivided into three horizontal north-south study areas. Three variables, temperature (T, °C), relative humidity (RH, %), and light intensity (LI, lumens m-2, or lux, lx), have been continuously measured at hourly intervals from June, 2olo to September, 2011 using HOBO H8 devices at lO fixed study sites. These hourly observations are aggregated to monthly, seasonal and annual mean values, and results are summarized to inaugurate a long-term climate change investigation. A region wide T difference in accordance with altitude, or lapse rate, over the interval is calculated as o.4°C l00 m-1. T lapse rates change seasonally, with winter lapse rates being greater than those of summer. RH is elevated in summer compared to other seasons. LI within forestland is lower during summer and higher during other seasons. The obtained results could closely relate to the vegetation type and structure and the terrain state since data loggers were located in forestland. 展开更多

Ultraviolet mutagenesis was applied to Nannochloropsis oculata and three mutants resistant to 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) were isolated. The cellular chlorophyll a and total lipid content of the wild are higher in the medium supplemented with DCMU than in the control without DCMU. Without DCMU, the growth rates and chlorophyll a contents of the mutants are similar to those of the wild. Significant changes of fatty acid content and composition have occurred in DCMU-resistant mutants growing in the medium supplemented with DCMU. The total lipid, palmitic acid (16∶0), palmitoleic acid (16∶1ω9) and oleic (18∶1ω9) contents decrease significantly, while the vaccenic acid (18∶1ω11) increases significantly and the EPA content of dried powder increases slightly in the mutants. The study may provide a basis to improve EPA content in Nannochloropsis oculata in the future. 展开更多

A rice initiation-type lesion mimic mutant (imi) was identified, which was isolated from an indica rice Zhongxian 3037 through γ radiation mutagenesis. Trypan blue staining and sterile culture revealed that the mutant spontaneously developed lesions on the leaves in a develop mentally regulated and light-dependent manner. Genetic analysis indicated that the lesion mimic trait was controlled by a single resessive locus. Using public molecular markers and an F2 population derived from lmi and 93-11, we mapped the lmi locus to the short arm of chromosome 8, nearby the centromere, between two SSR markers RM547 and RM331. The genetic distance was 1.2 and 3.2 Cm, respectively. Then according to the public rice genomic sequence between the two SSR markers, lmi was further finely tagged by three CAPS markers: C4135-8, C4135-9 and C4135-10. And lmi locus was a co-segregated with marker C4135-10, providing a starting point for imi gene cloning. 展开更多
