To elucidate the physiological function and fundamental mechanism,a goat genomic library, in which plague titer is 1\^15×10\+6 pfu/ml, was constructed. Then, with a probe derived from bovine EGP cDNA, the genomic library was screened and a 18 kb fragment containing the gEGP gene was obtained. After PCR with the primers based on the ovine EGP cDNA, two fragments of gEGP gene were obtained and then sequenced. The predicted exon sequence of gEGP gene shows 99 6%, 96 3%, 89 2%, 88 4%, 88 0%, 87 4%, 84 3%, 74 8% homology with the EGP cDNA of sheep, cow, pig, baboon, rhesus, human, hamster, mouse, respectively. Like the other members of EGP gene family, gEGP gene also has many properties of the chitinase super family, and demonstrates the phylogenetic and functional relativity of EGP family with chitinase family.展开更多

One-and two dimensional SDS-PAGE and Western blotting with monoclonal antibody or peanut agglutinin were used to characterize the glycoprotein,The specificity of the marking agents enabled the study to proceed without purification. EGP was found to include two proteins: one is acidic with a pH of 4.5 while the other is basic with a pH of 8.0 The molecular weights of their subunits are the same, that is 110 kD. This study may shed light on the mechanism of reproductive Process and be helpful to the development of a more cffective culture medium for in vitro culture of early embryo.展开更多

Sequences from chimpanzee, ora...DNA was extracted from lymphocytes of primates and PCR was used to amplify the estrous-associated glycoprotein DNA fragment. 537 kb DNA fragments from crab eating macaque were detected. Sequences from chimpanzee, orangutan and human were also amplified, but they were shorter than predicted. We were unable to amplify sequences from spider monkey, salmon, several rodents, bandicoot, sheep and pig . Analysis of the restriction map revealed several conserved RE sites in the amplified sequences of the primates. The results support the view that monkey has more close relationship with human than any other species at molecular level.展开更多
