背景:高尔基体膜蛋白1在人体组织中广泛存在,它的异常表达与癌症、病毒感染等多种疾病密切相关。研究表明,高尔基体膜蛋白1可调控一些纤维化细胞因子,但关于其在肾纤维化中的作用尚不清楚。目的:研究高尔基体膜蛋白1对单侧输尿管梗阻小... 背景:高尔基体膜蛋白1在人体组织中广泛存在,它的异常表达与癌症、病毒感染等多种疾病密切相关。研究表明,高尔基体膜蛋白1可调控一些纤维化细胞因子,但关于其在肾纤维化中的作用尚不清楚。目的:研究高尔基体膜蛋白1对单侧输尿管梗阻小鼠肾纤维化的影响。方法:选取6-8周龄的高尔基体膜蛋白1敲除小鼠(KO小鼠)和C57BL/6野生型小鼠(WT小鼠)各12只,分别对小鼠左侧肾脏行单侧输尿管结扎(单侧输尿管梗阻)手术,其右侧肾脏作为对照。单侧输尿管梗阻术后4 d取小鼠肾脏,采用苏木精-伊红染色和Masson染色观察肾组织病理、肾纤维化程度;流式细胞术检测肾脏巨噬细胞浸润比例;qRT-PCR检测肾组织细胞外基质成分及炎症因子mRNA表达。实验方案经上海交通大学动物实验伦理委员会批准。结果与结论:①WT-单侧输尿管梗阻组和KO-单侧输尿管梗阻组均可见明显肾损伤和肾纤维化,细胞外基质成分Ⅰ型胶原蛋白α1和纤维连接蛋白表达量显著升高,且KO-单侧输尿管梗阻组肾损伤、肾纤维化程度及细胞外基质表达量均高于WT-单侧输尿管梗阻组(P<0.05);WT-对照组和KO-对照组肾脏均未出现明显的肾损伤、肾纤维化及细胞外基质沉积;②与WT-单侧输尿管梗阻组相比,KO-单侧输尿管梗阻组的巨噬细胞浸润比例显著增多(P<0.05),其炎症因子白细胞介素6、肿瘤坏死因子α、白细胞介素1β以及趋化因子CCL5、单核细胞趋化蛋白1 mRNA表达明显升高(P<0.05)。WT-对照组和KO-对照组肾脏巨噬细胞浸润较少,炎症递质表达量低;③结果说明,高尔基体膜蛋白1在肾纤维化发展中起保护作用,其机制可能与巨噬细胞浸润、炎症递质的调控有关。 展开更多

Bilateral ureteral obstruction in children is a rare condition arising from several medical or surgical pictures.It needs to be promptly suspected in order to attempt a quick renal function recovery.In this paper we concentrated on uncommon causes of obstruction,with the aim of giving a summary of such multiple,rare and heterogeneous conditions joint together by the common denominator of sudden bilateral ureteral obstruction,difficult to be suspected at times.Conversely,typical and well-known diseases have been just run over.We considered pediatric cases of ureteral obstruction presenting as bilateral,along with some cases which truly appeared as single-sided,because of their potential bilateral presentation.We performed a review of the literature by a search on Pub Med,Cross Ref Metadata Search,internet and reference lists of single articles updated to May 2014,with no time limits in the past.Given that we deal with rare conditions,we decided to include also papers in non-English languages,published with an English abstract.For the sake of clearness,we divided our research results into 8 categories:(1) urolithiasis;(2) congenital urinary tract malformations;(3) immuno-rheumatologic causes of ureteral obstruction;(4) ureteral localization of infections;(5) other systemic infective causes of ureteral obstructions;(6) neoplastic intrinsic ureteral obstructions;(7) extrinsic ureteralobstructions; and(8) iatrogenic trigonal obstruction or inflammation.Of course,different pathogenic mechanisms underlay those clinical pictures,partly wellknown and partly not completely understood. 展开更多

目的研究丹皮酚对单侧输尿管阻梗后(UUO)小鼠肾纤维化、炎症以及细胞凋亡的抑制作用。方法通过单侧输尿管结扎术建立小鼠UUO模型,通过Masson染色研究UUO后小鼠肾脏组织中间质性胶原的沉积情况。通过免疫组化和免疫荧光实验研究小鼠肾组... 目的研究丹皮酚对单侧输尿管阻梗后(UUO)小鼠肾纤维化、炎症以及细胞凋亡的抑制作用。方法通过单侧输尿管结扎术建立小鼠UUO模型,通过Masson染色研究UUO后小鼠肾脏组织中间质性胶原的沉积情况。通过免疫组化和免疫荧光实验研究小鼠肾组织中胶原蛋白等纤维化相关蛋白的沉积情况,并且研究丹皮酚对UUO中炎症和肾脏细胞凋亡的影响。结果丹皮酚可明显降低UUO后小鼠肾组织中间质性胶原的沉积,降低胶原蛋白和纤维粘连蛋白在细胞质基质中的沉积。丹皮酚可明显降低肾组织中CD68和CD11b以及促凋亡因子Bax的表达,并可增加抗凋亡因子Bcl-2的含量。结论在本试验条件下,一定剂量的丹皮酚可明显降低UUO后小鼠肾脏中纤维化水平,抑制其炎症和细胞凋亡。 展开更多

Effect of salvianolic acid A and C compatibility on inflammatory cytokines in rats with unilateral ureteral obstruction 被引量:10

OBJECTIVE:To investigate the effect of salvianolic acid A and C component molecules,which are involved in drug compatibility,on inflammatory cytokine expression that affects human chemokine ligand 5(CCL5) and chemokine ligand 10(CXCL10)levels in rats with unilateral ureteral obstruction(UUO).METHODS:Fifty Sprague Dawley rats were randomly divided into five groups:normal,model,salvianolic acid A,salvianolic acid C and salvianolic acid A and C groups.The normal group was used as the control,and the other groups of rats had a UUO model established.The control group had free access to food and water,and the other groups received the corresponding drugs for 2 weeks.After the last administration,urine β_2-microglobulin(β_2-MG) and N-acetyl-β-D-glucosaminidase(NAG) levels were analyzed.After 24 h,all rats were sacrificed and the serum was analyzed for creatinine(Cr) and blood urea nitrogen(BUN) levels.Rat kidneys were removed,and CCL5 and CXCL10 inflammatory cytokine mRNA expression was measured using real-time fluorescent quantitative reverse transcription-polymerase chain reaction(RT-PCR).Kidney fibrosis was observed using hematoxylin-eosin(HE)staining and Masson trichrome staining.RESULTS:In the salvianolic acid A and salvianolic acid C treatment groups,serum Cr and urine NAG levels were significantly lower than in the model group(both P < 0.05).In all treatment groups,urine pYMG levels were significantly lower than in the model group(all P < 0.05).Compared with model group,the pathological changes and collagen deposition improved to varying degrees(both P <0.05).CCL5 and CXCL10 mRNA expression decreased to different degrees compared with the model group(both P < 0.05).CONCLUSION:Salvianolic acid A and C are component molecules of drug compatibility,and they may protect renal function and improve tubular function and renal pathology to a certain degree in UUO.This improvement may be related to a reduction in inflammatory cytokines CCL5 and CXCL10 secretion in the UUO rat kidney. 展开更多
