[Objective]The aim was to construct the fusion gene expression vector which consisted of GFP and TP gene of GGPPS from the Ginkgo biloba L.[Method]The transit-peptide(TP) sequence of GGPPS from cDNA of Ginkgo biloba...[Objective]The aim was to construct the fusion gene expression vector which consisted of GFP and TP gene of GGPPS from the Ginkgo biloba L.[Method]The transit-peptide(TP) sequence of GGPPS from cDNA of Ginkgo biloba L.was successfully cloned by using DNA recombination technology,which was then linked to the efficient plant expression vector p1304 + to construct the fusion gene expression vector p1304 +-TP.Then engineering strain EHA105-p1304 +-TP was constructed by transformed p1304 +-TP to Agrobacterium rhizogenes EHA105 using freeze-thaw method.[Result]The fusion gene expression vector which consisted of GFP and TP gene of GGPPS from the Ginkgo biloba L.and engineering strain EHA105-p1304 +-TP were successfully constructed.[Conclusion]It lays a foundation for further study of subcellular localization of TP transit peptide,which can help to clarify the molecular mechanism of a key step in biosynthesis of ginkgolides precursors,and also provides an important basis for the research on metabolic engineering of ginkgolide.展开更多

The structure of the composite was characterized by Fourier transform infrared spectroscopy, UV-Visible, and X-ray diffraction, which indicated that PANI and face-centered-cubic silver were synthesized under γ-ray irradiation. The reaction mechanism were discussed, which revealed that the PANI was formed by the reaction of aniline cation radicals formed by the reaction of aniline cation and -OH, and Ag was formed by the reaction of Ag+ and eaq. The morphology of the composite consisted of PANI nanofibers and Ag nanoparticles, and the mechanism of the morphology formation was discussed, which revealed that the rapid mixing like polymerization process might play an important role. It was revealed that the transport behavior of the composite well fitted with the variable-range-hopping model in 80-300 K and deviated from the model below 80 K.展开更多

The Qinghai-Tibet Railway(QTR) passes through 281 km of sandy land, 11.07 km of which causes serious sand damage to the railway and thus, the control of blown sand is important for the safe operation of the railway. C...The Qinghai-Tibet Railway(QTR) passes through 281 km of sandy land, 11.07 km of which causes serious sand damage to the railway and thus, the control of blown sand is important for the safe operation of the railway. Construction of the railway and sand prevention system greatly changed the blown sand transport of the primary surface. Effective and feasible sand-control measures include stone checkerboard barriers(SCBs), sand fences(SFs), and gravel coverings. This study simulated the embankments, SCBs and SFs of the QTR in a wind tunnel, and analyzed their respective wind profile, sand deposition, and sand-blocking rate(SBR) in conjunction with field data, aiming at studying the influence of Golmud-Lhasa section of the QTR and sand prevention system on blown sand transport. The results of wind tunnel experiments showed that wind speed increased by 67.7%–77.3% at the upwind shoulder of the embankment and decreased by 50.0%–83.3% at upwind foot of embankment. Wind speed decreased by 50.0%–100.0% after passing through the first SF, and 72.2%–100.0% after the first row of stones within the first SCB grid. In the experiment of sand deposition, the higher the wind speed, the lower the SBR of SCB and SF. From field investigation, the amount of sand blocked by the four SFs decreased exponentially and its SBR was about 50.0%. By contrast, SCB could only block lower amounts of sand, but had a higher SBR(96.7%) than SF. Although, results show that SFs and SCBs along the Golmud-Lhasa section of the QTR provide an obvious sand blocking effect, they lead to the deposition of a large amount of sand, which forms artificial dunes and becomes a new source of sand damage.展开更多

In this study, six workers in silver sport industry participating in the elderly job fair held in Ilsan in 2016 were selected as subjects and interviewed in depth. The sports equipment that the six subjects exhibited and their sales and PR (Public Relations) behaviors during the fair were observed, and they were interviewed about the current status of the sports goods industry, constraints as workers, and solutions to resolve the constraints. A senior sports goods developer analyzed sports goods development and PR process through a case study. The result of this study is as follows. The constraints of silver sport industry environment are constraints on investment and lack of research and development, legal and institutional constraints, and economic constraints. The constraints of the silver sport goods industrial field are anxiety for the production of unproven high-cost goods, constraints on unstable institutions, and constraints from the difficulties in collaboration. The solutions of such problems are as follows. First, it is needed to resolve the high production cost. Second, it is need to resolve the difficulties in delivery. The secular aberration drift observed as the apparent proper motion of extragalactic radio sources is caused by the acceleration of the coordinate origin,the Solar System barycenter.So,the acceleration can be estimate...The secular aberration drift observed as the apparent proper motion of extragalactic radio sources is caused by the acceleration of the coordinate origin,the Solar System barycenter.So,the acceleration can be estimated as a global parameter by using the 30 years global geodetic/astrometric VLBI data.This estimation is independent of any kinematic or dynamic model of the Milky Way or statistics hypothesis.The estimated acceleration in the direction of the Galactic center is (0.80±0.04) cm·s-1 ·yr-1,while the other two components are (0.0-1±0.06) and (0.52±0.05) cm ·s-1 ·yr-1 in the direction along the Solar motion in the Galactic plane and in the direction normal to the Galactic plane,respectively.The estimate of the acceleration independently provides another kinematics parameter without referring to any objects within the Galaxy.展开更多

Neurological examination reve...A 24-year-old male was transferred to the emergency department while being in the state of quadriplegia with a history of performing Parkour sport, which is also called double front flip. Neurological examination revealed that the patient's muscle power was 0/5 at all extremities. The patient did not show any sense of light touch or pain in his extremities. In radiological studies, cervical spine X-ray and CT scan images showed C4-C5 subluxation with bilateral locked facets and spinal cord injury. The results of this very rare case study revealed that exercising Parkour sport without taking into account safety standards could result in irreversible injuries to the cervical spinal cord with fatal outcome.展开更多
