[Objective] To explore a method for the rapid determination of protein con- tent in grains of Panicum miliaceum L. [Method] The near infrared transmittance spec- troscopy (NITS) was used to build the mathematical models for the quantitative analy- sis of protein content in the grains. Four combinations of treatment that first derivative and second derivative were respectively combined with partial least squares (PLS) and modified partial least squares (MPLS) were set to compare their effects on the original transmission spectrum. [Result] The predicting effects of the 4 combinations were similar. The optimal combination was first derivative with MPLS, in which the average determination coefficient of validation (RSQ) was 0.880 6, correlation coeffi- cient of cross validation (1-VR) was 0.857 0, standard error of calibration (SEC) was 0.342 4, standard error of cross validation (SECV) was 0.375 1, and the standard er- ror of prediction (SEP) was 0.454. [Conclusion] The constructed NITS model is a rapid way for the determination of protein content in grains of P. miliaceum. 展开更多

Functional Analysis of Homodimer Formation between Movement Proteins in Barley Yellow Dwarf Virus

[Objective] The aim was to investigate the dimer formation between the movement proteins(MP)in barely yellow dwarf virus by using the technology of bimolecular fluorescence complementation technology and to further study the relationship between MP homodimerization and viral movement.[Method] The DNA sequence of bimolecular fluorescent complementary vector containing cloning multiple cloning sites,35S promoter and terminator was cloned into the expression vector pCAMBIA1300,which replicates at a higher copy number in E.coli.Then,the BYDV-MP gene fragment was amplified in the presence of the whole BYDV-PAV cDNA sequence as template and the primers designed according to the BYDV-MP gene sequence from GenBank,cloned into the modified bimolecular fluorescent complementary vectors pCAMBIA1300-NE and pCAMBIA1300-CE.The resulting vectors were transformed into Agrobacterium by electroporation method and infiltrated into the tobacco leaf.Protein interactions were observed under fluorescence microscope.[Result] Yellow fluorescence could be viewed in the leaves co-infiltrated with Agrobacterium carrying pCAMBIA1300NE-MP and pCAMBIA1300CE-MP at 2-5 d post-infiltration,while yellow fluorescence could not be observed in negative control groups.[Conclusion] BYDV-MP formed homodimers in plant cells.The results can provide theoretical basis for further in-depth research about the movement process and mechanism of BYDV. 展开更多

Effects of windbreak width in wind direction on wind velocity reduction 被引量:2

The variations of drag force acting on the windbreak and the bulk drag coefficients for different windbreak widths were studied experimentally in the Eiffel-type non-circulating wind tunnel at the Hydraulic Engineering Laboratory, Saitama University, Japan, to elucidate the effects of windbreak width in the wind direction on wind velocity reduction behind a windbreak. The variations of flow field for different windbreak widths were studied numerically by using the two-dimensional Reynolds-averaged Navier-Stokes (RANS) equation with a k-ε turbulence closure model. Results show that the total drag force to wind increased with increasing windbreak width, but the bulk drag coefficient decreased slightly. The relationship between the bulk drag coefficient Cd and the windbreak width W and height H can be presented by the equation of Cd=kd (W/H)-b (kd, b: constants). The result of the numerical simulation shows that the windbreak width greatly affects the location and the value of the minimum wind velocity. The wind velocity decreased by 15%–22% as the windbreak width increased. 展开更多

In this paper, a novel liquid refractive index (R/) sensor based on fused tapered photonic crystal fiber (PCF) is proposed. It is fabricated by fusing and tapering a section of PCF which is spliced with two single-mode fibers (SMFs). Due to the fused biconical taper method, the sensor becomes longer and thinner, to make the change of the outside RI has more direct effects on the internal optical field of the PCF, which finally enhances the sensitivity of this sensor. Experimental results show that the transmission spectra of the sensor are red-shifted obviously with the increase of RI. The longer the tapered region of the sensor, the higher the sensitivity is. This sensor has the advantages of simple structure, easy fabri- cation, high performance and so on, so it has potential applications in RI measurement. 展开更多

By citrate sol-gel auto-combustion method,the nanophase M-type planar hexagonal ferrite is prepared.The transmission electron microscopy(TEM),X-ray diffraction(XRD) and thermal analysis are used to study the grain size,phase composition,microstructure and crystallization process.The results show that the nanophase M-type Sr-ferrite prepared by this method is single,and its grain size is smaller than 100 nm.Moreover,most of the grains present hexagonal sheet shape.Tests are carried out for its attenuation to 1.06 mm laser.It is found that the extinction capability of the nanophase M-type Sr-ferrite smoke is good,and its mass extinction coefficient is 1.628 m2/g. 展开更多

The detailed theoretical derivation to express the optical transform principle of the proposed method is presented. And excellent experimental results are demonstrated. It is shown that this approach works well and simplifies the experimental facility effectively, especially reducing the optical system length to half of that of the conventional method. In addition, the proposed method can realize the beam propagation trajectory control of Airy beam and generate Airy beam array. 展开更多
