酶萃取及组氨酸Heyns化合物加香对再造梗丝品质的影响

为改善再造梗丝产品香气平淡、杂气较多的问题,对其原有制备工艺进行优化调整,具体为以组合酶萃取代替温水萃取,辅以组氨酸Heyns化合物(His-H)进行回填加香,并采用核磁共振波谱(NMR)技术、气相色谱-质谱联用法等对工艺调整前后再造梗丝... 为改善再造梗丝产品香气平淡、杂气较多的问题,对其原有制备工艺进行优化调整,具体为以组合酶萃取代替温水萃取,辅以组氨酸Heyns化合物(His-H)进行回填加香,并采用核磁共振波谱(NMR)技术、气相色谱-质谱联用法等对工艺调整前后再造梗丝中试产品常规化学成分、细胞壁主要组分和感官品质的变化进行探讨。结果表明:萃取时使用的最优溶液应为质量分数0.8%E_(c)和质量分数0.4%E_(p)的组合酶溶液,萃取后产品质软色浅;回填阶段添加质量分数0.25%的His-H后,再造梗丝香气质提升,杂气减少,刺激性下降,感官品质整体优于对照梗丝;工艺调整后再造梗丝中试产品的常规化学成分保持协调,中性香气物质总含量提升约27.0%,细胞壁主要组分(纤维素、果胶和木质素)含量降低率超过15.0%,所制卷烟烟气透发性优于对照品,木质气下降,香气量明显增加,吸食品质明显提升。 展开更多

目的:研究经酶解-微波辅助萃取、超滤分级纯化的灰树花子实体多糖的免疫活性。方法:采用复合酶解结合微波辅助萃取方法提取灰树花子实体多糖,采用超滤工艺对提取多糖进行分级纯化;采用MTT法研究多糖对小鼠脾淋巴细胞、ConA/LPS诱导的小... 目的:研究经酶解-微波辅助萃取、超滤分级纯化的灰树花子实体多糖的免疫活性。方法:采用复合酶解结合微波辅助萃取方法提取灰树花子实体多糖,采用超滤工艺对提取多糖进行分级纯化;采用MTT法研究多糖对小鼠脾淋巴细胞、ConA/LPS诱导的小鼠脾淋巴细胞的增殖作用;建立荷瘤小鼠模型,研究灰树花多糖对肿瘤的抑制作用。结果:获得分子质量范围为<50kD的GFP-I、50~100kD的GFP-II和100~200kD的GFP-III;与RPMI-1640对照组相比,GFP-I、GFP-II和GFP-III对淋巴细胞增殖率分别增加26.558%~74.167%(P<0.001)、20.175%~152.075%(P<0.001)和26.473%~334.797%(P<0.001);对ConA诱导的淋巴细胞增殖率分别增加31.408%~122.894%(P<0.001)、53.075%~235.693%(P<0.001)和83.347%~367.241%(P<0.001);对LPS诱导的淋巴细胞增殖率分别增加20.526%~97.075%(P<0.001)、38.288%~211.348%(P<0.001)和54.454%~344.883%(P<0.001);相对于生理盐水(NS)组,在实验剂量范围内,GFP-I、GFP-II和GFP-III对S180肿瘤的抑制率分别达到10.15%~22.94%(P<0.001)、12.79%~30.10%(P<0.001),23.86%~51.65%(P<0.001)。结论:经复合酶-微波辅助萃取、超滤分级纯化后多糖具有显著免疫活性和抗肿瘤活性,其中GFP-III具有比GFP-I和GFP-II更强的免疫活性和抗肿瘤活性,而且活性与其浓度正相关,多糖可能以对T淋巴细胞增殖的促进作用为主。 展开更多

酶解辅助萃取法提取番茄红素以及D101大孔树脂动态分离纯化

番茄红素抗氧化能力极强。而番茄种植普遍,番茄中的番茄红素含量高且品质上乘,是公认的理想提取材料。本实验选用番茄为实验原料,采用酶解辅助萃取法提取番茄红素。在此基础上,用大孔树脂动态分离纯化获得番茄红素提取液并利用DPPH·... 番茄红素抗氧化能力极强。而番茄种植普遍,番茄中的番茄红素含量高且品质上乘,是公认的理想提取材料。本实验选用番茄为实验原料,采用酶解辅助萃取法提取番茄红素。在此基础上,用大孔树脂动态分离纯化获得番茄红素提取液并利用DPPH·探究番茄红素的抗氧化能力。研究结果表明:纤维素酶和果胶酶混合物(混合质量1∶1)加入量为0.025g/g(以1g新鲜番茄质量计),在45.7℃恒温反应1.5h,接着采用乙酸乙酯溶液对其浸取1h以得番茄红素粗提液,番茄红素得率可达0.801μg/g。动态分离纯化番茄红素吸附剂为D101大孔吸附树脂,洗脱液为乙酸乙酯,最佳洗脱时间为55min30s,最终番茄红素得率可达2.496μg/g,有较明显纯化效果,纯化倍数为3.12。番茄红素对DPPH·清除率较高,最高可达84.15%,验证了番茄红素的抗氧化能力。研究结果为番茄红素的提取与分离纯化提供了新思路。 展开更多

In this study, the effects of hot water temperature on clarity, total soluble solids, polyphenol oxidase (PPO) and color of banana juice during hot water ex-traction were discussed based on the theory in which heat treatment might induce the pectin and protein in banana pulp to form insoluble products. The results showed the hot water temperature had a significant effect on the formation of insol-uble polymers in banana pulp from pectin and protein. In 75 ℃ water, the pectin and protein in banana pulp were most inclined to form insoluble products. Under this condition, the clarity of banana juice was also highest. The light transmittance at 660 nm was close to 90%. In the banana juice, extracted by 75 ℃ water, the pectin and protein contents were lowest, and they were lower than 7.3 mg/100 ml and 12.9 mg/100 ml respectively. The 75 ℃ water could not inactivate completely the pectin in banana pulp due to its high heat resistance, Therefore, 0.05% L-cys-teine or ascorbic acid needed to be added into banana pulp to inhibit the browning of juice induced by residual PPO. 展开更多

Factors Affecting Trypsin Extraction by AOT Reversed Micelles and Observation by STM 被引量:3

In this article, the influence factors of trypsin extracted from crude pancreatin was investigated, and scanning turmeling microscope(STM) was used to observe the image of trypsin in butane-diacid-2-ethyl-hexyl-ester-sulfonic sodium (AOT)/iso-octane reversed micelles. The STM image showed that trypsins bounded in reversed micelles was rigid, which weakened its conjugative effect and caused maximum ultraviolet absorption and fluorescence emissive absorption moving toward blue waves. AOT concentration, pH and cations were the main influence factors of extraction. Specifically, extraction percentage of trypsin decreased with the increase of AOT concentration from 0.01 to 0.1mol·L^-1. When pH value is from 5.30 to 10.0, i.e. less than pI of trypsin, the extraction percentage is raised with the different increase of pI-pH, but when the pH value is less than 5.20, the extraction percentage is decreased with the acidity added. Besides, the extraction efficiency is negative, related with the concentrations of Ca^2+, Na^+, K^+ which were in the range of 0.2-1.0mol.L^-1, and influence of concentration of Ca^2+ is greater than that of Na^+, and K^+ which has the minimum impact with the same concentration. Finally, optimum conditions to extract trypsin were: AOT reversed micelles 0.05mol·L^-1, trypsin concentration in crude pancreatin solution 3mg·ml^-1, pH 5.2-- 5.3, ratio (by volume) of extraction phase to strip-extraction phase 1 : 1, and time of 5min. The corresponding percentage of extraction was 22.7% and specific activity was 78.9 N-benzoyl-L-arginlne ethyl ester (BAEE) U·mg^-1 protein, three times than that in crude pancreatin. There was no lipase and amylopsin activity was decreased to 1/5 of crude pancreatin. Partly purifying solution was treated by condition mentioned above with 0.05mol·L^-1 ceryl-trimethyl-ammonium bromide (CTAB), total extraction percentage of trypsin was 74.18% and specific activity was 3148.3 BAEE U·mg^-1, i.e. 48.16 times purer than that in crude pancreatin. Through sodium dodecyl sulfate-polyacryl amide gel electrophoresis (SDS-PAGE) and image analysis of extracted product, there were only three bands in the trypsin, while seven in crude pancreatin, and electrophoresis location of main bend was almost identical with the standard enzyme. 展开更多

The procedure of enzymatic aqueous extraction of soybean oil was assessed when two-step controlled enzymatic hydrolysis was applied. With aqueous extraction of soybean oil-containing protein, the highest yield of oil was 96.1% at the optimized conditions studied. Soybean oil-containing protein was hydrolyzed and resulted in releasing part of oil. The separated protein that contained 40% oil was enriched due to its adsorption capacity of released oil, the average oil extraction yeild reached 93.5%. Then the high oil content protein was hydrolyzed again to release oil by enzyme, the oil extraction yeild was 80.4%. As a result, high quality of soybean oil was obtained and the content of total oil yield was 74.4%. 展开更多

Eyestalk cDNA was obtained from total RNA by reverse transcription. Reverse transcriptase polymerase chain reaction (RT PCR) was initiated using eyestalk cDNA and degenerate primers designed from the amino acid sequence of molt inhibiting hormone from shrimp Penaeus japonicu s. A specific cDNA was obtained and cloned into a T vector for sequencing. The cDNA consisted of 201 base pairs and encoding for a peptide of 67 amino acid residues. The peptide of P. chinensis had the highest identity with molt inhibiting hormones of P. japonicus . The cDNA could be a partial gene of molt inhibiting hormones from P. chinensis . This paper reports for the first time cDNA encoding for neuropeptide of P. chinensis . 展开更多
