The experiment was conducted in cubic indoor fiberglass tanks, each holding 700 L in triplicate. Post-larvae with an average weight of 20.8 ± 0.20 mg were stocked at 80 PL m2. Five experimental isocaloric (15.06 MJ kgl digestible energy), and isonitrogenous (30.45% digestible protein) diets were formulated by blending of soybean oil and linseed oil to containing five dietary LA/LNA ratios (7.80, 2.75, 1.28, 0.65 and 0.30). The highest survival values were recorded for prawn PL fed diet containing 0.65 LA/LAN ratios. Growth indices of PL significantly increased (P 〈 0.05) with decreased dietary LA/LAN ratios to 0.65. The same trend was observed for the highest (P ≤ 0.05) protein efficiency ratio, protein productive value, fat retention, energy retention and best feed conversion ratio. The total whole tissue polyunsaturated fatty acid composition of M. rosenbergii PL was dominated by LA followed by LAN. Post larvae fed the diets containing higher LA/LNA ratios showed a higher tissue LA/LNA ratio. The obtained findings revealed that fatty acid patterns ofM. rosenbergii PL were influenced by fatty acid profiles of diets. METHODS: Using cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B(1), D(1), p16(ink4a) and p21(cip/waf1) of SGC-7901 cells which were treated with various c9, t11-CLA concentrations (25, 50, 100 and 200 micromol.L(-1))of c 9, t 11-CLA for 24 and 48h, with a negative control (0.1% ethane). RESULTS: The cell growth and DNA synthesis of SGC-7901 cells were inhibited by c9, t11-CLA.SGC-7901 cells. Eight day after treatment with various concentrations of c9, t11-CLA mentioned above, the inhibition rates were 5.92%, 20.15%, 75.61% and 82.44%, respectively and inhibitory effect of c9, t11-CLA on DNA synthesis (except for 25 micromol.L, 24h) showed significantly less (3)H-TdR incorporation than that in the negative controls (P【0.05 and P【0.01). Immunocytochemical staining demonstrated that SGC-7901 cells preincubated in media supplemented with different c9, t11-CLA concentrations at various times significantly decreased the expressions of PCNA (the expression rates were 7.2-3.0%, 24h and 9.1-0.9% at 48h, respectively), Cyclin A (11.0-2.3%, 24h and 8.5-0.5%,48h), B(1) (4.8-1.8% at 24h and 5.5-0.6% at 48h)and D(1) (3.6-1.4% at 24h and 3.7%-0 at 48h) as compared with those in the negative controls(the expressions of PCNA, Cyclin A, B(1) and D(1) were 6.5% at 24h and 9.0% at 48h, 4.2% at 24h and 5.1% at 48h, 9.5% at 24h and 6.0% at 48h,respectively)(P【0.01), whereas the expressions of P16(ink4a) and P21(cip/waf1), cyclin-dependent kinases inhibitors(CDKI), were increased. CONCLUSION: The cell growth and proliferation of SGC-7901 cell is inhibited by c9, t11-CLA via blocking the cell cycle, with reduced expressions of cyclin A,B(1) and D(1) and enhanced expressions of CDKI(P16(ink4a) and p21(cip/waf1)).展开更多

Methods GPR40/FFA1 expression was detected by immunofluorescence imaging. We employed linoleic acid (LA), a free fatty acid that has a high affinity to the rat GPR40, and examined its effect on cytosolic free calcium concentration ([Ca2+]i) in primary rat β-cells by Fluo-3 intensity under confocal microscopy recording. Downregulation of GPR40/FFA1 expression by antisense oligonucleotides was performed in pancreatic β-cells, and insulin secretion was assessed by enzyme-linked immunosorbent assay. Results LA acutely stimulated insulin secretion from primary cultured rat pancreatic islets. LA induced significant increase of [Ca2+]i in the presence of 5.6 mmol/L and 11.1 mmol/L glucose, which was reflected by increased Fluo-3 intensity under confocal microscopy recording. LA-stimulated increase in [Ca2+]i and insulin secretion were blocked by inhibition of GPR40/FFA1 expression in β-cells after GPR40/FFA1-specific antisense treatment. In addition, the inhibition of phospholipase C (PLC) activity by U73122, PLC inhibitor, also markedly inhibited the LA-induced [Ca2+]i increase. Conclusion LA activates GPR40/FFA1 and PLC to stimulate Ca2+ release, resulting in an increase in [Ca2+]i and insulin secretion in rat islet β-cells.展开更多

Methods: The inhibitory rate, cell growth curve and apoptotic morphological observation of MCF-7 cells were obtained by MTT assay, trypan blue staining and Hoechst33342 fluorescence staining. The apoptotic rate and cell cycle were detected with flow cytometry. Transcriptional level of genes was detected with RT-PCR semi-quantitative method, and Western blot was performed to detect proteins levels. Results: The two CLA isomers could reduce cell proliferation (P 〈 0.05), increase apoptotic rate (P 〈 0.05), and increase obviously the transcriptional and protein levels of PPARy (P 〈 0.01). The synchronism and correlation between the effects of CLA to PPARy and apoptotic proteins Bax, Bcl-2, Caspase 3 changes were found with the dose- and time-dependent manners. There was cooperative relation between the levels of PPARy and the rates of Bax/Bcl-2, Caspase 3 (small fragment) by experiments of PPARy inhibitor GW9662 and ligand Rosiglitazone. Conclusion: The apoptotic pathway of PPARy-Bcl-2-Caspase 3 signaling was found. The C9, t11-CLA and tl0, c12-CLA could inhibit MCF-7 cell proliferation and promote apoptosis via activating PPARy-Bcl-2-Caspase 3 pathway. CLA may be a kind of activator of PPARv.展开更多

The lipase Novozym 435 possessed the highest activity for the conjugated linoleic acid esterification during the synthesis of triglycerides. The triglycerides were synthesized by esterification of glycerol and conjugated linoleic acid (CLA) in a no-solvent system using lipase catalysis. We investigated the effects of temperature, enzyme concentration, water content, and time on esterification. Enzyme and water concentrations of up to 1% of the total reaction volume and a system temperature of 60℃ proved optimal for esterification. Similarly, when the esterification was carried out for 24 h, the reaction ratio improved to 94.11%. The esterification rate of the rotating screen basket remained high (87.28%) when the enzyme was re-used for the 5th time. We evaluated the substrate selectivity of lipase (NOVO 435) and determined that this lipase prefers the 10,12-octadacadienoic acid to the 9,11-octadecadienoic acid.展开更多

[Meth- od] Microwave power, microwave mode of action, the amount of catalyst and solvent were selected for single-factor-experiment. Then, based on the single-factor-experiment, the catalyst and solvent amount, and microwave function time were chosen for orthogonal experiment so as to deter- mine the optimal craft of preparing CLA from corn germ oil. UV absorption spectra and infrared absorption spectrum were used to analyse the struc- tural features of natural corn germ oil and CLA. [Resalt] The optimal preparation condition was as follow: the catalyst was 0.6 times heavier than oil, solvent was 3 times heavier than oil weight, microwave worked for ten minutes. Under the optimal condition, CLA was 56.3% and conversion rate was 99.47 %. The results indicated that non-conjugated double bonds of linoleic acid can be isomerized and changed into conjugated double bonds under the conditions of microwave irradiation,[ Conduslon] The study provided a new way for the preparation of CLA.展开更多

The purpose of this work was the study of the oxidative stability from different methods and the antioxidant content of the chia oils compared with other commercial oils. The oxidative stability of the oils was determined based on the oxidative stability index test (OSI test) conducted at 110 ~C and isothermal as well as non-isothermal differential scanning calorimeter (non-isothermal DSC) for the chia oil and the linseed oil. The OSI induction time of chia oil was compared with that of commercially available linseed, canola, sunflower and high-oleic sunflower oils, chia oil being the least stable oil among those studied. The inherent stability value and oxidazibility for linseed oil was lower than the chia oil A may be ascribed to a lower linolenic acid content of the former. The induction time (It) quotient at 110 ~C calculated for chia oil A and linseed oil were similar, suggesting a high degree of consistency between the results obtained by the two methods. The activation energy and specific reaction rate constant of chia and linseed oils were compared based on the results of isothermal and non-isothermal DSC. An apparent inconsistency in the experimental data results from the temperature-dependence of the activation energy of each fatty acid which can explain because the methods conditions were different.展开更多

Two hundred White Leghorn hens with 30 weeks of age were divided into five treatments with four replicates of l 0 birds each. Treatments were assigned randomly and consisted of 0.00%, 0.75%, 1.50%, 2.25% and 3.00% FO in commercial corn-soybean meal diets, in which the soybean oil was partially replaced. The experiment was conducted for 90 d. The main research findings were that feed intake, egg production rate, egg weight and feed conversion were not influenced by time and dietary treatment. Fatty acid content was significantly altered (P 〈 0.05) by FO, showing a progressive increase in egg n-3 fatty acid (especially docosahexaenoic acids (DHA) and eicosapentaenoic acids (EPA)) when FO was added. Levels of EPA and DHA were higher (P 〈 0.05) in the egg lipids of FO fed hens than those in the control group. However, no significant differences were observed either in egg weight or egg production among groups. The highest incorporation (P 〈 0.05) of total n-3 fatty acid content in eggs was obtained with 3% FO/kg. This increase was proportional to FO inclusion levels in the diets.展开更多

Since the IL can form strong hydrogen bonds with water, trace amount of water is a problematic impurity in the IL. Using the standard addition method (SAM), the densities were measured in the temperature range from 293.15 to 343.15±0.05 K. On the basis of the experimental data of density, the surface tension y, molar volume, and the molecular vol- ume Vm, molar enthalpy of vaporization AIgHm0, and the thermal expansion coefficients or, for [C7mim][Ala] were estimated using semi-empirical methods. Also the interstice model has been verified with good accordance. Then the [C7mim][Ala] was taken as catalyst in the transesterification reaction with fatty acid, where soybean oil was used as raw material and methanol as reactant. The amount of catalyst, alcohol consumption, reaction time, temperature and other factors of the reaction were inves- tigated. Using GC determination, the conversions were calculated by the internal standard method. The main ingredients of the products were identified by GC-MS as palm methyl ester and linoleic acid methyl ester only, which showed high selectivity in palmitic acid and linoleic acid.展开更多

Conclusion LA activates GPR40/FFA1 and PLC to stimulate Ca2+ release, resulting in an increase in [Ca2+]i and insulin secretion in rat islet β-cells.

文摘Objective: The aim of the study was to explore the activities of cis9, trans11-CLA (C9, t11-CLA) and transl0, cis12-CLA (t10, c12-CLA) inhibiting tumor, and investigate their relationships with PPARy and apoptotic proteins, and mechanism of anti-cancer. CLA may be a kind of activator of PPARv.
文摘The GC-MS spectrums of a sample from Amanitaceae family and Mycenaceae family were matched with their close match GC-MS spectrums from the spectrum library. The samples from Amanitaceae family (KMsp027) and Mycenaceae family (KMsp039) were found to contain fatty acid methyl esters (methyl palmitate and methyl linoleate). It is proposed that the presence of methyl linoleate and methyl palmitate in the samples may show antibacterial activities if tested against different bacterial strains.

文摘We compared four esterifiable enzymes. We evaluated the substrate selectivity of lipase (NOVO 435) and determined that this lipase prefers the 10,12-octadacadienoic acid to the 9,11-octadecadienoic acid.

文摘Iminodiacetic acid functionalized polyglycidyl methacrylate grafted-carbon fibers(PGMA-IDA/CFs) were prepared for Ni(II) removal from aqueous solutions. The effects of solution p H value, temperature and adsorption time were investigated. The maximum adsorption capacity of Ni(II) on PGMA-IDA/CFs is 0.923 mmol·L-1· g-1at pH 5.2 and 50 °C. Kinetic data indicate that the adsorption process matches the pseudo-second-order model and Elovich kinetic model. Thermodynamic data suggest that the adsorption process is endothermic spontaneous reaction.
文摘The effect of supplementation of conjugated linoleic acid (CLA) in diets on growth performance and total lactic bacteria (LAB) in small intestine of broiler. One hundred sixty eight broilers (Arbor Acres; 1 wk old) were randomly assigned to 8 groups. Each group (male 21 and female 21 broiler chickens) was further randomly divided into 3 replicates of 7 chickens were assigned to 4 dietary treatments (0%, 0.5%, 1.0%, and 1.5% CLA). Feed consumption and BW were recorded at every 7-day period. On day 56, and each week were collected at ileal content in small intestine for the total count of lactic acid bacteria (LAB). There were highly significant differences in body weight gain, average dairy feed intake, average dairy gain and feed conversion ratio among the treatments (P 〈 0.01). Moreover, our results revealed that the highest detection trend from the ileal occurred during the period in the supplementation of CLA.
文摘The objective of this study was to investigate the effect of Palm Oil By-pass Fat (POBF) on milk composition of early lactation Holstein during dry season, 24 postpartum Holstein (1 d-14 d) were selected and align to two groups according to CRD experimental design; each group included 12 Holstein cows. The control received by-pass fat 0 g/h.d, the experimental group received by-pass fat 300 g/h.d. The results shown, DMI and yield of milk did not be affected with supplementation of by-pass fat, however, milk protein increased by 6.71%, milk fat increased by 8.16%, lactose increased by 0.22%, SNF increased by 1.98%, TS increased 4.08%; digestibility of diet protein decreased by 6.62%, digestibility of EE decreased by 7.96%, digestibility of NDF and ADL decreased by 1.36%, 8.97% respectively, digestibility of DM and OM decreased by 4.42, 3.07 percentage units; amount of C4-C17 milk fatty acids showed a decreased tendency, that of CIs-C22 showed an increased tendency, the milk CLA increased significantly (P 〈 0.01). To sum up, supplementation of by-pass fat for early lactation Holstein is an important measure to balance energy and sustain milk yield and quality.
文摘[Objective] The optimal craft of preparing conjugated linoleic acid from corn germ oil by dint of microwave method was studied. The results indicated that non-conjugated double bonds of linoleic acid can be isomerized and changed into conjugated double bonds under the conditions of microwave irradiation,[ Conduslon] The study provided a new way for the preparation of CLA.
文摘The fatty acid composition as well as the antioxidant composition and content of two commercially available chia oils of different origins were studied. An apparent inconsistency in the experimental data results from the temperature-dependence of the activation energy of each fatty acid which can explain because the methods conditions were different.
文摘The study was aimed to evaluate the effect of replacement time and rate of soybean oil by flax oil (FO) on polyunsaturated fatty acid (PUFA) profile in egg of laying hens. This increase was proportional to FO inclusion levels in the diets.

文摘Amino acid ionic liquid (AAIL) based on alanine, 1-n-heptyl-3-methylimidazolium alanine ([C7mim][Ala]), has been prepared and characterized. The main ingredients of the products were identified by GC-MS as palm methyl ester and linoleic acid methyl ester only, which showed high selectivity in palmitic acid and linoleic acid.
