肿瘤酸度响应性聚(L-赖氨酸)-阿霉素键合药的制备与表征 被引量:3

用丁二酸酐(SA)和顺式乌头酸酐(CA)分别对阿霉素(DOX)进行修饰,得到非酸响应的SA-DOX(SAD)和酸响应的CA-DOX(CAD).通过SAD或CAD、端羧基化的聚乙二醇单甲醚(mPEG-COOH)与聚(L-赖氨酸)(PLL)的缩合反应,制得非酸响应的PLL-g-mPEG/SAD和酸... 用丁二酸酐(SA)和顺式乌头酸酐(CA)分别对阿霉素(DOX)进行修饰,得到非酸响应的SA-DOX(SAD)和酸响应的CA-DOX(CAD).通过SAD或CAD、端羧基化的聚乙二醇单甲醚(mPEG-COOH)与聚(L-赖氨酸)(PLL)的缩合反应,制得非酸响应的PLL-g-mPEG/SAD和酸响应的PLL-g-mPEG/CAD键合药.通过核磁共振氢谱和红外光谱表征键合药的化学结构,并通过紫外-可见分光光度计测定药物键合量.动态激光光散射研究结果表明,两亲性的PLL-DOX键合药可以在pH=7.4的磷酸缓冲溶液中自组装形成稳定的纳米微粒.体外释放实验及噻唑蓝检测结果表明,PLL-g-mPEG/SAD在实验pH范围和时间段内只释放出少量DOX,不具有酸响应特性,且对HeLa细胞增殖抑制作用较小.而PLL-g-mPEG/CAD在生理条件(pH=7.4)下相对稳定,在弱酸性条件(pH=5.3,6.8)下,CAD中酸响应的酰胺键能快速水解并释放出DOX,表现出较强的HeLa细胞增殖抑制效果. 展开更多

研究了色素亮蓝对氢离子浓度大于1mol/L溶液的颜色响应及其变色机理,并将其作为显色剂应用于高酸度试纸的研发。研究表明,固定于改性基纸上的亮蓝处于氢离子浓度为0.1~9.0mol/L范围的介质溶液中时,其颜色变化表现出明显的酸度... 研究了色素亮蓝对氢离子浓度大于1mol/L溶液的颜色响应及其变色机理,并将其作为显色剂应用于高酸度试纸的研发。研究表明,固定于改性基纸上的亮蓝处于氢离子浓度为0.1~9.0mol/L范围的介质溶液中时,其颜色变化表现出明显的酸度响应特征,即随着溶液酸度的提高,亮蓝逐渐由蓝色转化为蓝绿色、绿色、黄绿色直至黄色。该响应具有普适性,不受无机酸的种类与氧化性强弱的影响。采用分光光度法研究了溶液酸度对亮蓝光吸收特性的影响,提出其可能的变色机理。以亮蓝为显色剂开发出高酸度试纸,该试纸可以直接检测溶液中0.1~9.0mol/L范围内的氢离子平衡浓度,精确度为±1mol/L。 展开更多

Indoleamine 2, 3-dioxygenase (IDO) is a rate-limiting enzyme for the tryptophan catabolism. In human and murine cells, IDO inhibits antigen-specific T cell proliferation in vitro and suppresses T cell responses to fetal alloantigens during murine pregnancy. In mice, IDO expression is an inducible feature of specific subsets of dendritic cells (DCs), and is important for T cell regulatory properties. However, the effect of IDO and tryptophan deprivation on DC func- tions remains unknown. We report here that when tryptophan utilization was prevented by a pharmacological inhibitor of IDO, 1-methyl tryptophan (1MT), DC activation induced by pathogenic stimulus lipopolysaccharide (LPS) or inflam- matory cytokine TNF-α was inhibited both phenotypically and functionally. Such an effect was less remarkable when DC was stimulated by a physiological stimulus, CD40 ligand. Tryptophan deprivation during DC activation also regu- lated the expression of CCR5 and CXCR4, as well as DC responsiveness to chemokines. These results suggest that tryptophan usage in the microenvironment is essential for DC maturation, and may also play a role in the regulation of DC migratory behaviors. 展开更多

Some effective parameters on the copper extraction from Kiire chalcopyrite concentrate were optimized by using response surface methodology (RSM). Experiments designed by RSM were carried out in the presence of ammonium persulfate (APS) and different types of impeller in an autoclave system. Ammonium persulfate concentration and leaching temperature were defined numerically and three types of impellers were defined categorically as independent variables using experimental design software. The optimum condition for copper extraction from the chalcopyrite concentrate is found to be ammonium persulfate concentration of 277.77 kg/m3, leaching temperature of 389.98 K and wheel type of impeller. The proposed model equation using RSM has shown good agreement with the experimental data, with correlation coefficients R2 and RaZaj for the model as 0.89 and 0.84, respectively. 展开更多

Optimization for Determination of Esters in Biodiesel Oil by Spectrophotometric Method

The simple spectrophotometric method for the determination of esters has been developed. The procedure is based on the quantitative determination of ferric hydroxamate generated from the reaction of esters, hydroxylamine hydrochloride in basic solution at 120℃, and ferric chloride. The formed complex was monitored through absorbance at 517 nm. The parameters affecting the absorbance value were explored for optimum analytical performance. The optimum conditions were 1.50 mL of 0.5 M hydroxylamine hydrochloride, 0.40 mL of 6 M sodium hydroxide, 1.00 mL of 3 M hydrochloric acid and 0.50 mL of 5 % ferric chloride. The method exhibits linear response up to 7.50% wt. and a detection limit of 0.23% wt. The linear range and working range were used at the same range 1.25 to 7.50% wt. The proposed method can be applied to biodiesel (B 100) for measuring the esters quantity in the percentage unit of biodiesel oil. 展开更多
