为探究菊粉酶降低红肉风险物质N-羟乙酰神经氨酸(Neu5Gc)含量的效果和分子机制,本研究首先采用菊粉酶处理Neu5Gc标准品并对其进行液相质谱检测,其次选择外切型菊粉酶(蛋白数据序列号:1Y9G)和内切型菊粉酶(蛋白数据序列号:3SC7)与Neu5Gc... 为探究菊粉酶降低红肉风险物质N-羟乙酰神经氨酸(Neu5Gc)含量的效果和分子机制,本研究首先采用菊粉酶处理Neu5Gc标准品并对其进行液相质谱检测,其次选择外切型菊粉酶(蛋白数据序列号:1Y9G)和内切型菊粉酶(蛋白数据序列号:3SC7)与Neu5Gc进行分子对接和分子动力学模拟,并以氨基酸虚拟突变验证菊粉酶结合Neu5Gc的关键残基。最后利用密度泛函理论(DFT)和独立密度梯度模型(IGM)分析动力学平衡后Neu5Gc和菊粉酶活性残基复合物的分子间弱相互作用强度与分布。结果表明,外切和内切型菊粉酶对Neu5Gc标准品含量的平均降低率分别为55.33%和43.81%;理论计算表明,Neu5Gc主要以羟基氧、酰胺氮原子与菊粉酶活性位点残基发生弱相互作用而稳定结合到活性口袋中,水环境下对外切和内切菊粉酶的结合能分别为-22.71和-8.46 kJ·mol^-1。本研究结果为菊粉酶的理性设计和在红肉安全性控制中的应用提供了一定的参考。 展开更多

目的探讨川陈皮素(NOB)改善糖尿病认知功能障碍的作用及可能机制。方法 12周龄健康雄性SD大鼠40只,随机选取32只,腹腔注射1%链脲佐菌素(STZ)溶液建立糖尿病大鼠模型;余8只设为正常对照组给予等剂量缓冲液注射。造模成功后,随机分为糖尿... 目的探讨川陈皮素(NOB)改善糖尿病认知功能障碍的作用及可能机制。方法 12周龄健康雄性SD大鼠40只,随机选取32只,腹腔注射1%链脲佐菌素(STZ)溶液建立糖尿病大鼠模型;余8只设为正常对照组给予等剂量缓冲液注射。造模成功后,随机分为糖尿病组(n=8),NOB高剂量组(n=6,30 mg/kg)、中剂量组(n=6,20 mg/kg)和低剂量组(n=6,10 mg/kg),阳性对照组即α-硫辛酸组(n=6,60 mg/kg)。正常对照组、糖尿病组均予腹腔注射等量生理盐水,1次/d,连续8 w。22周龄时行Morris水迷宫实验测试大鼠学习记忆能力及空间探索能力比较各组认知功能,采用Western印迹方法检测海马CA1区γ-谷氨酰半胱氨酸合成酶(GCS)、血红素氧合酶(HO)-1、Caspase-12的表达。结果与正常对照组比较,糖尿病组水迷宫实验中潜逃潜伏期明显延长(P<0. 01)、在目标象限停留时间明显缩短(P<0. 01),提示大鼠学习记忆的能力明显下降;糖尿病组海马CA1区γ-GCS、HO-1、Caspase-12表达量增加(P<0. 01)。与糖尿病组比较,NOB治疗后各组潜逃潜伏期明显延长、在目标象限停留的时间明显缩短(P<0. 01,P<0. 05),海马CA1区γ-GCS、HO-1表达量增加(P<0. 05,P<0. 01)、Caspase-12表达量减少(P<0. 05,P<0. 01),以中、高剂量组效果显著,两组间比较差异无统计学意义(P>0. 05)。结论 NOB能够通过上调海马CA1区γ-GCS、HO-1表达、减少Caspase-12蛋白表达,增强抗氧化应激和抑制凋亡而改善糖尿病大鼠的学习和记忆能力。 展开更多

Determination and Analysis of Liposoluble Chemical Constituents from Ictalurus punctatus Oil

The lipids was extracted from the internal organs of Ictalurus punctatus by microwave, and its liposoluble chemical constituents was identified and quantified by gas chromatography (GC)/electron impact (EI) mass spectrometry (MS). Subsequent-ly, the acid catalysis method was used to convert fatty acids to methyl-esters. The total lipids in the pure fat of I. punctatus were found to be about 99%. Twenty-three fatty acids were identified. The major fatty acids found in the oils were normal mo-nounsaturated, polyunsaturated fatty acids and alkane paraffin. Unsaturated fatty acids comprised 72.84% of the total, and the major components of monounsaturated fatty acids were C18:1 (48.52%), while the polyunsaturated fatty acids were mainly composed of C18:2 (16.54%). Saturated fatty acids comprised approximately 19.21%of the total. The main components were C16:0 (14.46%) and C18:0 (4.17%), most of which were medium-long-chain fatty acids. Therefore, catfish oil can be used as an important dietary source of functional fatty acids. The difference between the pro-portion of unsaturated and saturated fatty acids appears to be significant compared to fish of the Cyprinidae family, as catfish skin and muscle have higher protein content and lower fatty content. This study confirms that catfish has excellent nutri-tional quality. 展开更多

A series of electron donors,including 1,1-cyclopentanecarboxylic acid diethyl ester (CPCADEE),1,1cyclopentanedimethanol acetic diester (CPDMAD),1,1-biethoxymethyl pentane (BEMP),2,2-diethyl diethylmalonate (DEDEM)and 2,2-diethyl-1,3-propanediol acetic diester (DEPDADE),were synthesized by diethyl malonate (DEM).The purities and structures of the above products were characterized by gas chromatography (GC) and gas chromatography-mass spectrometer (GC-MS),respectively.Furthermore,the possible optimal three-dimensional structures of these donors were simulated by means of Gaussian 03 and Chem 3D.Then these electron donors were coordinated with tetrachloro titanium (TiCl 4) and chloride magnesium (MgCl 2)to obtain the catalysts for the polymerization of propylene.The catalytic activities and properties of polypropylene are greatly improved by adding external donor(ED) when CPCADEE or DEPDADE is used as internal donor(ID).However,when BEMP was used as ID,the highest catalytic activity is obtained without adding ED,which can reduce production costs and simplify catalytic synthesis.The experiments indicate that BEMP has the shortest distance of oxygen atoms and the highest electronegativity. 展开更多

Relationship between Ethanol and 2-phenylethanol Stress Tolerance and Fatty Acid Compositions of Saccharomyces cerevisiae

In the present study, we investigated of the ethanol (Eth), 2-phenylethanol (2-PE) and ethanol/2-phenylethanol (Eth/2-PE) stress tolerance and established relationship between stress tolerance and fatty acid compositions of wine yeast strain S. cerevisiae UCM Y-524 in relation to different cultivation factors. Changes in cell membrane fatty acid profiles studied under different temperature and media. The active oxygenation, semi-aerobic cultivation supplemented Tween 80 conditions were used in different combinations. The concentrations of 2-PE and Eth, the fatty acids methyl esters (FAMES) in the media were measured by GC-MS analyses. 2-PE, Eth, FAMES were identified by NIST02 MS database at the 2-PE and Eth standard solution (Merck, Germany), bacterial FAMES standard (Supelco). Palmitoleic and oleic acid were dominated for S. cerevisiae UCM Y-524. The unsaturated/saturated fatty acid (UFA/SFA) ratio was higher than five for S. cerevisiae UCM Y-524 at 14 ~C in simple medium and about more than three for other conditions. It has been found that yeast cells grown in the presence of the 2-PE, Eth and Eth/2-PE appear to increase the amount of unsaturated fatty acids, especially oleic acid, in cellular lipids especially under active oxygenation supplemented with Tween 80. S. cerevisiae UCM Y-524 has a higher tolerance under 2-PE, Eth and Eth/2-PE stress at 28 ~C in a simple medium under aerobic conditions. A higher concentration of unsaturated fatty acids in the membrane S. cerevisiae UCM Y-524 and a lower temperature under aerobic conditions, has been considered to have a positive influence on the excretion of 2-phenylethanol. 展开更多

[Method] Taking grapes in Wuling Mountain as the row materials, Saccharomyces cerevisiae of different wines was chosen and brewed. The quality of grape wine was studied and the sensory evaluation was analyzed by dint of GC-MS. [Result] Result illustrated that the ethanol concentration of the wine brewed by exploration craft A was 68.5 g/L and that brewed by exploration craft B was 59.5 g/L; the higher alcohol content of craft A was 37.86% while that of craft B is 35.99%; craft A's percentage content of esters was 28.82% while that of B was 27.10%; craft A's percentage content of acids was 2.20% while that of B was 1.24%; the gamma-aminobutyric acid content of craft A was 33.2 g/100 ml while that of craft B was 35.4 g/100 ml. the essential amino-acid content of craft A was 11.8 g/100 ml while that of craft B was 13.9 g/100 ml .The result indicates that the wine brewed by craft A was aromatic and had special flavor. Lipid oxidation and degradation are generally recognized as unfavorable reactions in food processing. However, lipids can be utilized to produce meat-like flavorings through reaction with amino acids after degradation to carbonyls such as aldehydes and ketones. The fatty acids in chicken fat mainly comprise palmitic acid, oleic acid and linoleic acid. After being heated at 120 ~C for 3 h, the degraded carbonyls of chicken fat were reacted with cysteine, alanine, glycine and thiamine to form a chicken-like flavoring. The key aromatic components of the flavoring were identified by GC-MS. In the overall volatile profile, 22 kinds of compound were detected, among which 2-furanmethanol was formed by the Maillard reaction, 1-(2-furanyl)-ethanone by cyclization and 2,4,6-trimethyl-l,3,5-trithiane by the Strecker degradation of cysteine. Dihydro-2-methyl-3(2H)-furanone, dihydro-2-methyl- 3(2H)-thiophenone, 2-acetyl-lH-pyrroline and 4-methyl-5-thiazoleethanol were the degradation products of thiamine. The prepared chicken flavoring was added to comminuted chicken product (CCP). GC-MS analysis and sensory evaluation showed that the flavor and odor of the CCP were greatly enhanced. 展开更多

Microcapsules of volatile oil containing Herba Schizonepetae(VOHS) were prepared by emulsion solvent diffusion method to improve the drug loading and reduce the amount of pharmaceutical excipients.Orthogonal assay was applied to optimize the preparation condition of microcapsulation,and the results illustrated that the ratio of ethyl cellulose(EC) to VOHS influenced the property of VOHS microcapsule significantly.GC-MS analysis indicated that some volatile components with low concentration in VOHS were lost after microencapsulation.The microcapsules prepared with optimum condition had good fluidity,and the holes on the surface of the microcapsules contributed to the release of VOHS.The particles of the microcapsule conformed to a normal distribution with the diameter of 45—220 μm.In the simulated intestinal fluid containing 0.2% sodium dodecyl sulfate,pulegone in VOHS microcapsule showed a certain degree of slow release.Compared with β-cyclodextrin method,the microencapsulation used in the present work could reduce the amount of excipients and increase the drug loading.It was beneficial to reduce the dose of Chinese medicines containing volatile oils. 展开更多

Rosemary(Rosmarius officinalis L.), an endemic plant species in south region of China, is traditionally used as a spice. In this research, the anti-inflammatory activities of essential oil and the antibacterial activities of ethanol extraction were determined, respectively. Results showed that based on the GC-MS analysis there were 35 kinds of active ingredients in the essential oil in totally, mainly including D-limonene(24.158 ml/L), α-Pinene(23.325 ml/L), Camphor(9.855 ml/L),Camphene(7.076 ml/L), Verbenone(6.685 ml/L), Borneol(5.580 ml/L), etc. The LCUV determination indicated that the main components in the ethanol extractionwere rosmarinic acid(3 910 mg/kg) and carnosic acid(2 970 mg/kg). By mice peritoneal macrophage phagocytosis of chicken erythrocytes experiment, the essential oil of rosemary was shown having a significant role in anti-inflammation. And the ethanol extraction had broad-spectrum antibacterial effects, but had no effect on mold by the agar diffusion method of 8 bacteria. As a result, both rosemary essential oil and ethanol extraction had good potential medicinal values. 展开更多

In the present study, free fatty acids(FFAs, including palmitic acid, stearic acid, oleic acid, linoleic acid and arachidonic acid) in rabbit plasma were determined by gas chromatography-mass spectrometry(GC-MS) after trimethylsilylation derivatization using N,O-bis(trimethylsilyl)trifluoroacetamide(BSTFA) – trimethylchlorosilane(TMCS) as derivatization reagent. The experimental conditions, including extraction and silylation reaction, were investigated. The method was experimentally validated. The linearity between fatty acids’ peak areas and their concentrations was obtained with the corelative coefficient(r2) all more than 0.999, and the recoveries were between 82% and 111%. The intra-day variations of FFAs’ in plasma samples at different concentrations were all less than 6%. FFA analysis results of 16 rabbit plasma samples showed that the method could be well applied in the determination of plasma samples in vivo. In contrast to the traditional method of FFA derivatization, the established trimethylsilylation method presented simplicity, high specificity, and completely free from the interference of the esterified fatty acid, such as triacylglyceride.
