重复弱束缚应激对大鼠行为及睡眠的影响 被引量:1

抑郁症患者有不同程度的睡眠异常。这是早已肯定的事实,模拟人类抑郁症的动物模型,见于文献的已有20多种,而这些动物模型是不是也表现类似抑郁症患者的睡眠异常,有关报道很少。本文用重复弱束缚应激(Repeated Mild Restrained Stress.RM... 抑郁症患者有不同程度的睡眠异常。这是早已肯定的事实,模拟人类抑郁症的动物模型,见于文献的已有20多种,而这些动物模型是不是也表现类似抑郁症患者的睡眠异常,有关报道很少。本文用重复弱束缚应激(Repeated Mild Restrained Stress.RMRS)的大鼠作为抑郁症动物模型,进行了行为和睡眠特征的观察。其结果如下:1.RMRS 6天的大鼠(n=7)单个 REMS 平均持续时间比正常对照组(n=6)明显延长(P<0.01)。其总睡眠量、REMS 总量、慢波睡眠等与正常动物无明显差异。与正常动物相比,RMRS大鼠有类似抑郁症的行为改变,如激惹、摄食少、体重下降、开野实验活动明显减少(P<0.01)。2.在 RMRS 的时,给动物连续肌注抗抑郁剂氯丙咪嗪(Clomipramine.CLI;n=7)6天,可以抑制动物的部分行为异常和 REMS 持续时间的增加。结果提示:RMRS 模型在行为特征方面较好地模拟了人类抑郁症。 展开更多

目的:比较2种模拟人类抑郁症动物模型的行为改变和睡眠特征。方法:成年雄性SD大鼠34只,随机分为正常对照组(n=6),重复弱束缚应激(RMRS)+生理盐水(NS)注射组(n=7),RMRS+氯丙咪嗪(CLI)注射组(n=7),双侧嗅球切除(OB)+NS注射组(n=7)... 目的:比较2种模拟人类抑郁症动物模型的行为改变和睡眠特征。方法:成年雄性SD大鼠34只,随机分为正常对照组(n=6),重复弱束缚应激(RMRS)+生理盐水(NS)注射组(n=7),RMRS+氯丙咪嗪(CLI)注射组(n=7),双侧嗅球切除(OB)+NS注射组(n=7)和 OB+CLI注射组(n=7)。分别进行行为和睡眠特征的观察。结果;①RMRS的大鼠单个快动眼睡眠(REMS)平均持续时间比正常对照组明显延长(P<0.05)。其总睡眠量、REMS时间总量、慢波睡眠等与对照组比较差异无显著性,但在行为上有激惹、摄食少、体重下降、活动减少等改变;②OB大鼠的REMS时间总量及发生次数、单个REMS的平均持续时间比对照组均显著增加(P<0.05),同时大鼠表现出易怒、超敏等行为异常;③2种大鼠模型连续肌内注射抗抑郁剂氯丙咪嗪,可有效地抑制动物的某些行为异常和睡眠改变。结论:RMRS模型在行为及睡眠特征方面较好地模拟了人类抑郁症。 展开更多

Ela tablets contain an optimized extract of a Chinese herbal prescription. The original prescription has been used to treat erectile dysfunction(ED), while the detailed indications and specific treatment mechanisms of Ela tablets remain unclear. In the present study, we aimed to investigate whether Ela tablets could protect mice from ED caused by repeated restraint stress and to explore the possible mechanisms. Mice were restrained daily in centrifuge tubes with venting holes for 14 consecutive days and administered Ela tablets at 1 h before restraining. On the 14 th night, mating experiments were conducted. Thereafter, blood, testicular, and penile samples were collected. Serum testosterone levels were measured using a kit. Testicular tissue morphology was observed by H&E staining. The PDE5 expression in the corpus cavernosum was measured by Western blotting analysis. The results indicated that Ela tablets significantly shortened the sexual arousal time, increased the number of sexual encounters, maintained the normal morphology of testicular tissue, increased the serum testosterone levels, and decreased the PDE5 expression in the corpus cavernosum. Our data showed that Ela tablets elicited protective effects on the sexual ability of mice exposed to repeated restraint stress. Moreover, the underlying mechanism was related to the preservation of testicular tissue morphology, maintenance of testosterone levels, and inhibition of penile PDE5 expression. 展开更多
