This paper presents a method to study the free vibration of a plate with circular holes.The circular hole is regarded as a virtual small plate in which the mass density and Young's modulus are zero.Therefore,the f...This paper presents a method to study the free vibration of a plate with circular holes.The circular hole is regarded as a virtual small plate in which the mass density and Young's modulus are zero.Therefore,the free vibration problem of the circular hole plate can be transformed into the free vibration problem of the equivalent rectangular plate with non-uniform thickness.The model is derived from the spectral geometry method(SGM),and the displacement of the plate with circular holes is expanded by the modified Fourier series.Virtual springs are added to the boundary of the plate to simulate the boundary conditions of simply supported and fixed supports.The accuracy of this method is verified by comparison with the finite element calculation results.The relationship between modal numerical solutions of plates with circular holes and boundary conditions and geometry of the plate is studied.展开更多

It is ...Abstract: A simple optimized microplate-based method to assay endo-1,4-β-mannosidase activity was described as an improved high-throughput screening method. A series of experimental conditions were optimized. It is revealed that the optimum measurement procedure is as follows: adding 50μL of diluted enzyme sample and 50 μL substrate, incubating at 45 ℃ for exactly 5 min in micro-plate, mixing with 100 μL 3,5-dinitrosalicylic acid (DNS) reagent, maintaining at boiling point for 15 rain, cooling down to room temperature before determining the ABS value at 540 nm using an ELISA micro-plate reader. The reaction volume of the optimized microplate-assay is reduced to 200μL from 2 500 μL used in the standard β-mannanase macro-assay. The optimized micro-assay is significantly more sensitive in all of the 643 candidates during endo-1,4-β-mannosidase screening. Statistical analyses show that the sensitivity of the optimized micro-method is significantly greater than that of the macro-assay. The optimized method is convenient, fast, and cheap for high throughput enzyme screening.展开更多

The influence was quantified by means of stress conce...Finite element models were established to analyze the influence of soft filler on stress concentration for a rectangular plate with an elliptic hole in the center. The influence was quantified by means of stress concentration factor (SCF). Seven shape factors of the elliptic hole and three levels of elasticity modulus of the soft filler were considered. The reduction coefficient and sensitivity index of SCF are the two indicators in evaluating the influence of soft filler. It was found that the reduction coefficient of SCF increases significantly as the shape factor and the elasticity modulus of the filler increase, indicating that soft filler can reduce the concentrated stress effectively, especially when the shape factor is great. Analysis for the sensitivity index of SCF indicates that SCF is more sensitive to materials with small elasticity modulus than to materials with large one.展开更多
The present study experimentally investigated the near-field flow mixing characteristics of two turbulent jets issuing from equilateral triangular and circular orifice plates into effectively unbounded surroundings,re...The present study experimentally investigated the near-field flow mixing characteristics of two turbulent jets issuing from equilateral triangular and circular orifice plates into effectively unbounded surroundings,respectively.Planar particle image velocimetry(PIV) was applied to measure the velocity field at the same Reynolds number of Re=50,000,where Re = UeDe /with Ue being the exit bulk velocity and the kinematic viscosity of fluid,D e the equivalent diameters.The instantaneous velocity,mean velocity,Reynolds stresses were obtained.From the mean velocity field,the centreline velocity decay rate and half-velocity width were derived.Comparing the mixing characteristics of the two jets,it is found that the triangular jet has a faster mixing rate than the circular counterpart.The triangular jet entrainments with the ambient fluid at a higher rate in the near field.This is evidenced by a shorter unmixed core,faster Reynolds stress and centreline turbulence intensity growth.The primary coherent structures in the near field are found to break down more rapidly in the triangular jet as compared to the circular jet.Over the entire measurement region,the triangular jet maintained a higher rate of decay and spread.Moreover,all components of Reynolds stress of the triangular jet appear to reach their peaks earlier,and then decay more rapidly than those of the circular jet.In addition,the axis-switching phenomenon is observed in the triangular jet.展开更多
