以18个甘薯品种(系)为试验材料,设置对照和200 mmol L^(–1) NaCl浓度处理,通过苗期盐土栽培胁迫方式,对各处理下各品种(系)的茎叶鲜重、根系鲜重、茎叶干重、根系干重、叶片相对电导率、F_v/F_m、SPAD值、SOD酶活性、MDA含量、脯氨酸... 以18个甘薯品种(系)为试验材料,设置对照和200 mmol L^(–1) NaCl浓度处理,通过苗期盐土栽培胁迫方式,对各处理下各品种(系)的茎叶鲜重、根系鲜重、茎叶干重、根系干重、叶片相对电导率、F_v/F_m、SPAD值、SOD酶活性、MDA含量、脯氨酸含量、根系活力、根系Na^+和K^+含量、Na^+/K^+比值等14个生理指标进行测定,通过对各单项指标的耐盐系数进行相关分析、主成分分析、聚类分析和逐步回归等方法对品种(系)耐盐性进行综合评价。通过主成分分析,将盐胁迫处理下甘薯苗期的14个单项指标转换成5个彼此独立的综合指标;通过隶属函数分析,得到不同品种(系)苗期耐盐性综合评价值(D值),并通过聚类分析,将18个甘薯品种(系)划分为4种耐盐类型,其中盐敏感型4个、弱耐盐型3个、中度耐盐型7个和高度耐盐型4个。在此基础上,利用逐步回归方法建立了可用于甘薯苗期耐盐性评价的回归方程,同时筛选出茎叶鲜重、根系鲜重、茎叶干重、叶片SPAD值、SOD酶活性、MDA含量、脯氨酸含量、根系Na^+/K^+比值等8个可用于甘薯苗期耐盐性评价的生理指标。本研究可为甘薯耐盐新品种选育提供种质并为甘薯苗期耐盐性评价及耐盐机制研究提供理论依据。 展开更多

The identification of germplasm is an important step for effective utilization of the available germplasm. In previous studies, isoenzyme, RAPD and SSR techniques had been used to conduct the genetic identification of watermelon ( Citrullus lanatus (Thunb.) Mansf.), but their effectiveness was limited due to the extremely narrow genetic background among watermelon genotypes. In this research, amplified fragment length polymorphism (AFLP), which was reported as a reliable technique with high efficiency in detecting polymorphism, was used to conduct genetic analysis and variety identification of thirty genotypes of watermelon core collection that represent a wide range of breeding and commercially available germplasm. As a result, a DNA fingerprint based on 15 bands amplified with four primer combinations was developed. In this fingerprint, each genotype has its unique fingerprint pattern and can be distinguished from each other. Furthermore, in or der to facilitate the utilization of AFLP marker in practice, one specific AFLP band of genotype 'PI296341' coming from fragment amplified by primer combination E-AT/M-CAT was successfully converted into a sequence characterized amplified region (SCAR) marker. 展开更多

[Objective] The aim was to study wheat salt resistance appraisal indicators under the condition of saline water irrigation.[Method] A trial was conducted with five varieties irrigated with saline water at 1,2,4,6,and 8 g/L during 2009-2010 and 2010-2011 in the research station of Dry-land Farming Institute of Hebei Academy of Agricultural and Forestry Sciences.With standardized indicators for measuring treatment,the response relations among different salt stress levels and winter wheat growth index were analyzed in this study.[Result] Under the condition of different salinity of irrigation,relative plant height after jointing stage,relative leaf area index,relative dry matter weight,relative ear number per unit area showed significant differences among treatments and yield showed significant correlation,which can be taken as salt-resistance examination index of wheat.[Conclusion] The results showed that the relative height after jointing stage was recommended as the most practical salt-tolerance appraisal indices,because it was easy to be observed and sensitive to salt stress. 展开更多

DNA fingerprinting among members of the Chinese drug Pu Gong Ying(Taraxacum mongolicum Hand,-Mazz.)and six adulterants of Tu Gong Ying were demonstrated with random-primed polymerase chain reaction(PCR)including arbitrarily primed polymerase chain reaction(AP-PCR)and random amplified polymorphic DNA(RAPD).Distinctive,reproducible genomic fingerprints from DNA from 7 species belonged to Compositae were generated with two long(20 and 24 mer)and one short(10 mer)randomly chosen primers.The Pu Gong Ying can be differentiated from six species of Tu Gong Ying according to the banding pattems of their amplified DNA on agarose gels.The results showed that AP-PCR and RAPD methods can be used for identifying Chinese drugs.Moreover,the Similarity Indexes of the genomic DNA fingerprints showed that Pu Gong Ying and its adulterants are unrelated.Therefore,AP-PCR and RAPD methods can be used for identifying Chinese drugs. 展开更多

[Method] In the research, 36 wheat varieties in 2007-2009 were chosen and drought-tolerance degrees of wheat were graded and identified according to Winter-wheat Drought Tol- erance Evaluation Technical Standards (GB/T 21127-2007), and harvest wheat grains underwent spectrum collection, with a full-spectrum analyzer, to establish a database. [Result] Based on qualitative analysis and full-spectrum correlation research, the coef- ficient of determination (RSQ) and cross-validation coefficient of determination (1-VR) were concluded at 0.697 5 and 0.600 2, showing near-infrared diffuse reflectance spectroscopy is of significant differences among wheat varieties and of significant or extremely significant correlation with drought-tolerance indices. [Conclusion] The re- search indicates that to evaluate drought-tolerance of wheat with near-infrared diffuse reflectance spectroscopy is a rapid and feasible way, which is simple, convenient without damages on grains, and of practical values for construction wheat drought-tol- erance evaluation index system and identification of breeding materials. 展开更多

Batsch germplasms.[Method] Sixty peach genotypes,representing China common local cultivars and European samples were screened by microsatellites (simple sequence repeats,SSRs) and Inter-Simple Sequence Repeat (ISSR) markers.[Result] 26 reproducible bands were amplified by Nine SSR primers,and 24 of which were polymorphic; 236 bands were amplified by 30 ISSR primers,and 113 of which were polymorphic.31 genotypes were discriminated with 1-3 distinct polymorphic bands generated from the primers ISSR and SSR.Seven cultivar-specific ISSR fragments and two SSR unique alleles obtained from this study were available to be converted into Sequence Characterized Amplified Region (SCAR) markers.The genetic similarity coefficient (GS) estimated from these molecular data averaged were 0.939 (ranged from 0.856 to 0.983) for ISSR and 0.646 (ranged from 0.240 to 1.000) for SSR,respectively.The combined grouping association indicated that most local Chinese peach cultivars and exotic accessions were clustered together.This could be related to the mode of introduction and maintenance of the peach cultivars involving limited foundation germplasm,exchange of cultivars between plantations,and periodic development of new recombinant cultivars following sexual reproduction.[Conclusion] The results obtained in this work would help to improve the conservation,molecular identification and management of peach germplasm in breeding. 展开更多

Hook and Halodule pinifolia(Miki) Hartog) in the Vellar estuary.Experimental studies found that the strain PSSG6 was effective in phosphate solubilization with Phosphate Solubilization efficiency index E = 375 ± 8.54,followed by the strain PSSG5 with Phosphate Solubilization efficiency index E = 275 ± 27.3.Of the 6 strains isolated,the strains PSSG4 and PSSG5 be-longed to the genus Bacillus,and PSSG1,PSSG2 and PSSG3 were identified as Citrobacter sp.,Shigella sp.,and Klebsiella sp.,respectively,by conventional method,and PSSG6 was identified as Bacillus circulans using conventional and molecular methods. 展开更多

The morphological characters, physiological characters, yield traits and yield per plant of total 49 rice cultivars from Ningxia were investigated under conditions of water stress and non-water stress so as to determine the relationship between each trait and yield per plant under water stress and the relationship between each relative character and drought resistance coefficient under water stress and non-water stress. The correlation, grey correlation, stepwise regression and path analyses showed that the tiller number per plant, plant height, grain density, effective panicle number per plant and grain number per panicle, total 5 traits, were significantly correlated with the drought resistance of rice, and they could be used to identify the drought resistance of rice in Ningxia. In addition, the drought resistance of rice was graded qualitatively according to the subordinate function value of corresponding drought resistance coefficient. The results showed that among the 49 rice cultivars from Ningxia,6 rice cultivars were highly drought resistant, and 9 rice cultivars were moderately drought resistant, suggesting that the evaluation method was feasible and effective. 展开更多

Tea Germplasm resources are the material foundation for improving varieties and also the basic research which is good for developing and expanding tea well-bred varieties in tea field, especially in quality selection and biotechnology research. Furthermore, excellent tea germplasm is primary condition for tea breeding and quality. Therefore, identification and screening excellent tea germplasm become particularly important. We need accelerate develop high-quality tea varieties, and it need to base on exploring and studying tea germplasm. This paper reviews the research progress of tea germplasm on identification from morphological characteristic, biological characteristic, quality characteristic, resistance and heredity. However, there are still some problems on identification and screening of tea germplasm, no effective methods have been found so far which apply on early identification and genetic engineering, so we should make intensive and long lasting study tea germplasm and analyze systematically various identification, which are benefit for breeding high-quality seeds, shortening the breeding period and summing the new way of breeding. We expect them to play the key roles and provide the theory basis for genetic improvement, classification and breeding in tea plant. 展开更多

In reference to the method of the Conference Board,the coincident indexes for China are constructed from a sample period between January 1990 and May 2012 and by51 chosen component indicators.The resulting coincident indexes have higher correlations with gross domestic product(GDP) growth rates than the China Economic Monitoring and Analysis Center(CEMAC) coincident index over the sample period between February2005 and May 2012.The peaks and troughs of the growth rates in several indicators are identified.The total number of peaks and troughs in the resulting coincident index is the same with the CEMAC coincident index.Unfortunately,these troughs don't signify recessions in the Chinese economy because the financial system has not seen a negative growth rate over the specific sample periods.The impacts of the Southeast Asian Financial Crisis and subprime mortgage crisis on the business cycle could be dated via a smoother index from HP filtering to the coincident index. 展开更多

When the system is diagnosed as abnormal state, it needs predictive maintenance, lfthe system fails, an identical new one will replace it. The predictive maintenance cannot make the system "as good as new". Under these assumptions, the reliability index and the inspection-replacement policy of a system were studied. The explicit expression of the reliability index and the average income rate (i.e., the long-run average income per unit time) are derived by using probability analysis and vector Markov process method. The criterion of feasibility for the optimal inspection-replacement policy under the maximum average income rate is obtained. The numerical example shows the optimal inspection-replacement policy can raise the average income rate when the optimal inspection-replacement policy is feasible. 展开更多

In order to precisely predict the hazard degree of goaf(HDG), the RS-TOPSIS model was built based on the results of expert investigation. To evaluate the HDG in the underground mine, five structure size factors, i.e. goaf span, exposed area, goaf height, goaf depth, and pillar width, were selected as the evaluation indexes. And based on rough dependability in rough set(RS)theory, the weights of evaluation indexes were identified by calculating rough dependability between evaluation indexes and evaluation results. Fourty goafs in some mines of western China, whose indexes parameters were measured by cavity monitoring system(CMS), were taken as evaluation objects. In addition, the characteristic parameters of five grades' typical goafs were built according to the interval limits value of single index evaluation. Then, using the technique for order preference by similarity to ideal solution(TOPSIS), five-category classification of HDG was realized based on closeness degree, and the HDG was also identified.Results show that the five-category identification of mine goafs could be realized by RS-TOPSIS method, based on the structure-scale-effect. The classification results are consistent with those of numerical simulation based on stress and displacement,while the coincidence rate is up to 92.5%. Furthermore, the results are more conservative to safety evaluation than numerical simulation, thus demonstrating that the proposed method is more easier, reasonable and more definite for HDG identification. 展开更多

METHODS: Three primers, including (GACA)(4), (GTG)(5) and M13 core sequence (5'-GAGGGTG-GCGGTTCT-3'), were used to distinguish variations among 20 clinical isolates of T. rubrum and Trichophyton mentagrophytes. RESULTS: Different PCR-fingerprinting was seen between T. rubrum and T. mentagrophytes using three different primers. 2 stains of T. rubrum were identified among 6 supposed T. mentagrophytes. CONCLUSIONS: T. rubrum can be distinguished using PCR, and (GACA)(4) is the most suitable primer. 展开更多
