The results show that in the presence or absence of A. ferrooxidans, the oxidation reaction of pyrite is divided into two steps: the first reaction step involves the oxidation of pyrite to S, and the second reaction step is the oxidation of S to SO4^2-. The oxidation mechanism of pyrite is not changed in the presence of A. ferrooxidans, but the oxidation rate of pyrite is accelerated. With the extension of reaction time of A. ferrooxidan with pyrite, the polarization current density of pyrite increases and the breakdown potential at which the passive film dissolves decreases. The impedance in the presence ofA. ferrooxidans is obviously lower than that in the absence of A. ferrooxidans, further indicating that microorganism accelerates the corrosion process of pyrite.展开更多
A novel inorganic polymer flocculant,poly-ferric sulfate(BPFS) was prepared by oxidation of ferrous sulfate using domestic Thiobacillus ferrooxidans(T·f) under acid condition.The optimal conditions for the pr...A novel inorganic polymer flocculant,poly-ferric sulfate(BPFS) was prepared by oxidation of ferrous sulfate using domestic Thiobacillus ferrooxidans(T·f) under acid condition.The optimal conditions for the preparation were pH value of 1.5,(NH4)2SO4 dosage of 0.5 g/L,initial Fe2+ concentration of 45g/L,inoculum 10%,rotating speed of 120 r/min,reaction time of 5-6 d and reaction temperature of 30 ℃.Under the optimal conditions,the BPFS product with pH value of 1.5-2.2,basicity of 17.5%-22.7% and total iron content of 43.87-45.24 g/L was obtained.The application of the BPFS to three wastewaters was carried out,and the removal efficiencies of COD,decolorization and Zn2+ by BPFS can be reached 70%,90% and 99%,respectively.The result suggests that the BPFS is an excellent flocculant for water treatment.展开更多
According to the conserved sequence of the ferritin gene, a homologous sequence was obtained from the EST database through a BLAST search against the GenBank database. This sequence was amplified with the method of RT...According to the conserved sequence of the ferritin gene, a homologous sequence was obtained from the EST database through a BLAST search against the GenBank database. This sequence was amplified with the method of RT-PCR, false sequencing was corrected, and full length eDNA of the ferritin subunit from the Chinese sturgeon was obtained. After being submitted to the GenBank database, the sequence accession number EU348782 was assigned. With the length of 896 bp, this eDNA includes entire coding regions of 53 lbp, which encodes 176 amino acids (aa). The molecular weight was predicted to be 20339.9Mr and the theoretical isoelectric point 5.66. It shares 82.9% protein sequence homology with the ferritin of the Atlantic salmon. This gene is expressed in many organs of the Chinese sturgeon, for example, the liver, pancreas, muscle, brain, heart and gastric mucosa. The highest expression level was found in the pancreas and the heart, while the muscular tissue showed the lowest. Homology modeling was used to predict the 3-D structure of the protein, which included 5 alpha helices and 10 turns. The ferritin protein structure could be overlapped and showed high similarity with that of human, flog and bacteria. It was revealed that this kind of ferritin was highly conserved in structure and function.展开更多

The results show that A. cryptum is effective in enhancing the bioleaching of pyrite and biosolubilization of RP in the presence of At. ferrooxidans, although it could not oxidize pyrite and solubilize RP by itself. This effect is demonstrated experimentally that A. cryptum enhances a decrease in pH and an increase in redox potential, concentration of total soluble iron and planktonic part bacterial number in the broth during pyrite bioleaching processes by At. ferrooxidans. The mixed culture of At. ferrooxidans and A. cryptum leads to the most extensive soluble phosphate released at 30 °C. Pulp density exceeding 3% is shown to adversely influence the release of soluble phosphate by the consortium of At. ferrooxidans and A. cryptum. It is essential to add pyrite to the 9K basal salts medium for the biosolubilization of RP by the mixed culture of At. ferrooxidans and A. cryptum, and the percentage of soluble phosphate released is the greatest when the mass ratio of RP to pyrite is 1:2 or 1:3.展开更多
[Objective] With a strain of endophytic antagonistic bacteria with good disease control efficacy in pot as trial material,the paper was to explore the antagonistic mechanism of siderophore against Fusarium oxysporum.[...[Objective] With a strain of endophytic antagonistic bacteria with good disease control efficacy in pot as trial material,the paper was to explore the antagonistic mechanism of siderophore against Fusarium oxysporum.[Method] Whether the strain produced siderophore and its fluorescent property was judged by MSA(Modified Sugar-Aspartic acid) plate.The siderophore activity of strains in liquid MSA medium was determined through the absorbance values at particular wavelength.The inhibition effects of it siderophore against F.oxysporum under different concentrations of Fe3+ were compared.Then the strain was preliminarily identified based on morphological,physiological and biochemical characteristics [Result] The fluorescent siderophore produced by the endophytic bacteria in MSA medium had a strong inhibition effect against F.oxysporum.With the increasing concentration of Fe3+,the inhibition effect against the pathogen weakened.The preliminary analysis showed these strains belonged to Bacillus.[Conclusion] Bacillus could compete the absorption of Fe3+ by secreting siderophore to inhibit the growth of F.oxysporum.展开更多
The extracellular polymeric substances(EPS) of Acidithiobacillus ferrooxidans ATCC 23270,and iron and copper enclosed in EPS were extracted by ultrasonication and centrifugation methods to determine the interaction ...The extracellular polymeric substances(EPS) of Acidithiobacillus ferrooxidans ATCC 23270,and iron and copper enclosed in EPS were extracted by ultrasonication and centrifugation methods to determine the interaction mechanism of Cu2+,Fe3+ and EPS during bioleaching chalcopyrite.Generally,Cu2+ ions can stimulate bacteria to produce more EPS than Fe3+ ions.The mass ratio of Fe3+/Cu2+ enclosed in EPS decreased gradually from about 4:1 to about 2:1 when the concentration of Cu2+ ions increased from 0.01 to 0.04 mol/L.The amount of iron and copper bound together by EPS in ferrous-free 9K medium containing 1% chalcopyrite was about 2 times of that in 9K medium containing 0.04 mol/L Cu2+ ions.It was inferred that the EPS with jarosites on the surface of chalcopyrite gradually acted as a weak diffusion barrier for Cu2+,Fe3+ ions transference during bioleaching chalcopyrite.展开更多
Electrochemical measurements were carried out to elucidate decomposition mechanism of pentlandite using modified powder microelectrode with Acidithiobacillus ferrooxidans attached or without on the mineral powder surf...Electrochemical measurements were carried out to elucidate decomposition mechanism of pentlandite using modified powder microelectrode with Acidithiobacillus ferrooxidans attached or without on the mineral powder surface.Cyclic voltammetry(CV) results show that at a low potential of about-0.2 V(vs SCE),the pentlandite was transformed to an intermediated phase like Fe4.5-yNi4.5-xS8-z when Fe and Ni ions were evacuated from mineral lattice;when the potential was changed from-0.2 V to 0.2 V,the unstable violarite(Fe3Ni3S4) and FeNi2S4 were formed which was accompanied by element sulfur formed on the mineral surface;when the potential increased over 0.2 V,the unstable intermediated phase decomposed entirely;at a higher potential of 0.7 V,the evacuated ferrous ion was oxidized to ferric ion.The presence of Acidithiobacillus ferrooxidans made the oxidation peak current increase with initial peak potential negatively moving,and the bacteria also contributed to the sulfur removing from mineral surface,which was demonstrated by the reduction characteristic at potential ranging from-0.75 to-0.5 V.Leaching experiments and electrochemical results show that the solution acidity increasing when pH2 may impede the oxidation process slightly.展开更多
Iron L-edge and sulfur K-edge X-ray absorption near edge structure(XANES) spectroscopy analysis of pyrite leached by extreme thermophilic Archaea strain Acidianus manzaensis(A.manzaensis) was carried out.Leaching ...Iron L-edge and sulfur K-edge X-ray absorption near edge structure(XANES) spectroscopy analysis of pyrite leached by extreme thermophilic Archaea strain Acidianus manzaensis(A.manzaensis) was carried out.Leaching experiments show that the oxidation of pyrite can be accelerated by A.manzaensis.Leaching results show that with the increase of leaching time,pH value in the leaching solution gradually decreases,redox potential increases rapidly from day 0 to day 3,and then increases slowly.The SEM results show that the pyrite surfaces are corroded gradually by A.manzaensis,and the XRD results show that the leaching residues contain new compositions of jarosite and elemental sulfur(S0).The iron L-edge XANES spectroscopy analysis of pyrite during biooxidation indicates that pyrite is gradually converted to Fe(III)-containing species.The sulfur K-edge XANES spectroscopy analysis indicates that elemental sulfur is produced during bioleaching and maintains mass fractions of 3.2%-5.9%.Sodium thiosulfate was also detected from day 2 to day 4,indicating the existence of thiosulfate during biooxidation of pyrite.展开更多

The denomination on this group of Asplenium is very confused in the herbaria. We hop e by means of a biosystematic study to find out their genetic relationships in the reticulate evolution, and to raise a suggestion on their taxonomic treatment. Evidence from cytology, allozyme, morphology, and palynology shows that three ancestor diploids have formed Asplenium sarelii complex comprising 13 members. A. sarelii Hook. should be typified as a diploid. The so-called tetraploid 'A. sarelii' before is an allotetraploid that comes from the doubled hybrid between diploid A. sarelii and A. tenuicaule Hayata, which should be treated as a new species A. wudangense Z. R. Wang et X. Hou. A. pekinense Hance is an autotetraploid that comes from the doubled diploid ancestor A. sarelii. A. lushanense C. Chr., a diploid species and the only ancestor of A. yunnariense group, should not been sunk as a synonym of tetraploid A. yunnariense Franch. Most probably, A. varians Wall. ex Hook. et Grev. is an autotetraploid of A. tenuicaule Hayata. Three new natural tetraploid hybrids and their origins have been found out: they are A. x longmenense ( = A. pekinense x varians), A. x jingyunense ( = A. pekinense x yunnanense) and A. x kidoi ( = A. pekinense x wudangense). Three other new natural triploid hybrids have been found and their origins have been inferred: they are A. X huawuense ( = A. sarelii X wudangense), A. x luyunense ( = A. lushanense x yunnanense) and A. x teniuvaians ( = A. tenuicaule x varians). The method of allozyme comparion combined with cytological observation is employed to reveal the complicated relationships among the members of Asplenium sarelii complex in reticulate evolution and proved to be a highly effective tool to investigate the origin of polyploid and hybrid.展开更多
The changes of pH,redox potential,concentrations of soluble iron ions and Cu^2+ with the time of bioleaching chalcopyrite concentrates by acidithiobacillus ferrooxidans were investigated under the different condition...The changes of pH,redox potential,concentrations of soluble iron ions and Cu^2+ with the time of bioleaching chalcopyrite concentrates by acidithiobacillus ferrooxidans were investigated under the different conditions of initial total-iron amount as well as mole ratio of Fe(III) to Fe(II) in the solutions containing synthetic extracellular polymeric substances (EPS).When the solution potential is lower than 650 mV (vs SHE),the inhibition of jarosites to bioleaching chalcopyrite is not vital as EPS produced by bacteria can retard the contamination through flocculating jarosites even if concentration of Fe(III) ions is up to 20 g/L but increases with increasing the concentration of Fe(III) ions;jarosites formed by bio-oxidized Fe3+ ions are more easy to adhere to outside surface of EPS space on chalcopyrite;the EPS layer with jarosites acts as a weak diffusion barrier to further rapidly create a high redox potential of more than 650 mV by bio-oxidizing Fe^2+ ions inside and outside EPS space into Fe^3+ ions,resulting in a rapid deterioration of ion diffusion performance of the EPS layer to inhibit bioleaching chalcopyrite severely and irreversibly.展开更多
Anomozamites haifanggouensis (Kimura et al.) Zheng et Zhang comb. nov. is a characteristic Bennettitalean plant in Middle Jurassic flora. Only one specimen was collected from Haifanggou Formation (Middle Jurassic) in ...Anomozamites haifanggouensis (Kimura et al.) Zheng et Zhang comb. nov. is a characteristic Bennettitalean plant in Middle Jurassic flora. Only one specimen was collected from Haifanggou Formation (Middle Jurassic) in Daohugou village, Shantou town, Ningcheng County, Nei Mongol Autonomous Region, China. The stem of plant is slender and repeatedly branches as a dichasial system with a fertile shoot in the fork, forming equal and widely divergent branches, on the branches born with the fronds of Anomozamites type. Its reproductive organs consists of some microsporophylls and a few of the bracteoid small leaves (or receptacular leaves). Some scattered bracteoid leaves and microsporophylls were formerly considered by Pan as dicots. Since then, they were proposed to be a non-committal genus Pankuangia and described them as P. haifanggouensis by Kimura et al. In this study, we discover that these bracteoid small leaves and microsporophylls should be of Anomozamites. This discovery provides strong evidence for the 'Pankuangia' determination of the taxonomical position.展开更多
Responses of Prochlorococcus (Pro), Synechococcus (Syn), pico-eukaryotes (Euk) and heterotrophic bacteria (Bact) in pelagic marine ecosystems to external nutrient perturbations were examined using nitrogen- (N), phosp...Responses of Prochlorococcus (Pro), Synechococcus (Syn), pico-eukaryotes (Euk) and heterotrophic bacteria (Bact) in pelagic marine ecosystems to external nutrient perturbations were examined using nitrogen- (N), phosphorus- (P), iron- (Fe), and cobalt- (Co) enriched incubations in the South China Sea in November 1997. Variations in abundance of the 4 groups of microorganism and cellular pigment content of the autotrophs during incubation were followed by flow-cytometric measurements for seven days. During the incubation, Syn and Euk showed a relatively higher demand on Fe and N, while Pro required higher levels of Co and P. The Fe was inadequate for all the organisms in the deep euphotic zone (75 m) of the study area. The experimental results also implied that biological interaction among the organisms played a role in the community structure shift during the incubation. It seemed that besides the effects of temperature, there are some other physical and chemical limitations as well as impacts from biological interactions on Pro distribution in coast waters.展开更多

Bioleaching tests at different dosages of pyrolusite ore, pH and inoculation amounts of Acidithiobacillus ferrooxidans were studied. The results showed that the time of the bio-oxidation process was decreased obviously and the arsenic leaching rate reached 94.4% after the bioleaching. The bio-oxidation of arsenopyrite and the effective extraction of manganese from pyrolusite were achieved by the bioleaching process. After bioleaching, the leaching rate of gold from the reaction residues reached 95.8% by cyanide leaching. In the bio-oxidation process, pyrolusite increased the redox potential of the solution to accelerate the bioleaching rate. The experiment showed that there were two reaction modes in the bioleaching process.展开更多

The seedlings of Dendrobium officinale interplanted with Pleurotus os- treatus grew by 2.78 cm during 10 weeks on cultivation beds, and the seedlings grew by 2.68 cm in the cultivation beds without interplanting arrangement. These demonstrated that the interplanting of Dendrobium officinale and Pleurotus ostreatus has none adverse effects on growth of Dendrobium officinale.展开更多

The possibility of crystallization, and number, size and quality of crystals were obviously dependent on concentrations of NaCl, MgCl2, PEG 8000,Tris and Hepes buffer and on methods for crystallization. PEG concentration affected on the shape of the crystals. The optimal, concentrations of the chemicals for crystallization of MnFe protein were slightly different from those for crystallization of Delta nifZ MoFe protein from a nifZ deleted strain of Azotobacter vinelandii. SDS-PAGE showed that the protein from the dissolved crystals was almost the same as MnFe protein before crystallization, indicating that the crystal was formed from MnFe protein.展开更多

文摘The extracellular polymeric substances(EPS) of Acidithiobacillus ferrooxidans ATCC 23270,and iron and copper enclosed in EPS were extracted by ultrasonication and centrifugation methods to determine the interaction mechanism of Cu2+,Fe3+ and EPS during bioleaching chalcopyrite.Generally,Cu2+ ions can stimulate bacteria to produce more EPS than Fe3+ ions.The mass ratio of Fe3+/Cu2+ enclosed in EPS decreased gradually from about 4:1 to about 2:1 when the concentration of Cu2+ ions increased from 0.01 to 0.04 mol/L.The amount of iron and copper bound together by EPS in ferrous-free 9K medium containing 1% chalcopyrite was about 2 times of that in 9K medium containing 0.04 mol/L Cu2+ ions.It was inferred that the EPS with jarosites on the surface of chalcopyrite gradually acted as a weak diffusion barrier for Cu2+,Fe3+ ions transference during bioleaching chalcopyrite.

文摘Electrochemical measurements were carried out to elucidate decomposition mechanism of pentlandite using modified powder microelectrode with Acidithiobacillus ferrooxidans attached or without on the mineral powder surface.Cyclic voltammetry(CV) results show that at a low potential of about-0.2 V(vs SCE),the pentlandite was transformed to an intermediated phase like Fe4.5-yNi4.5-xS8-z when Fe and Ni ions were evacuated from mineral lattice;when the potential was changed from-0.2 V to 0.2 V,the unstable violarite(Fe3Ni3S4) and FeNi2S4 were formed which was accompanied by element sulfur formed on the mineral surface;when the potential increased over 0.2 V,the unstable intermediated phase decomposed entirely;at a higher potential of 0.7 V,the evacuated ferrous ion was oxidized to ferric ion.The presence of Acidithiobacillus ferrooxidans made the oxidation peak current increase with initial peak potential negatively moving,and the bacteria also contributed to the sulfur removing from mineral surface,which was demonstrated by the reduction characteristic at potential ranging from-0.75 to-0.5 V.Leaching experiments and electrochemical results show that the solution acidity increasing when pH2 may impede the oxidation process slightly.

文摘Iron L-edge and sulfur K-edge X-ray absorption near edge structure(XANES) spectroscopy analysis of pyrite leached by extreme thermophilic Archaea strain Acidianus manzaensis(A.manzaensis) was carried out.Leaching experiments show that the oxidation of pyrite can be accelerated by A.manzaensis.Leaching results show that with the increase of leaching time,pH value in the leaching solution gradually decreases,redox potential increases rapidly from day 0 to day 3,and then increases slowly.The SEM results show that the pyrite surfaces are corroded gradually by A.manzaensis,and the XRD results show that the leaching residues contain new compositions of jarosite and elemental sulfur(S0).The iron L-edge XANES spectroscopy analysis of pyrite during biooxidation indicates that pyrite is gradually converted to Fe(III)-containing species.The sulfur K-edge XANES spectroscopy analysis indicates that elemental sulfur is produced during bioleaching and maintains mass fractions of 3.2%-5.9%.Sodium thiosulfate was also detected from day 2 to day 4,indicating the existence of thiosulfate during biooxidation of pyrite.
文摘The series Variantia Ching et S. H. Wu mainly occur in China and its members are highly variable in morphology. The denomination on this group of Asplenium is very confused in the herbaria. We hop e by means of a biosystematic study to find out their genetic relationships in the reticulate evolution, and to raise a suggestion on their taxonomic treatment. Evidence from cytology, allozyme, morphology, and palynology shows that three ancestor diploids have formed Asplenium sarelii complex comprising 13 members. A. sarelii Hook. should be typified as a diploid. The so-called tetraploid 'A. sarelii' before is an allotetraploid that comes from the doubled hybrid between diploid A. sarelii and A. tenuicaule Hayata, which should be treated as a new species A. wudangense Z. R. Wang et X. Hou. A. pekinense Hance is an autotetraploid that comes from the doubled diploid ancestor A. sarelii. A. lushanense C. Chr., a diploid species and the only ancestor of A. yunnariense group, should not been sunk as a synonym of tetraploid A. yunnariense Franch. Most probably, A. varians Wall. ex Hook. et Grev. is an autotetraploid of A. tenuicaule Hayata. Three new natural tetraploid hybrids and their origins have been found out: they are A. x longmenense ( = A. pekinense x varians), A. x jingyunense ( = A. pekinense x yunnanense) and A. x kidoi ( = A. pekinense x wudangense). Three other new natural triploid hybrids have been found and their origins have been inferred: they are A. X huawuense ( = A. sarelii X wudangense), A. x luyunense ( = A. lushanense x yunnanense) and A. x teniuvaians ( = A. tenuicaule x varians). The method of allozyme comparion combined with cytological observation is employed to reveal the complicated relationships among the members of Asplenium sarelii complex in reticulate evolution and proved to be a highly effective tool to investigate the origin of polyploid and hybrid.

文摘The changes of pH,redox potential,concentrations of soluble iron ions and Cu^2+ with the time of bioleaching chalcopyrite concentrates by acidithiobacillus ferrooxidans were investigated under the different conditions of initial total-iron amount as well as mole ratio of Fe(III) to Fe(II) in the solutions containing synthetic extracellular polymeric substances (EPS).When the solution potential is lower than 650 mV (vs SHE),the inhibition of jarosites to bioleaching chalcopyrite is not vital as EPS produced by bacteria can retard the contamination through flocculating jarosites even if concentration of Fe(III) ions is up to 20 g/L but increases with increasing the concentration of Fe(III) ions;jarosites formed by bio-oxidized Fe3+ ions are more easy to adhere to outside surface of EPS space on chalcopyrite;the EPS layer with jarosites acts as a weak diffusion barrier to further rapidly create a high redox potential of more than 650 mV by bio-oxidizing Fe^2+ ions inside and outside EPS space into Fe^3+ ions,resulting in a rapid deterioration of ion diffusion performance of the EPS layer to inhibit bioleaching chalcopyrite severely and irreversibly.
文摘Anomozamites haifanggouensis (Kimura et al.) Zheng et Zhang comb. nov. is a characteristic Bennettitalean plant in Middle Jurassic flora. Only one specimen was collected from Haifanggou Formation (Middle Jurassic) in Daohugou village, Shantou town, Ningcheng County, Nei Mongol Autonomous Region, China. The stem of plant is slender and repeatedly branches as a dichasial system with a fertile shoot in the fork, forming equal and widely divergent branches, on the branches born with the fronds of Anomozamites type. Its reproductive organs consists of some microsporophylls and a few of the bracteoid small leaves (or receptacular leaves). Some scattered bracteoid leaves and microsporophylls were formerly considered by Pan as dicots. Since then, they were proposed to be a non-committal genus Pankuangia and described them as P. haifanggouensis by Kimura et al. In this study, we discover that these bracteoid small leaves and microsporophylls should be of Anomozamites. This discovery provides strong evidence for the 'Pankuangia' determination of the taxonomical position.
文摘Responses of Prochlorococcus (Pro), Synechococcus (Syn), pico-eukaryotes (Euk) and heterotrophic bacteria (Bact) in pelagic marine ecosystems to external nutrient perturbations were examined using nitrogen- (N), phosphorus- (P), iron- (Fe), and cobalt- (Co) enriched incubations in the South China Sea in November 1997. Variations in abundance of the 4 groups of microorganism and cellular pigment content of the autotrophs during incubation were followed by flow-cytometric measurements for seven days. During the incubation, Syn and Euk showed a relatively higher demand on Fe and N, while Pro required higher levels of Co and P. The Fe was inadequate for all the organisms in the deep euphotic zone (75 m) of the study area. The experimental results also implied that biological interaction among the organisms played a role in the community structure shift during the incubation. It seemed that besides the effects of temperature, there are some other physical and chemical limitations as well as impacts from biological interactions on Pro distribution in coast waters.

文摘Pyrolusite was added in the bioleaching process to enhance the bio-oxidation process. Bioleaching tests at different dosages of pyrolusite ore, pH and inoculation amounts of Acidithiobacillus ferrooxidans were studied. The results showed that the time of the bio-oxidation process was decreased obviously and the arsenic leaching rate reached 94.4% after the bioleaching. The bio-oxidation of arsenopyrite and the effective extraction of manganese from pyrolusite were achieved by the bioleaching process. After bioleaching, the leaching rate of gold from the reaction residues reached 95.8% by cyanide leaching. In the bio-oxidation process, pyrolusite increased the redox potential of the solution to accelerate the bioleaching rate. The experiment showed that there were two reaction modes in the bioleaching process.

文摘In summer, Cha No. 39 of 292 bags and 205 of 244 bags were inter- planted with Dendrobium officinale on cultivation bed, respectively, whose yields and biological efficiencies reached 102.46 kg and 103.98%, and 96.77 kg and 113.32%, accordingly. The seedlings of Dendrobium officinale interplanted with Pleurotus os- treatus grew by 2.78 cm during 10 weeks on cultivation beds, and the seedlings grew by 2.68 cm in the cultivation beds without interplanting arrangement. These demonstrated that the interplanting of Dendrobium officinale and Pleurotus ostreatus has none adverse effects on growth of Dendrobium officinale.
文摘Under a suitable condition of crystallization, dark brown short rhombohedron crystals could be obtained from nitrogenase MnFe protein purified from a mutant UW3 of Azotobacter vinelandii Lipmann grown in Mn-containing but Mo- and NH3-free medium. The possibility of crystallization, and number, size and quality of crystals were obviously dependent on concentrations of NaCl, MgCl2, PEG 8000,Tris and Hepes buffer and on methods for crystallization. PEG concentration affected on the shape of the crystals. The optimal, concentrations of the chemicals for crystallization of MnFe protein were slightly different from those for crystallization of Delta nifZ MoFe protein from a nifZ deleted strain of Azotobacter vinelandii. SDS-PAGE showed that the protein from the dissolved crystals was almost the same as MnFe protein before crystallization, indicating that the crystal was formed from MnFe protein.
