Methods Thirty Sprague-Dawley ( SD ) rats were divided into 5 groups randomly: control, 2-acetylaminofluorene ( 2-AAF ), 2-AAF + partial hepatectomy ( PH ), 2-AAF + carbon tetrachloride ( CCl4 ), and diabetic groups. As two-step collagenase perfusion protocol of Seglen, oval cells were isolated by Percoll density gradient centrifugation. Thy1. 1 positive cells were sorted by flow cytometry, and then cultured in Dulbecco's minimum Eagle's medium (DMEM). Immunofluorescence staining was applied to labelling Thyl. 1. Results Different rates of Thy1.1 positive oval cells were found in different rat model groups : 0. 5 % in 2-AAF, 0. 3% in 2-AAF + PH, 0. 2% in 2-AAF + CCl4, 0. 1% in diabetic, and 0. 0% in control. Isolated cells adhered to plate with fusiform or polygon as epithelial cells. Conclusion Progenitor stem cells exist in injured liver tissue including those from diabetic rats.展开更多
Objective: To investigate the expression level of E-cadherin in spinal plasma cell myeloma and its relationship with the clinicopathological indexes of spinal plasma cell myeloma. Methods: The expression of E-cadher...Objective: To investigate the expression level of E-cadherin in spinal plasma cell myeloma and its relationship with the clinicopathological indexes of spinal plasma cell myeloma. Methods: The expression of E-cadherin was detected in 41 cases of spinal plasma cell myeloma and 14 cases of simple bone cysts by using S-P (streptavidin-perosidase) immunohistochemical staining. The relationship between E-cadherin expression and the clinicopathological features was analyzed. Results: In spinal plasma cell myelomas, the positive expression rate of E-cadherin (46.34%) was higher than that in simple bone cyst tissues (all negative) (P〈0.01). And the expression of E-cadherin had no obvious relationship with patients' age, gender, clinical classification or M-component (P〉0.05), but had relationship with the classification of light chain. The expression rate in lambda chain subgroup was lower than that in the others (P〈0.05). We also found the E-cadherin negative patients had a higher overall survival rate than those who had strong E-cadherin expression (P〈0.05). Conclusion: E-cadherin expressed higher in spinal plasma cell myelomas than in simple bone cyst tissues and its expression had relationship with the light chain classification in spinal plasma cell myeloma. The patients with no stain of E-cadherin may have a poor prognosis. Detecting the type of light chain associated with expression of E-cadherin might be helpful in evaluating the outcome of spinal PCM patients.展开更多

Methods: Arabbit model of heavy TBI was developed by high velocity impact on the parietal bone with an iron stick. Totally 8 healthy adult New Zealand white rabbits were randomly divided into control group (n=2) and injury group (n=6). Four hours after injury, tissue specimens from the parietal lobe, temporal lobe, occipital lobe, cerebellum and brainstem were harvested to detect the expression of Hsp70 and Caspase-3 by immunohistochemistry. Besides, the gray values of cells positive for HspT0 and Caspase-3 were analyzed with an image analyzer. Results: Immunohistochemistry staining demonstrated a low level of Caspase-3 and Hsp70 expression in normal control group. While in injury group, both the Caspase-3 and Hsp70 expression was significantly elevated (P〈0.05). Positive cells gathered around the lesion focus. Occipital lobe and cerebellum had fewer positive cells while temporal and brainstem had the fewest. Conclusion: The expression of Caspase-3 and HspT0 at an early stage following severe TBI is characteristic and can be applied to estimate the time of injury.展开更多
