A new borderline discipline, biomechanopharmacology, is shaping by the efforts of applying biomechanics in pharmacological studies of ABCRBS herbs. Biomechanics is involved in modeling of blood stasis syndrome (BSS) with mechanical force induced injury and model evaluation by shear stress monitoring for blood coagulation. Investigations showed that tetramethylpyrazine (TMP) contained in Ligusticum chuanxiong Hort and diallyl trisulfide (DT) extracted from garlic demonstrated inhibiting characteristics on vWF mediated platelet activation and thrombus formation occurring under high shear rates. The effect of TMP on shear-induced platelet aggregation might be due to inhibition of calcium channel activity since it showed significant inhibition on intracellular level of calcium demonstrated by laser confocal microscope. The combined effects of TMP and shear stress on rat cerebral microvascular endothelial cell (rCMEC) were investigated by various doses of TMP incorporated with different levels of shear stress generated by a rotational cone-plate rheometer. The results indicated that apoptosis of rCMECs could be restrained by a combination of medial level of shear stress with a suitable dose of TMP. To study the influences of shear stress, pressure and TMP on angiogenesis of vascular endothelial cell, cultured rCMEC was pretreated in a flow chamber with independent adjustment for levels of shear stress and pressure, and then 3D cultured on Matrigel. The results indicate that combined effects of shear stress, pressure and TMP may influence angiogenesis significantly. We believe that research on interactions among blood shear stress, secretion of endothelial cell, and pharmacodynamics will be an interesting area of biomechanopharmacology. Herbs of ABCRBS and their extracts for protecting endothelial cells to maintain their normal functions are expected.展开更多

Methods: Cell proliferation was assessed by MTT assay, cell cycle distribution was detected by the flow cytometry (FCM). The expression of Cyclin A, CDK2 mRNA levels were examined by RT-PCR. Quantitative colorimetric assay was used to analyze Cyclin NCDK2 activity in HepG-2 cells. Results: Cinobufacini injection significantly inhibited HepG-2 cells proliferation in dose- and time-dependent ways; FCM analysis showed Cinobufacini injection induced cell cycle arrest at S phase; RT-PCR assay showed Cinobufacini injection down-regulated Cyclin A, CDK2 expression at mRNA levels; Quantitative colorimetric assay showed Cinobufacini injection deceased Cyclin NCDK2 activity in HepG-2 cells. The curative effect is not satisfactory as far as its treatment by western and traditional Chinese medicine is concerned, which has a long course of treatment and a high recurrent rate. 70 cases of external humeral epicondylitis were treated by massotherapy after local blocking from August 1995 to October 1997 at this hospital with satisfactory therapeutic effects.Clinical DataOf the 70 cases in this series treated by massotherapy after local blocking, 30 were males and 40 females, ranging in age from 19 to 65 years. 55 cases were 30 to 50 years old, 20 had the left elbow affected and 50 the right elbow. 7 cases had a history of trauma, 50 a history of chronic strain, and 13 the cause unknown. The shortest duration of disease was 15 days and the longest 24 months.Among 50 cases in the control group treated by massotherapy, 20 cases were males and 30 females, ranging in age from 16 to 58 years. 40 cases were 30 to 50 years old. 15 cases were affected on the left elbow and 35 on the right elbow. 6 cases had a history of trauma, 32 a history of chronic strain, and 12 the cause unknown. Elastic-plastic steel damper(EPSD) is a new device controlling seismic responses.The mechanical principle of EPSD was presented and a comparison was conducted between the theoretical formulas and finite element(FE) si...Elastic-plastic steel damper(EPSD) is a new device controlling seismic responses.The mechanical principle of EPSD was presented and a comparison was conducted between the theoretical formulas and finite element(FE) simulation of damper units.The verified force-displacement hysteretic curve of the damper system was obtained with reference to tests.The Nanjing Jiangxinzhou Bridge(NJB) was subsequently taken as the case to investigate the seismic response control effect of EPSDs on single-tower self-anchored suspension bridges.A 3-dimensional FE model of the bridge was established in ANSYS and the dynamic and static analyses of the bridge were conducted,the control effect of EPSDs under different seismic waves was further investigated through nonlinear time-history analysis based on the validated model.Results showed that both the simplified theoretical and FE simulation methods can preferable reflect the mechanical performance of EPSD,and that seismic responses of NJB with EPSDs are better than those with elastic connection device or fluid viscous damper.However,the control effect of EPSDs is influenced by seismic wave characteristics.展开更多
