奶牛饲料原料中霉菌毒素广泛存在,并严重地威胁着奶牛健康,影响着奶牛生产性能的发挥。本试验选择年龄、胎次、产奶量、泌乳天数接近的荷斯坦奶牛50头和带有永久性瘤胃瘘管的泌乳荷斯坦奶牛4头来探讨奶牛专用霉菌毒素吸附剂对奶牛粪便... 奶牛饲料原料中霉菌毒素广泛存在,并严重地威胁着奶牛健康,影响着奶牛生产性能的发挥。本试验选择年龄、胎次、产奶量、泌乳天数接近的荷斯坦奶牛50头和带有永久性瘤胃瘘管的泌乳荷斯坦奶牛4头来探讨奶牛专用霉菌毒素吸附剂对奶牛粪便中双歧杆菌数量和原料奶中霉菌毒素残留量的影响。结果表明,添加霉菌毒素吸附剂可降低非瘘管奶牛粪便中双歧杆菌的外排量(P<0.05)和原料奶中体细胞数。非瘘管奶牛试验组隐性乳房炎较试验前降低了75.27%;同时,对照组试验前后原料奶中霉菌毒素含量未发生较大变化,但试验组试验后奶中霉菌毒素含量较试验前显著降低(P<0.05);其中黄曲霉毒素代谢产物AFM1降低了97.50%,玉米赤霉烯酮毒素降低了80.60%,呕吐毒素降低了92.86%。因此,本研究表明,日粮添加霉菌毒素吸附剂可有效降低霉菌毒素对试验奶牛的毒害和在原料奶中的残留量,进而提高了原料奶质量和该牧场的整体经济效益。 展开更多

研究了不同微波有效功率下高水分稻谷的微波干燥特性,以及探讨了微波处理工艺对稻谷加工品质及微生物量的影响。结果表明,在较低的微波功率(485和927 W)下,处理初期稻谷温度迅速升高,水分下降缓慢,当温度达到65℃、含水量达到19. 7%左右... 研究了不同微波有效功率下高水分稻谷的微波干燥特性,以及探讨了微波处理工艺对稻谷加工品质及微生物量的影响。结果表明,在较低的微波功率(485和927 W)下,处理初期稻谷温度迅速升高,水分下降缓慢,当温度达到65℃、含水量达到19. 7%左右时,稻谷升温速度减小,而水分下降速度加快。微波功率增加可显著提高稻谷的升温速度和降水速率,但高功率微波干燥稻谷易产生焦糊现象。采用有效功率927 W的微波条件,2 min可将稻谷加热至60℃,稻谷含水量从21. 58%降低至19. 96%。通过4 h缓苏处理后,稻谷表面细菌量下降3. 6 log CFU/m L,表面霉菌量下降3. 3 log CFU/m L,对稻谷内部霉菌可实现95%灭菌,稻谷的出糙率和整精米率无显著下降(P> 0. 05),分别为83. 92%和68. 14%。与自然通风及热风处理稻谷至入仓偏高水分18. 34%相比,微波与热风联合处理只需20 min,其稻谷加工品质较高,杀菌效果也远远高于自然通风及热风处理。因此,适宜的微波处理在保障高水分稻谷加工品质的前提下,可显著缩短干燥时间,并获得高质量的杀菌效果,实现高水分稻谷快速安全入仓处置。 展开更多

Strain improvement and medium optimization to increase the productivity of spiramycin were carried out. Of oil tolerant mutant strains screened, one mutant, Streptomyces ambofaciens XC 2-37, produced 9% more spiramycin than the parent strain S. ambofaciens XC 1-29. The effects of soybean oil and propyl alcohol on spiramycin production with S. ambofaciens XC 2-37 were studied. The potency of S. ambofaciens XC 2-37 was improved by 61.8% with addition of 2% soybean oil in the fermentation medium and 0.4% propyl alcohol at 24 hours after incubation. The suitable time for feeding propyl alcohol is at 24 hours after incubation in flask fermentation and at 20 hours after incubation in fermentor fermentation. The new process with S. ambofaciens XC 2-37 was scaled up for industrial scale production of spiramycin in a 60 m3 fermentor in Xinchang Pharmaceutical Factory, Zhejiang Medicine Company, Ltd., China, and the potency and productivity of fermentation were improved by 42.9%. 展开更多

The present work studies the chemical constituents from marine-derived streptomyces 3320^# and their antitumor activities. The present work studies the chemical constituents from marine-derived streptomyces 3320^# and their antitumor activities. The n-BuOH extract of the ferment broth of 3320^# was chromatographed on silica gel, Sephadex LH-20, ODS columns and HPLC to separate the compounds with antitoumor activities. Their structures were identified using IR, UV, NMR, MS spectroscopic techniques and compared with published data. The antitumor activities of the isolates were assayed using SRB method and flow cytometry assay, accompanied with the morphological observation of the cells under light micro- scope against mammalian tsFT210 cells. Ten compounds, cyclo-(Ala-Leu) 1, cyclo-(Ala-Ile) 2, cyclo-(Ala-Val) 3, cyclo- (Phe-Pro) 4, cyclo-(Phe-Gly)5, cyclo-(Leu-Pro)6, 1-methyl-1, 2, 3, 4-tetrahydro-β-carboline-3-carboxylic acid 7, N-(4- hydroxyphenethyl) acetamide 8, 4-methyoxy-l-(2-hydroxy) ethylbenzene 9 and uridine 10, were isolated from the ferment broth of streptomyces 3320^# . Among them, compounds 6, 7, 8 and 10 showed potent cytotoxicity against the tsFT210 cell with the IC50 values of 3.6, 7.2, 5.2 and 1.6 mmol L-1, respectively. Compounds 8, 10 also exhibited apoptosis inducing activity under 2.0 mmol L-1. Compounds 6, 7, 8 and 10 are the principle bioactive constituents responsible for the antitumor activities of marine streptomyces 3320^#. 研究了紫苏精油对短期温度变化的稻谷品质的保持效果.方法:13%左右水分的稻谷分别经0、1μL/g和4μL/g终浓度的紫苏精油处理后使用恒温恒湿箱储藏,期间隔日调升0.5℃(储藏温度由13℃逐渐上升至25℃),储藏30 d,每10 d取样一次,检测稻谷... 研究了紫苏精油对短期温度变化的稻谷品质的保持效果.方法:13%左右水分的稻谷分别经0、1μL/g和4μL/g终浓度的紫苏精油处理后使用恒温恒湿箱储藏,期间隔日调升0.5℃(储藏温度由13℃逐渐上升至25℃),储藏30 d,每10 d取样一次,检测稻谷的不完善粒和脂肪酸值等品质指标,并采用平板计数法跟踪稻谷霉菌的带菌量.结果显示:未添加紫苏精油的常规组稻谷不完善粒数量增加约50%;脂肪酸值由初始的23.2(KOH/干基)/(mg/100 g)上升至33.9(KOH/干基)/(mg/100 g),增加46.1%;稻谷中的霉菌数量由3.6 log CFU/g增加至3.9 log CFU/g.1μL/g终浓度的紫苏精油处理组的不完善粒数量增加25%、脂肪酸值由23.2(KOH/干基)/(mg/100 g)上升至27.9(KOH/干基)/(mg/100 g),增加20.3%;稻谷中的霉菌数量由3.3 log CFU/g,增加至3.6 log CFU/g.4μL/g终浓度的紫苏精油处理组的不完善粒数量没有增加、脂肪酸值由23.2(KOH/干基)/(mg/100 g)上升至27.7(KOH/干基)/(mg/100 g),增加19.4%;稻谷中的霉菌数量由3.3 log CFU/g增加至20 d时的3.39 log CFU/g后降至30 d时的2.9 log CFU/g.试验结果表明:紫苏精油处理的稻谷品质优于未处理的稻谷,紫苏精油处理可一定程度上用于控制稻谷品质的劣变,为稻谷的短期保藏提供了一种可行的解决思路. 展开更多

To achieve higher antibiotic streptolydigin productivity through metabolic regulation, propionate was fed during the fermentation of Streptomyces lydicus AS 4.2501. The effects of propionate feeding on streptolydigin production and intracellular fluxes were investigated. The highest streptolydigin production yield of 95.10mg·L-1 was obtained when 2mmol·L-1 of sodium propionate was added at 60h of cultivation into shake-flask culture. This yield is 23.06% higher when compared to that of a batch culture without propionate supplementation. It was also found that when propionate was added, much more organic acids were excreted. Metabolic flux analysis was per-formed and it demonstrated that the carbon fluxes of the pentose phosphate pathway and the anaplerotic reaction were significantly increased after propionate feeding. The carbon flux from pyruvate to acetyl-CoA was determined to be 24.7, which was 12.27% higher than that in the batch culture. This study indicated that the glu-cose-6-phosphate and pyruvate nodes were potential bottlenecks for increasing streptolydigin productivity. Potential targets and strategies that could be manipulated through genetic and process engineering to increase the production of streptolydigin were also suggested. 展开更多

Development of a colorimetric assay for rapid quantitative measurement of clavulanic acid in microbial samples

We developed a colorimetric assay to quantify clavulanic acid (CA) in culture broth of Streptomyces clavuligerus, to facilitate screening of a large number of S. clavuligerus mutants. The assay is based on a β-1actamase-catalyzed reaction, in which the yellow substrate nitrocefin (λmax=390 nm) is converted to a red product (λmax=486 nm). Since CA can irreversibly inhibit β-1actamase activity, the level of CA in a sample can be measured as a function of the A390]A486 ratio in the assay mixture. The sensitivity and detection window of the assay were determined to be 50 μg L -1 and 50 μg L to 10 mg L-1, respectively. The reliability of the assay was confirmed by comparing assay results with those obtained by HPLC. The assay was used to screen a pool of 65 S. clavuligerus mutants and was reliable for identifying CA over-producing mutants. Therefore, the assay saves time and labor in large-scale mutant screening and evaluation tasks. The detection window and the reliability of this assay are markedly better than those of previously reported CA assays. This assay method is suitable for high throughput screening of microbial samples and allows direct visual observation of CA levels on agar plates. 展开更多
