In order to find the main factors that influence the urban traffic structure,a relational model between the travelers' characteristics and the trip mode choice is built.The data of urban residents' characteristics a...In order to find the main factors that influence the urban traffic structure,a relational model between the travelers' characteristics and the trip mode choice is built.The data of urban residents' characteristics are obtained from statistical data,while the trip mode split data is collected through a trip survey in Bengbu.In addition,the discrete choice model is adopted to build the functional relationship between the mode choice and the travelers' personal characteristics,as well as family characteristics and trip characteristics.The model shows that the relationship between the mode split and the personal,as well as family and trip characteristics is stable and changes little as the time changes.Deduced by the discrete model,the mode split result is relatively accurate and can be feasibly used for trip mode structure forecasts.Furthermore,the proposed model can also contribute to find the key influencing factors on trip mode choice,and restructure or optimize the urban trip mode structure.展开更多
An effort was made to couple FVCOM (a three-dimensional (3D),unstructured grid,Finite Volume Coastal Ocean Model) and FVCOM-SWAVE (an unstructured grid,finite-volume surface wave model) for the study of nearshore ocea...An effort was made to couple FVCOM (a three-dimensional (3D),unstructured grid,Finite Volume Coastal Ocean Model) and FVCOM-SWAVE (an unstructured grid,finite-volume surface wave model) for the study of nearshore ocean processes such as tides,circulation,storm surge,waves,sediment transport,and morphological evolution.The coupling between FVCOM and FVCOM-SWAVE was achieved through incorporating 3D radiation stress,wave-current-sediment-related bottom boundary layer,sea surface stress parameterizations,and morphology process.FVCOM also includes a 3D sediment transport module.With accurate fitting of irregular coastlines,the model provides a unique tool to study sediment dynamics in coastal ocean,estuaries,and wetlands where local geometries are characterized by inlets,islands,and intertidal marsh zones.The model was validated by two standard benchmark tests: 1) spectral waves approaching a mild sloping beach and 2) morphological changes of seabed in an idealized tidal inlet.In Test 1,model results were compared with both analytical solutions and laboratory experiments.A further comparison was also made with the structured grid Regional Ocean Model System (ROMS),which provides an insight into the performance of the two models with the same open boundary forcing.展开更多

The method is capable of simulating t...To evaluate the responses of fixed and pinned pile groups under torsion, a method is presented to analyze the nonlinear behavior of free-standing pile groups with rigid pile caps. The method is capable of simulating the nonlinear soil response in the near field usingp-y and r-θ curves, the far-field interactions through Mindlin's and Randolph's elastic solutions, and the coupling effect of lateral resistance on torsional resistance of the individual piles using an empirical factor. Based on comparisons of the solutions for fixed- and pinned-head, 1×2, 2×2, and 3×3 pile groups subjected to torsion, it was found that pile-cap connection significantly influences the torsional capacity of pile groups and the assignment of applied torques in the pile groups. In this study, the applied torques for the pinned-head pile groups are only 44%-64% of those for the corresponding fixed-head pile groups at a twist angle of 2^o. Such a difference is mainly due to the change of the lateral resistances of individual piles in the groups.展开更多

The results s...The decadal variability of the North Atlantic thermohaline circulation(THC) is investigated within a three-dimensional ocean circulation model using the conditional nonlinear optimal perturbation method. The results show that the optimal initial perturbations of temperature and salinity exciting the strongest decadal THC variations have similar structures: the perturbations are mainly in the northwestern basin at a depth ranging from 1500 to 3000 m. These temperature and salinity perturbations act as the optimal precursors for future modifications of the THC, highlighting the importance of observations in the northwestern basin to monitor the variations of temperature and salinity at depth. The decadal THC variation in the nonlinear model initialized by the optimal salinity perturbations is much stronger than that caused by the optimal temperature perturbations, indicating that salinity variations might play a relatively important role in exciting the decadal THC variability. Moreover, the decadal THC variations in the tangent linear and nonlinear models show remarkably different characteristics, suggesting the importance of nonlinear processes in the decadal variability of the THC.展开更多
AIM:To explore the function of Nonstructural 5A(NS5A) protein of genotype 2a(JFH1)in the replication and infection of hepatitis C virus(HCV).METHODS:Intergenotypic chimera FL-J6JFH/J4NS5A was constructed by inserting ...AIM:To explore the function of Nonstructural 5A(NS5A) protein of genotype 2a(JFH1)in the replication and infection of hepatitis C virus(HCV).METHODS:Intergenotypic chimera FL-J6JFH/J4NS5A was constructed by inserting NS5A gene from 1b stain HC-J4 by the overlapping polymerase chain reaction (PCR)method and the restriction enzyme reaction.In vitro RNA transcripts of chimera,prototype J6JFH and negative control J6JFH1(GND)were prepared and transfected into Huh-7.5 cells with liposomes.Immunofluorescence assay(IFA),fluorescence quantitative PCR and infection assay were performed to determine the protein expression and gene replication in Huh-7.5 cells.RESULTS:The HCV RNA levels in FL-J6JFH/J4NS5A chimera RNA transfected cells were significantly lower than the wild type at any indicated time point(2.58 ±5.97×106 vs 4.27±1.72×104,P=0.032).The maximal level of HCV RNA in chimera was 5.6±1.8× 104 GE/μg RNA at day 34 after transfection,while the wild type reached a peak level at day 13 which was 126 folds higher(70.65±14.11×105 vs 0.56±0.90 ×105,P=0.028).HCV proteins could also be detected by IFA in chimera-transfected cells with an obviously low level.Infection assay showed that FL-J6JFH/J4NS5A chimera could produce infectious virus particles,ranging from 10±5 ffu/mL to 78.3±23.6 ffu/mL,while that of FL-J6JFH1 ranged from 5.8±1.5×102 ffu/mL to 2.5±1.4×104 ffu/mL.CONCLUSION:JFH1 NS5A might play an important role in the robust replication of J6JFH1.The establishment of FL-J6JFH/J4NS5A provided a useful platform for studying the function of other proteins of HCV.展开更多
