Optimization of the Conditions of SRAP Molecular Marker Used in Analysis of Fagopyrum tataricum

[Objective] The aim was to investigate the optimal conditions of SRAP molecular marker used in the analysis on Fagopyrum tataricum(L.)Gaertn.[Method]SRAP-PCR amplification system on Fagopyrum tataricum was optimized by interactive orthogonal design L27(313)in 5 elements(Mg2+,dNTP,Taq DNA polymerase,template DNA and primer)at 3 levels.And the non-denaturing and denaturing PAGE detection methods were compared.The comparative test of DYCZ-24F and DYCZ-20C electrophoresis operating systems was carried out.[Result]The effects of four single-factor(Mg2+,dNTP,Taq DNA polymerase and primer)and two interactions(Mg2+×dNTP,Mg2+×Taq DNA polymerase)on tartary buckwheat SRAP-PCR were significant.An optimal reaction system was established containing 1.5 mmol/L Mg2+,0.2 mmol/L dNTP,1.5 u Taq DNA polymerase,40 ng DNA,0.25 μmol/L primer and 2 μl 10×buffer.Seven samples of tartary buckwheat were amplified using this system,and electrophoresis results showed clear bands,high level of polymorphism and good reproducibility.The PCR products were tested by denaturing and non-denaturing PAGE,and the results showed that the non-denaturing PAGE,DYCZ-24F operating system was more suitable for SRAP analysis.[Conclusion]This study established a foundation for the construction of SRAP genetic map of tartary buckwheat. 展开更多

The height-width ratio limited value for rubber bearing isolated structure computed by uniform design method 被引量:7

Rubber isolation is the most mature control technology in practical application, and is widely used by short rigid buildings. However, many high isolation buildings have been built around the world in recent years, which do not follow the existing criterions and codes. Many researchers began to research the special problems caused by larger height-width ratio isolation structures. The overturning effect of high height-width ratio structures with rubber bearing is firstly studied. Considering the main factors, such as the height-width ratio of structures, type of site, the designed basic acceleration of ground motion and the decouple factor in horizon, computing experiment is defined with the Uniform Design Method, which is also known as designing isolation structure. The forces of the bearing under edge of structures based on the position of the rubber bearing are calculated. The result indicates that the rubber bearings will lose its functionality under very high tension and compressing force of earthquake motion in horizon and vertical, when the height-width ratio is over a certain value. Thus, based on the calculation result of isolation structures defined in the uniform design method, regression analysis is conducted, and also the rubber edge force regression formula are gotten, which has higher correlation and smaller standard deviation. This formula can be used to roughly calculate whether the pull force occurs at the edge of the building. By the edge bearings of isolation structure minimum force formula, the height-width ratio limited value of the isolation structure is deducted when rubber bearing has minimum force of zero. 展开更多

Many engineering optimization problems frequently encounter continuous variables and discrete variables which adds considerably to the solution complexity. Very few of the existing methods can yield a globally optimal solution when the objective functions are non-convex and non-differentiable. This paper presents a hybrid swarm intelligence ap-proach (HSIA) for solving these nonlinear optimization problems which contain integer, discrete, zero-one and continuous variables. HSIA provides an improvement in global search reliability in a mixed-variable space and converges steadily to a good solution. An approach to handle various kinds of variables and constraints is discussed. Comparison testing of several examples of mixed-variable optimization problems in the literature showed that the proposed approach is superior to current methods for finding the best solution, in terms of both solution quality and algorithm robustness. 展开更多

The objective was to design adaptive nonlinear output feedback laws such that the closed-loop systems were globally asymptotically stable, while the estimated parameters remained bounded. The proposed systematic strategy combined input-state-scaling with backstepping technique. The adaptive output feedback controller was designed for a general case of uncertain chained system. Furthermore, one special case was considered. Simulation results demonstrate the effectiveness of the proposed controllers. 展开更多

The collected spectrum of the fiber Bragg grating(FBG) and the loss of the detected optical power are discussed with respect to the 3-dB bandwidth of a Fabry-Perot(F-P) type tunable optical filter(TOF),respectively.And the optimized parameters of the TOF are obtained consequently.It is demonstrated that the relationship between the transmission wave-length of the TOF and its drive voltage is nonlinear.A new method to compensate the nonlinearity of the TOF is proposed.The linear sweeping of the transmission wavelength of the TOF is achieved through modifying the drive voltage using interpolation algorithm.It is observed that the average error and the maximum error of the transmission wavelength are reduced sharply under linear fit.The dynamic strain sensing is realized by use of a reference FBG and moving averaging algorithm in this system. 展开更多

Support Vector Machines (SVMs) have been widely used in pattern recognition and have also drawn considerable interest in control areas. Based on a method of least squares SVM (LS-SVM) for multivariate function estimation, a generalized inverse system is developed for the linearization and decoupling control of a general nonlinear continuous system. The approach of inverse modelling via LS-SVM and parameters optimization using the Bayesian evidence framework is discussed in detail. In this paper, complex high-order nonlinear system is decoupled into a number of pseudo-linear Single Input Single Output (SISO) subsystems with linear dynamic components. The poles of pseudo-linear subsystems can be configured to desired positions. The proposed method provides an effective alternative to the controller design of plants whose accurate mathematical model is un- known or state variables are difficult or impossible to measure. A continuous-time nonlinear model predictive controller(NMPC) was designed for a boiler-turbine unit.The controller was designed by optimizing a receding-horizon performance index,with the nonlinear system approximated by its Taylor series expansion with a certain order,the magnitude saturation constraints on the inputs satisfied by increasing the predictive time,and the rate saturation conditions on the actuators satisfied by tuning the time constant of the reference trajectories in a reference governor.Simulation results showed that the controller can drive the drum pressure and output power of the nonlinear boiler-turbine unit to follow their respective reference trajectories throughout a varying operation range and keep the water level deviation within tolerances.Comparison of the NMPC scheme with the generic model control(GMC) scheme indicated that the responses are slower and there are more oscillations in the responses of the water level,fuel flow input and feed water flow input in the GMC scheme when the boiler-turbine unit is operating over a wide range. 展开更多

The three-dimensional (3D) deformation effect of the slope engineering under the step-by-step excavation for the Antaibao surface mine was analyzed using the FLAC^3D technique. An optimal excavated scheme with a relatively steeper slope angle of 47° instead of 30° was successfully implemented at the west wall in the geological section 73200 of the mine area, where the 3D effect of the nonlinear large deformation of the slope was taken into account. Based on the above research conclusion, put forward the countermeasures of shortening mining length, excavating by different regions, timely foot backfilling to protect the excavated slope, and monitoring and feedback adjustment by studying the nonlinear effect. The results show that these countermeasures are effective in controlling maximum deformation and increasing the stability of the slope. 展开更多
