The rigid and elastic wheels of tire-soil contact models are proposed based on the Bekker pressure model and the Jonasi shear soil model, and they are described in the form of S-function to enhance the calculation efficiency and simulation accuracy. Finally, the interaction of truck and soil is simulated by Adams-Maflab co-simulation to study the influence of soft terrain on the ride comfort of vehicles. The co-simulation results reveal that the terrain properties have a great influence on the ride comfort of vehicles as well as driving speed, road roughness and cargo weight. This co-simulation model is convenient for adding the factor of terrain deformation to the analysis of vehicle ride comfort. It can also be used to optimize suspension system parameters especially for off-road vehicles.展开更多

The as-synthesized folate-conjugated Fe3O4 n...Highly biocompatible superparamagnetic Fe3O4 nanoparticles were synthesized by amide of folic acid (FA) ligands and the NH2-group onto the surface of Fe3O4 nanoparticles. The as-synthesized folate-conjugated Fe3O4 nanoparticles were characterized by X-ray diffraction diffractometer, transmission electron microscope, FT-IR spectrometer, vibrating sample magnetometer, and dynamic light scattering instrument. The in vivo labeling effect of folate-conjugated Fe3O4 nanoparticles on the hepatoma cells was investigated in tumor-bearing rat. The results demonstrate that the as-prepared nanoparticles have cubic structure of Fe3O4 with a particle size of about 8 nm and hydrated diameter of 25.7 nm at a saturation magnetization of 51 A·m2/kg. These nanoparticles possess good physiological stability, low cytotoxicity on human skin fibroblasts and negligible effect on Wistar rats at the concentration as high as 3 mg/kg body mass. The folate-conjugated Fe3O4 nanoparticles could be effectively mediated into the human hepatoma Bel 7402 cells through the binding of folate and folic acid receptor, enhancing the signal contrast of tumor tissue and surrounding normal tissue in MRI imaging. It is in favor of the tumor cells labeling, tracing, magnetic resonance imaging (MRI) target detection and magnetic hyperthermia.展开更多

Objective: To observe the effect and toxicity of docetaxel with cisplatin in anthracyclines-resistant advanced breast cancer. Methods: Forty-five female patients received docetaxel 60 mg/m^2 on dl and cisplatin 30 m...Objective: To observe the effect and toxicity of docetaxel with cisplatin in anthracyclines-resistant advanced breast cancer. Methods: Forty-five female patients received docetaxel 60 mg/m^2 on dl and cisplatin 30 mg/m^2 on d1-d3 of every 28 days. Every patient was treated with at least 2 cycles and a median of 3 cycles (2-6 cycles ). Results: Five patients achieved complete response (11.1%) and 18 partial response (40.0%), 10 stable disease (22.2%). The overall response rate was 51.1%. The clinical disease control rate was 73.3%, median time to tumor progression (TTP) was 7.8 months (1.0-34.5 months), median survival time was 17.6 months (range 1.9-48.0 months), and one year survival rate was 65.2%. The main side effect was marrow suppression. The treatment was well tolerated with grades Ⅲ and Ⅳ leukopenia in nine (20%) and ten (22.2%) patients. Conclusion: Combinative chemotherapy of docetaxel and cisplatin has a good anti-tumor activity on refractory advanced breast cancer with manageable toxicity.展开更多

However, few studies have reported its effect on non-small cell lung cancer (NSCLC), an...YSY01-A, as a novel proteasome inhibitor, has shown remarkable proliferation inhibitory effect on certain types of tumor cells. However, few studies have reported its effect on non-small cell lung cancer (NSCLC), and its underlying mechanism remains unknown. In our present study, we aimed to figure out the inhibitory effects as well as the mechanism of proteasome inhibitor YSY01-A against A549 cells both individually and in combination with cisplatin. A549 cell proliferation inhibition was assessed by SRB assay. Its related protein expression levels were determined by western blot assay. Moreover, the change of intracellular cisplatin accumulation was examined by ICP-MS assay. The results suggested that YSY01-A significantly (P〈0.001) inhibited the proliferation of A549 cells (IC50 was 36.2 nM for 72 h) in a concentration-dependent and time-dependent manner. Compared with the negative control group, YSY01-A (60 nM, 48 h) down-regulated PI3K/Akt pathway in A549 cells by increasing the expression level of PTEN (P〈0.01), and decreasing the expression level of PI3K (P〈0.001) and p-Akt/Akt (P〈0.001). When combined with cisplatin, YSY01-A of different concentrations (5, 10, 20 nM) could significantly increase the inhibition effects on A549 cells compared with the cisplatin alone treatment, showing a synergistic effect. At the same time, YSY01-A could remarkably block the cisplatin-induced down-regulation of hCTR1 in a concentration-dependent manner and increase cisplatin uptake from 2.01 to 2.47 fold (P〈0.001). In conclusion, compound YSY01-A could significantly inhibit proliferation of NSCLC A549 cells, showing a strong synergistic effect when combined with cisplatin. Down-regulation of PI3K/Akt pathway might be the mechanism of inhibitory effect of YSY0 l-A, and the combination with cisplatin might increase the expression of CTR1 and intracellular cisplatin accumulation.展开更多
