配子体预发育技术在海带配子体育苗生产中的应用效果研究

为探究配子体预发育技术在海带配子体育苗生产中的作用,本研究将海带配子体置于水温(13±1)℃、光照强度37.5μmol·m^(-2)s^(-1)、接种密度10.0 g·L^(-1)条件下进行预发育,并在2个10 t育苗池中利用预发育的配子体进行海... 为探究配子体预发育技术在海带配子体育苗生产中的作用,本研究将海带配子体置于水温(13±1)℃、光照强度37.5μmol·m^(-2)s^(-1)、接种密度10.0 g·L^(-1)条件下进行预发育,并在2个10 t育苗池中利用预发育的配子体进行海带育苗生产试验。试验结果显示,海带配子体经过11 d预发育,精囊形成率达到(98.1±3.3)%,排卵率达到(1.8±0.7)%,孢子体形成率达到(1.2±0.5)%。在10 t培育池中,与正常配子体育苗比较,预发育配子体苗种达到商品规格的时间提前9.3%,体长达到≥0.2 cm的苗种密度提高28.4%,脱苗率降低18.6%,缺苗率降低70.8%。结果表明,在海带配子体育苗生产中,利用配子体预发育技术可缩短育苗池中海带苗种的培育时间,提高苗种质量。 展开更多

研究海带(Saccharina japonica)配子体预发育的适宜光照强度、培育密度和发育程度,为配子体预发育技术应用于海带配子体育苗提供技术支撑。以雌配子体发育率、排卵率、精囊形成率为考察指标,确定海带配子体预发育的适宜光照强度和培育密... 研究海带(Saccharina japonica)配子体预发育的适宜光照强度、培育密度和发育程度,为配子体预发育技术应用于海带配子体育苗提供技术支撑。以雌配子体发育率、排卵率、精囊形成率为考察指标,确定海带配子体预发育的适宜光照强度和培育密度;以出苗率和出苗时间为考察指标,确定海带配子体预发育的适宜发育程度。结果发现,在温度(13±1)℃、培育密度10.0 g·L^(-1)时,海带配子体在37.5~62.5μmol·m^(-2)s^(-1)光照条件下预发育速度显著快于25.0μmol·m^(-2)s^(-1);在(13±1)℃、光照强度37.5μmol·m^(-2)s^(-1)时,海带配子体在5.0~10.0 g·L^(-1)密度下发育速度显著快于15.0~20.0 g·L^(-1)密度条件;在培育密度达到15.0 g·L^(-1)及以上时,海带配子体只有部分发育,其余部分进行丝状体生长。海带配子体在温度(13±1)℃、光照强度37.5μmol·m^(-2)s^(-1)、培育密度10.0 g·L^(-1)条件下,预发育11~12 d后,进行出苗测试,培育5 d的出苗率达到96.0%以上,未进行预发育的配子体出苗测试9 d的出苗率为90.7%,预发育配子体比未预发育配子体提前4 d达到90.0%以上出苗率。试验结果表明,海带配子体预发育的适宜光照强度为37.5μmol·m^(-2)s^(-1)、适宜培育密度为10.0 g·L^(-1)、适宜预发育时间为11~12 d。 展开更多

AIM: To analyze the effectiveness of the endoscopic therapy and to identify prognostic factors for recurrent bleeding. METHODS: Retrospective study of patients with gas- trointestinal bleeding secondary to Dieulafoy's lesion (DL) from 2005 to 2011. We analyzed the demographic characteristics of the patients, risk factors for gastro- intestinal bleeding, endoscopic findings, characteristics of the endoscopic treatment, and the recurrence of bleeding. We included cases in which endoscopy de- scribed a lesion compatible with Dieulafoy. We exclud- ed patients who had potentially bleeding lesions such as angiodysplasia in other areas or had undergone other gastrointestinal endoscopic procedures. RESULTS: Twenty-nine patients with DL were identi- fied. Most of them were men with an average age of 71.5 years. Fifty-five percent of the patients received antiaggregatory or anticoagulant therapy. The most common location for DL was the stomach (51.7%). The main type of bleeding was oozing in 65.5% of cases. In 27.6% of cases, there was arterial (spurting) bleeding, and 6.9% of the patients presented with an adherent clot. A single endoscopic treatment was ap- plied to nine patients (31%), eight of them with adren- aline and one with argon, while 69% of the patients received combined treatment. Six patients (20.7%) presented with recurrent bleeding at a median of 4 d after endoscopy (interquartile range = 97.75). Within these six patients, the new endoscopic treatment ob- tained a therapeutic success of 100%. The presence of arterial bleeding at endoscopy was associated with a higher recurrence rate for bleeding (50% vs 33.3% for other type of bleeding) rP = 0.024, odds ratio (OR) = 8.5, 95% CI = 1.13-63.87]. The use of combined en- doscopic treatment prevented the recurrence of bleed- ing (10% vs 44.4% of single treatment) (P = 0.034, OR = 0.14, 95% CI = 0.19-0.99). CONCLUSION: Endoscopic treatment of DL is safe and effective. Adrenaline monotherapy and arterial (spurting) bleeding are associated with a high rate of bleeding recurrence. 展开更多

Predicting the Reproduction Strategies of Several Microalgae Through Their Genome Sequences 被引量:1

Documenting the sex and sexual reproduction of the microalgae is very difficult, as most of the results are based on the microscopic observation that can be heavily influenced by genetic, physiological and environmental conditions. Understanding the reproduction strategy of some microalgae is required to breed them in large scale culture industry. Instead of direct observation of sex and sexual reproduction under microscope, the whole set or the majority of core meiosis genes may evidence the sex and sexual reproduction in the unicellular algae, as the meiosis is necessary for maintaining the genomic stability and the advantages of genetic recombination. So far, the available genome sequences and bioinformatic tools (in this study, homolog searching and phylogenetic analysis) allow us to propose that at least 20 core meiosis genes (among them 〉6 must be meiosis specific) are enough for an alga to maintain its sexual reproduction. According to this assumption and the genome sequences, it is possible that sexual reproduction was carried out by Micromonas pusilla and Cyanidiosehyzon merolae, while asexual reproduction was adopted by Bigelowiella natans, Guillardia theta, Nannochloropsis gaditana, N. oeeanica, Chlorella variablis, Phaeodactylum tricornutum and Thalassiosira pseu- donana. This understanding will facilitate the breeding trials of some economic microalgae (e.g., N. gaditana, N. oceanica, C. vari- ablis and P. tricornutum). The Chaoyanggou oil field is a fractured low-permeability reservoir, where the distribution of oil and gas is controlled by the distribution and development of fractures.Based on outcrop, drilling core, thin section and log data, the development characteristics of fractures in this area are described.On this basis, the degree of fracture development was predicted by quantitative analysis of fracture strength and numerical simulation.The result shows that four groups of structural fractures, i.e., in near NS, and EW directions and in due NW and NE directions, were developed in the reservoir, with the nearly NS and EW fractures dominant, which are the along bedding decollement fractures formed by compressive folding action, while low angle shear fractures are related to thrusts.These fractures are mainly formed in the reversed tectonic stage at the end of the Mingshui formation during the Cretaceous period.The degree of fracture development is controlled by such factors as lithology, stratum thickness, faults, folds and depth.The fractures are developed with a clear zonation and are best developed in the northern zone, moderately developed towards the south and poorly developed in the middle zone.These prediction results are in good agreement with interpretation results from logs. 展开更多
