目的:研究白木香结香过程中苄基丙酮与浸出物含量(%)之间的相关性。方法:采用气相色谱法测定样品中苄基丙酮含量。按中国药典(2010年版)一部沉香鉴别项下相关要求测定样品醇溶性浸出物的含量,并进行理化鉴别;应用"小孔滴注法"... 目的:研究白木香结香过程中苄基丙酮与浸出物含量(%)之间的相关性。方法:采用气相色谱法测定样品中苄基丙酮含量。按中国药典(2010年版)一部沉香鉴别项下相关要求测定样品醇溶性浸出物的含量,并进行理化鉴别;应用"小孔滴注法"进行人工造香试验。结果:苄基丙酮浓度在5.780~104.0μg/mL(r=0.9997)及0.07500~28.20μg/mL(r=0.9995)范围内有良好的线性关系;加样回收率为99.9%~101.6%(n=6),RSD=0.65%。沉香商品药材中苄基丙酮与浸出物含量之间不呈正比关系。随着人工造香时间的增加,有些白木香结香部位的醇溶性浸出物和苄基丙酮含量均呈增加趋势;有些白木香结香部位醇溶性浸出物含量增加,而苄基丙酮含量呈下降趋势。结论:气相色谱法测定沉香中苄基丙酮含量操作简便,测定结果准确、重复性好;"小孔滴注法"人工造香技术快速可行,为沉香形成机理研究提供基础。 展开更多

This paper discusses on the findings of a survey on the present perspective of quality mission among the ma nufacturing practitioners in Hong Kong. Further, the quality approaches and techniques adopted in Hong Kong manufacturing to improve quality are looked into. The results of this study reveal that the present view on quality is to focus on customers, to improve the operational processes and to increase the total involvement among the staff in the company. In addition, it is found that the difference in working experience and types of industry would affect the perspective of quality as an individual and as a company to a certain extent. Moreover, it is interesting to find that SPC and QCC are widely used in manufacturing for controlling operational processes. TQM and ISO quality system are often adopted in large electronics companies. However, zero defect program is not common in the three industries while benchmarking emerged as a useful technique. 展开更多

镰刀菌诱导结香对白木香叶内生真菌分布和群落构成的影响研究 被引量:7

为了研究沉香形成前后白木香叶组织中内生真菌的分布和群落构成,采用石蜡永久制片法观察白木香叶组织显微结构,细胞化学法确定内生真菌的分布。通过改良的CTAB法对白木香进行总DNA提取,利用真核生物通用引物同时对植物和内生真菌rDNA IT... 为了研究沉香形成前后白木香叶组织中内生真菌的分布和群落构成,采用石蜡永久制片法观察白木香叶组织显微结构,细胞化学法确定内生真菌的分布。通过改良的CTAB法对白木香进行总DNA提取,利用真核生物通用引物同时对植物和内生真菌rDNA ITS序列进行PCR扩增,构建内生真菌rDNA ITS文库,RFLP分析和序列测定,用PUAP软件进行系统发育分析,构建最大简约树。结果表明:镰刀菌诱导结香前后,白木香内生真菌菌丝主要分布于海绵组织和韧皮部,结香后韧皮部内生真菌菌丝相对丰度明显增大;茎点霉是未结香白木香叶内生真菌优势种群,刺盘孢菌是已结香白木香叶内生真菌优势种群;刺盘孢菌是未结香和已结香白木香唯一共有内生真菌,且丰度差异较大。不依赖分离培养的分子生物学方法可快速直观地用于植物组织内生真菌的鉴定。白木香叶内生真菌具有一定的遗传多样性,沉香形成前后白木香内生真菌群落构成差异较大。 展开更多

目的白木香树在自然条件下不产生树脂,以自然、微生物或人工方式可使之形成沉香树脂。"小孔滴注法"接种镰刀菌人工诱导白木香结香,8个月后野外采集白木香结香部位(分泌黑色树脂的树干组织),提取其总RNA,为阐明沉香特征产物的代谢途... 目的白木香树在自然条件下不产生树脂,以自然、微生物或人工方式可使之形成沉香树脂。"小孔滴注法"接种镰刀菌人工诱导白木香结香,8个月后野外采集白木香结香部位(分泌黑色树脂的树干组织),提取其总RNA,为阐明沉香特征产物的代谢途径、揭示人工诱导白木香结香的分子机制奠定基础。方法 "小孔滴注法"人工造香,手工割取白木香结香部位。分别采用CTAB-Li Cl法、Trizol试剂法、RN09试剂盒法、改良异硫氰酸胍-CTAB法和Qiagen试剂盒法提取总RNA并检测完整性。结果与其它四种提取方法相比,Qiagen试剂盒法提取的RNA条带清晰,OD260/OD280比值在1.8-2.0的范围内。结论 Qiagen试剂盒法适用于镰刀菌人工诱导白木香结香部位总RNA提取,对其它次生代谢物质含量较高的植物中提取总RNA具有一定的借鉴意义。 展开更多

Research on structure of tectonically deformed coals(TDC) is a key issue in coal and gas outburst prevention and coalbed methane(CBM) exploitation.This paper presents a summary on the research progress in TDC's structural-genetic classification,tectonic strain influence on coal microstructure,coal porosity system,coal chemical structure and constituents,and their relationship with the excess coalbed methane.Previous studies suggested that tectonic deformation had significant influence on coal microstructure,coal super microstructure,and even chemical macromolecular structure.The main mechanisms of coal deformation are the tectonic stress degradation and polycondensation metamorphism(dynamical metamorphism).Besides,under different deformation mechanisms,the ultra-and micro-structure and chemical constituents of TDC presented distinct characteristics.Based on these achievements,we propose one possible evolutionary trend of TDC with different deformation mechanisms,and suggest that the coal and gas outburst in the TDC,especially in the mylonitic coals,may be not only controlled by geological structure,but also influenced by the tectonic stress degradation of ductile deformation.Therefore,further study on TDC should be focused on the controlling mechanism of deformation on structure and composition of coal,generation conditions and occurrence state of excess coalbed methane from deformation mechanism of coal. 展开更多
