驼乳清蛋白影响糖尿病大鼠胰腺β细胞功能的效果以及作用机制初探 被引量:5

目的观察驼乳清蛋白对于糖尿病模型大鼠胰腺β细胞功能的影响,并初步分析驼乳清蛋白的作用机制。方法选取实验用Wistar SPF级300~800 g的雄性大鼠10只以链尿佐菌素(STZ)诱发法建立糖尿病大鼠模型,分离筛选胰腺β细胞后进行体外培养,将... 目的观察驼乳清蛋白对于糖尿病模型大鼠胰腺β细胞功能的影响,并初步分析驼乳清蛋白的作用机制。方法选取实验用Wistar SPF级300~800 g的雄性大鼠10只以链尿佐菌素(STZ)诱发法建立糖尿病大鼠模型,分离筛选胰腺β细胞后进行体外培养,将以胎牛血清RPMI-1640作为培养基的胰腺β细胞作为模型对照组2只,将以乳清蛋白作为培养基的胰腺β细胞作为实验组3只,同时分离筛选正常大鼠的胰腺β细胞进行体外培养(空白对照组5只),应用MTT比色法对各组胰腺β细胞生长曲线进行绘制,并在干预后24 h、48 h、72 h、96 h分别进行葡萄糖依赖性促胰岛素释放肽(GIP)、胰岛素样生长因子-1(IGF-1)、胰高血糖素样多肽1(GLP-1)、白细胞介素-1α(IL-1α)以及超氧化物歧化酶(SOD)水平测定。结果①相同时间点,空白对照组胰腺β细胞贴壁率、细胞相对增殖速度GIP(78.50±1.10)pmol/L、IGF-1(38.65±3.25)μg/L、GLP-1(258.30±2.65)pmol/L、SOD(38.65±3.25)U/ml水平显著高于模型对照组和实验组(P<0.05),IL-1α(38.65±2.65)pg/ml显著低于模型对照组和实验组(P<0.05);②相同时间点,实验组胰腺β细胞贴壁率、细胞相对增殖速度GIP(60.45±1.89)pmol/L、IGF-1(164.76±2.82)μg/L、GLP-1(31.56±2.32)pmol/L、SOD(42.25±2.85)U/ml显著高于模型对照组(P<0.05),IL-1α(40.03±2.76)pg/ml显著低于模型对照组(P<0.05)。结论驼乳清蛋白能够促进胰腺β细胞生长,其可能成为2型糖尿病防治的新方法。 展开更多

Asthma is a chronic inflammatory disease with excessive irritability and airway narrowing and inflammation plays an important role in it.There are 300 million asthmatic people in the world currently.Main treatments for asthma include two groups of bronchodilators and inflammation controllers.Researches was continued in order to reach new treatments to reduce drug side effects and treatment-resistant cases or the types associated with weak treatment response.Today,World Health Organization recommends the application of traditional medicine especially in underdeveloped countries because of insufficient health resources and spread of diseases.Iranian traditional medicine(ITM)or Persian medicine is one of the oldest comprehensive traditional medicines with thousands years history which could help us to manage different diseases.The aim of this hypothesis is to investigate the camel milk as a complementary treatment of asthma because this chronic disease is sometimes resistant or response weakly to the treatment.In this article,the administration of camel milk in lung inflammatory diseases was studied by searching the PubMed and Scopus scientific databases.The results of this study indicated that camel milk due to having anti-inflammatory,immunomodulatory and anti-oxidant effects could decrease the levels of inflammatory factors such as tumor necrosis factorα,interleukin-17(IL-17),IL-6,IL-1B and transforming growth factor-β1 in a human and animal samples with inflammatory diseases.Besides,based on ITM,camel milk was used in treatment patients with asthma.But,clinical studies are needed to validate the effectiveness of camel milk in asthma and its mechanisms. 展开更多
