Determination and Analysis of Liposoluble Chemical Constituents from Ictalurus punctatus Oil

The lipids was extracted from the internal organs of Ictalurus punctatus by microwave, and its liposoluble chemical constituents was identified and quantified by gas chromatography (GC)/electron impact (EI) mass spectrometry (MS). Subsequent-ly, the acid catalysis method was used to convert fatty acids to methyl-esters. The total lipids in the pure fat of I. punctatus were found to be about 99%. Twenty-three fatty acids were identified. The major fatty acids found in the oils were normal mo-nounsaturated, polyunsaturated fatty acids and alkane paraffin. Unsaturated fatty acids comprised 72.84% of the total, and the major components of monounsaturated fatty acids were C18:1 (48.52%), while the polyunsaturated fatty acids were mainly composed of C18:2 (16.54%). Saturated fatty acids comprised approximately 19.21%of the total. The main components were C16:0 (14.46%) and C18:0 (4.17%), most of which were medium-long-chain fatty acids. Therefore, catfish oil can be used as an important dietary source of functional fatty acids. The difference between the pro-portion of unsaturated and saturated fatty acids appears to be significant compared to fish of the Cyprinidae family, as catfish skin and muscle have higher protein content and lower fatty content. This study confirms that catfish has excellent nutri-tional quality. 展开更多

A 3-month feeding experiment was conducted in an in-door seawater system to investigate the effect of dietary vitamin E (Ve) on the sperm quality of turbot (Scophthalmus maximus). D-α-tocopherol acetate was supplemented to the basal (control) diet (65.14 mg kg-l Ve) to obtain low and high levels of dietary Ve (244.60 mgkg-1, LVe; 721.60 mg kg-1, HVe). Compared with the con- trol, sperm concentration was significantly increased in Ve-supplemented groups (LVe and HVe); while relative sperm volume and testis-somatic index were significantly increased in group HVe only. Sperm motility duration was significantly longer in group HVe than in the control, but no significant difference was observed in percent motility among groups. Sperm size, the uniformity of mito- chondrial size, and the integrity of mitochondria cristae and plasma membrane were improved by dietary Ve, especially in HVe. The content of Ve in testis and liver as well as polyunsaturated fatty acids in sperm increased with dietary Ve. This study was conducted to compare lipid and fatty acid composition of cod,haddock and halibut.Three groups of cod(276 g±61 g),haddock(538 g±83 g)and halibut(3704 g±221 g)were maintained with commercial feeds mainly based on fish meal and marine fish oil for 12 weeks prior to sampling.The fatty acid compositions of muscle and liver were determined by GC/FID after derivatization of extracted lipids into fatty acid methyl esters(FAME).Lipids were also fractionated into neutral and polar lipids using Waters silica Sep-Pak(R).The phospholipid fraction was further separated by high-performance thin-layer chromatography(HPTLC)and the FAME profile was obtained.Results of the present study showed that cod and haddock were lean fish and their total muscle lipid contents were 0.8% and 0.7%,respectively,with phospholipid constituting 83.6% and 87.5% of the total muscle lipid,respectively.Halibut was a medium-fat fish and its muscle lipid content was 8%,with 84% of the total muscle lipid being neutral lipid.Total liver lipid contents of cod,haddock and halibut were 36.9%,67.2% and 30.7%,respectively,of which the neutral lipids accounted for the major fraction(88.1%-97.1%).Polyunsaturated fatty acids were the most abundant in cod and haddock muscle neutral lipid.Monounsaturated fatty acid level was the highest in halibut muscle neutral lipid.Fatty acid compositions of phospholipid were relatively constant.In summary,the liver of cod and haddock as lean fish was the main lipid reserve organ,and structural phospholipid is the major lipid form in flesh.However,as a medium-fat fish,halibut stored lipid in both their liver and muscle. 展开更多

To determine the replacement of fish oil with vegetable oils in the diet of juvenile Jade perch Scortum barcoo, four feeds with each a different oil (fish, sunflower, linseed and a mixture of 75% canola and 25% linseed oil), were fed to Jade perch reared in recirculating aquaculture systems (RAS). The trial lasted for 10 weeks and the fatty acid (FA) profile of both feed and fish muscle tissue were examined. There was no difference in growth, feed conversion rate (FCR) and mortality. The fish grew from 10 g to 110 g with a FCR of 1.25 and 0 mortality. The FA profile of the fish muscle tissue reflected the FA profile of the feed. The flesh of the fish that were fed the linseed oil diet, were extremely high in omega-3 (n-3) polyunsaturated FA (n-3 PUFA) with 3.75% of wet weight. This is one of the highest concentrations of n-3 PUFA ever recorded in fish flesh. In a finishing feeding test, the remaining vegetable oil fed fish were fed the fish oil diet for another two weeks immediately after the 10 weeks trial, to check for a possible recovery of n-3 highly unsaturated fatty acids (HUFA). The wash out rate of FA towards n-3 HUFA in the muscle tissue was about 25% over this two weeks period. 展开更多

Anchovy is a key species in the Yellow Sea ecosystem. An accurate estimate of anchovy abundance is vital for the management of the anchovy stock and measurement of the ecosystem response to changes in anchovy abundance. However, the acoustic fish abundance estimate may be biased by 30%-40% if the fat-content induced target strength variation is not taken into account. We measured the monthly variation in the fat content of anchovy (Engraulis japonicus) in the Yellow Sea, and evaluated the potential effect of variation in fat content on the acoustic assessment of anchovy abundance. The fat content of anchovy varied seasonally, with two maxima and two minima in a year. The highest fat content (14.75%) was measured in the pre-spawning period in May, and the lowest fat content (2.48%) was measured during the post-spawning period in October. Fat content appeared to correlate with water content, but not body size. Assuming that the target strength is decreased by 0.2dB for every 1% increase in fat content, the seasonal difference in the target strength of anchovy may be as high as 2.45 dB. Given this, the acoustic abundance estimate may be biased by between 43% and 76%. Our results highlight the need for more information on the changes in fat content of fishes whose abundance is estimated by acoustic surveys. 展开更多

Five treatments with rotifers and Artemia differing in n-3 HUFA were prepared by enriching them with various oil emulsions. Results indicated that dietary n-3 HUFA significantly influence fish n-3 HUFA levels and are essential for growth and survival of black sea bream larvae. The results also indicated that the incorporation of n-3HUFA TG (triacylglycerols) into tissues of larval black sea bream was more effective from natural fish oil in comparison with n-3 HUFA fatty acid ethyl esters from ethyl-esterified oil. 展开更多

Objective:This study aims to provide the material basis for the molecular mechanism of Yuxingcao(Houttuynia cordata Thunb,HCT)through the method of evidence-based study to summarize the natural constituents isolated from HCT.Methods:We searched CNKI,Wanfang,VIP,Pubmed and relevant conference compilations.The keywords were'Yuxingcao or Houttuynia cordata Thunb'and'components'or'ingredients'or'constituent'or'volatile oil'or'flavonoids'or'terpene'or'content',both in Chinese and English.According to the inclusion and exclusion criteria,the reported compositions and contents have been summarized,and SPSS software was used to draw the boxplot of contents.Results:A total of 603 natural compounds in 11 categories were obtained from pooled articles.In the diverse components,the number of aliphatic compounds(n=259)and terpenoids(n=158)are more than those of flavonoids(n=26),alkaloids(n=42)and aromatics(n=42).While,in the part of volatile oils of HCT,the largest components are aliphatic compounds(mainly distributed on the ground)and terpenoids(mainly distributed in the underground).Although,methyl n-nonylketone is distributed in the whole herb plant,a large proportion is present in the underground parts.As for non-volatiles,the flavonoid content(mainly distributed on the ground)was the highest,among which quercetin and its glycosyl derivatives were the prominent.Conclusion:The results of this study provide a more comprehensive material basis for the further study of HCT.It is also helpful to explain the mechanisms of anti-oxidant,anti-inflammatory,anti-bacterial,anti-viral and anti-cancer effects from the molecular target and molecular network levels. 展开更多

The objective of this study was to evaluate the effect of anthropogenic pollution on DNA damage and the fatty acid profile of the bottom dweller fish (Cirrhinus mr^gala), collected from the River Chenab, in order to assess the effect of the toxicants on the quality of the fish meat. The levels ofCd, Hg, Cu, Mn, Zn, Pb, Cr and Sn and of phenols from this river were significantly higher than the permissible limits set by the USEPA. Comet assays showed DNA damage in Cirrhinus mr^gala collected from three different sampling sites in the polluted area of the river. Significant differences were observed for DNA damage through comet assay in fish collected from polluted compared to control sites. No significant differences were observed for DNA damage between farmed and fish collected from upstream. The micronucleus assay showed similar trends. Fish from the highly polluted sites showed less number of fatty acids and more saturated fatty acids in their meat compared to fish from less polluted areas. Several fatty acids were missing in fish with higher levels of DNA in comet tail and micronucleus induction. Long-chain polyunsaturated fatty acids, eicosapentaenoic acid (20:5n-3) was found missing in the fish from polluted environment while it was found in considerable amount in farmed fish 7.8±0.4%. Docosahexaenoic acid (22:6n-3) also showed significant differences as 0.1±0.0 and 7.0±0.1% respectively, in wild polluted and fanned fishes. 展开更多

AIM:To investigate the liver-protecting effect of parenteral nutrition(PN) support with omega-3 fatty acids in a randomized controlled clinical trial.METHODS:Sixty-six patients with the diagnosis of end-stage liver disease or hepatic cellular carcinoma were admitted to the Affiliated Drum Tower Hospital,Nanjing University,China for orthotopic liver transplantation.The patients were randomly divided into two groups:PN group(n = 33) and polyunsaturated fatty acid(PUFA) group(n = 33).All patients received isocaloric and isonitrogenous PN for seven days after surgery,and in PUFA group omega-3 fish oil lipid emulsion replaced part of the standard lipid emulsion.Liver function was tested on days 2 and 9 after surgery.Pathological examination was performed after reperfusion of the donor liver and on day 9.Clinical outcome was assessed based on the post-transplant investigations,including:(1) post-transplant mechanical ventilation;(2) total hospital stay;(3) infectious morbidities;(4) acute and chronic rejection;and(5) mortality(intensive care unit mortality,hospital mortality,28-d mortality,and survival at a one-year post-transplant surveillance period).RESULTS:On days 2 and 9 after operation,a significant decrease of alanine aminotransferase(299.16 U/L ± 189.17 U/L vs 246.16 U/L ± 175.21 U/L,P = 0.024) and prothrombin time(5.64 s ± 2.06 svs 2.54 s ± 1.15 s,P = 0.035) was seen in PUFA group compared with PN group.The pathological results showed that omega-3 fatty acid supplement improved the injury of hepatic cells.Compared with PN group,there was a significant decrease of post-transplant hospital stay in PUFA group(18.7 d ± 4.0 d vs 20.6 d ± 4.6 d,P = 0.041).Complications of infection occurred in 6 cases of PN group(2 cases of pneumonia,3 cases of intra-abdominal abscess and 1 case of urinary tract infection),and in 3 cases of PUFA group(2 cases of pneumonia and 1 case of intra-abdominal abscess).No acute or chronic rejection and hospital mortality were found in both groups.The one-year mortality in PN group was 9.1%(3/33),one died of pulmonary infection,one died of severe intra-hepatic cholangitis and hepatic dysfunction and the other died of hepatic cell carcinoma recurrence.Only one patient in PUFA group(1/33,3.1%) died of biliary complication and hepatic dysfunction during follow-up.CONCLUSION:Post-transplant parenteral nutritional support combined with omega-3 fatty acids can significantly improve the liver injury,reduce the infectious morbidities,and shorten the post-transplant hospital stay. 展开更多

In the present study, the effects of season on fatty acid composition, total lipids, and ω3/ω6 ratios of northern pike muscle lipids in Klzlhrmak River (Klrlkkale, Turkey) were investigated. A total of 35 different fatty acids were determined in gas chromatography. Among these, palmitic, oleic, and palmitoleic acids had the highest proportion. The main polyunsaturated fatty acids (PUFAs) were found to be docosahexaenoic acid, eicosapentaenoic acid, and arachidonic acid. There were more PUFAs than monounsaturated fatty acids (MUFA) in all seasons. Similarly, the percentages of ω3 fatty acids were higher than those of total ω6 fatty acids in the fatty acid composition. ω3/ω6 ratios were calculated as 1.53, 1.32, 1.97, and 1.71 in spring, summer, autumn and winter, respectively. Overall, we found that the fatty acid composition and ω3/ω6 fatty acid ratio in the muscle of northern pike were significantly influenced by season. 展开更多

Lipid oxidation in salt-dried yellow croaker(Pseudosciaena polyactis) was evaluated during processing with commonly used analytical indices, such as the peroxide value(POV), the thiobarbituric acid reactive substances(TBARS) value, and oxidative-relative lipoxygenase(LOX) activity. Additionally, fatty acids were analyzed using gas chromatography-mass spectrometry. Both POV and TBARS increased significantly(P < 0.05) at the rinsing stage. POV reached its peak value of 3.63 meq O_2 per kg sample at the drying stage, whereas TBARS constantly increased from 0.05 to 0.20 mg MDA per kg sample. Processing of salt-dried yellow croaker had an extremely significant(P < 0.01) effect on LOX activity. Twenty-six fatty acids were identified. Combined eicosapentaenoic acid(EPA; C20:5n3) and docosahexaenoic acid(DHA; C22:6n3) content varied between(19.20 ±0.37) mg g^(-1) and(23.45 ± 1.05) mg g^(-1). The polyunsaturated fatty acid/saturated fatty acid(PUFA/SFA) ratio in yellow croaker was 0.73–1.10, and the n-6/n-3 PUFA ratio was approximately 0.13–0.20. The contents of most fatty acids varied significantly(P <0.05) during the different processing stages, and these differences were caused by lipid oxidation. Effect of Ice Storage on Fatty Acids Profile of Freshwater and Marine Fishes Available at Malaysia Wet Market

This study was conducted in the intention to identify and determine the fatty acid profile in freshwater fishes and marine fishes available at Malaysian wet market as well as its deterioration kept in ice storage up to 21 days. Three freshwater fishes: Oreochromis sp. commonly known as Tilapia, Monopterus alba commonly known as Belut, Pangasius hypothalamus commonly known as Patin and marine fish Rastrellinger kanagurta commonly known as Kembung, Thunnus tonggol commonly known as Tongkol and Epinephelus sp. commonly known as Kerapu with average weight of 500-600 g were tested with storage time of 1, 3, 6, 9, 12, 15. 18 and 21 days kept ice storage. The Fatty Acid Methyl Ester (FAMEs) were prepared using one step method and were identified by using gas chromatograph Agilent 6890N equipped with flame ionization detector. Separation was performed by injecting 1μL of a sample into the capillary column DB-225MS column (30 m length × 0.250 mm diameter × 0.25 μm film thickness). The fatty acids peaks were indentified using Agilent Technologies software 5988-5871EN and comparing their retention time against the authentic standard Supelco 37 Component FAME Mix. Statistical results showed that palmitic acid (CI6:0) were the major fatty acids among the saturated fatty acids (SFA) while oleic acid (C 18:1 n9) were the major monounsaturated fatty acids (MUFA) detected in each sample studied. Both linoliec acid (C 18:2n6) and arachadonic acid (C20:4n6) were predominant in total omega-6 polyunsaturated fatty acids (PUFA). The essential fatty acids show prominent in ecosapentanoic acid (C20:5n3) and docosahexanoic acid (C22:6n3). During ice storage polyunsaturated fatty acids (PUFA) and monounsaturated fatty acids (MUFA) decrease as the time of storage increase. There is significant decrease in polyene index (PI) index of all the species throughout the storage time, The PI in freshwater fishes decrease by 70% in Tilapia, 90.1% in Belut and 90.9% in Patin, while for marine fishes, PI in Kembung decreases by 67.8%, 85.7% in Kerapu and 65.93% in Tongkol during kept in ice storage. During ice storage, the percentage of EPA, DHA and ALA decrease in Tilapia (55.36%, 13.86% and 75.41%), Belut (78.55%, 87.74 % and 74.53%) and Patin (82.45%, 40.0% and 82.10%), respectively. The same result were also observed in the marine fishes where the percentage ofEPA, DHA and ALA decrease in Kembung (94.27%, 49.03% and 88.52%), in Kerapu (73.03%, 88.18% and 38.73%) and Tongkol (70.05%, 38.24% and 57.14%), respectively. 展开更多

This study investigated the effect of dietary lipid on the growth, fatty acid composition and Δ5 fatty acyl desaturase genes(Fads) expression of juvenile abalone(Haliotis discus hannai Ino) hepatopancreas. Six purified diets were formulated to contain tripalmitin(TP), olive oil(OO, 72.87% 18:1n-9), grape seed oil(GO, 68.67% 18:2n-6), linseed oil(LO, 70.48% 18:3n-3), ARA oil(AO, 41.81% ARA) or EPA oil(EO, 44.09% EPA and 23.67% DAH). No significant difference in survival rate was observed among abalone fed with different diets. Weight gain rate(WGR) and daily growth rate of shell length(DGRSL) were significantly increased in abalone fed with diets containing OO, AO and EO, but decreased in abalone fed with LO diet(P < 0.05) in comparison with those fed with TP. High level of dietary 18:2n-6 resulted in higher content of n-6 polyunsaturated fatty acids(PUFAs) in abalone fed with GO than those fed with TP, OO, LO and EO(P < 0.05). n-3 PUFAs in abalone fed with LO was significantly higher than those in abalone fed with TP, OO, GO and AO(P < 0.05). The highest contents of 20:1n-9 and 22:1n-9 were observed in abalone fed with OO. The expression of Δ5 Fads in hepatopancreas of abalone was enhanced by high concentration of 18:3n-3 and suppressed by dietary LC-PUFAs; however it was not affected by dietary high concentration of 18:1n-9 or 18:2n-6. These results provided valuable information for understanding the synthesis of LC-PUFAs and nutritional regulation of Δ5 Fads expression in abalone. 展开更多

Extruded soybean meal (ESBM) was evaluated as a protein source for partial replacement of fish meal (FM) in diets of juvenile Litopenaeus vannarnei. In the control diet (Diet 1), FM protein was replaced with increasing dietary levels of ESBM (4.28%, 8.40%, 12.62%, 16.82%, and 25.26%) at 10%, 20%, 30%, 40%, and 60% levels (Diets 2 to 6, respectively). An eight-week feeding trial was conducted on 720 juvenile shrimp (0.67 g ± 0.01 g mean initial Weight), and nutrient digestibility of the six diets was determined. ESBM could replace 20% of FM without causing a significant reduction in growth of shrimp, but other dietary treatments strongly affected whole body composition. Crude protein content of the whole body fed Diet 6 was significantly lower than that fed Diet 2 (P〈0.05), while crude lipid content of the whole body fed Diet 5 or 6 was significantly higher than that fed Diet 2 (P〈0,05). Protein digestibilities of Diets 5 and 6 were significantly lower than that of Diet 1 (P〈0.05). Digestibility of lipids ranged from 96.97% in Diet 6 to 98.34% in Diet 3, whereas dry matter digestibility decreased with increasing replacement level. This study indicates that 20% FM replacement with ESBM in the basic diet containing 40% protein and 30% FM is optimal for juvenile L. vannamei. 展开更多

The results showed, UFA (Unsaturated Fatty Acid) and SFA (Saturated Fatty Acid) were 74/09 and 21/63%, respectively in fresh tissue. So that, DHA (C22:6) oleic acid (C 18:1c) had high amounts (15/07, 20/57) UFA and palmitic acid (C16:0) was the most (13/09%) SFA. The effects of freezing on fish tissue showed that UFA and SFA contents have reached to 58/79 and 22/17%, respectively at the end of cold storage. Also ω-3 and ω-6 series of fatty acids was 24/22 and 15/56% in fresh tissue, but their contents decreased to 8/68 and 5/11% at the end of period. Among, the fatty acids C22:6, C 18:1 c and C 16:0 had the most changes. The changes of fatty acids were significantly at 95% level expected for C18:0. 展开更多

Abstract A 60-day feeding experiment was conducted to investigate the influence of dietary omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) on growth, nutritional composition and immune function of marine fish Sebastiscus marmoratus. Five diets containing 3.6, 10.2, 18.2, 26.5, or 37.0 g/kg n-3 LC-PUFA were prepared. The results reveal significant influences of dietary n-3 LC-PUFA on the final weight, weight gain, specific growth rate, feed conversion ratio, and condition factor. As dietary n-3 LC- PUFA increased, weight gain and specific growth rate increased and were significantly higher in groups fed 18.2, 26.5 and 37.0 g/kgthan in groups fed 3.6 and 10.2 g/kg (P〈0.05); there was no significant difference between groups fed 18.2, 26.5, or 37.0 g/kg (P〉0.05). With increasing dietary n-3 LC-PUFA, eicosapentaenoic acid and docosahexenoic acid content in muscle and liver increased significantly, immunoglobulin class M content gradually increased from 9.1 to 14.8 Ixg/L, and lysozyme activity content increased from 1 355 to 2 268 U/mL. Broken line model analysis according to weight gain indicated that a dietary n-3 LC-PUFA level of 18.2 g/kg is essential for normal growth at a fat level of 125 g/kg. Therefore, appropriate dietary n-3 LC-PUFA not only promote growth and improve the n-3 LC-PUFA content, but also enhance immune function in S. marmoratus. 展开更多
