Freeze drying and frozen preservation way was used to preserve a moderately thermophilic culture for bioleaching of chalcopyrite concentrate.After preservation of 15 months,the cell viability rate decreases to 22% wit...Freeze drying and frozen preservation way was used to preserve a moderately thermophilic culture for bioleaching of chalcopyrite concentrate.After preservation of 15 months,the cell viability rate decreases to 22% with a cell density of 7×107 mL-1.When the growth time was extended from 8 days to 14 days,cell density would increase in a large scale to about 3×108 mL-1.In the bioleaching experiments,unpreserved and preserved cultures were compared for dissolving chalcopyrite concentrate.Before 44 days,the unpreserved culture can reach a high copper extraction of about 17.4 g/L.While the preserved culture shows a rather low copper extraction,which is only 9.7 g/L.When the bioleaching time was extended to 80 days,copper extraction by preserved culture increases remarkably,and the concentration of copper finally achieves up to 18.3 g/L.On the other hand,copper extraction by the unpreserved culture does not show remarkable increase from 44th to the 80th day,and finally the total copper extraction is 19.8 g/L.As a result,total copper extraction in 80 days by preserved culture approaches that by unpreserved culture and freeze drying and frozen preservation even after 15 months does not bring much decrease of bioleaching ability.展开更多

H...Compacted loess is widely used as fills of road embankments in loess regions of northern China.Generally, densely-compacted loess can satisfy the requirements of embankment strength and postconstruction deformation. However, uneven subsidence, pavement cracks and other related damages can affect the integrity of loess subgrade after several years of operation,and even cause some hazards, especially in North China, where the strong freeze-thaw erosion occurs. In this study, cyclic freeze-thaw tests for both densely and loosely compacted loess samples were performed to determine the variation in engineering properties such as volume, void ratio, collapsible settlement,microstructure, and the related mechanisms were addressed. The experimental results showed that an obvious water migration and redistribution occurred within the samples during freeze-thaw cycles. Ice lenses and fissures could be identified in the upper frozen layers of the samples. After freeze-thaw cycles,the dry densities of the upper layers of samples changed significantly due to strong freeze-thaw erosion. The dry densities decreased for the dense sample and increased for the loose sample. It can be found that dense samples become loose, while loose samples became dense with the increasing number of freeze-thaw cycles. Their related void ratios changed reversely. Both void ratios tended to fall into a certain range, which verified the concept of a residual void ratio proposed by Viklander. The loosening process of densely compacted samples involves the formation of large pores, volume increase and density reduction as well as the related changes in mechanical properties because freeze-thaw cycles may be important contribution to problems of loess road embankments.Adverse effects of freeze-thaw cycles, therefore,should be taken into account in selecting loess parameters for the stability evaluation of road embankment in seasonally frozen ground regions.展开更多
A mixed culture of bioleaching microorganisms was enriched.Then the mixed culture was preserved by liquid nitrogen cryopreservation for 6 months and 12 months,respectively,using PEG-2000 as the protective agent.The ch...A mixed culture of bioleaching microorganisms was enriched.Then the mixed culture was preserved by liquid nitrogen cryopreservation for 6 months and 12 months,respectively,using PEG-2000 as the protective agent.The chalcopyrite leaching ability,activity and diversity of the mixed culture before and after preservation were compared.The results showed that the copper extraction rate was 95.7%in chalcopyrite bioleaching within 20 d by the original culture.After cryopreservation for 6 months and 12 months,the copper extraction rate of the mixed culture was 94.9%within 25 d and 93.6%within 35 d,respectively.The cell viability achieved 87%and 41%after being preserved for 6 months and 12 months,respectively.Furthermore,the ecology analysis identified Acidithiobacillus ferrooxidans,Acidithiobacillus caldus,Sulfobacillus thermotolerans and Pseudomonas aeruginosa in the original mixed culture.After cryopreservation for 12 months,the composition of community changed,but the predominant microorganisms still existed.展开更多
