[Method] The anatomical structure of infected and healthy leaves of tomato were observed and compared by using paraffin section method. The activity changes of SOD, POD and CAT in the infected leaves of tomato were determined. [ Result] The results revealed that there were some differences in anatomical structure between healthy and infected leaves. Some cells of infected leaves were damaged so that the leaves curled and became yellow, which affected the normal function of organs. Compared with control, enzyme activities in the tomato plants infected by TYLCV were enhanced at the early periods and higher than that in control, then started to decline at the middle and late periods but lower than that in control.[ Conclusion] After infection by TYLCV, the leaf anatomical structure of tomato was changed greatly and the protective enzyme system was damaged severely, and affected the normal physJological metabolic functions of tissues and organs in tomato in further.展开更多

[Method] In this paper, broad bean rust, fusarium rhizome rot, broad bean zonate spot, yellow leaf curl virus and normal leaves were studied using Fourier transform infrared spectroscopy combined with chemometrics. [Result] The spectra of the samples were similar, only with minor differences in absorption inten- sity of several peaks. Second derivative analyses show that the significant difference of all samples was in the range of 1 200-700 cm2. The data in the range of 1 200- 700 cm' were selected to evaluate correlation coefficients, hierarchical cluster analy- sis (HCA) and principal component analysis (PCA). Results showed that the correla- tion coefficients are larger than 0.928 not only between the healthy leaves, but also between the same diseased leaves. The values between healthy and diseased leaves, and among diseased leaves, are all declined. HCA and PCA yielded about 73.3% and 82.2% accuracy, respectively. [Conclusion] This study demonstrated that FTIR techniques might be used to detect crop diseases.展开更多

A wheat (Triticum aestivum L cv. Xiaoyan 54) VDE cDNA was obtained using RT-PCR method. Its deduced protein sequence shares high identity with that of Arabidopsis and rice. Southern blot revealed that there are three copies of VDE gene per haploid genome of wheat. VDE transcript levels were higher in green leaf than in root, seed and etiolated leaf. Results showed that D1 protein contents of PSⅡ in photosynthetic apparatus dropped, the generation of antheraxanthin (A) and zeaxanthin (Z) of xanthophyll cycle were inhibited partly, PSⅡ photochemical efficiency (F v/F m)and non-photochemical quenching (q N) were also decreased obviously. In addition, endogenous active oxygen scavenger—superoxide dismutase (SOD) reduced, superoxide anion radical (O -· 2) and malondialdehyde (MDA) accumulated, as a result, photooxidation of leaves occurred under chilling temperature and strong light stress conditions. Obvious differences in the changes of the above mentioned physiological parameters between indica and japonica rice were observed. Experiments in leaves treated with inhibitors under chilling temperature and strong light conditions showed that indica rice was more sensitive to chilling temperature with strong light and subjected to photooxidation more than japonica rice. Notable positive correlation between D1 protein contents and F v/F m or (A+Z)/(A+Z+V), and a marked negative correlation between F v/F m and MDA contents were obtained by regression analysis in indica and japonica rice during chilling temperature and strong light conditions. According to the facts mentioned above, it was inferred that PSⅡ photochemical efficiency(F v/F m) was the key index to forecast for the prediction of photooxidation under stress circumstances and the physiological basis were the synthetic capacity of D1 protein and the protection of xanthophyll cycle.展开更多

The structures were determined on the basis of spectroscopic methods (UV, IR, FAB_MS, 1H_NMR and 13 C NMR). Antifungal assay of all compounds showed that metabolite 1, quercitrin and quercetin were inhibitory to the growth of phytopathogenic fungus Helminthosporium sativum Pamel King et Bakke with minimum inhibitory concentrations (MICs) of 25, 50 and 50 μg/mL, respectively. Moreover, glycoside 1 and quercitrin were also shown to be able to inhibit the growth of Fusarium graminearum Schw. with MIC of 100 μg/mL. Resurrection plants which are able to quickly reactivate after falling into a period of anabiosis caused by dehydration have been very rare among angiosperms, especially among dicotyledons whose chlorophyll content an...Resurrection plants which are able to quickly reactivate after falling into a period of anabiosis caused by dehydration have been very rare among angiosperms, especially among dicotyledons whose chlorophyll content and chloroplast structure little changed in the course of desiccation, therefore has been called homoiochlorophyllous desiccation-tolerant plants (HDTs). Another type of resurrection angiosperms that lost its chlorophyll dining desiccation is called poikilochlorophyllous desiccation-tolerant plants (PDTs). HDTs have been received more attention because of simplicity of protection mechanism which is much easy to the study and utilization of the desiccation tolerance of resurrection angiosperms. Recent advances in studies of photosynthesis of resurrection angiosperms indicate that photochemical activities are sensitive indicators for the study of physiological state of resurrection angiosperms during desiccation and rehydration. Photochemical activities of resurrection angiosperms are inhibited with loss of water similar to those of general plants, however, the magic thing is that they could reactivate rapidly during rehydration even losing more than 95% water. Up-regulations in xanthophyll cycle and antioxidative systems as well as preservation in integrity and stability of photosynthetic membranes during desiccation may be very important to desiccation tolerance of resurrection angiosperms. Photo...The purpose of the present studies was analysis of the age induced changes in photochemical efficiency and xanthophyils cycle pigments of the primary cabbage (Brassica oleracea L. cv. Capitata f. alba) leaves. Photochemical efficiency of photosystem Ⅱ (PS Ⅱ) was studied by a pulse amplitude modulated chlorophyll fluorescence apparatus, chlorophyll concentration was analysis spectrophotometrically and xanthophyll cycle pigments were estimated by high-pressure liquid chromatography (HPLC). Leaf senescence was accompanied with a decrease both in chlorophylls concentration, the photochemical efficiency and rate constant for PS Ⅱ photochemistry whereas non-photochemical parameters increased. Excitation pressure (1-qP) which is a measure of relative lumen acidification increased by 1.2x in aging leaves. The maximum quantum yield of PS Ⅱ showed no significant change. The level of de-epoxidised xanthophylls increased but the concentration of mono- and di-epoxy xanthophylls decreased in aging leaves. A linear relationship between the excitation pressure and the depoxidation state of the xanthophyll cycle pigments and lutein, during the onset of senescence suggests that excitation pressure can be used as a sensor for monitoring the onset of senescence as well for the de-epoxidation state of the xanthophylls responsible for non-photochemical quenching in stressed leaves.展开更多
The chemical components of the essential oils in the barks and leaves of Eucommia ulmoides Oliver were analyzed and compared by chromatograms and mass spectra technique, heuristic evolving latent projections (HELP), a...The chemical components of the essential oils in the barks and leaves of Eucommia ulmoides Oliver were analyzed and compared by chromatograms and mass spectra technique, heuristic evolving latent projections (HELP), alternative moving window factor analysis (AMWFA) algorithms and normalization method based on the peak areas; the flavones in the barks and leaves of Eucommia ulmoides Oliver were separated on an ODS column by gradient elution carried out with the flow phase consisting of water, methanol and phosphoric acid (0.1%), and their contents were quantitatively determined by standard curve method and diode array detection (DAD) at 362 nm. The results show that 68 and 73 compounds respectively from essential oils of the barks and leaves of Eucommia ulmoides Oliver are identified, and there are 33 mutual compounds among 108 compounds determined. The total contents of these volatile components of the two samples possess 92.9% and 97.75% of the gross of the relevant essential oils, respectively; the contents of the rutin, quercetin and kaempferol in the barks and leaves of Eucommia ulmoides Oliver are 0.016 9, 0.003 6, 0.002 1 and 0.064 4, 0.030 2, 0.010 0 mg/g, respectively, and the determination recoveries are 95.2%-106.2%. The comparative analysis shows that for the barks and leaves of Eucommia ulmoides Oliver, there are significant differences in their components of the relevant essential oils and flavones.展开更多

The results show that the presence of S. metallicus effectively enhances the dissolution of the mineral. The yield of zinc increases from 0.5 g/L in sterile control to 2.7 g/L in bioleaching. The pyrite in the concentrate facilitates zinc dissolution in the early stage, but has hindrance role in the late stage for the formation of jarosite. Sulfur speciation analyses show that jarosite and elemental sulfur are main products in bioleaching process, and the accumulation ofjarosite is mainly responsible for the decline of leaching efficiency.展开更多

This technology can effectively separate the starch and cellulose from Dioscorea Zingiberensis, thus the output of diosgenin is raised by 20%. Meanwhile using the suspension after separation, the synergy of enzymes, which are under the best condition for enzymolysis is studied, and an enzymolysis model, which uses two enzymes at the same time, is established. This model provides a theoretical basis for finding new enzyme resources. What's more orthogonal experiments show that by using this model, either the output of diosgenin can be increased by 50.58% or the amount of acid used can be reduced by 83% which means less pollution.展开更多

Mild extraction technique involving the use of HPLC-DAD-MS was used. The structures of the flavonoids were determined on the basis of mass spectroscopy. Separation of the crude extract by paper chromatography (PC) on forestall as solvent system gave one major yellowish brown spot which had Rf value of 3.9. The Rf value and maximum absorption from UV spectroscopy were the same as those of quercetin standard. The most prominent compound was quercetin followed by three others: kaempferol-3,7-diglucoside, luteolin-7-glucoside, and isorhmnetin-3 -sulphate.展开更多

The PAL activities and flavonoids content were measured by the UV-Vis spectrophotometer. Changes of the whole year were also studied. The results showed that four groups of metal ion treatments had significant effects on the PAL activities and flavonoids contents in Ginkgo leaves. The treatments with 0.1% and 0.3% FeSO4, 0.1%, 0.2% and 0.3% ZnSO4, 0.1%, 0.3% and 0.5% MnSO4, 0.1% and 0.4% CuSO4 not only enhanced flavonoids contents, but also extended the optimum harvest dates of ginkgo leaves. The optimum combination was 0.1% FeSO4, 0.2% ZnSO4, 0.3% MnSO4, and 0.4% CuSO4. It could improve flavonoids content up to 1.619 4%.展开更多

Excitation pressure (1-qP) which is a measure of relative lumen acidification increased by 1.2x in aging leaves. The maximum quantum yield of PS Ⅱ showed no significant change. The level of de-epoxidised xanthophylls increased but the concentration of mono- and di-epoxy xanthophylls decreased in aging leaves. A linear relationship between the excitation pressure and the depoxidation state of the xanthophyll cycle pigments and lutein, during the onset of senescence suggests that excitation pressure can be used as a sensor for monitoring the onset of senescence as well for the de-epoxidation state of the xanthophylls responsible for non-photochemical quenching in stressed leaves.

文摘The chemical components of the essential oils in the barks and leaves of Eucommia ulmoides Oliver were analyzed and compared by chromatograms and mass spectra technique, heuristic evolving latent projections (HELP), alternative moving window factor analysis (AMWFA) algorithms and normalization method based on the peak areas; the flavones in the barks and leaves of Eucommia ulmoides Oliver were separated on an ODS column by gradient elution carried out with the flow phase consisting of water, methanol and phosphoric acid (0.1%), and their contents were quantitatively determined by standard curve method and diode array detection (DAD) at 362 nm. The results show that 68 and 73 compounds respectively from essential oils of the barks and leaves of Eucommia ulmoides Oliver are identified, and there are 33 mutual compounds among 108 compounds determined. The total contents of these volatile components of the two samples possess 92.9% and 97.75% of the gross of the relevant essential oils, respectively; the contents of the rutin, quercetin and kaempferol in the barks and leaves of Eucommia ulmoides Oliver are 0.016 9, 0.003 6, 0.002 1 and 0.064 4, 0.030 2, 0.010 0 mg/g, respectively, and the determination recoveries are 95.2%-106.2%. The comparative analysis shows that for the barks and leaves of Eucommia ulmoides Oliver, there are significant differences in their components of the relevant essential oils and flavones.

文摘Sulfur speciation transformation during bioleaching of pyrite-containing sphalerite concentrate by thermophile Sulfolobus metallicus (S. metallicus) at 65 ℃ was investigated by X-ray diffraction (XRD), diffuse reflectance Fourier transform infrared spectroscopy (FT-IR) and sulfur K-edge X-ray absorption near edge structure spectroscopy (XANES). The results show that the presence of S. metallicus effectively enhances the dissolution of the mineral. The yield of zinc increases from 0.5 g/L in sterile control to 2.7 g/L in bioleaching. The pyrite in the concentrate facilitates zinc dissolution in the early stage, but has hindrance role in the late stage for the formation of jarosite. Sulfur speciation analyses show that jarosite and elemental sulfur are main products in bioleaching process, and the accumulation ofjarosite is mainly responsible for the decline of leaching efficiency.

文摘Based on a through study of the chemical compositions of Dioscorea Zingiberensis, a new separation technology is presented in this paper. This technology can effectively separate the starch and cellulose from Dioscorea Zingiberensis, thus the output of diosgenin is raised by 20%. Meanwhile using the suspension after separation, the synergy of enzymes, which are under the best condition for enzymolysis is studied, and an enzymolysis model, which uses two enzymes at the same time, is established. This model provides a theoretical basis for finding new enzyme resources. What's more orthogonal experiments show that by using this model, either the output of diosgenin can be increased by 50.58% or the amount of acid used can be reduced by 83% which means less pollution.
文摘Eight flavonoid derivatives: rutin, quercetin-3-glucoside, quercetin, luteolin-7-glucoside, isorhmnetin-3-sulphate, kaempferol-3,7-diglucoside,'luteolin and kaempferol have been extracted and characterized from Nipa palm. Mild extraction technique involving the use of HPLC-DAD-MS was used. The structures of the flavonoids were determined on the basis of mass spectroscopy. Separation of the crude extract by paper chromatography (PC) on forestall as solvent system gave one major yellowish brown spot which had Rf value of 3.9. The Rf value and maximum absorption from UV spectroscopy were the same as those of quercetin standard. The most prominent compound was quercetin followed by three others: kaempferol-3,7-diglucoside, luteolin-7-glucoside, and isorhmnetin-3 -sulphate.

文摘The enzyme activities of phenylalanine ammonia-lyase from six years old Ginkgo biloba leaves in the potted orchard were induced by four groups of metal ions respectively. The PAL activities and flavonoids content were measured by the UV-Vis spectrophotometer. Changes of the whole year were also studied. The results showed that four groups of metal ion treatments had significant effects on the PAL activities and flavonoids contents in Ginkgo leaves. The treatments with 0.1% and 0.3% FeSO4, 0.1%, 0.2% and 0.3% ZnSO4, 0.1%, 0.3% and 0.5% MnSO4, 0.1% and 0.4% CuSO4 not only enhanced flavonoids contents, but also extended the optimum harvest dates of ginkgo leaves. The optimum combination was 0.1% FeSO4, 0.2% ZnSO4, 0.3% MnSO4, and 0.4% CuSO4. It could improve flavonoids content up to 1.619 4%.
文摘Phytochemical investigation of Artemisia gmelinii Web. ex Stechm. led to the isolation of 11 known flavonoids. Their structures were identified as genkwanin(1), hispidulin(2), 3'-hydroxy-genkwanin(3), chrysoeriol(4), apigenin(5), 5,7,3',4'-tetrahydroxy-6,5'-dimethoxy-flavone(6), kumatakenin(7), quercetin(8), patuletin(9), quercetagetin-3,6,7-trimethylether(10) and 7,3',4'-trihydroxy-3-methoxy-flavone(11) by spectroscopic methods. Ten compounds(2–11) were isolated from this title plant for the first time, and five compounds(3, 6, 7, 10 and 11) were isolated from genus Artemisia for the first time.
