Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0.5 g· mL^- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV·h^-1, and the volume of aq. 70% (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h^-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporous resin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.展开更多

The apparent molecular mass of the vitellin is 546 ku based on the data of native-PAGE. Under denatured condition (SDS-PAGE), it was found that vitellin was composed of four polypeptides each at 120, 100, 65, and 55 ku. One disulfide bond was detected in the binding of polypeptide subunits. The purified vitellin, contained 4.47% phosphor and 10.6% polysaccharides, and was identified as glyco-lipo-carotenoprotein, according to the PAGE staining data. The purified vitellin can be used as antigen to raise polyclonal antisera in further application.展开更多
In order to determine various traits which need to be improved for improving the productive life span and longevity, purebred Sahiwal cows available at bull mother experimental farm and cattle breeding farm located at...In order to determine various traits which need to be improved for improving the productive life span and longevity, purebred Sahiwal cows available at bull mother experimental farm and cattle breeding farm located at Veterinary College, Durg, Chhattisgarh, India were used. For present investigation, 17 linear type traits were measured, scaled and scored as per the guidelines of International Committee for Animal Recording (2001). The average score point (ASP) under 1-9 point scale score system along with respective observed group for different linear type traits were stature (6.88; taller), chest width (5.30; intermediate), body depth (4.11; intermediate), rump angle (4.27; intermediate), rump width (5.09; intermediate), rear leg set-side view (4.55; intermediate), rear leg set-rear view (5.95; intermediate), foot angle (5.66; intermediate), udder depth (5.71; intermediate), rear udder height (4.90; intermediate), udder balance (5.27; intermediate), udder cleft (3.75; intermediate), fore udder attachment (5.55; intermediate), teat length (3.54; intermediate), fore teat placement (5.33; intermediate), rear teat placement (6.37; intermediate) and teat thickness (2.76; narrow). For most of the traits, ASP which lies near midpoint (i.e. near five) is known to be ideal for dairy type cattle. Moreover, some traits also showed the presence of undesirable ASP. Hence, the traits such as body depth, rump angle, foot angle, udder depth, rear udder height, udder cleft, fore udder attachment, teat length, and teat thickness needs to be improved for improving the production sustainability and herd life of Sahiwal cattle. Thus, present investigation gives explicit clue to incorporate these conformation traits in selection program of this valuable germplasm commonly found in Southern part of Asia (India, Pakistan, Srilanka, etc.).展开更多
The interaction among different livelihood capitals is a key to generate a deeper understanding of the livelihood sustainability.In this paper,we use net income per capita(economic capital),meat and milk production pe...The interaction among different livelihood capitals is a key to generate a deeper understanding of the livelihood sustainability.In this paper,we use net income per capita(economic capital),meat and milk production per capita(physical capital),and areas of fenced pasture,livestock shelter,grassland rodent control and planted grassland(physical capital) as proxy indicators of livelihood promotion,livelihood provision,and livelihood protection respectively.By developing a correlation model between pastoralists' livelihood protection and improvement,we found that(1) there is a statistically significant correlation between the pastoralists' livelihood protection and promotion;(2) based on the maximum effect of pastoralists' livelihood promotion and provision,there is a benchmark in the effect of the intervention intensity of livelihood capital(grassland resource protection) on livelihood improvement;(3) on basis of two indicators,i.e.net income per capita and meat production per capita,the reasonable scales of fenced pasture,livestock shelter and planted grassland are less than 843,860 and 46 thousand hectares(hm2) per year respectively.With the marginal effect of livelihood protection,moderately decreased areas of fenced pasture and planted grassland,and increased area of livestock shelter is a critical to ensure pastoralist's livelihood sustainability.展开更多

The results showed that the molecular weight range of antimicrobial polypeptide (CMC...The preliminary purification and antimicrobial mechanism of antimicrobial peptide from Antarctic Krill were studied in this paper. The results showed that the molecular weight range of antimicrobial polypeptide (CMCC-1) obtained by cation exchange chromatography was between 245-709D as detected by molecular sieve chromatography, and the minimum inhibition concentration (MIC) of CMCC-1 against Staphylococcus aureus was 5.0mgmL^-1. The antimicrobial mechanism of CMCC-1 was studied with S. aureus as indicator bacterium. Compared with control group, the results of the experimental group in which S. aureus was treated with CMCC-1 were as follows: l) CMCC-1 could inhibit cell division at logarithmic phase. 2) The protein and reducing sugar con- tent, and the conductivity of culture medium increased, and the activity of alkaline phosphatase and [3-galactosidase could be detected in the culture medium. 3) Observation under scanning electron microscope revealed that somatic morphology became irregular, and then somatic surface became coarse. The cell became much smaller, and most somatic ceils gathered. The boundary between cells became dim and finally fused as a whole. 4) Observation under transmission electron microscope showed that the surface of S. aureus became rough and the reproducing ability was restrained. The cell wall became thin and the cytoplasm shrunk. Substances inside cell leaked out, which caused cells death. 5) SDS-PAGE analysis showed that some bands disappeared, and the residual bands became vague. 6) The genomic DNA electrophoresis results showed that the genomic DNA bands ofS. aureus were not degraded but the brightness significantly reduced. Thus, it is supposed that CMCC-1 could destroy the cell wall and membrane of S. aureu, increase the cell membrane permeability and the leaking-out of intracellular substances, and thus cause the death ofS. aureu.展开更多

First, the total flavone was extracted from the defatted powder of ginkgo L. biloba leaves. Effects of solvents and operation conditions were examined to get a relative high yield and purity in this step. The crude extract was further purified by resin HZ-841. Both adsorption and elution process were studied to obtain an optimized conditions, i.e., pH, flow rate, concentration. A yellow powder was obtained, of which 37.3% was flavones, obviously higher than the basic international standard of 24%.展开更多

Verticillium dahliae toxins (VD-toxins) can be purified by dif...The complexes excreted by VerticiUium dahliae are phytotoxins, which are responsible for most of the symptoms associated with Verticillium wilt disease. Verticillium dahliae toxins (VD-toxins) can be purified by different methods. In the present study, we reported a simpler, more effective method to purify VD-toxins. The supematant of V. dahliae culture was frozen, lyophilized and dialyzed by 1 kDa Dialysis Membranes (MWCO). We also partially identified the characteristics of the purified VD-toxins. The results showed that the components of VD-toxins include glycoprotein within 35.8-83.2 kDa. The phytotoxic activity of VD-toxins was remained after VD-toxins were pretreated by high temperature, Concanavalin-A, and proteinase E, respectively. These data suggest that VD-toxins are heat-stable, and the protein fraction and glycosyl are both important contributors to the phytotoxic activity. VD-toxins purified effectively from the culture filtrates of V. dahliae may help in further understanding the mechanisms of interactions between V. dahliae and plants.展开更多
