The survival thermal maximum for L. crocea was 33.0℃, the 50% critical thermal maximum (50% CTMax) was 35.5℃, and the critical thermal maximum (CTMax) was 36.0℃. Three microsatellite markers (LYC0148, LYC0200 and LYC0435), associated with thermal tolerance were screened and identified using a Bulked Segregation Analysis (BSA) method. These markers have six amplified fragments in which four are related to thermal tolerance. These fragments were cloned and sequenced, and the results showed the core motif were all "AC" repeats. For LYC0148 and LYC0200, the lengths of fragments are 18l bp and 197 bp, respectively. For LYC0435, which has two fragments, the fragment lengths are 112 bp and 100 bp. The results provide useful molecular markers for thermal-tolerance breeding of large yellow croaker in the near future.展开更多

The berry was extracted by ethanol-water solution. MTT assay and growth rate method were applied to determine antibacterial ability in vitro of the extract solution. The antibacterial and antifungal assay indicated that the extract exhibited different degree of inhibitory activity. The result showed that the extract at a concentration of 1 250 mg/L had the maximum inhibitory effect against Escherichia coli, Bacillus subtilis, and Staphylococcus aureus. Inhibitory effect against B. subtilis was much larger than that against the other two bacterial species. MIC values(minimum inhibitory concentration) of the extract on three bacterial species were 5 000, 1 250, and 1 000 mg/L respectively. IC_(50)(50% inhibiting concentration) evaluation of Trichoderma viride, Rhizopus stolonifer, Aspergillus niger, Penicillium expansum, and Vintage Red indicated that the berry had the strongest inhibitory effect against P. expansum(5 520 mg/L), while it had the weakest inhibitory effect against R. stolonifer(18 870 mg/L). The results proved that seabuckthorn berry had highly effective and comprehensive antimicrobial function.展开更多

METHODS: Twenty Sprague-Dawley rats were divided into high-fat diet and normal control groups of 10 rats each. The composition of microbiota in the mucosa from the distal esophagus was analyzed based on se- lective culture. A variety of Lactobacillus species were identified by molecular biological techniques. Bacterial DNA from Lactobacillus colonies was extracted, and 165 rDNA was amplified by PCR using bacterial uni- versal primers. The amplified 16S rDNA products were separated by denaturing gradient gel electrophoresis (DGGE). Every single band was purified from the gel and sent to be sequenced. RESULTS: Based on mucosal bacterial culturing in the distal esophagus, Staphylococcus aureus was absent, and total anaerobes and Lactobacillus species were de- creased significantly in the high-fat diet group compared with the normal control group (P 〈 0.01). Detailed DGGE analysis on the composition of Lactobacillus species in the distal esophagus revealed that Lactobacillus crispa- tus, Lactobacillus gasseri (L. gasser/] and Lactobacillus reuteri (L. reuterl] comprised the Lactobacillus species in the high-fat diet group, while the composition of Lactobacillus species in the normal control group consisted of L. gasseri, Lactobacillus jensenii and L. reuteri. The authors used a stump of Bifidobacterium breve isolated from the new saddles...The aim of this study is to demonstrate the influence of Spirulina platensis on the growth of Bifidobacteium breve in the infantile milk. The authors used a stump of Bifidobacterium breve isolated from the new saddles which was born after identification and characterization. The follow-up of the kinetics of growth of B. breve in the milk enriched by various concentrations of Spirulina (0, 1, 5 and 10 mg/mL) was realized, and the authors estimated the antimicrobial effect of the cultures realised on three pathogenic bacteria: a bacteria isolated from saddles "Salmonella ssp.", two reference stumps "Staphylococcus aureus ATCC25923" and "Escherichia coli ATCC25922". The growth rate was 0.11 hl for the culture of B. breve associated with Spirulina (10 mg/mL), 0.09 hl for the culture of B. breve enriched with Spirulina (5 mg/mL) and 0.08 hl for the culture of B. breve enriched with Spirulina (1 mg/mL). In the case of the pure culture of B. breve, the growth rate was 0.07 h1. The authors observed inhibitory effects of B. breve in the various cultures towards the considered pathogenic bacteria. The optimal antimicrobial activity was observed in the preculture bifid with the algae culture (10 mg/mL) towards Salmonella ssp. Only 2 of these were detected at the same locus at four environments, 1 was significant at three environments, 6 were significant at two environments and 1 was significant at one environment. Seven QTLs located on chromosome 1(two), 2(two),5,6,8 were found for low temperature chlorosis resistance and five QTLs located on chromosome 3, 4, 7, 8, 11 resistant to chilling injury. The amount of variation explained by individual QTL ranged from 4.85%to 49.34%. There was no linkage relationship among the three characters, which indicates seedling cold tolerance is a complex character and is controlled by different QTLs.展开更多

Landfill Leachate comprises a mass of different organic and inorganic components which can be dissolved or suspended. Regardless of the nature of the components, it poses a potential pollution on local land and surface waters. The present work was undertaken to assess, isolate, identify and characterize the hydrocarbon degrading bacteria associated with environmental samples collected from discharge of Ain Temouchent, Algeria. The samples (landfill leachate) were chemically and microbiologically analyzed using standard techniques. These organisms were further studied to determine the level of biodegrading activities on hydrocarbons as the single carbon source using enrichment medium under different physical and chemical parameters (pH, temperature, NaCI and tolerance of crude oil...), The identification of seven strains isolated named as Pseudomonas aeruginosa, Pseudomonas fluorescens, Pseudornonas sp., Staphylococcus aureus, Staphylococcus xylosus, Staphylococcus hominis and Staphylococcus warnerii. These strains growth on a neutral medium and temperature between 30 ℃ and 37 ℃, 5 at 6% of NaCI concentration, and can use a concentration of 5 at 10% of crude oil as sole source of carbon.展开更多

Virulence survey of cereal rust fungi have traditionally used differential host genotype and virulent physiologic race 134 E 134 AT was received to evaluate the segregating lines. A set of 35 segregating lines and three additional susceptible cultivars were added, making a total of 38 eultivars-lines. Spreader plants served as a source of inoculums. The plants were evaluated after two months based on infection percentage and the disease scoring scales. The results indicated that, there are significant changes in susceptibility assessments of the segregating lines in contrast to cultivated varieties to the yellow rust P. striiformis as far as the disease incidence and rust development is concerned in these experiments. The selected differential genotypes associated with various levels of resistance. The infection percentages were varied on the lines and the varieties, whereas the severity percentages were entirely different. The lowest severity is of line 38 with 34.70% and the highest in line 1 with 93.49% indicating 2.7 folds reduction in severity. For discrimination among the lines, the results on scoring scales gave distinctions of five classes of resistance, in which, the highest score is 29.93 and the lowest one 11.46. The cluster analysis showed almost similar results and confirmed, the analysis by Duncan's Multiple-lest rang as the above five distinctive groups.展开更多

The antimicrobial activity and minimal inhibitory concentration of B. articulata, B. trimera and B. crispa aqueous and ethanoli...Plants of Baccharis (Asteraceae) genus are commonly known in Argentina as "carqueja". The antimicrobial activity and minimal inhibitory concentration of B. articulata, B. trimera and B. crispa aqueous and ethanolic extracts were evaluated by using the micro-well dilution method. Previously, the components of extracts were analyzed by spectroscopial means. Gram-positive bacteria were more sensitive to Baccharis species extracts than Gram-negative bacteria. Out of 3 plant species, B. trimera showed significant antibacterial activity and aqueous and ethanolic extracts were active against Staphylococcus aureus (MIC = 2,500 μg/mL and 1,250 μg/mL, respectively) and Listeria monocytogenes (MIC = 625 μg/mL and 625 μg/mL, respectively). All ethanolic extracts inhibited the growth of the selected Gram-positive (MIC values ranged between 625 μg/mL and 1,250 μg/mL). Therefore, all Gram-negative bacteria were resistant to the ethanolic and aqueous extracts tested. One flavone, genkawanin, was identified from the three ethanolic extracts as the responsible of antibacterial activity. Two terpenes, hawtriwaic acid and bacrispine, were identified from ethanolic extract of B. crispa and B. trimera as the responsibles of antibacterial activity. These preliminary studies corroborated the antimicrobial activity of the selected plants used in folklore medicine. Therefore, they could be potential sources of new antimicrobial agents used in treatment of infectious diseases.展开更多
Juveniles of the shore crab Hemigrapsus oregonensis are highly variable in color, ranging from the typical yellowish- green of adults to pure white and myriad patterns of white mottling and other disruptive markings, ...Juveniles of the shore crab Hemigrapsus oregonensis are highly variable in color, ranging from the typical yellowish- green of adults to pure white and myriad patterns of white mottling and other disruptive markings, but large individuals with white coloration appear to be very rare. Using image analysis to quantify the relative "whiteness" of beaches, we sampled crabs from nine locations in Washington State that varied widely in their amount of shell fragments and other light-colored material. The total proportion of white individuals in the different locations was strongly correlated to the proportion of white material on the beaches, but there was a striking difference between sexes. Although white specimens of both sexes declined significantly at sizes above 10 mm carapace width, white females generally persisted throughout the entire female size range on lighter-colored beaches while white males larger than 10 mm were virtually absent from all of the sampled populations. Pure white males held on dark backgrounds in captivity remained white, as they lack the dark chromatophores in their hypoderrnis needed to change color; off-white males became darker and in some cases lightened up again when transferred back to a white background. Behavioral differences between the sexes may result in differential mortality of white individuals by visual predators [Current Zoology 61 (4) 729-738, 2015 ].展开更多
