晚明泰州经济的衰败很大程度上是国家黄、运治理政策形塑的结果。明代治黄以保运为先,造成淮河入海通路被侵占,遂使淮扬运道、祖陵与下河州县同罹水患。在治水技术无法提供兼顾三者的解决方案时,明廷鉴于祖陵、运道对皇室与国计的特殊意... 晚明泰州经济的衰败很大程度上是国家黄、运治理政策形塑的结果。明代治黄以保运为先,造成淮河入海通路被侵占,遂使淮扬运道、祖陵与下河州县同罹水患。在治水技术无法提供兼顾三者的解决方案时,明廷鉴于祖陵、运道对皇室与国计的特殊意义,选择将下河作为排泄区。这种角色设定导致泰州水害频发,破坏了原先优良的农作环境,当地经济也随之陷入困境。泰州士绅既无力扭转大的水利格局,也未能说服国家提供更多的赋税优惠,水利与税收分配的主导权始终掌握在国家手中。这表明在地方与国家核心利益发生冲突时,地方社会所能体现出来的自主性与能动性有限,很难抵制国家行为对地方民生的侵害。晚明泰州的遭遇显示帝制国家对地方社会保有较强的穿透力与影响力,地方史研究亦需充分考量"国家"要素。 展开更多

[Objective]The aim of this study was to identify transient expression of movement protein (MP) gene in Nicotinana benthaminana rapidly and further investigate the function of this exogenous gene. [Method]The movement protein gene of barley yellow dwarf virus (BYDV) was cloned into potato virus X (PVX) viral vector of pGR107,and PVX-recombinant vector was obtained. After electroporation of Agrobacterium tumefaciens,PVX was inoculated into the lower leaves of tobacco by Agrobacterium infiltration assay to observe the infection of virus on tobacco. [Result]After infection for 7 days,upper non-inoculated leaves of tobacco infected by the PVX-recombinant vector showed the virus infection symptoms,while the control group had no viral infection phenomenon. Daily follow-up observations for two groups revealed that tobacco infected by PVX-recombinant vector had severe symptoms of virus infection and curling leaves,or even led to necrosis both in infiltrated and systemic leaves in late period. However,tobacco infected by PVX vector had only slight symptoms of virus infection and could recover from infection. RT-PCR of the infected tobacco indicated that exogenous gene BYDV-MP had a normal transcription and expression in tobacco. [Conclusion]As a determinant factor for viral disease,BYDV-MP promotes the systemic infection rate of PVX and its symptom. In addition,it is feasible to express exogenous MP gene in Nicotiana benthaminan via PVX expression vector. 展开更多

Tobacco ( Nicotiana tabacum L.) “NC89” plants were transformed with deletion mutant of cucumber mosaic virus (CMV) movement protein (MP) gene and full_length CMV MP gene, respectively. The transformed plants were analyzed with polymerase chain reaction (PCR), PCR_Southern, Southern and Western blots. R 0 generation of the transgenic plants were inoculated with CMV. Five out of 10 lines of tobacco plants (BMPK) transformed with CMV MP deletion mutant gene showed high resistance to CMV infection and remained symptomless for up to 50 days post_inoculation. In contrast, tobacco plants (BMPR) transformed with full_length CMV MP gene did not show resistance to CMV infection. However, most of the infected full_length CMV MP gene transgenic plants recovered by showing none or very mild mosaic symptoms in 40 days post_inoculation. The results of R 1 generation of the BMPK transgenic plants tested under field conditions showed that all 5 lines of transgenic plants could delay the virus disease development. 展开更多

The electron transport properties in Ge are calculated by full band Monte Carlo technique with anisotropic scattering consideration.The calculation procedures are as follows:the full band structure is calculated by nonlocal empirical pseudopotential approach;the relative value of density of state (DOS) is computed by counting the number of states located in a certain region of the energy;the phonon dispersion curve is obtained from an adiabatic bond charge model;the electron phonon scattering rates are approximated by the nonparabolic model derived from Fermi’s golden rule at low energy region and scaled by DOS at higher energy region;the energy and momentum conservations are employed for choosing the final state after scattering.The validity of this Monte Carlo simulator and the physical models that are used is fully confirmed by comparing the program output to experimental results listed in references.As this Monte Carlo model can accurately reproduce the velocity and energy characteristics of electrons in Ge and the DOS scaled scattering rate can significantly reduce the computational cost for scattering rates,this approach is suitable for device simulation. 展开更多

Objective This study was conducted to examine the absorption and translocation of conjugated bile acids(BAs)in Calculus bovis and its substitutes to detect differences in these materials.Methods A Caco-2 monolayer cell model was used to compare the apparent permeability coefficient(Papp)value and efflux ratio(ER)of BAs in natural cow-bezoar(NCB),artificial cow-bezoar(ACB),and in vitro cultured cow-bezoar(Ivt-CCB).Papp and ER values were determined by liquid chromatography-mass spectrometry.Samples were separated on an analytical column.Results The distribution of BAs in NCB was significantly different from that in ACB and Ivt-CCB.The percentages of conjugated BAs were significantly higher in NCB than in the two substitutes.The distribution differences of conjugated and unconjugated BAs can be used to distinguish costly NCB from relatively inexpensive substitutes.Conclusion The transport characteristics of BAs in Ivt-CCB were more consistent with NCB than with ACB,even when the proportions of BAs in Ivt-CCB were closer to those of ACB. 展开更多

In this paper, the development strategiesof shipping centers and transportation network are discussed based on economic globalizationtendency. The results argue that a united transportation network should be built in order to promotethe economic competition of Northeast Asia in the world. As a key component of the economiccooperation, a hierarchical shipping centers network should be established with Hong Kong, Shanghai,Pusan, Kobe, and Tokyo as cores. Dissolved nutrient concentration in the Huanghe (Yellow) River at Lijin was monitored during a water-sediment regulation period and a subsequent rainstorm from 14 June to 19 July, 2005. This study provides detailed information on nutrient concentrations in the Huanghe River during the water-sediment regulation and rainstorm periods, and is of significance for the downstream area of the Huanghe River and the Bohai Sea. The average concentrations of nitrate, nitrite and ammonia were 304.7 μmol/L, 0.19 μmol/L, and 1.10 μmol/L, respectively, while the average concentrations of dissolved inorganic phosphorus (DIP) and dissolved silicate (DSi) were 0.23 gmol/L and 122.9 ktmol/L, respectively. Nutrient concentrations during the water-sediment regulation period were mainly influenced by the dilution effect, floodplain effect and sediment resuspension while dilution and erosion effects were the main factors during the rainstorm. The fluxes of dissolved inorganic nitrogen (DIN), DIP and DSi during the water-sediment regulation and rainstorm periods accounted for 20.4%, 19.5%, 16.7% and 4.97%, 6.45%, 5.47% of the annual nutrient fluxes, respectively. Discharge was the main factor influencing the fluxes of nutrients during both the water- sediment regulation and the rainstorm periods. 展开更多

Objective:Lotus leaf is a traditional Chinese herb that has been used successfully for centuries for relieving edema by inducing diuresis.Based on its good clinical evidence and anti-hypertensive effectiveness,this study aimed to investigate the potential mechanism of the hyperuricemic inhibitory effects of lotus leaf crude extract(LL)and lotus leaf total alkaloids fraction(LA).Methods:The xanthine oxidase(XOD)inhibitory effect of LL and LA was analyzed in vitro by determining mRNA expression and protein expression levels of hepatic XOD.The hyperuricemic inhibitory effect of the lotus leaf was analyzed in vivo in a potassium oxonate(PO)-induced rat model by determining mRNA expression for renal urate transporters.Results:At a concentration of 40mg/mL,LL and LA suppressed XOD enzymatic activity by 37.35%±9.50%and 47.73%±8.32%,respectively.Both LL and LA administration significantly reduced the concentration of uric acid in the serum and liver of PO-induced hyperuricemic rats.Both LL and LA administration could inhibit XOD mRNA and protein expression,activate renal organic anion transporter 1/3 mRNA expression,and inhibit renal urate reabsorption by decreasing renal GLUT9 and renal urate transporter 1.Conclusions:Insight was gained into the mechanism behind the hyperuricemic inhibitory effects of LL and LA.Our results suggest that they act on two targets:decreasing the production of uric acid by inhibiting mRNA and protein expression of XOD in the liver,and regulating the mRNA expression of renal urate transporters in the kidneys. 展开更多
