Study on compound Duzhong Jiangu granule in the treatment of knee osteoarthritis based on transcriptome analysis strategy

Duzhong Jiangu Granule in the treatment of primary osteoarthritis(POA)model of postmenopausal kidney deficiency type in Hartley female guinea pigs after ovariectomy,and the correlation between gene expression of bone marrow tissue,cartilage tissue,and knee osteoarthritis.Methods:383-months-old Hartley female guinea pigs after one week of adaptive feeding were weighed about 400 g±20 g,numbered,and sorted by ear tag.Six of them were selected as normal groups by looking up random number tables.The remainder were removed from the bilateral ovaries to construct the postmenopausal kidney deficiency model,and castrated guinea pigs were used to construct the postmenopausal kidney deficiency POA model.After the modeling cycle,a guinea pig from the blank group and a guinea pig from the model group were sacrificed and the right knee was observed.The model was established and the experiments continued.There were five guinea pigs in the blank group,and the remaining model guinea pigs were randomly divided into model control group,high-dose group of compound Duzhong Jiangu granules,middle-dose group of compound Duzhong Jiangu granules,low-dose group of compound Duzhong Jiangu granules and design group,with 5 guinea pigs in each group.Blanks and model groups were given a cellulose sodium solution by gavage.The guinea pigs were sacrificed after 30 days of intragastric administration.The left knee cartilage and bone marrow of the blank group,model group,middle dose group,and high dose group of compound Duzhong Jiangu granule were collected and applied to transcriptome sequencing,and the sequencing data were analyzed,including differential gene expression analysis,functional enrichment analysis of database established by Gene Ontology federation(GO)and Kyoto Encyclopedia of Gene and Genome(KEGG)pathway enrichment analysis.The complete specimens of the right knee joint were collected,and the morphological changes of the cartilage of the right knee joint in each group were observed by saffron rapid green staining,and the subchondral bone was quantitatively analyzed by Micro CT so that the expression of TRAF6,MIP-1βand IL-1βprotein in NF-kappa B signaling pathway was detected by Western Blot technique(WB).Results:The results of Safranin Fast Green staining showed that Compound Duzhong Jiangu Granules could effectively reduce the degree of morphological damage of articular cartilage in guinea pigs with the POA model.According to the analysis results of the subchondral bone structure under Micro CT,Compound Duzhong Jiangu Granules can improve the bone condition of the POA model,thus delaying the process of degenerative changes of the knee joint.From the results of transcriptome analysis,Compound Duzhong Jiangu Granules can inhibit the expression of related genes in POA model guinea pigs.According to the results of Wester Bolt verification,Compound Duzhong Jiangu Granules can effectively improve knee osteoarthritis.Conclusions:The effect of Compound Duzhong Jiangu Granules on OA is obvious,and its mechanism may be related to the expression of genes GZMK,Jchain,igkc,IGHV3-74,IGHV3-11,IGHV4-1,CCL5,and IGKV1–39. 展开更多

In this study, rabbit models of optic nerve injury were reproduced by the clamp method. After modeling, rabbit models were given one injection of 50 ng recombinant human ciliary neurotrophic factor into the vitreous body and/or intragastric injection of 4 g/kg compound light granules containing Radix Angelicae Sinensis and Raidix Paeoniae Alba at 4 days after modeling, once per day for 30 consecutive days. After administration, the animals were sacrificed and the intraorbital optic nerve was harvested. Hematoxylin-eosin staining revealed that the injured optic nerve was thinner and optic nerve fibers were irregular. After treatment with recombinant human ciliary neurotrophic factor, the arrangement of optic nerve fibers was disordered but they were not markedly thinner. After treatment with compound light granules, the arrangement of optic nerve fibers was slightly disordered and their structure was intact. After combined treatment with recombinant human ciliary neurotrophic factor and compound light granules, the arrangement of optic nerve fibers was slightly disordered and the degree of injury was less than after either treatment alone. Results of tensile mechanical testing of the optic nerve showed that the tensile elastic limit strain, elastic limit stress, maximum stress and maximum strain of the injured optic nerve were significantly lower than the normal optic nerve. After treatment with recombinant human ciliary neurotrophic factor and/or compound light granules, the tensile elastic limit strain, elastic limit stress, maximum stress and maximum strain of the injured optic nerve were significantly increased, especially after the combined treatment. These experimental findings indicate that compound light granules and ciliary neurotrophic factor can alleviate optic nerve injury at the histological and biochemical levels, and the combined treatment is more effective than either treatment alone. 展开更多

BACKGROUND Recent research suggests that although prokinetic agents,acid suppressors,and radical treatment for Helicobacter pylori infection may be effective in patients with functional dyspepsia(FD),a large proportion of patients still fail to respond to these treatments or may suffer from severe adverse reactions.Many traditional Chinese medicinal herbs can regulate the status of the entire body and have special advantages in the treatment of functional diseases.The present study was designed to verify the efficacy of Biling Weitong Granules(BLWTG),a traditional Chinese medicinal herbal compound formula,in alleviating epigastric pain syndrome(EPS)in FD patients,in an attempt to provide an effective prescription for the clinical treatment of this disease.AIM To evaluate the clinical efficacy and safety of BLWTG in treating EPS in patients with FD.METHODS In this multicenter,stratified,randomized,double-blind,placebo-controlled,parallel group clinical trial,eligible patients were randomized into the BLWTG and placebo groups who were treated for 6 wk.Efficacy indicators including the severity and frequency of EPS and the time to pain resolution and safety indicators including adverse events were observed and compared.RESULTS The baseline demographic data and clinical characteristics,such as epigastric pain symptoms,pain intensity,and frequency of attacks,were matched between the two groups before randomization.After 6 wk of treatment and after the center effect was eliminated,the epigastric pain was significantly improved in 28.33%and 85.59%of the patients in the placebo and BLWTG groups,respectively(P<0.05).At 6 wk,the resolution rate of epigastric pain was 15%and 69.49%in the placebo and BLWTG groups,respectively(P<0.05).The differences of total FD clinical score between these two groups were significant(P<0.05)at 2,4,and 6 wk(P<0.05).The scores of each item and the total score in the Functional Digestive Disorders Quality of Life Questionnaire showed significant differences between the two groups at 6 wk after both the center and interaction effects were eliminated(P<0.05).There was no significant difference in the incidence of adverse events between the two groups,and no serious adverse event was noted during the observation.CONCLUSION Compared with placebo,BLWTG markedly improved EPS in FD patients without causing serious adverse reactions. 展开更多

Objective:In this study,we used HepG2 human hepatocellular carcinoma cells to study the effects of Compound Xishu Granule(CXG)on cell proliferation,apoptosis,and the cell cycle in vitro.We also used a xenograft tumor model to study the anti-tumor effects of CXG and related mechanisms in vivo.Methods:The effect of CXG on cell viability was measured using Cell Counting Kit-8 and a colony formation assay.The effect of CXG on apoptosis and the cell cycle was analyzed using flow cytometry.The in vivo anti-tumor effect of CXG was assessed by measuring the volume change in xenograft tumors after drug administration.The CXG anti-tumor mechanism was studied using western blotting assay to detect cell cycle and apoptotic associated proteins.Results:CXG suppressed HepG2 cell proliferation in a time-and dose-dependent manner in vitro.Colony formation experiments showed that CXG administration for 24 h significantly reduced HepG2 cell formations(P<.01).Flow cytometric analysis showed that CXG treatment for 48 h promoted apoptosis and blocked HepG2 cells in the G2/M phase.Western blotting results showed that Bax was significantly upregulated and Bcl-2 was down-regulated in graft tumor tissues and HepG2 cells after CXG administration,which increased the Bax/Bcl-2 ratio.PLK1,CDC25 C,CDK1,and Cyclin B1 expression were upregulated.CXG had a good inhibitory effect on graft tumor growth in vivo.Conclusion:CXG has good anti-tumor effects in vitro and in vivo.In vitro,CXG promoted HepG2 cell apoptosis and induced G2/M phase arrest.In vivo,CXG significantly inhibited graft tumor growth.The CXG mechanism in treating hepatocellular carcinoma may be that CXG can induce abnormal apoptotic and cell cycle associated protein expression,leading to mitotic catastrophe and apoptosis. 展开更多

Objective:To investigate the clinical study of Qiming granule combined with compound anisodine injection in the treatment of diabetic retinopathy.Methods:104 patients with nonproliferative diabetic retinopathy admitted to department of ophthalmology in Xishui Hospital Affiliated to Hubei University of science and technology from September 2016 to May 2018 were selected as the study subjects.According to the principle of treatment,they were divided into 52 cases in the control group and 52 cases in the treatment group,the clinical efficacy,blood sugar and lipid levels,hemorheological changes and adverse reactions were compared between the two groups.Results:The total effective rate difference between the treatment group(92.31%)and the control group(71.15%)was statistically significant(χ^(2)=7.792,P=0.005).After treatment,the two groups of fasting plasma glucose(FPG),hemoglobin A1c(HbA1c),total cholesterol(TC),low density lipoprotein-cholesterol(LDL-C),whole blood high-viscosity,whole blood cut viscosity,whole blood low-cut viscosity,plasma viscosity,number of microangioma and area of the hemorrhage were significantly lower than before treatment(P<0.05),and the levels of FPG,HbA1c,TC,LDL-C,whole blood highviscosity,whole blood cut viscosity,whole blood low-cut viscosity,plasma viscosity,number of microangioma and area of the hemorrhage in the treatment group were significantly lower than those in the control group(χ^(2)=0.122,P=0.727).Conclusions:Qiming granule combined with compound anisodine injection can effectively improve the clinical efficacy of patients with non-proliferative DR,and its mechanism may be related to improve blood sugar and blood lipid levels and correct blood rheology disorder,reduce microangioma and hemorrhage.It has clinical promotion significance. 展开更多

目的构建粒子群反向传播(Particle swarm optimization-back propagation,PSO-BP)神经网络对中药复方颗粒剂安慰剂制备着色剂的用量进行预测,为中药复方颗粒剂安慰剂颜色的模拟提供一种新思路。方法运用BP神经网络建立样品颜色参数L、a^... 目的构建粒子群反向传播(Particle swarm optimization-back propagation,PSO-BP)神经网络对中药复方颗粒剂安慰剂制备着色剂的用量进行预测,为中药复方颗粒剂安慰剂颜色的模拟提供一种新思路。方法运用BP神经网络建立样品颜色参数L、a^(*)、b^(*)与色素质量分数的模型,利用粒子群算法的全局搜索能力优化BP神经网络权重和偏置,防止模型出现局部最小值,再采用线性降低权系数法和引入变异算子提高粒子群算法的全局寻优能力;以颜色综合评价指标(ΔE)为客观评价标准,验证试验结果。结果训练结果表明,改进的PSO-BP神经网络拟合精度最高达到98.31%;预测结果表明,改进的PSO-BP神经网络的预测误差最小,平均绝对百分比误差(MAPE)、均方根误差(RMSE)和平均色差(ΔE)分别为0.4115、2.1646、2.56;制备3种颗粒的验证样品进行验证,验证样品与模型药物的ΔE分别为1.73、2.63、4.11,肉眼直观评价其中两组与模型药物色差较小。结论基于改进粒子群优化算法的BP神经网络可模拟中药复方颗粒剂安慰剂制备着色剂用量预测,可作为安慰剂配色研究的推荐优化模型。 展开更多

复方栀子根颗粒联合替诺福韦酯对慢性乙型肝炎患者血清细胞因子水平的影响 被引量:1

目的:观察复方栀子根颗粒联合替诺福韦酯(TDF)治疗慢性乙型肝炎(CHB)患者的临床疗效及对细胞因子的影响。方法:纳入CHB初治患者80例,采用临床随机对照研究,除去脱落及剔除病例,共纳入分析75例(治疗组35例、对照组40例)。对照组患者给予... 目的:观察复方栀子根颗粒联合替诺福韦酯(TDF)治疗慢性乙型肝炎(CHB)患者的临床疗效及对细胞因子的影响。方法:纳入CHB初治患者80例,采用临床随机对照研究,除去脱落及剔除病例,共纳入分析75例(治疗组35例、对照组40例)。对照组患者给予口服TDF,治疗组患者在对照组基础上加用复方栀子根颗粒,两组患者均在治疗4周、24周时比较肝功能(ALT、AST、GGT)变化,在治疗24周比较病毒学指标(HBV DNA、HBsAg、HBeAg)、细胞因子(IL-6、IL-10、IL-17、IL-23)水平及临床疗效判定。结果:两组患者治疗24周后肝功能、病毒学指标、细胞因子水平均较治疗前显著降低(P<0.05)。组间比较,治疗4周后治疗组转氨酶下降水平更低,差异有统计学意义(Z=-2.90,P=0.004;Z=-3.26,P=0.001;Z=-2.24,P=0.025);治疗组24周时IL-6、IL-23水平较对照组低,差异有统计学意义(Z=-2.14,P=0.033;Z=-2.10,P=0.036);治疗组总有效率97.14%(34/35),对照组95.00%(38/40),治疗组总有效率高于对照组(χ2=9.49,P=0.019);治疗组较对照组不良反应更少,差异有统计学意义(χ2=6.19,P=0.044)。结论:复方栀子根颗粒联合TDF治疗早期能更快降低转氨酶水平,降低促炎细胞因子水平及提高临床综合疗效优于单用TDF,并减轻临床不良反应。 展开更多

利用网络药理学和生物信息学方法揭示复方木鸡颗粒治疗肝癌的作用机制。通过TCMSP、Swiss Target Prediction数据库分别获取复方木鸡颗粒的活性成分以及相关靶点;在GEO数据库筛选肝癌的相关靶点;利用Venny 2.1在线平台获取药物与疾病的... 利用网络药理学和生物信息学方法揭示复方木鸡颗粒治疗肝癌的作用机制。通过TCMSP、Swiss Target Prediction数据库分别获取复方木鸡颗粒的活性成分以及相关靶点;在GEO数据库筛选肝癌的相关靶点;利用Venny 2.1在线平台获取药物与疾病的共同靶点;由Cytoscape 3.8.2绘制药物–成分–靶点–疾病网络;用STRING数据库构建PPI网络;通过DAVID数据库进行GO功能富集和KEGG通路富集分析;利用Kaplan Meier-Plotter数据库对关键基因的表达量及预后关联性进行分析;运用AutoDock软件对关键成分和靶点进行分子对接验证。复方木鸡颗粒中共获得37个活性成分,筛选出57个共有靶点,涉及生物过程98条、细胞成分17条、分子功能37条,介导15条信号通路。生存期分析结果显示,ESR1、CYP3A4、G6PD基因的表达量与肝癌患者的生存期具有相关性。分子对接表明筛选的6个活性成分与6个关键靶点蛋白之间具有较好的结合能力。复方木鸡颗粒中的大豆皂苷C、柚皮素、β-谷固醇、汉黄芩素等活性成分可以作用于ESR1、CYP3A4、G6PD等靶点,可能通过P53、戊糖磷酸途径、MAPK等信号通路发挥治疗肝癌的作用。 展开更多

目的基于网络药理学方法及实验验证探讨复方金钱草颗粒(JQC)对前列腺增生(BPH)的防治作用及机制。方法(1)通过TCMSP数据库检索广金钱草、车前草、光石韦、玉米须的活性成分,同时通过文献检索进行补充;利用PubChem、Swiss Target Predict... 目的基于网络药理学方法及实验验证探讨复方金钱草颗粒(JQC)对前列腺增生(BPH)的防治作用及机制。方法(1)通过TCMSP数据库检索广金钱草、车前草、光石韦、玉米须的活性成分,同时通过文献检索进行补充;利用PubChem、Swiss Target Prediction、CTD数据库筛选JQC活性成分作用靶点。通过GeneCards、CTD、DRUGBANK、DisGeNET数据库筛选BPH疾病相关靶点。对JQC活性成分作用靶点与BPH疾病相关靶点进行映射取交集,即得JQC治疗BPH的潜在作用靶点。利用STRING 11.5数据库对JQC治疗BPH的潜在作用靶点进行蛋白互作(PPI)网络分析。利用Metascape平台对潜在作用靶点进行GO功能及KEGG通路富集分析。采用Cytoscape 3.9.1软件构建“药物-成分-靶点-通路”网络,预测JQC治疗BPH的关键活性成分及核心靶点。利用AutoDock Vina软件进行关键活性成分与核心靶点的分子对接验证。(2)将60只SD雄性大鼠随机分为空白组、模型组、前列舒通胶囊组(0.324 g·kg^(-1))及JQC高、中、低剂量组(3.24、1.62、0.81 g·kg^(-1)),每组10只。采用去势联合注射丙酸睾酮诱导建立BPH大鼠模型。灌胃给药,每天1次,连续给药30 d。摘取大鼠前列腺组织,称定质量并计算前列腺湿质量指数;采用ELISA法检测血清COX-2、VEGF、PGE2的含量;HE染色法观察大鼠前列腺组织的病理变化;Western Blot法检测前列腺组织中AKT1、p-AKT1、PI3K、p-PI3K、COX-2、VEGF蛋白表达水平。结果(1)共得到JQC活性成分作用靶点260个,疾病相关靶点1584个,JQC治疗BPH的潜在作用靶点(交集靶点)125个。PPI网络分析发现AKT1、TNF、TP53、IL1β、EGFR、CASP3、PTGS2等靶点可能是JQC治疗BPH的重要靶点。JQC治疗BPH的潜在作用靶点主要与IL-17、HIF-1、TNF及PI3K-Akt通路相关。得到关键活性成分为木犀草素、芹菜素、车前子苷、芒果苷、夏佛塔苷、异牡荆苷,核心靶点为PTGS2、AR、CASP3、IL6、PTGS1、AKT1等。共有24对“关键活性成分-核心靶点”的分子结合能均≤-6 kcal·mol^(-1);PTGS2蛋白受体与木犀草素、芹菜素等配体结合活性最好;AKT1与车前子苷、芒果苷、木犀草素、芹菜素、夏佛塔苷、异牡荆苷等均具有较好的亲和力。(2)与模型组比较,JQC高、中剂量组大鼠的前列腺湿质量与湿质量指数均明显降低(P<0.05,P<0.01);大鼠前列腺腺体基底细胞肥大有明显缓解,腺腔大小相对规则,上皮细胞增生情况均有所改善;血清COX-2、VEGF、PGE2水平明显降低(P<0.05,P<0.01);前列腺组织中的p-AKT1、p-PI3K、PI3K、COX-2、VEGF蛋白表达明显下调(P<0.05,P<0.01)。结论JQC对BPH具有明显防治作用,其机制可能是通过车前子苷、芒果苷、木犀草素等活性成分来调控PI3K/Akt信号通路,抑制COX-2的表达,减少PGE2释放,进而导致VEGF表达下调,抑制前列腺细胞增殖。 展开更多

复方仙鹤颗粒治疗儿童难治性过敏性紫癜的临床研究

目的:观察复方仙鹤颗粒治疗难治性过敏性紫癜的临床疗效及其对免疫、炎症、凝血相关指标的影响。方法:将120例过敏性紫癜患儿,按照随机数字表法分为观察组和对照组,每组60例,两组患儿均给予西医常规治疗,在此基础上对照组口服槐杞黄颗粒... 目的:观察复方仙鹤颗粒治疗难治性过敏性紫癜的临床疗效及其对免疫、炎症、凝血相关指标的影响。方法:将120例过敏性紫癜患儿,按照随机数字表法分为观察组和对照组,每组60例,两组患儿均给予西医常规治疗,在此基础上对照组口服槐杞黄颗粒,观察组加用复方仙鹤颗粒口服,连续治疗8周后评价两组患儿的疗效,观察两组患儿皮疹消退时间,复发次数,复发间隔时间,每2周检测尿常规;治疗前后两组患儿分别进行安全性指标(血常规、肝肾功能、电解质)、免疫相关指标(细胞因子、免疫球蛋白IgA、IgE)、炎性和凝血相关指标[C反应蛋白(CRP)、抗链球菌溶血素O(ASO)、D二聚体(D-D)]检测;并在6个月内进行随访,比较两组复发率。结果:观察组总有效率为86.7%,对照组为63.3%,观察组总有效率显著高于对照组(P<0.05)。与对照组相比,观察组治疗后皮疹持续时间缩短,复发次数减少、复发间隔时间更长(P<0.05);但两组患儿治疗后肾脏损伤改善比较无统计学意义(P>0.05)。治疗后,两组患者IL-2水平均高于治疗前,IL-4、IL-6、IL-10水平均低于治疗前(P<0.05),且治疗后观察组IL-2水平高于对照组,IL-4、IL-6、IL-10水平显著低于对照组(P<0.05),TNF-α水平两组之间差异无统计学意义(P>0.05);治疗后两组IgA水平均低于治疗前(P<0.05),且观察组IgA水平显著低于对照组(P<0.05),IgE水平两组之间差异无统计学意义(P>0.05);治疗后两组ASO、CRP、D-D水平均低于治疗前(P<0.05),且观察组ASO、CRP、D-D水平显著低于对照组(P<0.05);观察组复发率低于对照组(P<0.05)。结论:对于难治性过敏性紫癜,应用复方仙鹤颗粒联合西药治疗,临床疗效显著而且安全性好,特别是在治疗皮肤型过敏性紫癜较有优势,可缩短过敏性紫癜患者的皮疹持续时间,有效降低复发率,改善长期预后。作用机制可能与调节Th1/Th2免疫平衡,控制炎症反应,改善血液高凝状态有关。 展开更多

不同APP添加量对复合肥料造粒性能的影响

聚磷酸铵(APP)作为近几年备受关注的新型肥料,在农业应用中研究较多且效果显著,但作为原料在复合肥料生产中的研究报道较少。为探究APP与其他肥料的复配和造粒性能,采用APP、磷酸一铵(MAP)、磷酸二铵(DAP)等原料通过复配再造粒制备复合... 聚磷酸铵(APP)作为近几年备受关注的新型肥料,在农业应用中研究较多且效果显著,但作为原料在复合肥料生产中的研究报道较少。为探究APP与其他肥料的复配和造粒性能,采用APP、磷酸一铵(MAP)、磷酸二铵(DAP)等原料通过复配再造粒制备复合肥料。肥料制备共设计两个方案,其中方案I以APP、MAP、尿素和K_(2)SO_(4)为原料,方案II以APP、DAP、尿素和K_(2)SO_(4)为原料,同时两个方案又分别设定不同的APP添加量作为变量,且对原料配伍性能、造粒过程现象、加水量、成粒率、肥料p H、颗粒强度、颗粒分散性及肥料吸湿性等造粒性能进行了评价。结果表明:APP与其他原料复配再造粒能够制备2~4.75 mm的颗粒产品;当聚合磷质量分数≥40%时,复配性能良好;不同磷源配伍对复合肥料造粒性能有明显影响,方案II的产品成粒率、颗粒强度和分散性均优于方案I,但方案II的吸湿性较强;适宜的APP添加量有利于复合肥料造粒,两个方案均显示在聚合磷质量分数为20%~40%时,肥料产品的成粒率较高,方案I>67%,方案II>77%。 展开更多

目的探究中药复方消银颗粒对咪喹莫特诱导的银屑病小鼠的改善作用及其潜在的信号通路。方法采用咪喹莫特乳膏涂抹构建银屑病小鼠模型,随机分为以下5组:对照组(control)、咪喹莫特(IMQ)组、中药复方消银颗粒低剂量组、中药复方消银颗粒... 目的探究中药复方消银颗粒对咪喹莫特诱导的银屑病小鼠的改善作用及其潜在的信号通路。方法采用咪喹莫特乳膏涂抹构建银屑病小鼠模型,随机分为以下5组:对照组(control)、咪喹莫特(IMQ)组、中药复方消银颗粒低剂量组、中药复方消银颗粒高剂量组、阿维A组。观察不同组小鼠皮损严重程度,分析各组小鼠皮损面积和严重程度指数(psoriasis area and severity index,PASI)评分,处死小鼠后收集血清,HE染色观察各组小鼠组织形态。CCK-8检测含药血清处理后HaCaT细胞增殖活力,流式细胞仪检测不同处理组HaCaT细胞凋亡情况,Western blot检测HaCaT细胞内JAK/STAT3通路的激活情况。结果与对照组相比,模型组小鼠出现了明显的银屑病病变,红斑、鳞屑明显增加,表皮厚度增加,而中药复方消银颗粒与阿维A作用组小鼠PASI评分明显降低,症状明显减轻。不同浓度含药血清(2.5%、5%和10%)作用HaCaT细胞后,可抑制其增殖并促进凋亡。此外5%、10%含药血清作用细胞后,能显著抑制JAK、p-JAK、STAT3、p-STAT3蛋白水平。结论中药复方消银颗粒通过抑制JAK/STAT3通路激活在缓解银屑病中发挥重要效应。 展开更多
