BACKGROUND Yigong San(YGS)is a representative prescription for the treatment of digestive disorders,which has been used in clinic for more than 1000 years.However,the mechanism of its anti-gastric cancer and regulate ...BACKGROUND Yigong San(YGS)is a representative prescription for the treatment of digestive disorders,which has been used in clinic for more than 1000 years.However,the mechanism of its anti-gastric cancer and regulate immunity are still remains unclear.AIM To explore the mechanism of YGS anti-gastric cancer and immune regulation.METHODS Firstly,collect the active ingredients and targets of YGS,and the differentially expressed genes of gastric cancer.Secondly,constructed a protein-protein interaction network between the targets of drugs and diseases,and screened hub genes.Then the clinical relevance,mutation and repair,tumor microenvironment and drug sensitivity of the hub gene were analyzed.Finally,molecular docking was used to verify the binding ability of YGS active ingredient and hub genes.RESULTS Firstly,obtained 55 common targets of gastric cancer and YGS.The Kyoto Encyclopedia of Genes and Genomes screened the microtubule-associated protein kinase signaling axis as the key pathway and IL6,EGFR,MMP2,MMP9 and TGFB1 as the hub genes.The 5 hub genes were involved in gastric carcinogenesis,staging,typing and prognosis,and their mutations promote gastric cancer progression.Finally,molecular docking results confirmed that the components of YGS can effectively bind to therapeutic targets.CONCLUSION YGS has the effect of anti-gastric cancer and immune regulation.展开更多

The formation of reservoir space went through the cooling and solidification stage (including blast fragmentation, crystallization differentiation and solidification) and the epidiagenesis stage (including metasomatism, filling, weathering and leaching, formation fluid dissolution and tectonism). Primary pores were formed at the solidification stage, which laid the foundation for the development and transformation of effective reservoirs. Secondary pores were formed at the epidiagenesis stage, with key factors as weathering and leaching, formation fluid dissolution and tectonism. In China, Mesozoic-Cenozoic volcanic rocks developed in the Songliao Basin and Bohai Bay Basin in the east and Late Paleozoic volcanic rocks developed in the Junggar Basin, Santanghu Basin and Ta- rim Basin in the west. There are primary volcanic reser- voirs and secondary volcanic reservoirs in these volcanic rocks, which have good accumulation conditions and great exploration potential.展开更多
Background:Baoyuan decoction is used clinically as an adjuvant treatment for lung cancer.However,the underlying mechanism remains unclear.Therefore,this study aimed to explore the mechanism of action of Baoyuan decoct...Background:Baoyuan decoction is used clinically as an adjuvant treatment for lung cancer.However,the underlying mechanism remains unclear.Therefore,this study aimed to explore the mechanism of action of Baoyuan decoction in lung cancer treatment using network pharmacology and molecular docking technology.Methods:The Traditional Chinese Medicines Systems Pharmacology Database and Analysis Platform and SwissTargetPrediction databases were used to screen the active ingredients of Baoyuan decoction and their relevant targets.Lung cancer-related targets were obtained from the GeneCards,Online Mendelian Inheritance in Man,and DrugBank databases.Protein-protein interaction network of the common targets was constructed using the STRING database and analyzed using Cytoscape software 3.10.1.Furthermore,Gene Ontology enrichment,Kyoto Encyclopedia of Genes and Genomes pathway analyses and visualization of common genes were performed using the R software.Finally,molecular docking of the selected key ingredients and targets was performed,and the results were verified using AutoDock Vina software.Results:We identified 142 potential active ingredients,3624 potential lung cancer-related targets,and 341 common drug targets.A total of 72 core targets were identified,of which AKT1,TP53,interleukin-6,epithelial growth factor receptor,and signal transducer and activator of transcription 3 were key.A total of 4116 items were obtained via Gene Ontology enrichment analyses.Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses revealed 189 related signaling pathways,including the PI3K-Akt,AGE-RAGE signaling pathways in diabetic complications,FOXO,and TH signaling pathways,which are involved in cell proliferation,autophagy,metastasis,invasion,radiation resistance,and chemotherapy resistance in the lung cancer microenvironment.The molecular docking results suggested that the key ingredients had a strong affinity for key targets.Conclusion:This study demonstrates that Baoyuan decoction plays a key therapeutic role in a complex manner involving multiple ingredients,targets,and pathways in lung cancer.Our findings are anticipated to provide new ideas for follow-up experimental research and clinical application.展开更多
Background:To explore the potential mechanism of action of the active ingredients of Smilacis Glabrae Rhizoma(SGR)in the treatment of migraine using network pharmacology and in vivo experiments.Methods:Through the sea...Background:To explore the potential mechanism of action of the active ingredients of Smilacis Glabrae Rhizoma(SGR)in the treatment of migraine using network pharmacology and in vivo experiments.Methods:Through the search of Traditional Chinese Medicines Systems Pharmacology Database and Analysis Platform,Genecards,Drugbank and other databases,we obtained active ingredients,targets of SGR and related disease targets of migraine,and took the intersection for protein-protein interactions analysis.After constructing the network diagram,network topology analysis was performed to derive the core targets and key active ingredients,and Gene Ontology enrichment and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed.Finally,molecular docking was performed and validated by in vivo experiments.In vivo experiments,18 male BALB/c mice were selected,and the SGR group was fed with SGR drinking tablet concentrate,and nitroglycerin injection was used to construct a mouse model of migraine.Enzyme-linked immunosorbent assay test was used to detect the levels of TNF-α,IL-1β,IL-6,and AKT1 in plasma.Results:The results showed that the core targets of SGR for the treatment of migraine were TNF-α,IL-1β,IL-6,and AKT1.These core targets and key active ingredients had better binding ability.Compared with the blank group,the number of head scratching in the model group increased.Compared with the model group,there was a significant reduction of the number of head scratching in the SGR group.In comparison with the blank group,the protein level in the plasma in the model group was markedly higher.Compared with the model group,the protein level in the SGR group was significantly lower.Conclusion:SGR has the characteristics of improving migraine based on multi-targets,multi-components and multi-pathways,and the mechanism of action may be related to the inhibition of the release of inflammatory factors,neuron protection,and interference with apoptosis and other processes.展开更多
The molecular mechanism underlying Corydalis Yanhusuo’s therapeutic potential in prostate cancer(PCa)treatment was elucidated using network pharmacology and molecular docking.Nineteen active ingredients,399 drug targ...The molecular mechanism underlying Corydalis Yanhusuo’s therapeutic potential in prostate cancer(PCa)treatment was elucidated using network pharmacology and molecular docking.Nineteen active ingredients,399 drug targets,1790 disease targets and 143 intersection targets were identified.Ten core targets were screened from the protein-protein interaction network.Enrichment analysis revealed 133 GO terms and 114 KEGG pathways.Corydalis Yanhusuo may potentially treat prostate cancer through pathways such as the Rap1 signaling pathway,phospholipase D signaling pathway,Ras signaling pathway,VEGF signaling pathway and JAK-STAT signaling pathway.Significant differences in expression were observed for EGFR,PDGFRA,PIK3CA,PIK3CD,PIK3CG and PIK3R1.Molecular docking and dynamics simulation analysis showed low binding energy between active components and the six core genes of Corydalis Yanhusuo,indicating a favorable docking effect.This study shows that Corydalis Yanhusuo exhibits promise in prostate cancer treatment through a synergistic“multi-component-multi-target-multi-pathway”effect.展开更多
Catastrophic geological disasters frequently occur on slopes with obliquely inclined bedding structures(also referred to as obliquely inclined bedding slopes),where the apparent dip sliding is not readily visible.This...Catastrophic geological disasters frequently occur on slopes with obliquely inclined bedding structures(also referred to as obliquely inclined bedding slopes),where the apparent dip sliding is not readily visible.This phenomenon has become a focal point in landslide research.Yet,there is a lack of studies on the failure modes and mechanisms of hidden,steep obliquely inclined bedding slopes.This study investigated the Shanyang landslide in Shaanxi Province,China.Using field investigations,laboratory tests of geotechnical parameters,and the 3DEC software,this study developed a numerical model of the landslide to analyze the failure process of such slopes.The findings indicate that the Shanyang landslide primarily crept along a weak interlayer under the action of gravity.The landslide,initially following a dip angle with the support of a stable inclined rock mass,shifted direction under the influence of argillization in the weak interlayer,moving towards the apparent dip angle.The slide resistance effect of the karstic dissolution zone was increasingly significant during this process,with lateral friction being the primary resistance force.A reduction in the lateral friction due to karstic dissolution made the apparent dip sliding characteristics of the Shanyang landslide more pronounced.Notably,deformations such as bending and uplift at the slope’s foot suggest that the main slide resistance shifts from lateral friction within the karstic dissolution zone to the slope foot’s resistance force,leading to the eventual buckling failure of the landslide.This study unveils a novel failure mode of apparent dip creep-buckling in the Shanyang landslide,highlighting the critical role of lateral friction from the karstic dissolution zone in its failure mechanism.These insights offer a valuable reference for mitigating risks and preventing disasters related to obliquely inclined bedding landslides.展开更多

Traditional mechanical rock breaking method is labor-intensive and low-efficient,which restrictes the development of deep resources and deep space.As a new rock-breakage technology,microwave irradiation is expected to...Traditional mechanical rock breaking method is labor-intensive and low-efficient,which restrictes the development of deep resources and deep space.As a new rock-breakage technology,microwave irradiation is expected to overcome these problems.This study examines the failure characteristics,weakening law,and breakdown mechanism of deep sandstone(depth=1050 m)samples in a microwave field.The macroscopic and microscopic properties were determined via mechanical tests,mesoscopic tests,and numerical simulations.Microwave application at 1000 W for 60 s reduced the uniaxial compressive strength of the sandstone by 50%.Thermal stress of the sandstone was enhanced by uneven expansion of minerals at the microscale.Moreover,the melting of some minerals in the high-temperature environment changed the pore structure,sharply reducing the macroscopic strength.The temperature remained high in the lower midsection of the sample,and the stress was concentrated at the bottom of the sample and along its axis.These results are expected to improve the efficiency of deep rock breaking,provide theoretical and technical support for similar rock-breakage projects,and accelerate advances in deep-Earth science.展开更多

By examining structures,sediments,reservoirs and accumulation assemblages in the Deyang-Anyue rift and its surrounding area,four new understandings are obtained.First,during the initiation period of Deyang-Anyue rift,...By examining structures,sediments,reservoirs and accumulation assemblages in the Deyang-Anyue rift and its surrounding area,four new understandings are obtained.First,during the initiation period of Deyang-Anyue rift,multiple groups of faults developed in the rift due to the effect of tensile force,bringing about multiple mound and shoal belts controlled by horsts in the second member of the Sinian Dengying Formation;in the development stage of the rift,the boundary faults of the rift controlled the development of mound and shoal belts at the platform margin in the fourth member of Dengying Formation;during the shrinkage period of the rift,platform margin grain shoals of the Cambrian Canglangpu Formation developed in the rift margin.Second,four sets of large-scale mound and shoal reservoirs in the second member of Dengying Formation,the fourth member of Dengying Formation,Canglangpu Formation and Longwangmiao Formation overlap with several sets of source rocks such as Qiongzhusi Formation source rocks and Dengying Formation argillaceous limestone or dolomite developed inside and outside the rift,forming good source-reservoir-cap rock combinations;the sealing of tight rock layers in the lateral and updip direction results in the formation model of large lithologic gas reservoirs of oil pool before gas,continuous charging and independent preservation of each gas reservoir.Third,six favorable exploration zones of large-scale lithologic gas reservoirs have been sorted out through comprehensive evaluation,namely,mound and shoal complex controlled by horsts in the northern part of the rift in the second member of Dengying Formation,isolated karst mound and shoal complex of the fourth member of Dengying Formation in the south of the rift,the superimposed area of multi-stage platform margin mounds and shoals of the second and fourth members of Dengying Formation and Canglangpu Formation in the north slope area,the platform margin mounds and shoals of the second and fourth members of Dengying Formation in the west side of the rift,the platform margin mound and shoal bodies of the fourth member of Dengying Formation in the south slope area,etc.Fourth,Well Pengtan-1 drilled on the mound and shoal complex controlled by horsts of the second member of Dengying Formation in the rift and Well Jiaotan-1 drilled on the platform margin mound and shoal complex of the North Slope have obtained high-yield gas flows in multiple target layers,marking the discovery of a new gas province with reserves of(2-3)×10^(12) m^(3).This has proved the huge exploration potential of large lithologic gas reservoir group related to intracratonic rift.展开更多
Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effectiv...Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effective,but its underlying molecular mechanisms remain unclear.In this study,we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology,and provided some novel ideas for managing this disease.Methods:In the present study,we focused on the underlying mechanisms between DPS and endometriosis,via oral bioavailability screening,drug similarity assessment,target identification,network analysis,drug relocation,and molecular docking using network pharmacology.Results:By bioinformatics,54 active components of DSP were predicted from the 534 components provided by the TCMSP database.We then found 85 validated targets and 6059 predicted targets for herbal components,and 2241 targets for endometriosis.After mapping,there were 255 targets in common.After that,network analysis was performed to screen key networks and core targets.Ninetynine indirect targets were collected from 14 direct ones.GO and KEGG analyses identified 63 enriched functions and 10 pathways(P<.01).According to the disease target,dipyridamole could undergo drug repositioning for endometriosis(score?1).The mechanism of action of dipyridamole on endometriosis was compared with that of DSP.Finally,eight targets,namely,AR,ESR1,HMGCR,NOS2,NR3C1,PPARG,PTGES,and PTGS2,were validated by molecular docking,suggesting that the probable molecular mechanisms by which DSP treats endometriosis are through seven validated pathways:hsa05200,hsa04915,hsa01100,hsa00900,hsa04920,hsa01130,and hsa00590.Conclusion:In this study,we uncovered the mechanism by which DSP can treat endometriosis using a drugecomponentetarget interaction network.This approach can provide some novel ideas for the development of Chinese medicine and the clinical management of endometriosis.展开更多
文摘A recent study by Wang et al,published in the World Journal of Psychiatry,provided preventative and therapeutic strategies for the comorbidity of obesity and depression.The gut-brain axis,which acts as a two-way communication system between the gastrointestinal tract and the central nervous system,plays a pivotal role in the pathogenesis of these conditions.Evidence suggests that metabolic byproducts,such as short-chain fatty acids,lipopolysaccharide and bile acids,which are generated by the gut microbiota,along with neurotransmitters and inflammatory mediators within the gut-brain axis,modulate the host's metabolic processes,neuronal regulation,and immune responses through diverse mechanisms.The interaction between obesity and depression via the gut-brain axis involves disruptions in the gut microbiota balance,inflammatory immune responses,and alterations in the neuroendocrine system.Modulating the gut-brain axis,for example,through a ketogenic diet,the use of probiotics,and the supplementation of antioxidants,offers new remedial approaches for obesity and depression.Future research that explores the mechanisms of the gut-brain axis is needed to provide more evidence for clinical treatment.

文摘This review comments on the article“To explore the mechanism of Yigong San anti-gastric cancer and immune regulation”.We are interested that the article applied network pharmacology and bioinformatics techniques to elucidate the mechanism of action of Yigong Sang,a traditional Chinese medicine(TCM),in the treatment of gastric cancer(GC).The mechanism of action of Yigong Sang in the treatment of GC has not yet been elucidated because it is composed of multiple Chinese medicines with multiple components and multiple targets.The emergence of network pharmacology and bioinformatics analysis helps explain the mechanism of action of TCM in preventing and treating GC,and provides a possibility for TCM to transform from empirical to evidence-based medicine.This is of great significance for the application of TCM in oncology,new drug development,formula optimization,and the improvement of clinical efficacy.

文摘Cells,tissues,and organs are constantly subjected to the action of mechanical forces from the extracellular environment-and the nervous system is no exception.Cell-intrinsic properties such as membrane lipid composition,abundance of mechanosensors,and cytoskeletal dynamics make cells more or less likely to sense these forces.Intrinsic and extrinsic cues are integrated by cells and this combined information determines the rate and dynamics of membrane protrusion growth or retraction(Yamada and Sixt,2019).Cell protrusions are extensions of the plasma membrane that play crucial roles in diverse contexts such as cell migration and neuronal synapse formation.In the nervous system,neurons are highly dynamic cells that can change the size and number of their pre-and postsynaptic elements(called synaptic boutons and dendritic spines,respectively),in response to changes in the levels of synaptic activity through a process called plasticity.Synaptic plasticity is a hallmark of the nervous system and is present throughout our lives,being required for functions like memory formation or the learning of new motor skills(Minegishi et al.,2023;Pillai and Franze,2024).

文摘BACKGROUND Yigong San(YGS)is a representative prescription for the treatment of digestive disorders,which has been used in clinic for more than 1000 years.However,the mechanism of its anti-gastric cancer and regulate immunity are still remains unclear.AIM To explore the mechanism of YGS anti-gastric cancer and immune regulation.METHODS Firstly,collect the active ingredients and targets of YGS,and the differentially expressed genes of gastric cancer.Secondly,constructed a protein-protein interaction network between the targets of drugs and diseases,and screened hub genes.Then the clinical relevance,mutation and repair,tumor microenvironment and drug sensitivity of the hub gene were analyzed.Finally,molecular docking was used to verify the binding ability of YGS active ingredient and hub genes.RESULTS Firstly,obtained 55 common targets of gastric cancer and YGS.The Kyoto Encyclopedia of Genes and Genomes screened the microtubule-associated protein kinase signaling axis as the key pathway and IL6,EGFR,MMP2,MMP9 and TGFB1 as the hub genes.The 5 hub genes were involved in gastric carcinogenesis,staging,typing and prognosis,and their mutations promote gastric cancer progression.Finally,molecular docking results confirmed that the components of YGS can effectively bind to therapeutic targets.CONCLUSION YGS has the effect of anti-gastric cancer and immune regulation.
文摘The complex morphological,anatomical,physiological,and chemical mechanisms within the aging brain have been the hot topic of research for centuries.The aging process alters the brain structure that affects functions and cognitions,but the worsening of such processes contributes to the pathogenesis of neurodegenerative disorders,such as Alzheimer's disease.Beyond these observable,mild morphological shifts,significant functional modifications in neurotransmission and neuronal activity critically influence the aging brain.Understanding these changes is important for maintaining cognitive health,especially given the increasing prevalence of age-related conditions that affect cognition.This review aims to explore the age-induced changes in brain plasticity and molecular processes,differentiating normal aging from the pathogenesis of Alzheimer's disease,thereby providing insights into predicting the risk of dementia,particularly Alzheimer's disease.

文摘Regulated cell death is a form of cell death that is actively controlled by biomolecules.Several studies have shown that regulated cell death plays a key role after spinal cord injury.Pyroptosis and ferroptosis are newly discovered types of regulated cell deaths that have been shown to exacerbate inflammation and lead to cell death in damaged spinal cords.Autophagy,a complex form of cell death that is interconnected with various regulated cell death mechanisms,has garnered significant attention in the study of spinal cord injury.This injury triggers not only cell death but also cellular survival responses.Multiple signaling pathways play pivotal roles in influencing the processes of both deterioration and repair in spinal cord injury by regulating pyroptosis,ferroptosis,and autophagy.Therefore,this review aims to comprehensively examine the mechanisms underlying regulated cell deaths,the signaling pathways that modulate these mechanisms,and the potential therapeutic targets for spinal cord injury.Our analysis suggests that targeting the common regulatory signaling pathways of different regulated cell deaths could be a promising strategy to promote cell survival and enhance the repair of spinal cord injury.Moreover,a holistic approach that incorporates multiple regulated cell deaths and their regulatory pathways presents a promising multi-target therapeutic strategy for the management of spinal cord injury.

文摘Characterized by complex lithology and strong heterogeneity, volcanic reservoirs in China developed three reservoir space types: primary pores, secondary pores and fractures. The formation of reservoir space went through the cooling and solidification stage (including blast fragmentation, crystallization differentiation and solidification) and the epidiagenesis stage (including metasomatism, filling, weathering and leaching, formation fluid dissolution and tectonism). Primary pores were formed at the solidification stage, which laid the foundation for the development and transformation of effective reservoirs. Secondary pores were formed at the epidiagenesis stage, with key factors as weathering and leaching, formation fluid dissolution and tectonism. In China, Mesozoic-Cenozoic volcanic rocks developed in the Songliao Basin and Bohai Bay Basin in the east and Late Paleozoic volcanic rocks developed in the Junggar Basin, Santanghu Basin and Ta- rim Basin in the west. There are primary volcanic reser- voirs and secondary volcanic reservoirs in these volcanic rocks, which have good accumulation conditions and great exploration potential.
文摘Background:Baoyuan decoction is used clinically as an adjuvant treatment for lung cancer.However,the underlying mechanism remains unclear.Therefore,this study aimed to explore the mechanism of action of Baoyuan decoction in lung cancer treatment using network pharmacology and molecular docking technology.Methods:The Traditional Chinese Medicines Systems Pharmacology Database and Analysis Platform and SwissTargetPrediction databases were used to screen the active ingredients of Baoyuan decoction and their relevant targets.Lung cancer-related targets were obtained from the GeneCards,Online Mendelian Inheritance in Man,and DrugBank databases.Protein-protein interaction network of the common targets was constructed using the STRING database and analyzed using Cytoscape software 3.10.1.Furthermore,Gene Ontology enrichment,Kyoto Encyclopedia of Genes and Genomes pathway analyses and visualization of common genes were performed using the R software.Finally,molecular docking of the selected key ingredients and targets was performed,and the results were verified using AutoDock Vina software.Results:We identified 142 potential active ingredients,3624 potential lung cancer-related targets,and 341 common drug targets.A total of 72 core targets were identified,of which AKT1,TP53,interleukin-6,epithelial growth factor receptor,and signal transducer and activator of transcription 3 were key.A total of 4116 items were obtained via Gene Ontology enrichment analyses.Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses revealed 189 related signaling pathways,including the PI3K-Akt,AGE-RAGE signaling pathways in diabetic complications,FOXO,and TH signaling pathways,which are involved in cell proliferation,autophagy,metastasis,invasion,radiation resistance,and chemotherapy resistance in the lung cancer microenvironment.The molecular docking results suggested that the key ingredients had a strong affinity for key targets.Conclusion:This study demonstrates that Baoyuan decoction plays a key therapeutic role in a complex manner involving multiple ingredients,targets,and pathways in lung cancer.Our findings are anticipated to provide new ideas for follow-up experimental research and clinical application.

文摘Background:To explore the potential mechanism of action of the active ingredients of Smilacis Glabrae Rhizoma(SGR)in the treatment of migraine using network pharmacology and in vivo experiments.Methods:Through the search of Traditional Chinese Medicines Systems Pharmacology Database and Analysis Platform,Genecards,Drugbank and other databases,we obtained active ingredients,targets of SGR and related disease targets of migraine,and took the intersection for protein-protein interactions analysis.After constructing the network diagram,network topology analysis was performed to derive the core targets and key active ingredients,and Gene Ontology enrichment and Kyoto Encyclopedia of Genes and Genomes enrichment analyses were performed.Finally,molecular docking was performed and validated by in vivo experiments.In vivo experiments,18 male BALB/c mice were selected,and the SGR group was fed with SGR drinking tablet concentrate,and nitroglycerin injection was used to construct a mouse model of migraine.Enzyme-linked immunosorbent assay test was used to detect the levels of TNF-α,IL-1β,IL-6,and AKT1 in plasma.Results:The results showed that the core targets of SGR for the treatment of migraine were TNF-α,IL-1β,IL-6,and AKT1.These core targets and key active ingredients had better binding ability.Compared with the blank group,the number of head scratching in the model group increased.Compared with the model group,there was a significant reduction of the number of head scratching in the SGR group.In comparison with the blank group,the protein level in the plasma in the model group was markedly higher.Compared with the model group,the protein level in the SGR group was significantly lower.Conclusion:SGR has the characteristics of improving migraine based on multi-targets,multi-components and multi-pathways,and the mechanism of action may be related to the inhibition of the release of inflammatory factors,neuron protection,and interference with apoptosis and other processes.

文摘The molecular mechanism underlying Corydalis Yanhusuo’s therapeutic potential in prostate cancer(PCa)treatment was elucidated using network pharmacology and molecular docking.Nineteen active ingredients,399 drug targets,1790 disease targets and 143 intersection targets were identified.Ten core targets were screened from the protein-protein interaction network.Enrichment analysis revealed 133 GO terms and 114 KEGG pathways.Corydalis Yanhusuo may potentially treat prostate cancer through pathways such as the Rap1 signaling pathway,phospholipase D signaling pathway,Ras signaling pathway,VEGF signaling pathway and JAK-STAT signaling pathway.Significant differences in expression were observed for EGFR,PDGFRA,PIK3CA,PIK3CD,PIK3CG and PIK3R1.Molecular docking and dynamics simulation analysis showed low binding energy between active components and the six core genes of Corydalis Yanhusuo,indicating a favorable docking effect.This study shows that Corydalis Yanhusuo exhibits promise in prostate cancer treatment through a synergistic“multi-component-multi-target-multi-pathway”effect.

文摘Catastrophic geological disasters frequently occur on slopes with obliquely inclined bedding structures(also referred to as obliquely inclined bedding slopes),where the apparent dip sliding is not readily visible.This phenomenon has become a focal point in landslide research.Yet,there is a lack of studies on the failure modes and mechanisms of hidden,steep obliquely inclined bedding slopes.This study investigated the Shanyang landslide in Shaanxi Province,China.Using field investigations,laboratory tests of geotechnical parameters,and the 3DEC software,this study developed a numerical model of the landslide to analyze the failure process of such slopes.The findings indicate that the Shanyang landslide primarily crept along a weak interlayer under the action of gravity.The landslide,initially following a dip angle with the support of a stable inclined rock mass,shifted direction under the influence of argillization in the weak interlayer,moving towards the apparent dip angle.The slide resistance effect of the karstic dissolution zone was increasingly significant during this process,with lateral friction being the primary resistance force.A reduction in the lateral friction due to karstic dissolution made the apparent dip sliding characteristics of the Shanyang landslide more pronounced.Notably,deformations such as bending and uplift at the slope’s foot suggest that the main slide resistance shifts from lateral friction within the karstic dissolution zone to the slope foot’s resistance force,leading to the eventual buckling failure of the landslide.This study unveils a novel failure mode of apparent dip creep-buckling in the Shanyang landslide,highlighting the critical role of lateral friction from the karstic dissolution zone in its failure mechanism.These insights offer a valuable reference for mitigating risks and preventing disasters related to obliquely inclined bedding landslides.

文摘Traditional mechanical rock breaking method is labor-intensive and low-efficient,which restrictes the development of deep resources and deep space.As a new rock-breakage technology,microwave irradiation is expected to overcome these problems.This study examines the failure characteristics,weakening law,and breakdown mechanism of deep sandstone(depth=1050 m)samples in a microwave field.The macroscopic and microscopic properties were determined via mechanical tests,mesoscopic tests,and numerical simulations.Microwave application at 1000 W for 60 s reduced the uniaxial compressive strength of the sandstone by 50%.Thermal stress of the sandstone was enhanced by uneven expansion of minerals at the microscale.Moreover,the melting of some minerals in the high-temperature environment changed the pore structure,sharply reducing the macroscopic strength.The temperature remained high in the lower midsection of the sample,and the stress was concentrated at the bottom of the sample and along its axis.These results are expected to improve the efficiency of deep rock breaking,provide theoretical and technical support for similar rock-breakage projects,and accelerate advances in deep-Earth science.

文摘This editorial takes a deeper look at the insights provided by Soresi and Giannitrapani,which examined the therapeutic potential of glucagon-like peptide-1 receptor agonists(GLP-1RAs)for metabolic dysfunction-associated fatty liver disease.We provide supplementary insights to their research,highlighting the broader systemic implications of GLP-1RAs,synthesizing the current understanding of their mechanisms and the trajectory of research in this field.GLP-1RAs are revolutionizing the treatment of type 2 diabetes mellitus and beyond.Beyond glycemic control,GLP-1RAs demonstrate cardiovascular and renal protective effects,offering potential in managing diabetic kidney disease alongside renin–angiotensin–aldosterone system inhibitors.Their role in bone metabolism hints at benefits for diabetic osteoporosis,while the neuroprotective properties of GLP-1RAs show promise in Alzheimer's disease treatment by modulating neuronal insulin signaling.Additionally,they improve hormonal and metabolic profiles in polycystic ovary syndrome.This editorial highlights the multifaceted mechanisms of GLP-1RAs,emphasizing the need for ongoing research to fully realize their therapeutic potential across a range of multisystemic diseases.

文摘Background:Explore the anti-tumor mechanism of herb pair Pinellia ternate-Magnolia officinalis(BX-HP)in liver cancer through network pharmacology using molecular docking methods.Method:The active ingredients and corresponding targets of the herb pair Pinellia ternate-Magnolia officinalis were obtained from the HERB database.The relevant targets for liver cancer were obtained from GeneCards,DisGeNET,TTD,and Drugbank databases.Obtain common targets between herb pair Pinellia ternate-Magnolia officinalis and liver cancer through the Bioinformatics platform,establish a PPI network diagram using STRING software,and perform GO functional enrichment and KEGG pathway enrichment analysis on the DAVID platform.AutoDockTools 1.5.7 software and molecular dynamics simulation analysis are used to evaluate the binding of components to target proteins.HERB database,SwissTargetPrediction database,SwissADME database,UniProt database,GeneCards database,TTD database,DRUGBANK database,DisGeNET database,String,DAVID.Bioinformatics platform,PDB database,PubChem and TCMSP database.Result:A total of 22 active ingredients with a Probability>0.1 targets in Magnolia officinalis were screened,26 active ingredients with a Probability>0.1 targets in Pinellia ternata,ten vital active ingredients,corresponding to 979 and 803 targets with a Probability>0.1 targets,2536 liver cancer-related targets,and 279 targets in the herb pair Pinellia ternata-Magnolia officinalis.The GO functional enrichment analysis resulted in 1297 entries,namely 971 biological process entries,118 cell localization entries,and 208 molecular function entries.Three signaling pathways were annotated through the KEGG pathway.Based on molecular docking,ten vital active ingredients and five target proteins were validated to exhibit an excellent binding affinity.The above data indicates that combining the herb pair Pinellia ternata-Magnolia officinalis may treat liver cancer through specific targets and signaling pathways.Conclusion:Herb pair Pinellia ternata-Magnolia officinalis has a synergistic effect on treating liver cancer through multicomponent,multitarget,and multi-pathway approaches.This study provides a sufficient theoretical basis for subsequent research.

文摘By examining structures,sediments,reservoirs and accumulation assemblages in the Deyang-Anyue rift and its surrounding area,four new understandings are obtained.First,during the initiation period of Deyang-Anyue rift,multiple groups of faults developed in the rift due to the effect of tensile force,bringing about multiple mound and shoal belts controlled by horsts in the second member of the Sinian Dengying Formation;in the development stage of the rift,the boundary faults of the rift controlled the development of mound and shoal belts at the platform margin in the fourth member of Dengying Formation;during the shrinkage period of the rift,platform margin grain shoals of the Cambrian Canglangpu Formation developed in the rift margin.Second,four sets of large-scale mound and shoal reservoirs in the second member of Dengying Formation,the fourth member of Dengying Formation,Canglangpu Formation and Longwangmiao Formation overlap with several sets of source rocks such as Qiongzhusi Formation source rocks and Dengying Formation argillaceous limestone or dolomite developed inside and outside the rift,forming good source-reservoir-cap rock combinations;the sealing of tight rock layers in the lateral and updip direction results in the formation model of large lithologic gas reservoirs of oil pool before gas,continuous charging and independent preservation of each gas reservoir.Third,six favorable exploration zones of large-scale lithologic gas reservoirs have been sorted out through comprehensive evaluation,namely,mound and shoal complex controlled by horsts in the northern part of the rift in the second member of Dengying Formation,isolated karst mound and shoal complex of the fourth member of Dengying Formation in the south of the rift,the superimposed area of multi-stage platform margin mounds and shoals of the second and fourth members of Dengying Formation and Canglangpu Formation in the north slope area,the platform margin mounds and shoals of the second and fourth members of Dengying Formation in the west side of the rift,the platform margin mound and shoal bodies of the fourth member of Dengying Formation in the south slope area,etc.Fourth,Well Pengtan-1 drilled on the mound and shoal complex controlled by horsts of the second member of Dengying Formation in the rift and Well Jiaotan-1 drilled on the platform margin mound and shoal complex of the North Slope have obtained high-yield gas flows in multiple target layers,marking the discovery of a new gas province with reserves of(2-3)×10^(12) m^(3).This has proved the huge exploration potential of large lithologic gas reservoir group related to intracratonic rift.

文摘Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effective,but its underlying molecular mechanisms remain unclear.In this study,we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology,and provided some novel ideas for managing this disease.Methods:In the present study,we focused on the underlying mechanisms between DPS and endometriosis,via oral bioavailability screening,drug similarity assessment,target identification,network analysis,drug relocation,and molecular docking using network pharmacology.Results:By bioinformatics,54 active components of DSP were predicted from the 534 components provided by the TCMSP database.We then found 85 validated targets and 6059 predicted targets for herbal components,and 2241 targets for endometriosis.After mapping,there were 255 targets in common.After that,network analysis was performed to screen key networks and core targets.Ninetynine indirect targets were collected from 14 direct ones.GO and KEGG analyses identified 63 enriched functions and 10 pathways(P<.01).According to the disease target,dipyridamole could undergo drug repositioning for endometriosis(score?1).The mechanism of action of dipyridamole on endometriosis was compared with that of DSP.Finally,eight targets,namely,AR,ESR1,HMGCR,NOS2,NR3C1,PPARG,PTGES,and PTGS2,were validated by molecular docking,suggesting that the probable molecular mechanisms by which DSP treats endometriosis are through seven validated pathways:hsa05200,hsa04915,hsa01100,hsa00900,hsa04920,hsa01130,and hsa00590.Conclusion:In this study,we uncovered the mechanism by which DSP can treat endometriosis using a drugecomponentetarget interaction network.This approach can provide some novel ideas for the development of Chinese medicine and the clinical management of endometriosis.
