In order to determine the applicability of microsatellite primers developed from common carp ( Cyprinuscario ) for genomic analysis in mud carp ( Cirrhina molitorella ), 24 primer pairs from common carp were desig...In order to determine the applicability of microsatellite primers developed from common carp ( Cyprinuscario ) for genomic analysis in mud carp ( Cirrhina molitorella ), 24 primer pairs from common carp were designed to amplify microsatellite loci in the mud carp containing CA, GA, AT and GGGA sequences. Thirteen primers (54%) successfully amplified specific products in the mud carp and 11 primers (48%) showed high polymorphism in the mud carp population. The results indicated that the average number of alleles per locus in the mud carp stocks was 5.2. Average heterozygosity (Ho), unbiased expected heterozygosity (He) and polymorphism information content (PIC) in the wild population were 0.61 ± 0.2, 0.8 ± 0.09 and 0.72 ± 0.1 respectively. Several Hardy-Weinberg departure value were significandy departed from Hardy-Weinberg equilibrium. The study showed that microsatellite primers from a species of Cyprinidae can be used for mud carp genetic analysis without much cost or time input.展开更多

Here we reported the development of microsatellite markers and their utilization in mud carp (Cirrhina molitorella). An (CA)15 enriched library has been constructed for mud carp, using the magnetic beads enrichment procedure. Sequence analysis of 60 randomly picked positive colonies indicate that 56 (93.3%) of the colonies contain microsatellites. Microsatellite polymorphism was assessed using 10 mud carp individuals, and 12 microsatellite loci turned out to be polymorphic. We utilized these loci to study the genetic diversity of a wild population (WM) and a cultured population (CM) of the mud carp. A total of 109 alleles were detected with an average of 9.08 alleles per locus. The mean value of the observed heterozygosity of WM and CM was 0.6361 and 0.6417, respectively, and significant decrease of genetic diversity in CM was not observed. The genetic distance between the two populations was 0.1546 and the value of Gsr was 0.0473. This showed that there existed a slight genetic differentiation between WM and CM.展开更多
Analysis of fish meat using gas chromatography is described.Flavor is the sensation arising from the interplay of the signals of sensing smell, taste and irritating stimuli from food stuff.For human, flavor and nutrit...Analysis of fish meat using gas chromatography is described.Flavor is the sensation arising from the interplay of the signals of sensing smell, taste and irritating stimuli from food stuff.For human, flavor and nutrition are inseparable.In fish, trace amount of volatile organic compounds (VOCs) are the major compounds to affect consumer’s preference, which are produced during storage and spoilage.In the present study, volatile compounds were extracted by Likens-Nickerson concurrent distillation apparatus from wild and farmed Cirrhina mrigala and Cyprinus carpio. The quantitative and qualitative estimation of volatiles was made by gas chromatography.Wild and farmed fish of different fish sizes were compared for these compounds (appearing in the form of peaks), which were identified from their retention time by comparing with the standards.Fifteen major VOCs were found in these species which included hexadecane, 3-octanol, hexanal, decane, 3-hexene-1-ol, 2-undecanone, 2-heptanone, butanal, 2-nonanone, 1-heptanal, furaldehyde, 3-methyl-1-butanal, trans-3-hexene-1-ol, octanal and decanal.These compounds varied qualitatively and quantitatively in both wild and farmed fish of different fish sizes.展开更多
