目的:对新基因mRSD-9功能进行初步研究。方法:应用免疫共沉淀结合免疫荧光共定位技术对mRSD-9和endophlin 3的相互作用进行验证;ATP/GTP结合实验及体外网格蛋白释放实验对mRSD-9参与细胞内吞进行初步研究。结果:(1)过量表达的Myc-mRSD-... 目的:对新基因mRSD-9功能进行初步研究。方法:应用免疫共沉淀结合免疫荧光共定位技术对mRSD-9和endophlin 3的相互作用进行验证;ATP/GTP结合实验及体外网格蛋白释放实验对mRSD-9参与细胞内吞进行初步研究。结果:(1)过量表达的Myc-mRSD-9能被Flag-endophilin 3共沉淀,反之,过表达的Flag-endophilin 3也能被Myc-mRSD-9共沉淀。并且在激光共聚焦显微镜下mRSD-9-GFP融合蛋白与endophilin3-RFP融合蛋白在CHO细胞中共定位。(2)mRSD-9蛋白可以和ATP/GTP结合,而且结合是特异的,而突变体ΔmRSD-9则不能。(3)转染mRSD-9组细胞内吞转铁蛋白比周围未转染的细胞明显减少。而转染pDsRed1-N1-ΔmRSD-9组细胞内的转铁蛋白明显增多。结论:mRSD-9基因编码的蛋白可能对内吞具有抑制作用。 展开更多

Intelligent GPS-Less Speed Detection and Clustering in VANET

Vehicular Ad Hoc Network (VANET) has emerged as a new wireless network for vehicular communications. To provide a flexible and high reliable communication service in VANET, vehicles are clustered to construct many small networks (clusters) so that channel interferences and flooding messages can be limited. This research presents a novel Multi-Resolution Relative Speed Detection (MRSD) model to improve the clustering algorithm in VANET without using Global Positioning System (GPS). MRSD uses the Moving Average Convergence Divergence (MACD), the Momentum of Received Signal Strength (MRSS), and Artificial Neural Networks (ANNs) to estimate the motion state and the relative speed of a vehicle based purely on Received Signal Strength. The proposed MRSD model is accurate with the assistance of the intelligent classification, and incurs less overhead in the cluster head election than that of other algorithms. 展开更多
