目的:探讨鱼腥草注射液及聚山梨酯80注射液的过敏原性。方法:以Ⅰ型超敏反应为模型,豚鼠腹腔注射致敏,在末次致敏后的第11天用致敏量的2倍量进行激发,观察过敏反应情况。结果:含聚山梨酯80的鱼腥草注射液、香丹注射液、聚山梨酯80注射... 目的:探讨鱼腥草注射液及聚山梨酯80注射液的过敏原性。方法:以Ⅰ型超敏反应为模型,豚鼠腹腔注射致敏,在末次致敏后的第11天用致敏量的2倍量进行激发,观察过敏反应情况。结果:含聚山梨酯80的鱼腥草注射液、香丹注射液、聚山梨酯80注射液对豚鼠过敏实验均出现阳性结果,其中聚山梨酯80含量达5mg/ml时为明显阳性,聚山梨酯80含量在3mg/ml以下时为弱阳性,不含聚山梨酯80的香丹注射液豚鼠过敏实验结果为阴性;不同来源的聚山梨酯80均可引起豚鼠过敏反应,制备工艺中加热灭菌处理对聚山梨酯80的过敏反应没有影响。结论:含聚山梨酯80的鱼腥草注射液及香丹注射液均可引起豚鼠过敏反应,其原因可能为其中所含聚山梨酯80所致,过敏反应强弱与聚山梨酯80浓度有关,与加热灭菌与否无关。 展开更多

目的 观察复方鱼腥草合剂治疗急喉痹(急性咽炎)外感风热证引起的咽喉肿痛的有效性及安全性。方法 将264例急性咽炎的患者,随机按2:1分入试验组和对照组,试验组口服复方鱼腥草合剂,30 mL/次,3次/d,连续服药7 d。对照组口服复方鱼腥草合... 目的 观察复方鱼腥草合剂治疗急喉痹(急性咽炎)外感风热证引起的咽喉肿痛的有效性及安全性。方法 将264例急性咽炎的患者,随机按2:1分入试验组和对照组,试验组口服复方鱼腥草合剂,30 mL/次,3次/d,连续服药7 d。对照组口服复方鱼腥草合剂模拟剂,且用法用量与试验组完全相同。观察两组治疗期间咽痛消失率,临床缓解情况以及中医证候积分。结果 治疗3 d,试验组和对照组的咽痛消失率分别为40.48%(68/168)和16.87%(14/83)(P<0.05)。两组率差为23.61%(95%CI:12.92%,34.29%),试验组显著优于对照组。咽痛中位消失时间试验组为95.81 h,小于对照组的110.25 h(P<0.05)。两组咽痛VAS评分AUC的分析,试验组为(9.87±7.41),对照组为(12.87±7.36),两组组间比较差异有统计学意义。治疗3 d、5 d,试验组痊愈率分别为23.81%(40/168)、61.31%(103/168)。治疗3 d、5 d,对照组痊愈率分别为7.23%(6/83)、 42.17%(35/83)。试验组的痊愈率显著高于对照组。结论 复方鱼腥草合剂可以有效提高外感风热证引起的咽喉肿痛的痊愈率,缓解病痛,缩短治疗时间,并且没有明显副作用。 展开更多

目的:研究鱼腥草素对乳鼠离体培养的心肌细胞及成纤维细胞增殖及抗氧化作用的影响。方法:取新生SD乳鼠左心室,采用差速贴壁法分离心肌细胞及成纤维细胞,培养在96孔培养板中,在培养成功的心肌细胞及成纤维细胞培养液中加入异丙肾上腺素(I... 目的:研究鱼腥草素对乳鼠离体培养的心肌细胞及成纤维细胞增殖及抗氧化作用的影响。方法:取新生SD乳鼠左心室,采用差速贴壁法分离心肌细胞及成纤维细胞,培养在96孔培养板中,在培养成功的心肌细胞及成纤维细胞培养液中加入异丙肾上腺素(ISO)造模,采用不同浓度的鱼腥草素干预,孵育培养2天,采用MTT法测定成纤维细胞的增殖,采用考马斯亮兰试剂测定心肌细胞中蛋白质含量,并测定其一氧化氮(NO)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的含量。结果:心肌细胞培养液中,10-5mol/L ISO能诱导心肌细胞肥大,SOD及GSH-Px活力明显降低(p<0.05);10-5~10-4mol/L鱼腥草素组蛋白含量则显著降低(p<0.05),SOD活力及GSH-Px均较模型组显著升高(p<0.05);成纤维细胞培养液中,10-5mol/L ISO能诱导成纤维细胞增殖,SOD、NO及GSH-Px含量明显降低(p<0.05);鱼腥草素对成纤维细胞的增殖有抑制作用,对SOD、NO及GSH-Px含量较模型组显著升高(p<0.05)。结论:10-5~10-4mol/L鱼腥草素对ISO诱导的乳鼠心肌细胞肥大及成纤维细胞的增殖有一定对抗作用,其抗心肌肥大作用可能与其抗氧化及影响NO水平有关。 展开更多

[Objective] This study was to identify the expression of exogenous antimicrobial peptide in transgenic Houttuynia cordata Thunb. plants,and analyze their resistance to stem rot disease. [Methods] SDS-PAGE and Western blot analysis were employed to detect expression of exogenous antimicrobial peptide in transgenic H. cordata plants. Both wild type and transgenic H. cordata plants were inoculated with different concentrations of Rhizoctonia solani spores for detecting their resistance. [Results] The exogenous antimicrobial peptide was detected at translation level. The optimal parameters for detecting the resistance of transgenic H. cordata plants to R. solani was inoculation of spores at a concentration of 3×105 ind./ml and cultured for three days. The results showed that resistance of transgenic H. cordata plants to R. solani was enhanced in comparison with CKs. [Conclusion] Expression of exogenous antimicrobial peptide can enhance the resistance of transgenic H. cordata plants to stem rot disease. 展开更多

Objective:This study aims to provide the material basis for the molecular mechanism of Yuxingcao(Houttuynia cordata Thunb,HCT)through the method of evidence-based study to summarize the natural constituents isolated from HCT.Methods:We searched CNKI,Wanfang,VIP,Pubmed and relevant conference compilations.The keywords were'Yuxingcao or Houttuynia cordata Thunb'and'components'or'ingredients'or'constituent'or'volatile oil'or'flavonoids'or'terpene'or'content',both in Chinese and English.According to the inclusion and exclusion criteria,the reported compositions and contents have been summarized,and SPSS software was used to draw the boxplot of contents.Results:A total of 603 natural compounds in 11 categories were obtained from pooled articles.In the diverse components,the number of aliphatic compounds(n=259)and terpenoids(n=158)are more than those of flavonoids(n=26),alkaloids(n=42)and aromatics(n=42).While,in the part of volatile oils of HCT,the largest components are aliphatic compounds(mainly distributed on the ground)and terpenoids(mainly distributed in the underground).Although,methyl n-nonylketone is distributed in the whole herb plant,a large proportion is present in the underground parts.As for non-volatiles,the flavonoid content(mainly distributed on the ground)was the highest,among which quercetin and its glycosyl derivatives were the prominent.Conclusion:The results of this study provide a more comprehensive material basis for the further study of HCT.It is also helpful to explain the mechanisms of anti-oxidant,anti-inflammatory,anti-bacterial,anti-viral and anti-cancer effects from the molecular target and molecular network levels. 展开更多

AIM: To evaluate the effects of frying oil and Houttuynia cordata Thunb (H. cordata), a vegetable traditionally consumed in Taiwan, on the xenobiotic-metabolizing enzyme system of rodents. METHODS: Forty-eight Sprague-Dawley rats were fed with a diet containing 0%, 2% or 5% H. cordata powder and 15% fresh soybean oil or 24-h oxidized frying oil (OFO) for 28 d respectively. The level of microsomal protein, total cytochrome 450 content (CYP450) and enzyme activities including NADPH reductase, ethoxyresorufin 0-deethylase (EROD), pentoxyresorufin 0-dealkylase (PROD), aniline hydroxylase (ANH), aminopyrine demethylase (AMD), and quinone reductase (QR) were determined. QR represented phase Ⅱ enzymes, the rest of the enzymes tested represented phase Ⅰ enzymes. RESULTS: The oxidized frying oil feeding produced a significant increase in phase Ⅰ and Ⅱ enzyme systems, including the content of CYP450 and microsomal protein, and the activities of NADPH reductase, EROD, PROD, ANH, AMD and QR in rats (P<0.05). In addition, the activities of EROD, ANH and AMD decreased and QR increased after feeding with H. cordata in OFO-fed group (P<0.05). The feeding with 2% H. cordata diet showed the most significant effect. CONCLUSION: The OFO diet induces phases I and II enzyme activity, and the 2% H. cordata diet resulted in a better regulation of the xenobiotic-metabolizing enzyme system. 展开更多

OBJECTIVE: To study antimicrobial effect of Sodi- um houttuyfonate (SH) on Staphylococcus epider- midis (SE) and Candida albicans (CA). METHODS: The prepared strain broths (OD600=0.05) containing SE and CA were firstly used to test the minimal inhibitory concentrations (MICs) of SH, azithromycin (AZM) and fluconazole (FLU) by mi- cro-dilution method. Then the biofilms of SE and CA were matured in 96-well plates, and co-cultured with SH, AZM and FLU for 1, 2 and 3 days to assess the antibiofilm efficacies of the agents with differ- ent concentrations by crystal violet staining meth- od. At last, the treated biofilms of SE and CA by 2× MIC agents were observed by scanning electronic microscope. RESULTS: The MlCs of SE and CA were 256 and 1024 μg/mL, respectively. After the 1st, 2nd and3rd day of medications, the suppressions of biofilm were about 60% (P〈0.01), 76% (P=0.000) and 75% (P=0.000) by 2×MIC SH, the suppressions of biofilm were about 90% (P=0.000), 88% (P=0.000) and 90% (P=0.000) by 2×MIC SH, which could be testified by scanning electron microscope results. However, the inhibitions of biofilm attachment had no significant difference for SE by SH and azithromycin and CA by SH and fluconazole. CONCLUSION: SH had widely anti-pathogenic ef- fect on pathogenic biofilm formation of either bac- teria or fungus, had more influence on enclosed cells of SE and CA than the traditional antibiotics, revealing its target might be the extracellular poly- meric substances, and was more active to inhibit the growth of CA than SE. 展开更多

OBJECTIVE: To investigate the in vivo inhibitory effects of sodium houttuyfonate(SH) on symptom pattern of Qi-deficiency in rats induced by infection of bacterial biofilm on rat respiratory tract.METHODS: Symptom pattern is a term used in Traditional Chinese Medicine(TCM) to define a cluster of symptoms in a medical condition. Based on the pattern, TCM therapies are administered. The symptom pattern used in this study was lung-Qi deficiency pattern identified in rats, which was induced by nasal intubation drip of Pseudomonas aeruginosa(P. aeruginosa)(two strains) to form bacterial biofilm on airway combined with stimulation of cold and fatigue. We measured the variations of the symptoms of the pattern, weight, spleen and thymus index, blood gas, lung bronchial tissue pathology and cytokine of rat in different treatments and control groups.RESULTS: The rats of SH-treatment groups had not showed typical symptoms comparing with model group in the early stage of infection. The weight,spleen and thymus index of the SH-treatment groups were significantly higher comparing with untreated model group. The SH-treatment groups also showed higher O2 partial pressure and lower CO2 partial pressure than model group. Furthermore, we found that the bronchopulmonary section of SH-treatment groups not showed typical pathogenic variation in model group. The comparison of cytokine concentration in different groups indicated that SH could prevent the over-production of cytokine to reduce the inflammation occurrence.CONCLUSION: In the early stage of airway infection by biofilm of P. aeruginosa, application of SH can prevent the occurrence of lung-Qi deficiency pattern. 展开更多

OBJECTIVE:To assess the total and soluble oxalate contents of commonly used Chinese medicinal herbs.METHODS:Twenty-two Chinese medicinal herbs were extracted in both acid and water prior to determination of total and soluble oxalate,respectively.Oxalate was assayed in herbal extracts using a well-established enzymatic procedure.RESULTS:Among the 22 medicinal herbs,there was significant variation in oxalate content;Houttuynia cordata contained the highest amount of soluble oxalate(2146 mg/100 g) and Selaginella doederleinii contained the lowest amount(71 mg/100g).CONCLUSION:The results indicated that different Chinese medicinal herbs,even from the same family,contain significantly different amounts of oxalate.In susceptible individuals,the use of medicinal herbs with the highest oxalate contents could increase risk of kidney stone formation. 展开更多
