Thus, we attempted to find significant patterns in global HIV-...The epidemiology of HIV-1 varies in different areas of the world, and it is possible that this complexity may leave unique footprints in the viral genome. Thus, we attempted to find significant patterns in global HIV-1 genome sequences. By applying the rule inference algorithm RIPPER (Repeated Incremental Pruning to Produce Error Reduction) to multiple sequence alignments of Env sequences from four classes of compiled datasets, we generated four sets of signature patterns. We found that these patterns were able to distinguish southeastern Asian from non- southeastern Asian sequences with 97.5% accuracy, Chinese from non-Chinese sequences with 98.3% accuracy, African from non-African sequences with 88.4% accuracy, and southern African from non-southern African sequences with 91.2% accuracy. These patterns showed different associations with subtypes and with amino acid positions. In addition, some signature patterns were characteristic of the geographic area from which the sample was taken. Amino acid features corresponding to the phylogenetic clustering of HIV-1 sequences were consistent with some of the deduced patterns. Using a combination of patterns inferred from subtypes B, C, and all subtypes chimeric with CRF01_AE worldwide, we found that signature patterns of subtype C were extremely common in some sampled countries (for example, Zambia in southern Africa), which may hint at the origin of this HIV-1 subtype and the need to pay special attention to this area of Africa. Signature patterns of subtype B sequences were associated with different countries. Even more, there are distinct patterns at single position 21 with glycine, leucine and isoleucine corresponding to subtype C, B and all possible recombination forms chimeric with CRF01_AE, which also indicate distinct geographic features. Our method widens the scope of inference of signature from geographic, genetic, and genomic viewpoints. These findings may provide a valuable reference for epidemiological research or vaccine design.展开更多

Methods.Rat kidney transplantation was performed according to the procedure of Kamada with some modification.Experimental rats were divided into 5 groups.The survival time of recipient rats and function of grafts after renal transplantation were observed.The sections of renal graft were stained for monoclonal antibody ICAM 1 and LFA 1, and then quantification of ICAM 1 and LFA 1 expression was accomplished by computer image analysis. Results.ICAM 1 and LFA 1 increased significantly in the renal allograft rejection group as compared with the non rejection groups(P<0 05). Conclusion.Both biopsy of renal graft and monitoring of ICAM 1 and LFA 1 are useful tools in diagnosing and treating acute rejection.展开更多

It is unclear, however, whether Vpr-induced yeast ...Human immunodeficiency virus type 1 (HIV-1) Vpr induces cell death in mammalian and fssion yeast cells, suggesting that Vpr may affect a conserved cellular process. It is unclear, however, whether Vpr-induced yeast cell death mimics Vpr-mediated apoptosis in mammalian cells. We have recently identified a number of Vpr suppressors that not only suppress Vpr-induced cell death in fission yeast, but also block Vpr-induced apoptosis in mammalian cells. These findings suggest that Vpr-induced cell death in yeast may resemble some of the apoptotic processes of mammalian cells. The goal of this study was to develop and validate a fission yeast model system for future studies of apoptosis. Similar to Vpr-induced apoptosis in mammalian cells, we show here that Vpr in fission yeast promotes phosphatidylserine externalization and induces hyperpolarization of mitochondria, leading to changes of mitochondrial membrane potential. Moreover, Vpr triggers production of reactive oxygen species (ROS), indicating that the apoptotic-like cell death might be mediated by ROS. Interestingly, Vpr induces unique morphologic changes in mitochondria that may provide a simple marker for measuring the apoptotic-like process in fission yeast. To verify this possibility, we tested two Vpr suppressors (EF2 and Hspl6) that suppress Vpr-induced apoptosis in mammalian cells in addition to a newly identified Vpr suppressor (Skpl). All three proteins abolished cell death mediated by Vpr and restored normal mitochondrial morphology in the yeast cells. In conclusion, Vpr-induced cell death in fission yeast resembles the mammalian apoptotic process. Fission yeast may thus potentially be used as a simple model organism for the future study of the apoptotic-like process induced by Vpr and other proapoptotic agents.展开更多

Objective The effects of Shuang Dan Ming Mu capsule on expression of VEGF-a,VEGF-b,VEGF-c and the VEGF receptor,Flk-1,were examined in a diabetic retinopathy rats model.Methods Forty Sprague-Dawley(SD)rats were random...Objective The effects of Shuang Dan Ming Mu capsule on expression of VEGF-a,VEGF-b,VEGF-c and the VEGF receptor,Flk-1,were examined in a diabetic retinopathy rats model.Methods Forty Sprague-Dawley(SD)rats were randomized into Groups A(blank),B(model),C(Shuang Dan Ming Mu)and D(positive control)group,with each group containing10rats and20eyes.Rats from groups B,C and D were administered one dose of50mg/kg streptozotocin(STZ)by tail vein injection to establish a diabetic rat model.One week after model preparation,medication was continuously administered by gavage.After gavage for8weeks,the animals were sacrificed and retinal expression of VEGF-a,VEGF-b,VEGF-c and Flk-1was quantified by immunohistochemical analysis.Results At week8of drug administration after model preparation,the average protein expression grayscale values for VEGF-a,VEGF-b,VEGF-c and Flk-1in the rats model,Shuang Dan Ming Mu and positive control groups were all lower than those in the normal group,while the mean optical density values were higher than those in the normal group.When the model group was compared to the normal group,the difference was extremely significant(P<0.01).The mean grayscale values of VEGF-a,VEGF-b,VEGF-c and Flk-1in the Shuang Dan Ming Mu and positive control groups were all higher than those in the model group,while the mean optical density values were lower than those in the model group(P<0.05or0.01).Conclusion Shuang Dan Ming Mu capsule can significantly decrease the expression of VEGF-a,VEGF-b,VEGF-c and Flk-1in the retinas of diabetic model rats and exhibit some protective effects in their retinas.展开更多

The results showed that the planting method for fresh edible "faba bean/spring maize+soybean-autumn maize/autumn soybean" was much easier to operate with the input-output ratio of about185.6%, and its yield and economic benefits were 2.09 and 1.83 times of that of the planting mode for three crops a year, significantly improving the agricultural yield and income of farmers. In addition, the cropping index of the planting mode for fresh edible fresh edible "faba bean/spring maize +soybean-autumn maize/autumn soybean" reached up to 350%, and planting faba bean once a year and soybean twice a year could make the biological fixation amount of nitrogen increase 350-450kg/m^2, which equaled to up to 700 kg/m^2 of urea, showing significant ecological and social benefits. Based on the comparison results, the high-yield culture techniques of the planting mode of fresh edible "faba bean/spring maize+soybean-autumn maize/autumn soybean" were summarized.展开更多

To investigate the validity of two dynamic modulus predictive models( Witczak 1-37 A viscosity-based model and Witczak 1-40 D shear modulus-based model) in the context of Jiangsu, and evaluate the effect of differen...To investigate the validity of two dynamic modulus predictive models( Witczak 1-37 A viscosity-based model and Witczak 1-40 D shear modulus-based model) in the context of Jiangsu, and evaluate the effect of different mixture design variables( aggregate gradations, binder type, and volumetric properties) on dynamic modulus E*, asphalt mixtures commonly used in the local surface layer, including Sup-13 and AC-13, are prepared in the laboratory and their dynamic modulus E*values are predicted based on the above mentioned models. The corresponding asphalt tests, including viscosity and dynamic shear modulus tests, are also carried out to obtain the prediction model parameters. The test results showthat binder type and asphalt content have a significant impact on dynamic modulus.There is a good correlation between the E*values based on above two predictive models and the measured E*, while a relatively lower bias can be expected from Witczak 1-37 A model. The test results can be used for the calibration of dynamic modulus with higher accuracy.展开更多
AIM:To develop a prognostic approach for gastrointestinal stromal tumors(GISTs) using a cluster of indicators and follow-up information.METHODS:One hundred and four GISTs that had not been subjected to targeted therap...AIM:To develop a prognostic approach for gastrointestinal stromal tumors(GISTs) using a cluster of indicators and follow-up information.METHODS:One hundred and four GISTs that had not been subjected to targeted therapies were collected and classified by NIH risk assessment and anatomic location.By immunohistochemistry,the expressions of PTEN,Ki-67,CD44s matrix metalloproteinase(MMP)-9 and TIMP-1 were detected on tissue microarray.Univariate and multimarker survival analyses were performed and then a COX hazard proportion model was constructed to evaluate a cluster of predictors of GIST.RESULTS:Our data showed small intestinal GIST are more aggressive than gastric GIST.The NIH risk assessment correlated with disease-free survival foreither gastric GIST or small intestinal GIST.Immunohistochemical analysis revealed that Ki-67 labeling indexes(LIs) < 5% predicted higher disease-specific survival(DSS) in gastric and small intestinal GIST.CD44s positivity and PTEN LIs ≥ 50% correlated with higher DSS in gastric GIST.MMP-9 and TIMP-1 had no correlation with survival.Multimarker analysis revealed that the expression pattern of PTEN LIs ≥ 50% combined with Ki-67 LIs < 5% and CD44s positivity reliably predicted favorable outcomes for gastric GIST(P = 0.009),as did the combination of PTEN LIs ≥ 50% and Ki-67 LIs < 5% for small intestinal GIST(P = 0.011).Authors also found that high NIH risk grade was correlated with DSS in patients with gastric GIST and disease-free survival in patients with small intestinal GIST.CONCLUSION:PTEN LIs ≥ 50%,Ki-67 LIs < 5% and CD44s positivity provides an accurate,favorable prognosis for gastric GIST.PTEN LIs ≥ 50% and Ki-67 LIs < 5% does the same for small intestinal GIST.Ki-67 LIs enhances the NIH assessment.展开更多

These queue models consist of mathematical formulas and relationships that can be used to determine the operating characteristics (performance measures) for a waiting line. Schematic and transition diagrams of different single server queue models were shown. Relationships between birth-death processes, waiting lines (queues) and transition diagrams were given. While M/M/I/K queue model states was limited by K customers and had (K+I) states, M/M/1/1 queue model had only two states. M/G/1/∝/∝ and M/M/1/∝/∝ shared similar characteristics. Resource provisioning is a critically important problem for AAL applications in cloud data centers(CDCs). This paper focuses on modeling and analysis of multi-tier AAL applications, and aims to optimize resource provisioning while meeting requests' response time constraint. This paper models a multi-tier AAL application as a hybrid multi-tier queueing model consisting of an M/M/c queueing model and multiple M/M/1 queueing models. Then, virtual machine(VM) allocation is formulated as a constrained optimization problem in a CDC, and is further solved with the proposed heuristic VM allocation algorithm(HVMA). The results demonstrate that the proposed model and algorithm can effectively achieve dynamic resource provisioning while meeting the performance constraint.展开更多

The chosen analogues were 4-(a...Dummy molecularly imprinted polymers (DMIPs) for 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) were produced using three structural analogues as dummy template molecules. The chosen analogues were 4-(acetymethylamino)-1-(3-pyridyl)-butanol, 4- (methylamino)-1-(3-pyridyl)-1-butanol, and 1-(3-pyridyl)-1,4,-butanediol. The molecular recognition characteristics of the produced polymers were evaluated by X-ray photoelec- tron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FT-IR). Interactions between NNAL and methacrylic acid should be cooperative hydrogen bonds while the ni- trogen atom of the pyridine ring and the oxygen atom of the nitroso group in NNAL are two of the hydrogen-bond acceptors. It was further demonstrated that DMIP synthesized by 4-(acetymethylamino)-1-(3-pyridyl)-butanol had the best binding performance by XPS and FT-IR. Then dummy molecularly imprinted solid phase extraction (DMISPE) was developed for the determination of the analyte using the hit polymer as the sorbing material. Under optimal conditions, the recovery of NNAL dissolved in standard solution reached 93%. And the investigated polymer exhibited much higher binding of NNAL when nicotine was acted as the competitive molecule. Also the proposed method was applied to the measurement of NNAL spiked in blank urine samples with recoveries ranging from 87.2% to 101.2%.展开更多

Model 1 determines the searching area. We predict the trajectory of floats generated after the disintegration of the plane by using RBF neural network model, and then determine the searching area according to the trajectory. With the pass of time, the searching area will also be constantly moving along the trajectory. Model 2 develops a maritime search plan to achieve the purpose of completing the search in the shortest time. We optimize the searching time and transform the problem into the 0-1 knapsack problem. Solving this problem by improved genetic algorithm, we can get the shortest searching time and the best choice for the search power.展开更多

The highest T/NT reached 7.1. Some new results about the recursive expression of the transient queue size distribution at any epoch n+ and the recursive formulae of the equilibrium distribution are obtained. Furthermore, the recursive formulae of the equilibrium queue size distribution at epoch n-, and n are obtained, too. The important relations between stationary queue size distributions at different epochs are discovered (being different from the relations given in M/G/I queueing system). 文摘Objective The effects of Shuang Dan Ming Mu capsule on expression of VEGF-a,VEGF-b,VEGF-c and the VEGF receptor,Flk-1,were examined in a diabetic retinopathy rats model.Methods Forty Sprague-Dawley(SD)rats were randomized into Groups A(blank),B(model),C(Shuang Dan Ming Mu)and D(positive control)group,with each group containing10rats and20eyes.Rats from groups B,C and D were administered one dose of50mg/kg streptozotocin(STZ)by tail vein injection to establish a diabetic rat model.One week after model preparation,medication was continuously administered by gavage.After gavage for8weeks,the animals were sacrificed and retinal expression of VEGF-a,VEGF-b,VEGF-c and Flk-1was quantified by immunohistochemical analysis.Results At week8of drug administration after model preparation,the average protein expression grayscale values for VEGF-a,VEGF-b,VEGF-c and Flk-1in the rats model,Shuang Dan Ming Mu and positive control groups were all lower than those in the normal group,while the mean optical density values were higher than those in the normal group.When the model group was compared to the normal group,the difference was extremely significant(P<0.01).The mean grayscale values of VEGF-a,VEGF-b,VEGF-c and Flk-1in the Shuang Dan Ming Mu and positive control groups were all higher than those in the model group,while the mean optical density values were lower than those in the model group(P<0.05or0.01).Conclusion Shuang Dan Ming Mu capsule can significantly decrease the expression of VEGF-a,VEGF-b,VEGF-c and Flk-1in the retinas of diabetic model rats and exhibit some protective effects in their retinas.

文摘The high-efficiency planting mode for five crops a year of fresh edible "faba bean/spring maize+soybean-autumn maize/autumn soybean" was introduced, and its yield and economic benefits were compared with the planting mode of three crops a year of "faba bean-spring maize/red bean". The results showed that the planting method for fresh edible "faba bean/spring maize+soybean-autumn maize/autumn soybean" was much easier to operate with the input-output ratio of about185.6%, and its yield and economic benefits were 2.09 and 1.83 times of that of the planting mode for three crops a year, significantly improving the agricultural yield and income of farmers. In addition, the cropping index of the planting mode for fresh edible fresh edible "faba bean/spring maize +soybean-autumn maize/autumn soybean" reached up to 350%, and planting faba bean once a year and soybean twice a year could make the biological fixation amount of nitrogen increase 350-450kg/m^2, which equaled to up to 700 kg/m^2 of urea, showing significant ecological and social benefits. Based on the comparison results, the high-yield culture techniques of the planting mode of fresh edible "faba bean/spring maize+soybean-autumn maize/autumn soybean" were summarized.

文摘A new limited memory symmetric rank one algorithm is proposed. It combines a modified self-scaled symmetric rank one (SSR1) update with the limited memory and nonmonotone line search technique. In this algorithm, the descent search direction is generated by inverse limited memory SSR1 update, thus simplifying the computation. Numerical comparison of the algorithm and the famous limited memory BFGS algorithm is given. Comparison results indicate that the new algorithm can process a kind of large-scale unconstrained optimization problems.

文摘To investigate the validity of two dynamic modulus predictive models( Witczak 1-37 A viscosity-based model and Witczak 1-40 D shear modulus-based model) in the context of Jiangsu, and evaluate the effect of different mixture design variables( aggregate gradations, binder type, and volumetric properties) on dynamic modulus E*, asphalt mixtures commonly used in the local surface layer, including Sup-13 and AC-13, are prepared in the laboratory and their dynamic modulus E*values are predicted based on the above mentioned models. The corresponding asphalt tests, including viscosity and dynamic shear modulus tests, are also carried out to obtain the prediction model parameters. The test results showthat binder type and asphalt content have a significant impact on dynamic modulus.There is a good correlation between the E*values based on above two predictive models and the measured E*, while a relatively lower bias can be expected from Witczak 1-37 A model. The test results can be used for the calibration of dynamic modulus with higher accuracy.

文摘AIM:To develop a prognostic approach for gastrointestinal stromal tumors(GISTs) using a cluster of indicators and follow-up information.METHODS:One hundred and four GISTs that had not been subjected to targeted therapies were collected and classified by NIH risk assessment and anatomic location.By immunohistochemistry,the expressions of PTEN,Ki-67,CD44s matrix metalloproteinase(MMP)-9 and TIMP-1 were detected on tissue microarray.Univariate and multimarker survival analyses were performed and then a COX hazard proportion model was constructed to evaluate a cluster of predictors of GIST.RESULTS:Our data showed small intestinal GIST are more aggressive than gastric GIST.The NIH risk assessment correlated with disease-free survival foreither gastric GIST or small intestinal GIST.Immunohistochemical analysis revealed that Ki-67 labeling indexes(LIs) < 5% predicted higher disease-specific survival(DSS) in gastric and small intestinal GIST.CD44s positivity and PTEN LIs ≥ 50% correlated with higher DSS in gastric GIST.MMP-9 and TIMP-1 had no correlation with survival.Multimarker analysis revealed that the expression pattern of PTEN LIs ≥ 50% combined with Ki-67 LIs < 5% and CD44s positivity reliably predicted favorable outcomes for gastric GIST(P = 0.009),as did the combination of PTEN LIs ≥ 50% and Ki-67 LIs < 5% for small intestinal GIST(P = 0.011).Authors also found that high NIH risk grade was correlated with DSS in patients with gastric GIST and disease-free survival in patients with small intestinal GIST.CONCLUSION:PTEN LIs ≥ 50%,Ki-67 LIs < 5% and CD44s positivity provides an accurate,favorable prognosis for gastric GIST.PTEN LIs ≥ 50% and Ki-67 LIs < 5% does the same for small intestinal GIST.Ki-67 LIs enhances the NIH assessment.
文摘A transition diagram is used to describe the behavior of systems. Birth-death equations were derived from transition diagram depicting the state of the birth-death processes. Queue models and characteristics of queue models are also derivable from birth-death processes. These queue models consist of mathematical formulas and relationships that can be used to determine the operating characteristics (performance measures) for a waiting line. Schematic and transition diagrams of different single server queue models were shown. Relationships between birth-death processes, waiting lines (queues) and transition diagrams were given. While M/M/I/K queue model states was limited by K customers and had (K+I) states, M/M/1/1 queue model had only two states. M/G/1/∝/∝ and M/M/1/∝/∝ shared similar characteristics. Many ideal queuing situations employ M/M/1 queueing model.
文摘Cloud computing provides the essential infrastructure for multi-tier Ambient Assisted Living(AAL) applications that facilitate people's lives. Resource provisioning is a critically important problem for AAL applications in cloud data centers(CDCs). This paper focuses on modeling and analysis of multi-tier AAL applications, and aims to optimize resource provisioning while meeting requests' response time constraint. This paper models a multi-tier AAL application as a hybrid multi-tier queueing model consisting of an M/M/c queueing model and multiple M/M/1 queueing models. Then, virtual machine(VM) allocation is formulated as a constrained optimization problem in a CDC, and is further solved with the proposed heuristic VM allocation algorithm(HVMA). The results demonstrate that the proposed model and algorithm can effectively achieve dynamic resource provisioning while meeting the performance constraint.
文摘Dummy molecularly imprinted polymers (DMIPs) for 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL) were produced using three structural analogues as dummy template molecules. The chosen analogues were 4-(acetymethylamino)-1-(3-pyridyl)-butanol, 4- (methylamino)-1-(3-pyridyl)-1-butanol, and 1-(3-pyridyl)-1,4,-butanediol. The molecular recognition characteristics of the produced polymers were evaluated by X-ray photoelec- tron spectroscopy (XPS) and Fourier transform infrared spectroscopy (FT-IR). Interactions between NNAL and methacrylic acid should be cooperative hydrogen bonds while the ni- trogen atom of the pyridine ring and the oxygen atom of the nitroso group in NNAL are two of the hydrogen-bond acceptors. It was further demonstrated that DMIP synthesized by 4-(acetymethylamino)-1-(3-pyridyl)-butanol had the best binding performance by XPS and FT-IR. Then dummy molecularly imprinted solid phase extraction (DMISPE) was developed for the determination of the analyte using the hit polymer as the sorbing material. Under optimal conditions, the recovery of NNAL dissolved in standard solution reached 93%. And the investigated polymer exhibited much higher binding of NNAL when nicotine was acted as the competitive molecule. Also the proposed method was applied to the measurement of NNAL spiked in blank urine samples with recoveries ranging from 87.2% to 101.2%.
文摘In this paper, we construct two models for the searching task for a lost plane. Model 1 determines the searching area. We predict the trajectory of floats generated after the disintegration of the plane by using RBF neural network model, and then determine the searching area according to the trajectory. With the pass of time, the searching area will also be constantly moving along the trajectory. Model 2 develops a maritime search plan to achieve the purpose of completing the search in the shortest time. We optimize the searching time and transform the problem into the 0-1 knapsack problem. Solving this problem by improved genetic algorithm, we can get the shortest searching time and the best choice for the search power.
文摘In this study, the human breast cancer-bearing nude mice model has been established and the radioim-munoimaging was carried out by using human anti-human monoclonal antibodies. The results showed that the breast tumor-bearing nude mice received 131I-McAb-CM-l had a clear image of the xenograft during 4 to 6 days after injection and at the same time T/NT all over 1. 0. The highest T/NT reached 7.1. It demonstrates that McAb-CM-1 can specially combine with breast cancer tissues and hopefully it could be used clinically to improve accurate rate of the early breast cancer diagnosis.

文摘This paper considers the Geom/G/1 queueing model with feedback according to a late arrival system with delayed access (LASDA). Using recursive method, this paper studies the transient property of the queue size from the initial state N(0+) = i. Some new results about the recursive expression of the transient queue size distribution at any epoch n+ and the recursive formulae of the equilibrium distribution are obtained. Furthermore, the recursive formulae of the equilibrium queue size distribution at epoch n-, and n are obtained, too. The important relations between stationary queue size distributions at different epochs are discovered (being different from the relations given in M/G/I queueing system). The model discussed in this paper can be widely applied in all kinds of communications and computer network.
