通过2-甲基-5-氨基喹啉和取代水杨醛缩合反应,合成了10个含喹啉基片段的水杨醛席夫碱类化合物。通过1HNMR、13CNMR、IR、LC-MS和元素分析技术确证了产物的结构,并测定了目标化合物在不同浓度时淬灭1,1-二苯基-2-三硝基苯肼(DPPH)自由基... 通过2-甲基-5-氨基喹啉和取代水杨醛缩合反应,合成了10个含喹啉基片段的水杨醛席夫碱类化合物。通过1HNMR、13CNMR、IR、LC-MS和元素分析技术确证了产物的结构,并测定了目标化合物在不同浓度时淬灭1,1-二苯基-2-三硝基苯肼(DPPH)自由基活性。结果表明,所有化合物在测试浓度0.02~0.10 g/L时都表现出一定的淬灭DPPH自由基活性,清除率主要集中在19%~35%之间,但是4-溴-2-甲氧基-6-{[(2-甲基-5-喹啉基)亚氨基]甲基}苯酚在浓度0.06 g/L时表现出特别优异的活性,清除率高达43.1%。目标化合物质量浓度的变化对活性的影响不明显或者没有明显的变化规律。 展开更多

The three water-soluble polysaccharides (PI, PII and Pill), the ethyl acetate extraction (EA) and its five fractions of petroleum ether (PeF), ethyl acetate (EaF), butanol (BuF), methanol (MeF) and water (WtF) from marine agrobacterium 1202 were prepared and then subjected to antioxidant activity evaluation. The antioxidant activity was tested using the 1, 1-Diphenyl-2-picryl-hydrazyl (DPPH) free radical-scavenging assay, the hydrogen peroxide-induced luminot chemiluminescence (CL) assay and hydroxyl free radical-initiated chemiluminescence (CL) assay. All the fractions exerted significant inhibitory effects on hydrogen peroxide and hydroxyl free radical. The extracts of EA, EaF and BuF could also showed inhibition on DPPH. Among of them, the extracts of EaF showed full of antioxidant activity on the three tested system. The result suggested that the marine agrobacterium 1202 is a potential source of natural antioxidantagent. 展开更多

目的研究普洱茶中茶氨酸的含量及其抗氧化活性。方法采用高效液相色谱方法对普洱茶中茶氨酸含量进行测定,分析条件为:Aglient TC-C18(4.6 mm×250 mm,5μm);流动相:(A)0.1%磷酸水溶液(B)甲醇;流速:1.000 0 m L/min;进样体... 目的研究普洱茶中茶氨酸的含量及其抗氧化活性。方法采用高效液相色谱方法对普洱茶中茶氨酸含量进行测定,分析条件为:Aglient TC-C18(4.6 mm×250 mm,5μm);流动相:(A)0.1%磷酸水溶液(B)甲醇;流速:1.000 0 m L/min;进样体积:10μL;柱温:25℃;检测波长:203 nm。对分析条件进行优化和验证,构建了茶氨酸含量精确分析方法,进而通过清除1,1-二苯基-2-苦基肼基自由基(DPPH)法来测定其抗氧化活性。结果:30个普洱茶氨酸含量范围为7.350 1~14.235 3 mg/g,平均值为9.638 3 mg/g,抗氧化性在29.06%~72.28%之间。结论高效液相色谱法能够准确测定普洱茶中茶氨酸含量,对普洱茶抗氧化活性研究有利于对普洱茶保健疗效的进一步鉴定和提高。 展开更多

【目的】在一次对虫生真菌代谢物进行大规模的清除自由基活性物质筛选中,发现一种被毛孢(Hirsutella sp.)菌株RCEF0881发酵液中存在有较强的清除自由基活性物质。本研究目的是初步搞清这些活性成分的具体组成,并制备出一定量的纯品用于... 【目的】在一次对虫生真菌代谢物进行大规模的清除自由基活性物质筛选中,发现一种被毛孢(Hirsutella sp.)菌株RCEF0881发酵液中存在有较强的清除自由基活性物质。本研究目的是初步搞清这些活性成分的具体组成,并制备出一定量的纯品用于进一步的结构鉴定。【方法】用有机溶剂法提取活性成分;用二苯基苦基苯肼自由基(DPPH)酶标仪法和薄层色谱法进行活性测定;用高分辨液质联用方法进行活性成分初步分析和鉴定;用反相制备色谱法制备活性组分。【结果】提取实验结果表明具清除自由基活性的物质能较好地被乙酸乙酯提取出来;液相色谱-质谱-活性测定分析表明提取物中活性组分的可能分子式分别为C7H6O4、C8H8O3和C12H14N2O。结合色谱特性、紫外光谱特征、质谱碎片和数据库查询可初步推断它们分别为二羟基苯甲酸、羟基甲基苯甲酸和生物碱类物质,但具体结构还有待于进一步确认。从高效液相色谱和质谱离子流的峰面积可知上述3种活性物质中C12H14N2O的含量最高。本研究成功地用反相制备色谱制备出该天然活性组分的纯品。该3种清除自由基活性物质都是首次发现存在于虫生真菌的代谢物中。 展开更多

探讨了压力、温度和时间对厚朴超临界流体萃取工艺的影响,确定了最佳萃取工艺条件是:萃取压力25 MPa、萃取温度40℃、萃取时间3 h。在该工艺条件的基础上加入10%乙醇作夹带剂,不仅大大提高了萃取率,而且大大缩短了萃取时间。用不同极性... 探讨了压力、温度和时间对厚朴超临界流体萃取工艺的影响,确定了最佳萃取工艺条件是:萃取压力25 MPa、萃取温度40℃、萃取时间3 h。在该工艺条件的基础上加入10%乙醇作夹带剂,不仅大大提高了萃取率,而且大大缩短了萃取时间。用不同极性溶剂索氏抽提与超临界流体萃取相比较,并用OSI法和DPPH自由基清除法对其提取物的抗氧化活性进行了测定。结果发现,OSI法和DPPH自由基清除法实验结果一致,其抗氧化活性的顺序是:BHT>乙酸乙酯提取物>氯仿提取物>超临界CO2萃取物(加入10%乙醇)>石油醚提取物>超临界CO2萃取物(未加夹带剂)>乙醇提取物。厚朴中的抗氧化成分主要是小极性物质,主要存在于石油醚、氯仿和超临界CO2的提取物中。 展开更多

Five hydroxystilbene derivatives were synthesized and their structures were determined using high-resolution mass spectrometry, ^1H NMB, and IR. In this study, the antioxidant activities of eight hydroxystilbenes (five synthesized hydroxystilbene derivatives and three other compounds) were evaluated with the free radiced scavenging model 1,1-diphenyl-2-picrylhydraxyl. It was observed that there exists a very significant linear rehtionship between the reciprocal of the EC50 value of each of the compounds and the maximum wavelength of the ultraviolet absorption peaks, It is believed that this result will be helpful for the further synthesis of such antioxidants. 展开更多

Objective: Anabasis aretioides (Coss & Moq.), a Saharan plant belonging to Chenopodiaceae family, is widely distributed in semi-desert areas from the Tafilalet region of Morocco. This plant is extensively used by local population against diabetes and cardiovascular disorders. The purpose of the study was to investigate the effect of the aqueous A. aretioides extract on lipid metabolism in normal and strepto-zotocin (STZ)-induced diabetic rats and to identify the polyphenolic compounds present. In addition, the in vitro antioxidant activity of the aqueous A. aretioides extract was also evaluated. Methods: The effect of an aerial part aqueous extract (APAE) ofA. aretioides (5 mg/kg of lyophilized A. are-tioides APAE) on plasma lipid profile was investigated in normal and STZ-induced diabetic rats (n = 6) after once daily oral administration for 15 days. The aqueous extract was tested for its 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. Polyphenolic compounds in the extracts were defini-tively characterized by high-performance liquid chromatography-diode array detection-electrospray ionization-mass spectrometry. Results: In diabetic rats, oral administration ofA. aretioides APAE provoked a significant decrease in both plasma cholesterol and triglyceride levels from the first to the second week (P 〈 0.01). A significant decrease on plasma triglyceride levels was also observed in normal rats (P 〈 0.01), where the reduction was 53%. In addition, the phytochemical analysis revealed the presence of 12 polyphenolic compounds. Moreover, according to the DPPH radical-scavenging activity, the aqueous extract showed an in vitro antioxidant activity. Conclusion: Aqueous A. aretioides APAE exhibits lipid-lowering and in vitro antioxidant activities. Many polyphenols were present in this extract and these phytoconstituents may be involved in the pharmaco-logical activity of this plant. 展开更多

Turmeric is a spice widely used to enhance the taste and color of certain meats. We investigated the antioxidant capacities of turmeric ethanol soaking extracts by utilizing the scavenging properties of hydroxyl radical and 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radical. The results showed that the optimum extraction parameters were: extraction time 4 h, sample-solvent ratio 1:84.12, solvent 72.34% ethanol solution, and water bath temperature 81.3 ℃. The optimum ethanol soaking extraction parameters were: extraction time 4 h, sample-solvent ratio 1:90.74, solvent 80.46% ethanol solution, water bath temperature 88.2 ℃. With these parameters, the hydroxyl radical scavenging rate and the DPPH free radical clearance of crude extracts from turmeric can reach about 93.78% and 40.69%, respectively. These results indicate that ethanol soaking extraction may be a useful method for extracting antioxidants from turmeric. 展开更多

Objective:To analyze the methanol extract of Trigonella foenum-graecum(T.foenum-graecum)for antioxidant,phytotoxic and cytotoxic activity.Methods:The powder of T.foenum-graecum was extracted in diluted methanol with the help of random shaking method.All extracts of the plant were measured for cytotoxic activity(beside brine shrimp and antioxidant activity vs.1,1-diphenyl-2-picrylhydrazyl free radical).Results:Various concentrations of methanolic extract of T.foenum-graecum were observed as 36.16%to 54.12%with rising concentrations of 50 to 1000μg/mL.Significantly phytotoxic activity(100 and 1000μg/mL)reduced the growth of roots(radicals)and shoots(hypocotyls)of rice when compared to control after 3 and 7 days’treatment.At a concentration of 10μg/mL,the survival rate of cytotoxic activity of brine shrimp was maximum and at a concentration of 250μg/mL,the death rate of brine shrimp was maximum.Conclusions:T.foenum-graecum has potential activity against free radical mediated sickness and thus it is possible to treat cancer. 展开更多

Objective:To quantify total phenols,flavonoids and to investigate the in vitro antioxidant power of Haplophyllum tuberculatum(H.tuberculatum)leaves extracts of varying polarities.Methods:The authenticated sample of H.tuberculatum(50 g)leaves was dried under shade,powdered and extracted exhaustively with ethanol by cold percolation method.The alcoholic extract was further partitioned into petroleum ether,acetone,chloroform and methanol to obtain the fractions of varying polarities which were subjected to qualitative phytochemical testing.Total phenolics and flavonoids content in the acetone,chloroform and methanol extracts were quantified by using standard colorimetric methods.Petroleum ether extract was omitted because it did not show the presence of either tannins or flavonoids.In vitro antioxidant activity and total antioxidant capacity were determined by using 1,1 diphenyl-2-picrylhydrazyl radical and phosphomolybdenum reagent.Ascorbic acid was used as a reference antioxidant for comparison purpose.Results:Qualitative phytochemical results of leaves extracts confirmed the presence of major secondary plant metabolites.The extraction of phenolic compounds varied considerably according to the polarity of solvent.The most polar fractions i.e.methanol were observed to have the highest phenolic content(561.22 mg/g of gallic acid equivalent)and flavonoids(165.54 mg/g of quercetin equivalent).Although the free radical scavenging activity of leaves fractions was noted to be slightly lower than the reference compound,a direct relationship was observed between phenolic content and in vitro antioxidant activity.On the other hand,leaves fractions exhibited significant total antioxidant capacity as ascorbic acid equivalent.Conclusions:The aerial part of H.tuberculatum is rich in phenolic compounds which might play a vital role in the discovery of natural antioxidants. 展开更多
