Application of sulfuric acid immobilized on silica gel as an efficient and benign catalyst has been explored in the syn-thesis of 2,4,5-Triaryl-1H-imidazoles via condensation reaction of benzil or benzoin, aldehyde and ammonium acetate. The key advantages of this process are high yields, cost effectiveness of catalyst, easy work-up, purification of products by non-chromatographic method and the reusability of the H2SO4.SiO2 catalyst. 展开更多

以1-肼羰亚甲基-2-三氟甲基苯并[d]咪唑(1)为原料,与芳基异硫氰酸酯在无水乙醇中反应得酰氨基硫脲2a~2d,继而在氢氧化钠水溶液中合环得4,5-二氢-3-(2-三氟甲基苯并[d]咪唑-1-亚甲基)-4-芳基-1,2,4-三唑-5-硫酮(3a~3d),然后分别采用超... 以1-肼羰亚甲基-2-三氟甲基苯并[d]咪唑(1)为原料,与芳基异硫氰酸酯在无水乙醇中反应得酰氨基硫脲2a~2d,继而在氢氧化钠水溶液中合环得4,5-二氢-3-(2-三氟甲基苯并[d]咪唑-1-亚甲基)-4-芳基-1,2,4-三唑-5-硫酮(3a~3d),然后分别采用超声辐射法和常规加热法与四种胺反应合成了16个未见报道的Mannich碱4a~4d,5a~5d,6a~6d和7a~7d.与常规加热法对比,超声辐射法具有操作简单,反应时间短,条件温和,产率高,副反应少等优点,为此类化合物的合成提供了一种有效的新方法.目标化合物的结构经元素分析,IR和1H NMR确证. 展开更多

The present work describes a facile,one-pot three component synthesis of a series of 3-[(4,5-diphenyl-2-substituted aryl/heteryl)-1H-imidazol-1-yl]-1H-1,2,4-triazole-5-carboxylic acid derivatives M(1-15).Benzil,aromatic aldehydes and 3-amino-l,2,4-triazole-5-carboxylic acid was refluxed in ethanol using cerric ammonium nitrate(CAN) as a catalyst to give the title compounds in good yields.The compounds were evaluated for their in vitro antifungal and antibacterial activity.Compounds M1,M9,and M15 were found to be equipotent against Candida albicans when compared with fluconazole.Compounds M2.M5,and M14 showed higher activity against Streptococcus pneumoniae.Escherichia coli and Streptococcus pyogenes,respectively,compared with ampicillin.Docking study of the newly synthesized compounds was performed,and the results showed good binding mode in the active sites of C albicans enzyme cytochrome P450 lanosterol 14α-demethylase.The results of in vitro antifungal activity and docking study showed that synthesized compounds had potential antifungal activity and can be further optimized and developed as a lead compound. 展开更多

Compounds 1 and 2 are one-dimensional triple-stranded chain. Compounds 3a and 3b are polymorphous and construct one-dimensional single chain. The conformational analysis is performed. Phosphorus pentoxide supported on silica gel (P2O5/SiO2) has been used as an efficient and reusable catalyst for the one-pot pseudo four-component synthesis of 2,4,5-trisubstituted imidazoles from benzil or benzoin, aldehydes, and ammonium acetate. It was also used for four-component preparation of 1,2,4,5-tetrasubstituted imidazoles from benzil or benzoin, aldehydes, primary amine, and ammonium acetate under thermal solvent-free conditions. The remarkable features of this new procedure are high conversions, cleaner reaction, simple experimental and work-up procedures and also the catalyst can be easily separated from the reaction mixture and reused several times without any loss of its activity. 展开更多
