AIM:To evaluate effects of endogenous dopamine induced by low concentration atropine eye drops on choroidal neovascularization(CNV)in high myopia mice.METHODS:The C57BL/6J mice were deprived of the right eye for 4wk,and the high myopia was diagnosed by optometry,the diopter was less than-6.00 D,and CNV was induced by 532 nm laser.The changes of dopamine D1 receptor(DRD1),dopamine D2 receptor(DRD2),and vascular endothelial growth factor A(VEGFA)were detected by Western blot technology at 0.5,1,2h,and 7d after 0.01%,0.05%,and 0.1%atropine eye drops,respectively,the area of CNV was measured.RESULTS:Significant increases were observed on the expression of DRD2 in mouse high myopia model at 0.5,1,2h,7d with 0.05%and 0.1%atropine eye drops(P<0.05).Significant decreases were observed on the expression of DRD1 and VEGFA in mouse high myopia model at 0.5,1,2h,7d with 0.05%and 0.1%atropine eye drops(P<0.05).The area of CNV induced by laser in the drug-treated group was significantly smaller than that in the control group,and the higher the concentration,the more significant the inhibitory effect(P<0.05).CONCLUSION:The 0.01%,0.05%,0.1%atropine eye drops can decrease the level of VEGFA and inhibit high myopia CNV indirectly by up-regulating the level of DRD2 and down-regulating the level of DRD1,and the effect of 0.05%and 0.1%atropine eye drops is more significant. 展开更多

目的研究1∶25000阿托品眼药水对树脂透镜诱导型中度近视豚鼠屈光度、眼轴长度、视网膜形态学及视网膜细胞凋亡的影响。方法按照体重将2周龄雄性豚鼠随机分为3组:正常组,模型组,实验组,每组10只。豚鼠给予-20D透镜连续固定在右眼建立离... 目的研究1∶25000阿托品眼药水对树脂透镜诱导型中度近视豚鼠屈光度、眼轴长度、视网膜形态学及视网膜细胞凋亡的影响。方法按照体重将2周龄雄性豚鼠随机分为3组:正常组,模型组,实验组,每组10只。豚鼠给予-20D透镜连续固定在右眼建立离焦性近视模型。造模成功当天,实验组给予1∶25000阿托品眼药水点眼,每5 d停药1 d,连续给药1个月。检查3组豚鼠的眼轴和屈光度;用缺口末端标记法检测细胞凋亡变化。结果正常组、模型组和实验组的右眼屈光度分别为1. 53±0. 52,-4. 00±0. 57和-3. 93±0. 49,右眼眼轴分别为10. 56±0. 03,14. 02±0. 24和13. 96±0. 57,模型组与正常组对比,这2项指标的差异均有统计学意义(均P <0. 05);实验组与模型组比较,这2项指标的差异均有统计学意义(均P <0. 05)。模型组和实验组的视网膜细胞凋亡率分别为(11. 82±0. 12)%和(7. 43±0. 08)%,实验组明显下降,差异有统计学意义(P <0. 05)。结论 1∶25000阿托品眼药水通过对视网膜细胞凋亡的修复,降低凋亡率,对视网膜功能有较好的保护作用。 展开更多
