BACKGROUND Leukemia stem cells(LSCs)are found to be one of the main factors contributing to poor therapeutic effects in acute myeloid leukemia(AML),as they are protected by the bone marrow microenvironment(BMM)against...BACKGROUND Leukemia stem cells(LSCs)are found to be one of the main factors contributing to poor therapeutic effects in acute myeloid leukemia(AML),as they are protected by the bone marrow microenvironment(BMM)against conventional therapies.Gossypol acetic acid(GAA),which is extracted from the seeds of cotton plants,exerts anti-tumor roles in several types of cancer and has been reported to induce apoptosis of LSCs by inhibiting Bcl2.AIM To investigate the exact roles of GAA in regulating LSCs under different microenvironments and the exact mechanism.METHODS In this study,LSCs were magnetically sorted from AML cell lines and the CD34+CD38-population was obtained.The expression of leucine-rich pentatricopeptide repeat-containing protein(LRPPRC)and forkhead box M1(FOXM1)was evaluated in LSCs,and the effects of GAA on malignancies and mitochondrial RESULTS LRPPRC was found to be upregulated,and GAA inhibited cell proliferation by degrading LRPPRC.GAA induced LRPPRC degradation and inhibited the activation of interleukin 6(IL-6)/janus kinase(JAK)1/signal transducer and activator of transcription(STAT)3 signaling,enhancing chemosensitivity in LSCs against conventional chemotherapies,including L-Asparaginase,Dexamethasone,and cytarabine.GAA was also found to downregulate FOXM1 indirectly by regulating LRPPRC.Furthermore,GAA induced reactive oxygen species accumulation,disturbed mitochondrial homeostasis,and caused mitochondrial dysfunction.By inhibiting IL-6/JAK1/STAT3 signaling via degrading LRPPRC,GAA resulted in the elimination of LSCs.Meanwhile,GAA induced oxidative stress and subsequent cell damage by causing mitochondrial damage.CONCLUSION Taken together,the results indicate that GAA might overcome the BMM protective effect and be considered as a novel and effective combination therapy for AML.展开更多
Objective:To investigate the effect of isoimperatorin on histopathological and biochemical changes in acetic acid-induced colitis rats.Methods:Colitis was induced by intracolonic administration of acetic acid solution...Objective:To investigate the effect of isoimperatorin on histopathological and biochemical changes in acetic acid-induced colitis rats.Methods:Colitis was induced by intracolonic administration of acetic acid solution(4%v/v)in rats.Rats were divided into six groups including the sham group,the negative control group,the dexamethasone-treated group,and the groups treated with isoimperatorin(0.1,1,and 10 mg/kg/d by gavage).The treatments were administered for three days and then colonic status was assessed by macroscopic,histopathological,and biochemical analyses.Results:Isoimperatorin significantly alleviated colonic damage in a dose-dependent manner and improved histological changes in rats with acetic acid-induced colitis.It also significantly reduced myeloperoxidase,TNF-α,IL-1β,and malodialdehyde levels.Conclusions:Isoimperatorin alleviates acetic acid-induced colitis in rats and may be a potential therapeutic agent for the treatment of colitis.展开更多

The results show that HFAA, La(FAA) 3 and Eu(FAA) 3 with the concentrations of 1 0×10 -5 ~1 0×10 -2 μg·ml -1 inhibit the colony formation of leukemia cells(L 1210 ) and the growth of transplanted tumor sarcoma 180(S 180 ), hepatic carcinoma(HEPA) and ehrlich ascites tumor(EC) as well. The maximum inhibitory rate of Eu(FAA) 3 for S 180 is 38 4%, that HFAA and La(FAA) 3 for EC are 22 4% and 43 4%, respectively. The life prolongation rate of Eu(FAA) 3 for HEPA bearing mice is as long as 284%.展开更多

Its crystal structure was d...A new energetic compound based on the tetrazole-1-acetic acid (tza) and potassium(I) salt, K2(tza)2(H2O), was synthesized and characterized by elemental analysis and FT-IR spectrum. Its crystal structure was determined by single-crystal X-ray diffraction analysis. The results show that the crystal belongs to the orthorhombic system, space group Pna21 with a = 1.11972(17) nm, b = 0.46647(7) nm, c = 2.5158(4) nm, V = 1.3140(3) nm3, K2C6H8N8O5, Mr = 350.40 g·mol-1, Dc = 1.771 g·cm^-3, μ(MoKα) = 0.759 mm^-1, F(000) = 712, Z = 4, R = 0.023 and wR = 0.0527 for 2961 observed reflections (I 〉 2σ(I)). The K(I) cation is six-coordinated with four O atoms from three carboxylate groups, one O atom from one H2O molecule and one N atom from tetrazolyl ring, in which each tza is coordinated in a tridentate chelating bridging coordination mode. The thermal decomposition mechanism of the title complex was studied by DSC and TG-DTG techniques. Under nitrogen atmosphere at a heating rate of 10 K·min-1, the thermal decomposition of the complex contains one main exothermic process between 191.7 and 243.8 ℃ in the DSC curve. Its combustion heat was mensurated by oxygen bomb calorimetry. The non-isothermal kinetics parameters were calculated by the Kissinger's method and Ozawa-Doyle's method, respectively. The sensitivity properties of K2(tza)2(H2O) were also determined with standard methods, which was very sensitive to flame.展开更多

It is known that the consumption of bacteria such as lactobacilli and bifidobacteria has beneficial effects on human immune function. Most of them are Gram-positive bacteria, and there are few reports on Gram-negative...It is known that the consumption of bacteria such as lactobacilli and bifidobacteria has beneficial effects on human immune function. Most of them are Gram-positive bacteria, and there are few reports on Gram-negative bacteria. In this study, we evaluated the effects of intake of Gluconacetobacter hansenii GK-1 (GK-1), Gram-negative acetic acid bacteria, for 12 weeks on physical condition and immune indices. We conducted a randomized, double-blind, placebo-controlled, parallel-group study in 100 healthy adults. The subjects were randomized into the GK-1 and the placebo groups. The diary-administered physical condition survey was conducted during the study period. The evaluation of salivary sIgA levels, NK-cell activity, and serum IFN-γ levels and quality of life survey was conducted before, in 6 weeks, and 12 weeks after the start of ingestion. Based on the physical condition survey, the cumulative onset-days of symptoms were significantly suppressed in the GK-1 group compared to the placebo group regarding the evaluation of 13 symptoms related to immunity, every 3 weeks. Additionally, salivary sIgA levels per hour were significantly increased in the GK-1 group compared with the placebo group at 6 and 12 weeks. Despite no significant differences in the NK-cell activity, serum IFN-γ levels or quality of life survey between the groups. Serum IFN-γ levels in the GK-1 group were significantly elevated at 12 weeks after the start of ingestion compared with those before ingestion. In conclusion, intake of GK-1 was shown to increase salivary sIgA levels and improve physical condition. This suggested that oral intake of GK-1 may help maintain the immune system.展开更多

Perovskite oxide catalysts with the composition LaMoxV1-xOn (x = 0.1, 0.3, 0.5, 0.7, and 0.9) have been synthesized by the sol-gel method and then used in the ethane dry reforming reaction for the direct synthesis of acetic acid. The influence of the nature of the metallic source (metal, nitrate, acetylacetonate, and ammonium) on gel formation has been studied by Fourier-transform infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA-DTA). After calcination, the obtained perovskites were characterized by X-ray diffraction (XRD) and energy-dispersive X-ray spectrometry (EDS) coupled with scanning electron microscopy (SEM). The catalysts were then subjected to thermo-programmed reduction (TPR). The surface area (BET) was found to increase from 2.6 m^2/g (x = 0.1) to 5.1 m2/g (x = 1.0) with increasing molybdenum content following calcinations at 750 °C, and pure LaMoxV1-xOn perovskite was obtained with good homogeneity. The catalysts have been characterized by XRD, SEM, EDS, and carbon analysis (CA). The results indicate that through this synthesis it is possible to obtain highly crystalline, homogeneous and pure solids, with well-defined structures. The direct synthesis of acetic acid from ethane over the perovskite catalysts was studied at temperatures between 450 and 850 °C and elevated pressures between 1 and 8 bar. It was found that the yield of acetic acid and the selectivity of its formation could be increased by incorporating more molybdenum into the perovskite structure. The experimental studies have shown that the calcination temperature and the molybdenum content have a significant influence on the catalytic activity. Amongst the catalysts tested, LaMo0.7V0.3O4.2 exhibited the best activity and stability.展开更多

METHODS: Chronic gastric lesions were induced in male Sprague-Dawley (S...AIM: To assess the synergistic action of famotidine (FMD) and chlorpheniramine (CPA) on acetic acid-induced chronic gastric ulcer in rats. METHODS: Chronic gastric lesions were induced in male Sprague-Dawley (SD) rats by serosal application of the acetic acid. Forty SD rats were randomly divided into blank group (n = 8), control group (n = 8), FMD group (n = 8), CPA group (n = 8), and FMD+CPA group (n = 8). Each group was given intraperitoneally (i.p.) 0.5 mL/100 g distilled water, 9 g/L NaCl saline, 4 mg/kg FMD, 10 mg/kg CPA, 4 mg/kg FMD+10 mg/kg CPA, respectively, daily for 10 d, On d 10, ulcer area was determined by planimetry, The level of myeloperoxidase (MPO) in the liver homogenation was determined by biochemical methods and the plasma levels of 6-ketoprostaglandin F1 alpha (6-keto-PGFla)and IL-8 were determined by radioimmunoassay. RESULTS: The synergistic effects of FMD+CPA group on the lesion, IL-8, 6-keto-PGFla and MPO were confirmed. The effect of FMD+CPA group was significantly different as compared to the control and FMD groups. The lesion (mm2) was reduced from 40.18±2.6 in control group to 6.83±2.97 in PMD+CPA group, P〈0.01, and from 32.9 ±3.27 in FMD group to 6.83±2.97 in PMD+CPA group, P〈0.01. The plasma levels of IL-8 decreased from 0.69± 0.11 ng/L in control group to 0.4±0.04 ng/L in PMD+CPA group, P〈0.01, and from 0.51±0.08 ng/L in FMD group to 0.4±0.04 ng/L in PMD+CPA group, P〈0.05. The level of 6-keto-PGF1, increased from 7.55±1.65 ng/L in control group to 16.62±0.97 ng/L in PMD+CPA group,P〈0.01, and from 13.15±1.48 ng/L in FMD group to 16.62±0.97 ng/L in PMD+CPA group,P〈0.05. The levels of MPO in the liver homogenate decreased from 9.12±2.05 u/Lin control group to 4.33±0.95 u/L in PMD+CPA group, P〈0.01, and from 8.3±1.29 u/L in FMD group to 4.33±0.95 u/L, P〈0.01. CONCLUSION: The synergistic action of FMD and CPA on acetic acid-induced chronic gastric ulcer in rats decreases the incidence of ulcer and also enhances the healing of ulcer.展开更多

In this study, two important auxins were selected as model analytes, namely indole-3- butyric acid (IBA) and 1-naphthylacetic acid (NAA). They could be extracted and concentrated due to their π-π stacking interactions with MWCNT. Then HPLC-UV was introduced to detect IBA and NAA after sample pretreatment. Factors that may affect the enrichment efficiency were investigated and optimized. Comparative studies showed that MWCNT was superior to CI 8 for the extraction of the two analytes. Validation experiments showed that the optimized method had good linearity (0.9998 and 0.9960), high recovery (81.4%-85.4%), and low detection limits (0.0030 mg/L and 0.0012 mg/L). The results indicated that the novel method had advantages of convenience, good sensitivity, high efficiency, and it was feasible for the determination of auxins in plant samples.展开更多

The direct synthesis of acetic acid from ethane over the perovskite catalysts was studied at temperatures between 450 and 850 °C and elevated pressures between 1 and 8 bar. It was found that the yield of acetic acid and the selectivity of its formation could be increased by incorporating more molybdenum into the perovskite structure. The experimental studies have shown that the calcination temperature and the molybdenum content have a significant influence on the catalytic activity. Amongst the catalysts tested, LaMo0.7V0.3O4.2 exhibited the best activity and stability.

文摘AIM: To assess the synergistic action of famotidine (FMD) and chlorpheniramine (CPA) on acetic acid-induced chronic gastric ulcer in rats. METHODS: Chronic gastric lesions were induced in male Sprague-Dawley (SD) rats by serosal application of the acetic acid. Forty SD rats were randomly divided into blank group (n = 8), control group (n = 8), FMD group (n = 8), CPA group (n = 8), and FMD+CPA group (n = 8). Each group was given intraperitoneally (i.p.) 0.5 mL/100 g distilled water, 9 g/L NaCl saline, 4 mg/kg FMD, 10 mg/kg CPA, 4 mg/kg FMD+10 mg/kg CPA, respectively, daily for 10 d, On d 10, ulcer area was determined by planimetry, The level of myeloperoxidase (MPO) in the liver homogenation was determined by biochemical methods and the plasma levels of 6-ketoprostaglandin F1 alpha (6-keto-PGFla)and IL-8 were determined by radioimmunoassay. RESULTS: The synergistic effects of FMD+CPA group on the lesion, IL-8, 6-keto-PGFla and MPO were confirmed. The effect of FMD+CPA group was significantly different as compared to the control and FMD groups. The lesion (mm2) was reduced from 40.18±2.6 in control group to 6.83±2.97 in PMD+CPA group, P〈0.01, and from 32.9 ±3.27 in FMD group to 6.83±2.97 in PMD+CPA group, P〈0.01. The plasma levels of IL-8 decreased from 0.69± 0.11 ng/L in control group to 0.4±0.04 ng/L in PMD+CPA group, P〈0.01, and from 0.51±0.08 ng/L in FMD group to 0.4±0.04 ng/L in PMD+CPA group, P〈0.05. The level of 6-keto-PGF1, increased from 7.55±1.65 ng/L in control group to 16.62±0.97 ng/L in PMD+CPA group,P〈0.01, and from 13.15±1.48 ng/L in FMD group to 16.62±0.97 ng/L in PMD+CPA group,P〈0.05. The levels of MPO in the liver homogenate decreased from 9.12±2.05 u/Lin control group to 4.33±0.95 u/L in PMD+CPA group, P〈0.01, and from 8.3±1.29 u/L in FMD group to 4.33±0.95 u/L, P〈0.01. CONCLUSION: The synergistic action of FMD and CPA on acetic acid-induced chronic gastric ulcer in rats decreases the incidence of ulcer and also enhances the healing of ulcer.

文摘In this work, a new sample pretreatment method prior to HPLC separations was developed for the determination of auxins in plant samples, Owing to its large surface area and high adsorption capacity, multi-walled carbon nanotube (MWCNT) was chosen as the adsorbent for the extraction of auxins from plant samples. In this study, two important auxins were selected as model analytes, namely indole-3- butyric acid (IBA) and 1-naphthylacetic acid (NAA). They could be extracted and concentrated due to their π-π stacking interactions with MWCNT. Then HPLC-UV was introduced to detect IBA and NAA after sample pretreatment. Factors that may affect the enrichment efficiency were investigated and optimized. Comparative studies showed that MWCNT was superior to CI 8 for the extraction of the two analytes. Validation experiments showed that the optimized method had good linearity (0.9998 and 0.9960), high recovery (81.4%-85.4%), and low detection limits (0.0030 mg/L and 0.0012 mg/L). The results indicated that the novel method had advantages of convenience, good sensitivity, high efficiency, and it was feasible for the determination of auxins in plant samples.
