Testosterone is closely associated with lipid metabolism and known to affect body fat composition and muscle mass in males.However,the mechanisms by which testosterone acts on lipid metabolism are not yet fully unders...Testosterone is closely associated with lipid metabolism and known to affect body fat composition and muscle mass in males.However,the mechanisms by which testosterone acts on lipid metabolism are not yet fully understood,especially in teleosts.In this study,cyp17a1-/-zebrafish(Danio rerio)exhibited excessive visceral adipose tissue(VAT),lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis(DNL)enzymes.The assay for transposase accessible chromatinwithsequencing(ATAC-seq)results demonstrated that chromatin accessibility of DNL genes was increased in cyp17a1-/-fish compared to cyp17a1+/+male fish,including stearoyl-CoA desaturase(scd)and fatty acid synthase(fasn).Androgen response element(ARE)motifs in the androgen signaling pathway were significantly enriched in cyp17a1+/+male fish but not in cyp17a1-/-fish.Both androgen receptor(ar)-/-and wildtype(WT)zebrafish administered with Ar antagonist flutamide displayed excessive visceral adipose tissue,lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis enzymes.The Ar agonist BMS-564929 reduced the content of VAT and lipid content,and down-regulated acetyl-CoA carboxylase a(acaca),fasn,and scd expression.Mechanistically,the rescue effect of testosterone on cyp17a1-/-fish in terms of phenotypes was abolished when ar was additionally depleted.Collectively,these findings reveal that testosterone inhibits lipid deposition by down-regulating DNL genes via Ar in zebrafish,thus expanding our understanding of the relationship between testosterone and lipid metabolism in teleosts.展开更多

In this study, we examined the effects of E2 treatment on the dynamics of E2 and T during gonadal differentiation in the olive flounder Paralichtbys olivaceus using histology and radioimmunoassay (RIA). Flounder larvae were divided into five groups (G0~G4), and fed with 0 (control), 0.2, 2, 20 and 100 mg E2/kg feed from 35 to 110 day post hatching (dph). Fish growth in the G1 and G2 groups was not significantly different from that of the control group (P〉0.05), while fish in the G3 and G4 groups were less active and showed growth depression and high mortality. The gonads of fish in the G3 and G4 groups were smaller and surrounded by hyperplastic connective tissue. The frequency of females in the G0-G4 groups was 54.5%, 75.0%, 100%, 100% and 93.3%, respectively. The RIA analyses of E2 and T showed that T levels decreased during gonadal differentiation, and increased slightly at the onset of ovarian differentiation, while E2 levels increased gradually and peaked at the onset of ovarian differentiation in the control group. In the E2-treated groups, T levels decreased before the onset of ovarian differentiation. E2 levels were high on the 48 dph, but declined to a lower level on the 54 dph, and then increased gradually during gonadal differentiation. And a sharp increase of E2 levels were observed in all E2-treated groups at the onset of ovarian differentiation. The data suggest that T and E2 play important roles during gonadal differentiation, and an E2 dose of 2 mg/kg feed could induce sex reversal in P olivaceus.展开更多
This study was carried out in 2022 at the Itato agriculture community estate of fish hatchery in eastern Senegal with the aim of finding an alternative method to 17-α-methyl testosterone (MT) by using Garcinia kola ...This study was carried out in 2022 at the Itato agriculture community estate of fish hatchery in eastern Senegal with the aim of finding an alternative method to 17-α-methyl testosterone (MT) by using Garcinia kola “Bitter kola” powder to obtain Oreochromis niloticus male sex populations. 1800 fry of average size between 0.01 and 0.02 g were distributed in 12 aboveground tanks of 1 m3 each at a rate of 150 individuals/tank, following 6 duplicate treatments. The negative (B1 and B2) and positive (B3 and B4) control diets, to which between 0 and 60 mg/kg of 17alpha-methyl testosterone had been added respectively, were mixed with industrial feed containing 40% protein;the tank diets (B5 and B6), (B7 and B8), (B9 and B10) and (B11 and B12) contained 10 g, 20 g, 30 g and 40 g of Garcinia kola powder respectively for 1 kg of industrial feed con-taining 40% protein. These different treatments resulted in high proportions of males in the order of 97.03%;98.09% for the hormone and proportions of 94.03%;93.26% for 20 g of Garcinia kola extracts. In terms of weight growth, the best average weights for the hormone and Garcinia kola were 0.896 g and 0.980 g respectively. With regard to fish survival, the following results were recorded: 95.53% and 94.56% respectively for additions of 20 g and 30 g of Garcinia kola extracts to the feed supplied to the fish. On the other hand, a significant difference in the mortality rate of 35% and 5% respectively for the conventional hormone and 20 g of Garcinia kola extracts was observed. To conclude the study, it is recommended that further studies be carried out to determine the quality of the flesh, their life stages and the profile of the amino acids present in their proteins.展开更多

In the influents, estrone (E 1), androsterone (A), androstenedione (AD), BPA (bisphenol A), NP (nonylphenol) and daidzein (DZ) were detected in high amounts with subsequent 24 h-average concentrations of 350, 1000, 29, 1300, 3900, and 5700 ng/L in K-WWTP and of 310, 620, 59, 1600, 2600, and 8400 ng/L in S-WWTP. The estrogenic (androgenic) activity as 17β-estradiol (E2) equivalents (EEQ) or testosterone (Te) equivalents (TEQ) was consequently 620 ng E2/L (570 ng Te/L) and 580 ng E2/L (800 ng Te/L) for the two WWTPs. The removal efficiencies of the above mentioned sole target chemicals were 51%-100% for K-WWTP and 55.6%-100% for S-WWTP. The removal efficiencies of EEQ were about 73% for both WWTPs, while the removal efficiencies of TEQ were 62.1% for K-WWTP and 98.4% for S-WWTP. In addition, chemical-derived EEQ were about 1.2%-52.4% of those by ER-binding assay for K-WWTP and the corresponding ratios were 1.3%-83.3% for S-WWTP, while chemical derived TEQ were less than 3% of values measured by the AR-binding assay for both WWTPs.展开更多

Isolated rat Leydig ceils were treated with annexin A5. Testosterone production was detected by chemiluminescence assay. The protein and mRNA of Steroidogenic acute regulatory (STAR), P450scc, 3β-hydroxysteroid dehydrogenase (3β-HSD), 17β-hydroxysteroid dehydrogenase (17β-HSD), and 17β-hydroxylase were examined by Western blotting and semi-quantitative RT-PCR, respectively. Annexin A5 significantly stimulated testosterone secretion from rat Leydig cells in dose- and time-dependent manners and increased mRNA and protein expression of STAR, P450scc, 3β-HSD, and 17β-HSD but not 17α-hydroxylase. Annexin A5 knockdown by siRNA significantly decreased the level of testosterone and protein expression of P450scc, 3β-HSD, and 17β-HSD. The significant activation of ERK1/2 signaling was observed at 5, 10, and 30 min after annexin A5 treatment. After the pretreatment of Leydig cells with ERK inhibitor PD98059 (50 μmol l^-1) for 20 min, the effects of annexin A5 on promoting testosterone secretion and increasing the expression of P450scc, 3β-HSD, and 17β-HSD were completely abrogated (P 〈 0.05). Thus, ERK1/2 signaling is involved in the roles of annexin A5 in mediating testosterone production and the expression of P450scc, 3β-HSD, and 17β-HSD in Leydig cells.展开更多
Objective:To investigate the effects of a single oxytocin injection on plasma steroid concentrations,testicular blood flow measures(resistive and pulsatility indexes),and testicular echogenicity in Baladi goats.Method...Objective:To investigate the effects of a single oxytocin injection on plasma steroid concentrations,testicular blood flow measures(resistive and pulsatility indexes),and testicular echogenicity in Baladi goats.Methods:Twelve Baladi goat bucks were randomly allocated into 2 groups and received an intravenous injection of either 0.7 IU/kg oxytocin or normal saline 0.9%.Venous blood samples were collected,and testicular blood flow Doppler parameters(i.e.,peak systolic velocity,end-diastolic velocity,time average maximum velocity,resistive index,and pulsatility index)were assessed for supratesticular arteries in the left and right testes immediately before(0),and at 5,30,60,and 120 min after injection.Results:Plasma concentrations of testosterone significantly decreased in the oxytocin group at 60 min post-treatment compared to the control group,whereas plasma concentrations of estradiol-17βwere not affected(P>0.05).Both resistive index and pulsatility index declined in the oxytocin group at 60 min post-treatment compared to the control group(P<0.05).Testicular pixel intensity increased at 30 min post-treatment in the oxytocin group compared to the control group(P<0.05).Conclusions:Oxytocin treatment significantly increases testicular blood flow and decreases plasma testosterone concentrations in male Baladi goats.展开更多

In this article we will review how we have explored the evidence of different sex hormones on blood vessels since 1996. The results showed that low-dose HRT significantly protected cardiovascular system in postmenopausal women.However, the high-dose 17β-estradiol (E2), with or without progesterone (P) or testosterone (T), or any hormone alone, as their concentrations increase, may even inhibit the protective effects of low-dose HRT on blood vessels.展开更多
