A new image thresholding method is introduced, which is based on 2-D histgram and minimizing the measures of fuzziness of an input image. A new definition of fuzzy membership function is proposed, it denotes the characteristic relationship between the gray level of each pixel and the average value of its neighborhood. When the threshold is not located at the obvious and deep valley of the histgram, genetic algorithm is devoted to the problem of selecting the appropriate threshold value. Globozoospermia is a severe form of teratozoospermia characterized by round-headed spermatozoa with an absent acrosome, an aberrant nuclear membrane and midpiece defects. Globozoospermia is diagnosed by the presence of 100% round-headed spermatozoa on semen analysis, and patients with this condition are absolutely infertile. The objective of this study was to investigate the differences in protein expression between human round- headed and normal spermatozoa. Two-dimensional (2-D) fluorescence difference gel electrophoresis (DIGE) coupled with mass spectrometry (MS) was used in this study. Over 61 protein spots were analysed in each paired normal/round-headed comparison, using DIGE technology along with an internal standard. In total, 35 protein spots identified by tandem mass spectrometry (MS/MS) exhibited significant changes (paired t-test, P 〈 0.05) in the expression level between normal and round-headed spermatozoa. A total of nine proteins were found to be upregulated and 26 proteins were found to be downregulated in round-headed spermatozoa compared with normal spermatozoa. The differentially expressed proteins that we identified may have important roles in a variety of cellular processes and structures, including spermatogenesis, cell skeleton, metabolism and spermatozoa motility. 展开更多

Eddy-current (EC) testing is an effective electromagnetic non-destructive testing (NDT) technique.Planar eddy-current sensor arrays have several advantages such as good coherence,fast response speed,and high sensitivity,which can be used for micro-damage inspection of crucial parts in mechanical equipments and aerospace aviation.The main purpose of this research is to detect the defect in a metallic material surface and identify the length of a crack using planar eddy-current sensor arrays in different directions.The principle and characteristics of planar eddy-current sensor arrays are introduced,and a crack length quantification algorithm in different directions is investigated.A damage quantitative detection system is established based on a field programmable gate array and ARM processor.The system is utilized to inspect the micro defect in a metallic material,which is carved to micro crack with size of 7mm(length)×0.1mm(width)×1mm(depth).The experimental data show that the sensor arrays can be used for the length measurement repeatedly,and that the uncertainty of the length measurement is below ±0.2mm. 展开更多

Improvement of resolution in 2-D gel analysis by simple pre-treatment of human urine with EDTA

Regardless of thermodynamic stability, less complex proteome than plasma, convenient and noninvasive sampling of urine, reproducibility and comparability of the data among laboratories are challenges for the urine proteome analysis. Pretreatment of urine with 10 mM EDTA in conjunction with desalting methods to remove interfering molecules significantly improved the presentation of urine proteome on 2-D gels. Statistical analysis of the average ratio of the spot numbers on 2-D gels between EDTA-treated and non-treated samples were 1.35 ± 0.167, 1.26 ± 0.091, and 1.24 ± 0.095 for dialysis, ultrafiltration, and desalting column, respectively, with the p-value of 5.85 × 10–6 for the overall comparison. This result came from the fact that EDTA-treated samples showed better resolutions with less streaks and clearer spots on 2-D gels than the control samples. The results suggest that a simple and non-fractionating EDTA-treatment can be employed to the procedures for the high resolution 2-D gel analysis and other urinary proteomic analysis. 展开更多

In order to fight against this aging sign, we develope... Facial sagging is a major aging change that affects the overall shape of the face. Sagging can negatively impact self-perception and consequently quality of life. In order to fight against this aging sign, we developed a combination of three active ingredients: Tetrahydroxypropyl Ethylenediamine (THPE), Ononis Spinosa extract and Glycerin. In a clinical study, we assessed the efficacy of this association on facial sagging and wrinkles, with a special focus on immediate and lasting effects. Thirty-nine women applied the tested product daily during an eight-week treatment period. Clinical grading by experts and a new, easy to operate, 2-D imaging method were performed at different time points. The 2-D imaging method measured the upper eyelid lifting effect. The results showed an immediate and significant improvement in sagging and wrinkle parameters;these benefits were still observed seven hours after the first application. Results also showed a significant long-term improvement. The lifting effect calculated from the 2-D pictures was 1.08 mm immediately after application;the same method showed a 1.80 mm lifting effect following the eight-week treatment period. These results suggest that a skin care product combining THPE, Ononis Spinosa and Glycerin properties may be used to reduce sagging as well as wrinkles with both immediate and long term benefits. 展开更多

Aim: To extend the analysis of the proteome of human spermatozoa and establish a 2-D gel electrophoresis (2-DE) reference map of human spermatozoal proteins in a pH range of 3.5-9.0. Methods: In order to reveal more protein spots, immobilized pH gradient strips (24 cm) of broad range of pH 3-10 and the narrower range of pH 6-9, as well as different overlapping narrow range pH immobilized pH gradient (IPG) strips, including 3.5-4.5, 4.0-5.0, 4.5-5.5, 5.0-6.0 and 5.5-6.7, were used. After 2-DE, several visually identical spots between the different pH range 2-D gel pairs were cut from the gels and confirmed by mass spectrometry and used as landmarks for computer analysis. Results: The 2-D reference map with pH value from 3.5 to 9.0 was synthesized by using the ImageMaster analysis software. The overlapping spots were excluded, so that every spot was counted only once. A total of 3 872 different protein spots were identified from the reference map, an approximately 3-fold increase compared to the broad range pH 3-10 IPG strip (1 306 spots). Conclusion: The present 2-D pattern is a high resolution 2-D reference map for human fertile spermatozoal protein spots. A comprehensive knowledge of the protein composition of human spermatozoa is very meaningful in studying dysregulation of male fertility. 展开更多

The image segmentation difficulties of small objects which are much smaller than their background often occur in target detection and recognition. The existing threshold segmentation methods almost fail under the circumstances. Thus, a threshold selection method is proposed on the basis of area difference between background and object and intra-class variance. The threshold selection formulae based on one-dimensional (1-D) histogram, two-dimensional (2-D) histogram vertical segmentation and 2-D histogram oblique segmentation are given. A fast recursive algorithm of threshold selection in 2-D histogram oblique segmentation is derived. The segmented images and processing time of the proposed method are given in experiments. It is compared with some fast algorithms, such as Otsu, maximum entropy and Fisher threshold selection methods. The experimental results show that the proposed method can effectively segment the small object images and has better anti-noise property. 展开更多

采用以肿瘤免疫学与蛋白质组学(proteomics)研究技术有机地结合为基础的血清蛋白质组学研究体系(serologicproteomeanalysis ,SERPA)筛选肺癌分子标志物.对10例人肺鳞癌组织,应用双向凝胶电泳(two dimensionalelectrophoresis ,2 DE)... 采用以肿瘤免疫学与蛋白质组学(proteomics)研究技术有机地结合为基础的血清蛋白质组学研究体系(serologicproteomeanalysis ,SERPA)筛选肺癌分子标志物.对10例人肺鳞癌组织,应用双向凝胶电泳(two dimensionalelectrophoresis ,2 DE)技术对同一肺鳞癌组织的细胞总蛋白同时进行电泳后获得3张相同的凝胶,其中一块2 DE凝胶经银染显色作为平行胶,其余两块2 DE凝胶经电转膜将凝胶中的蛋白质转至硝酸纤维素(NC)膜上,然后分别与肺癌患者的自身血清以及正常对照血清进行Western印迹分析,获取Western印迹反应图谱.经计算机图像分析识别差异反应的蛋白质,然后与平行胶比较找出相应的差异反应蛋白质点.获得了分辨率较高的人肺鳞癌组织与患者的自身血清以及正常对照血清的Western印迹反应图谱;图像分析共识别36±8个差异反应的蛋白质;在平行胶上找到了匹配的差异反应蛋白质点.对2 0个差异蛋白质点进行了肽质指纹图分析,鉴定出14个与细胞生长增殖、细胞代谢、细胞周期调控、信号转导等有关的肺鳞癌相关抗原.通过血清蛋白质组技术对肺鳞癌组织进行的研究,建立了分辨率较高的人肺鳞癌组织与患者的自身血清以及正常血清的Western印迹反应图谱,成功鉴定14个肺鳞癌相关抗原,为进一步筛选用于肺鳞癌诊断。 展开更多

Since the logarithmic form of Shannon entropy has the drawback of undefined value at zero points,and most existing threshold selection methods only depend on the probability information,ignoring the within-class uniformity of gray level,a method of reciprocal gray entropy threshold selection is proposed based on two-dimensional(2-D)histogram region oblique division and artificial bee colony(ABC)optimization.Firstly,the definition of reciprocal gray entropy is introduced.Then on the basis of one-dimensional(1-D)method,2-D threshold selection criterion function based on reciprocal gray entropy with histogram oblique division is derived.To accelerate the progress of searching the optimal threshold,the recently proposed ABC optimization algorithm is adopted.The proposed method not only avoids the undefined value points in Shannon entropy,but also achieves high accuracy and anti-noise performance due to reasonable 2-D histogram region division and the consideration of within-class uniformity of gray level.A large number of experimental results show that,compared with the maximum Shannon entropy method with 2-D histogram oblique division and the reciprocal entropy method with 2-D histogram oblique division based on niche chaotic mutation particle swarm optimization(NCPSO),the proposed method can achieve better segmentation results and can satisfy the requirement of real-time processing. 展开更多

It is integrated from a CsI(Na) crystal pixel array, an optical fiber array, an image intensifier and an ICCD camera. The 2-D positions of one cosmic ray track is determined by the location of a fired CsI(Na) pixel. The scintillation light of these 1.0× 1.0 mm CsI(Na) pixels is delivered to the image intensifier through fibers. The light information is recorded in the ICCD camera in the form of images, from which the 2-D positions can be reconstructed. The background noise and cosmic ray images have been studied. The study shows that the cosmic ray detection efficiency can reach up to 11.4%, while the false accept rate is less than 1%. 展开更多

The 2-D nature of the algorithm provides it an advantage of circumventing the roundabout transforming procedures between two dimensional (2-D) image deta and one-dimensional (l-D) signal. Moreover the combination of the Newton (fixed-point algorithm) and natural gradient algorithms in this composite algorithm increases its efficiency and robustness. The convincing results of a successful example in functional magnetic resonance imaging (fMRI) show the potential application of composite 2-D ICA in the brain activity detection. 展开更多

Hemodynamics is thought to be an important factor in the pathogenesis and treatment of aneurysms. To date, the bulk of investigations into hemodynamics have been conducted by making use of mathematically idealized models for rigid aneurysms and associated arteries. However the walls of aneurysms and associated arteries are elastic in vivo. This study shows the differences of the simulation between elastic and rigid wall models. The numerical simulation of elastic aneurysm model is made fi'om a representative Digital Subtraction Angiography (DSA) image and calculated with CFD software to get the wall deformation and the velocity field. Then the results are analyzed. By comparing the simulation results of the two models from their velocity vectors and shear stress distribution, many differences can be noted. The main difference exists in the distribution of velocity magnitude at some sections, with one outlet having obviously off-center distribution for the elastic wall model. The currents of the distribution of wall shear stress along the wall of aneurysm simulated in rigid and elastic wall models were similar. But there were apparent differences between the two models on the values of wall shear stress especially at the neck of aneurysm. The off-center distribution of velocity magnitude affects the distribution of wall shear stress and the exchange of substance through the wall. The analysis demonstrated clearly that the results of 2-D elastic numerical simulation were in good agreement with the clinical and pathological practice. The results of this study play an important role in the formation, growth, rupture and prognosis of an aneurysm on clinic application. 展开更多
