A 24-membered ring macrolide compound,macrolactin A has potential applications in pharmaceuticals for its anti-infectious and antiviral activity.In this study,macrolactin A was produced by a marine bacterium,which was identified as Bacillus subtilis by 16S ribosomal RNA(rRNA) sequence analysis.Electrospray ionization mass spectrometry(ESI/MS) and nuclear magnetic resonance(NMR) spectroscopy analyses were used to characterize this compound.To improve the production,response surface methodology(RSM) involving Box-Behnken design(BBD) was employed.Faeces bombycis,the main by-product in sericulture,was used as a nitrogen source in fermentation.The interactions between three significant factors,F.bombycis,soluble starch,and(NH4)2SO4 were investigated.A quadratic model was constructed to fit the production and the factors.Optimum medium composition was obtained by analysis of the model.When cultivated in the optimum medium,the production of macrolactin A was increased to 851 mg/L,2.7 times as compared to the original.This study is also useful to find another way in utilizing F.bombycis. 展开更多

【目的】研究前体调控对Bacillus sp. 108菌株发酵合成抑制甘蔗鞭黑粉菌活性物质24元大环内酯产量的影响,为Bacillus sp.108菌株发酵工艺规模化放大及甘蔗黑穗病的防治提供新途径。【方法】利用单因素优化方法,考察乙酸钠、丙酸钠、正... 【目的】研究前体调控对Bacillus sp. 108菌株发酵合成抑制甘蔗鞭黑粉菌活性物质24元大环内酯产量的影响,为Bacillus sp.108菌株发酵工艺规模化放大及甘蔗黑穗病的防治提供新途径。【方法】利用单因素优化方法,考察乙酸钠、丙酸钠、正丙醇、异亮氨酸和缬氨酸对Bacillus sp. 108菌株发酵合成24元大环内酯的作用,确定有效前体的最佳添加浓度和添加时间;采用平板对峙法测定Bacillus sp. 108菌株发酵产物24元大环内酯对甘蔗鞭黑粉菌的抑制作用。【结果】乙酸钠是Bacillus sp. 108菌株发酵合成24元大环内酯的最佳前体,在发酵第96 h时添加1.0%乙酸钠,24元大环内酯的产量达104.45μg/mL,比未添加前体发酵的对照组提高58.74%。Bacillus sp. 108菌株发酵粗提物对甘蔗鞭黑粉菌菌丝生长具有较强的抑制作用,半数有效浓度(EC_(50))0为157.94μg/mL。【结论】乙酸钠是Bacillus sp.108菌株发酵合成24元大环内酯较适宜的前体物质,24元大环内酯对甘蔗鞭黑粉菌菌丝生长具有明显的抑制作用,具有开发成微生物农药的潜力。 展开更多

One new indoloditerpene,6,7-dehydropaxilline(1),one new indole-diketopiperazine,spirotryprostatin F(2),and one new 13-membered macrolide,N-demethylmelearoride A(3),as well as 8 known alkaloids(4-11),were isolated from the solid cultures of the endophytic fungus,Penicillium brefeidianum.The structures and absolute configurations of compounds 1-3 were elucidated by comprehensive spectroscopic analysis and the circular dichroism(CD) exciton chirality method.All the metablites were evaluated for their cytotoxic activites against three human tumor cell lines.Compound 2 exhibited cytotoxicities against HepG2 and MDA-MB-231 cells with IC(50) values of 14.1 μmol/L and 35.5μmol/L,respectively.Compound 3 showed moderate activity against HepG2 cells with IC(50) values of 36.6 μmol/L. 展开更多
