A new phloretin derivative 1 3-[2-(4-hydroxy-phenyl)-ethyl]-benzo[d] isoxazole-4,6-diol (yield 63%) was synthesized from phloretin by carbonyl nucleophilic addition condensation reaction. Its structure was characterized by 1H NMR, 13C NMR and HR-MS. The phloretin, compound 1, resveratrol and acetylated resveratrol were determined by comparing them with paclitaxel. Anti-tumor activity of alcohol on SPC-A1, EC109, A549, MCF-7 and MDA-MB-231 cell lines. Compound 1 showed better antitumor activity than docetaxel against A549 tumor cells. 展开更多

New cobalt(II) complex, [Co(O<sub>2</sub>C<sub>15</sub>H<sub>11</sub>N<sub>2</sub>S)<sub>2</sub>(OH<sub>2</sub>)<sub>2</sub>]∙2H<sub>2</sub>O (1∙2H<sub>2</sub>O), has been synthesized upon reaction of cobalt chloride hexahydrate (Co(Cl)<sub>2</sub>∙6H<sub>2</sub>O) with 3-methyl-1-Phenyl-4-(2-thienoyl)-pyrazol-5-one (referred as HL) in ethanol at room temperature. Single crystal X-ray diffraction (XRD), spectroscopic methods, and microelemental analyses were used to characterize 1∙2H<sub>2</sub>O. Compound 1∙2H<sub>2</sub>O crystallizes in the orthorhombic crystal system with a Pbca space group and with the cobalt atom being pseudo-octahedral coordinated. The broth microdilution technique was used to screen the free ligand (HL) and the complex (1∙2H<sub>2</sub>O) for antimicrobial activities. HL has a low activity (MIC > 100 μg/mL) on all microorganisms, whereas compound 1∙2H<sub>2</sub>O displayed moderate activity (10 ∙2H<sub>2</sub>O exhibited bactericidal and fungicidal activity respectively on all the bacteria and yeasts tested. These findings reveal that the antimicrobial activity of HL was enhanced upon coordination to Co(II) ion against all microorganisms (bacteria and fungus). 展开更多

超高效液相色谱串联线性离子阱质谱法检测减肥类产品中N-丁基-3-(三氟甲基)-α-甲基苯乙胺

目的建立减肥类产品(含中成药、保健食品及食品)中N-丁基-3-(三氟甲基)-α-甲基苯乙胺的检测方法。方法采用超高效液相色谱串联线性离子阱质谱法,色谱柱为Waters ACQUITY UPLC®HSS T3柱(100 mm×2.1 mm,1.8μm),流动相为0.1%... 目的建立减肥类产品(含中成药、保健食品及食品)中N-丁基-3-(三氟甲基)-α-甲基苯乙胺的检测方法。方法采用超高效液相色谱串联线性离子阱质谱法,色谱柱为Waters ACQUITY UPLC®HSS T3柱(100 mm×2.1 mm,1.8μm),流动相为0.1%甲酸水溶液-0.1%甲酸-甲醇(梯度洗脱),流速为0.3 mL/min,检测波长为215 nm,柱温为30℃,进样量为1μL;电喷雾离子源、正电子模式(ESI+)。根据保留时间及离子对进行定性分析;利用多反应监测模式(MRM)拟合标准曲线,计算检出N-丁基-3-(三氟甲基)-α-甲基苯乙胺的含量。结果N-丁基-3-(三氟甲基)-α-甲基苯乙胺质量浓度在0.1~50 ng/mL范围内与定量离子对峰面积线性关系良好(r=1.0000);检测限为0.4 ng/g,定量限为1.2 ng/g;精密度、稳定性、重复性试验结果的RSD均小于3.0%;平均加样回收率为109.67%,RSD为5.46%(n=9)。结论该方法可特异、快速地检出减肥类产品中添加的N-丁基-3-(三氟甲基)-α-甲基苯乙胺,从而可应用于大批量样品的筛查与确证。 展开更多

Novel hexadecyl 3- { 4-[2-hydroxy-3(isopropylamino)propoxy]phenyl }propionate (HPP)was synthesized and its effect on delivery of liposomes into cultured cardiomyocytes was examined. The structure of HPP was characterized by IH NMR, 1R and MS. The amount of cardiomyocytes uptake of HPP-liposome was 3.9-fold higher than plain-liposome, and the increase was 6.2-fold when hypoxia happens. It indicated that HPP was a potential ligand for liposome targeting to ischemic myocardium. 展开更多

X-ray diffraction analysis reveals that the single crystal contains strong non-classical hydrogen bonds. The extraction behavior of rare earths was studied by using paraffin with ceresin as a diluent containing 1-phenyl-3-methyl-4-benzoyl-pyrazolone-5.In solid phase,the composition of complexes is REP_3.The equilib- rium extraction constants and pH_(1/2) values of solid-liquid extraction are higher than those of normal liquid-liquid extraction.The extraction efficiency tends to maximum when the ratio of phases is 1:1.When the extraction temperature is higher than the melting point of paraffin and the extraction time is over 10 min,the extraction efficiency keeps constant.Moreover,the relationship among separation factor,equilibrium extrac- tion constant,pH_(1/2) value and atomic number was obtained.The mechanism of solid-liquid extraction is analogous to that of liquid-liquid extraction. 展开更多

BACKGROUND: To date, a complete protein expression profile of the midbrain substantia nigra in a mouse model of chronic Parkinson's disease, induced by 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP), does not exist. In addition, there are no reports of analysis of differential protein expression. OBJECTIVE: To separate and evaluate MPTP-induced differential protein expression through the use of proteomics in the substantia nigra of a mouse model of chronic Parkinson's disease. DESIGN: Randomized controlled animal study. SETTING: Department of Neurology, the First Affiliated Hospital, Chongqing Medical University. MATERIALS: Sixteen 8-10-week old, healthy, male, C57BL mice, weighing 20-25 g, and of clean grade, were provided by the Experimental Animal Center of Chongqing Medical University. The experimental animals were disposed according to ethical criteria. MPTP was provided by Sigma Company, USA; Pdquest 2D image analysis software and gelatum/irradiance image analysis system (ChemiDoc XRS) by Bio-Rad, USA; and Voyager DE-PROMALD1-TOF-MS mass spectroscopy analyzer by AB1 Company, USA. METHODS: This study was performed in Chongqing Neurological Laboratory between November 2006 and July 2007. Mice were randomly divided into model and control groups, with 8 mice in each group. Mice in the model group were received a subcutaneous injection of MPTP (25 mg&g), twice a week, for five successive weeks, to establish a chronic Parkinson's disease model. Mice in the control group received the same volume of a subcutaneous saline injection at the same time points. Mice were sacrificed by anesthesia to rapidly obtain the midbrain for protein separation of the substantia nigra. MAIN OUTCOME MEASURES: (1) 2-ED handbook (Bio-Rad Company) was referenced for two-dimensional electrophoresis, (2) PDQUEST8,0 analytical electrophoresis pattern was adopted to evaluate differential protein expression. (3) Peptide mass finger print map and data were retrieved on http://www.prospector.ucsf.edu to compare differential substantia nigral protein expression in the two groups. RESULTS: Two-dimensional gel electrophoresis of substantia nigra tissue indicated that there were 33 differential protein expressions between the two groups. Three new proteins were evaluated, including α -enolase, which exhibited regulated expression, tumor necrosis factor ligand superfamily member 4, and cyclin-dependent kinase inhibitor 1B. Using 2'-Hydroxyacetophenone and as the raw material, 1-( 2-Hydroxyphenyl )-3-Phenyl-1, 3- Propanedione (HPPPD) was synthesized by esterification reaction and Fries rearrangement reaction. And the complexes of HPPPD with europium were also synthesized. The compounds were characterized by IR, thermo-gravimetric analysis and ^1H-NMR. The effects of phenolic hydroxyl on the luminescence properties of complexes were studied. The results show that the complexes emit strong red fluorescence and the fluorescence intensity of the ternary complex is better than that of the duality. But the complexes of HPPPD with Sm( Ⅲ ), Tb ( Ⅲ ), Dy ( Ⅲ ) emit weak fluorescence. The phenomenon was interpreted by electron-effect and energy-matching mechanism. Energy transfer mechanism of the luminescence in the complex was discussed. 展开更多

The paper describes the solvent extraction of trace thorium from hydrochloric acid media by1-phenyl-3-methyl-4-benzoyl-5-pyrazolone (PMBP) using a radioactive tracer technique. The percent extraction ofthorium was studied as a function of acidity, PMBP concentration and equilibrium time. The back-extraction behaviorof thorium from the organic phase was also tested. Separation of thorium was performed from fission products pro-duced in 14 MeV neutron bombardment of natural uranium by employing the PMBP extraction procedure. Thegamma-ray spectra of the separated thorium fractions show that thorium can be separated from most of fission prod-ucts and a large amount of uranium. 展开更多

The title compound 3a-hydroxy-3-phenyl-1,2,3,3a,8a-pentahydrocyclopenta[α]- inden-8-one 2 (C18H16O2, Mr = 264.31) has been obtained by the treatment of 2-(3-iodopropyl)-2- phenylindan-1,3-dione 1 with tributyltin hydride in refluxing benzene, and its crystal structure was determined by single-crystal X-ray diffraction. The analysis was carried out by direct and Fourier methods and the structure was refined by full-matrix least-squares computations. The title com- pound crystallizes in monoclinic, space group P21/n with a = 9.947(3), b = 10.336(3), c = 13.239(4) ?, β = 95.565(6)o, V = 1354.7(7) ?3, Z = 4, Dc = 1.296 g/cm3, μ(MoKα) = 0.083 mm-1, F(000) = 560, the final R = 0.0749 and wR = 0.1191. X-ray analysis revealed that the bond length of O(2)– C(12) is 1.219(2) ?, slightly longer than that of the normal C=O bond (1.119 ?). The dihedral angle between planes II and III is almost 180°, while plane II is almost vertical to plane I consisting of C(1), C(3), C(4) and C(5) with the deviation of C(2) being 0.6269 ?. In addition, there exist inter- molecular hydrogen bonds between O(2) and H(1)–O(1) in the crystal structure. 展开更多

Three novel complexes, namely {[La(tapipa)(4,4′-bibp)·(H2O)2]·H2O}n(1),{[La-(tapipa)(H2O)4(OH)]·3 H2O}n(2),[Co(tapipa)(1,4-bbib)]n(3)(tapipa = 5-(3-(tetrazo-5-yl)phenyl)isophthalic acid, 4,4′-bibp = ... Three novel complexes, namely {[La(tapipa)(4,4′-bibp)·(H2O)2]·H2O}n(1),{[La-(tapipa)(H2O)4(OH)]·3 H2O}n(2),[Co(tapipa)(1,4-bbib)]n(3)(tapipa = 5-(3-(tetrazo-5-yl)phenyl)isophthalic acid, 4,4′-bibp = 4,4′-bis(imidazolyl)biphenyl, and 1,4-bbib = 1,4-bis(benzimidazo-1-ly)benzene) have been synthesized under solvothermal conditions and characterized by PXRD, IR,TGA, and so on. Complex 1 presents a 2 D layer structure, which is finally packed into a 3 D supramolecular structure by means of hydrogen bonds;complexes 2 and 3 exhibit 1 D chains and 2 D wavy-type structures, respectively, which are expanded into 3 D supramolecular structures through hydrogen bonds and π-π interactions. Furthermore, the luminescence properties of complex 1 and the variable-temperature magnetic susceptibilities of 3 were investigated.
