目的探讨长链非编码RNA GSTM3TV2在胰腺癌中的表达及其对胰腺癌细胞化疗耐药的调控作用和可能机制。方法收集2016年7—8月在北京协和医院基本外科接受手术治疗的15例胰腺导管腺癌患者的癌组织和其配对的癌旁组织,以及人胰腺癌细胞株AsP... 目的探讨长链非编码RNA GSTM3TV2在胰腺癌中的表达及其对胰腺癌细胞化疗耐药的调控作用和可能机制。方法收集2016年7—8月在北京协和医院基本外科接受手术治疗的15例胰腺导管腺癌患者的癌组织和其配对的癌旁组织,以及人胰腺癌细胞株AsPC-1、BxPC-3、MIAPaCa-2、PanC-1、SU86.86、T3M4、耐药株AsPC-1/GR、耐药株MIAPaCa-2/GR和人正常胰腺上皮细胞株hTERT-HPNE。采用实时定量PCR法检测GSTM3TV2在胰腺癌组织及细胞中表达情况。通过脂质体转染法使胰腺癌细胞中过表达或抑制表达GSTM3TV2后,采用CCK-8法和流式细胞仪检测GSTM3TV2对胰腺癌细胞化疗敏感性的影响。构建GSTM3TV2稳定过表达胰腺癌细胞株,建立裸鼠皮下移植瘤模型,在体内水平进一步观察GSTM3TV2对胰腺癌细胞化疗敏感性的影响。同时,采用免疫印迹法检测GSTM3TV2在胰腺癌细胞化疗耐药过程中可能的分子机制。结果GSTM3TV2在胰腺癌组织中的表达水平(0.493±0.084)高于癌旁组织(0.084±0.019)(t=5.146,P<0.05);同时,GSTM3TV2在胰腺癌耐药细胞株AsPC-1/GR(210.799±19.788)和MIAPaCa-2/GR(122.408±23.419)中的表达水平高于其亲本株(3.793±0.615和5.179±1.095)(t=21.800,P<0.05;t=-18.490,P<0.05)。体内实验结果提示,经化疗药物治疗后,稳定过表达GSTM3TV2的胰腺癌裸鼠皮下移植瘤的体积[(1 059.609±102.498)mm3]大于对照组[(566.414±81.087)mm3](t=4.230,P<0.05)。免疫印迹法检测结果显示,过表达GSTM3TV2可促进细胞增殖相关蛋白Cyclin D1、Cyclin E1、CDK6等的表达,抑制细胞凋亡相关蛋白cleaved caspase-3、cleaved PARP等的表达可增加上皮间质转化相关蛋白Vimentin、N-cadherin、ZEB1、Snail及Slug等表达;反之则抑制GSTM3TV2的表达水平。结论长链非编码RNA GSTM3TV2在胰腺癌组织中呈高表达,可降低胰腺癌细胞的化疗敏感性,其调控化疗耐药的机制可能与胰腺癌细胞增殖、凋亡及上皮间质转化信号转导通路的异常活化相关。 展开更多

改良TVS RT 3D-HyCoSy联合SIS诊断不孕症患者宫腔粘连与输卵管通畅性的价值分析 被引量:1

目的:探讨改良经阴道实时三维子宫输卵管超声造影技术(TVS RT 3D-HyCoSy)联合宫腔水造影(SIS)诊断不孕症患者宫腔粘连与输卵管通畅性的价值。方法:纳入我院2019年7月到2021年7月于我院接受检查的114例不孕症患者,均接受改良TVS RT 3D-Hy... 目的:探讨改良经阴道实时三维子宫输卵管超声造影技术(TVS RT 3D-HyCoSy)联合宫腔水造影(SIS)诊断不孕症患者宫腔粘连与输卵管通畅性的价值。方法:纳入我院2019年7月到2021年7月于我院接受检查的114例不孕症患者,均接受改良TVS RT 3D-HyCoSy与SIS检查。与腹腔镜检查(LPS)及宫腔镜检查(HS)对比,记录改良TVS RT 3D-HyCoSy单独及联合SIS检查诊断卵管通畅性结果与诊断效能、宫腔粘连结果与诊断效能。结果:114例患者共有228条输卵管,LPS、改良TVS RT 3D-HyCoSy单独及联合SIS检查输卵管通畅的阳性、阴性结果分别为87、141条,77、151条,84、144条,改良TVS RT 3D-HyCoSy单独或联合SIS检查输卵管通畅性的诊断效能对比差异无统计学意义(P>0.05);改良TVS RT 3D-HyCoSy单独或联合SIS检查输卵管通畅性与LPS的一致性均较高(Kappa=0.905、0.896,P均<0.001)。HS、改良TVS RT 3D-HyCoSy单独或联合SIS检查宫腔粘连结果分别为:阳性43例,阴性71例,阳性49例、阴性65例,阳性47例、阴性67例,改良TVS RT 3D-HyCoSy单独检查的准确度、灵敏度、特异度低于联合SIS检查(P<0.05);改良TVS RT 3D-HyCoSy单独诊断宫腔粘连的一致性中等(Kappa=0.600,P<0.001),联合SIS检查诊断宫腔粘连的一致性较好(Kappa=0.890,P<0.001)。结论:改良TVS RT 3D-HyCoSy单独与SIS均能有效诊断输卵管通畅性,但两者联合诊断宫腔粘连的价值更好。 展开更多

Now depth estimation still ... Depth estimation is an active research area with the developing of stereo vision in recent years. It is one of the key technologies to resolve the large data of stereo vision communication. Now depth estimation still has some problems, such as occlusion, fuzzy edge, real-time processing, etc. Many algorithms have been proposed base on software, however the performance of the computer configurations limits the software processing speed. The other resolution is hardware design and the great developments of the digital signal processor (DSP), and application specific integrated circuit (ASIC) and field programmable gate array (FPGA) provide the opportunity of flexible applications. In this work, by analyzing the procedures of depth estimation, the proper algorithms which can be used in hardware design to execute real-time depth estimation are proposed. The different methods of calibration, matching and post-processing are analyzed based on the hardware design requirements. At last some tests for the algorithm have been analyzed. The results show that the algorithms proposed for hardware design can provide credited depth map for further view synthesis and are suitable for hardware design. 展开更多

MAX phases(Ti_(3)SiC_(2),Ti_(3)AIC_(2),V_(2)AlC,TiqAlN_(3),etc.)are layered ternary carbides/nitrides,which are generally processed and researched as structure ceramics.Selectively removing A layer from MAX phases,MXenes(Ti_(3)C_(2),V_(2)C,Mo_(2)C,etc.)with two-dimensional(2D)structure can be prepared.The MXenes are electrically conductive and hydrophilic,which are promising as functional materials in many areas.This article reviews the milestones and the latest progress in the research of MAX phases and MXenes,from the perspective of ceramic science.Especially,this article focuses on the conversion from MAX phases to MXenes.First,we summarize the microstructure,preparation,properties,and applications of MAX phases.Among the various properties,the crack healing properties of MAX phase are highlighted.Thereafter,the critical issues on MXene research,including the preparation process,microstructure,MXene composites,and application of MXenes,are reviewed.Among the various applications,this review focuses on two selected applications:energy storage and electromagnetic interference shielding.Moreover,new research directions and future trends on MAX phases and MXenes are also discussed. 展开更多
