清肾颗粒对5/6肾切除大鼠肾组织线粒体自噬及肾小管上皮-间质细胞转化的影响 被引量:1

目的:探讨清肾颗粒对5/6肾切除大鼠肾组织线粒体自噬及肾小管上皮细胞转分化(Epithelial-mesenchymal,EMT)的影响。方法:雄性SD大鼠30只,随机分为假手术组、模型组、清肾颗粒组,每组各10只。除假手术组外,其余20只大鼠均制作5/6肾切除... 目的:探讨清肾颗粒对5/6肾切除大鼠肾组织线粒体自噬及肾小管上皮细胞转分化(Epithelial-mesenchymal,EMT)的影响。方法:雄性SD大鼠30只,随机分为假手术组、模型组、清肾颗粒组,每组各10只。除假手术组外,其余20只大鼠均制作5/6肾切除模型。清肾颗粒各组分别给予相应剂量清肾颗粒水溶液;假手术组、模型组以生理盐水灌胃。连续给药8周后,无菌采取腹主动脉血,代谢笼留取24h尿液,摘取左侧肾脏。检测血、尿肌酐浓度,并计算内生肌酐清除率;Western blot法检测肾组织中Pink1、Parkin、LC3-Ⅱ、α-SMA;免疫荧光法检测肾组织中LC3-Ⅱ和线粒体膜蛋白VDAC1共定位表达;HE和Masson染色光镜观察大鼠肾脏病理改变;透射电镜检测肾小管上皮细胞中线粒体的超微结构。结果:与假手术组比较,模型组大鼠的线粒体自噬相关蛋白Pink1、Parkin、LC3Ⅱ表达量均显著下降,肾小管EMT标志蛋白α-SMA显著升高(P<0.05)。与模型组比较,清肾颗粒组大鼠的Pink1、LC3Ⅱ蛋白表达量显著升高,α-SMA表达量显著降低(P<0.05),Parkin亦有所升高,但差异无统计学意义(P>0.05)。结论:清肾颗粒能够上调NRK-52E细胞内miR-23b-5p表达,并通过增强PINK1/Parkin通路介导的线粒体自噬活性,抑制NRK-52E细胞转分化进程。 展开更多

目的观察Smad3基因敲除对5/6肾切除慢性肾纤维化小鼠模型的影响。方法清洁级WT-C57及Smad3-KO(以下简写S3-KO)小鼠各24只,分为假手术野生组(SWT)、假手术Smad3-KO组(SS3-KO)和模型野生组(MWT)、模型Smad3-KO组(MS3-KO)4组,模型组以plat... 目的观察Smad3基因敲除对5/6肾切除慢性肾纤维化小鼠模型的影响。方法清洁级WT-C57及Smad3-KO(以下简写S3-KO)小鼠各24只,分为假手术野生组(SWT)、假手术Smad3-KO组(SS3-KO)和模型野生组(MWT)、模型Smad3-KO组(MS3-KO)4组,模型组以platt法建立慢性肾脏纤维化模型,造模后4周,检测各组小鼠血中肌酐(Scr)、尿素氮(BUN)、24h尿蛋白(U-Pro/24h)含量、病理形态学变化;免疫荧光染色法观察肾组织中α-平滑肌肌动蛋白(α-SMA),I型胶原蛋白(CollageI),纤维连接蛋白(FN)的表达面积变化;蛋白免疫印迹法(WB)和实时荧光定量PCR(RT-PCR)法检测各组小鼠肾组织TGF-β1、p-Smad3、α-SMA、CollageI、FN蛋白及mRNA的表达水平;免疫组化法观察肾组织内中性粒细胞阳性表达个数的变化。结果与假手术组比较,MWT组P-smad3蛋白,Smad3mRNA明显增高(P<0.01),MS3-KO组虽有增高,但无显著性差异(P>0.05)。HE染色SWT,SS3-KO组肾小球形态基本正常,天狼星染色肾组织间质无或有少许红色胶原纤维沉积。MWT,MS3-KO组肾小球明显异常,天狼星染色MWT,MS3-KO组间质能够见到明显的胶原纤维,MS3-KO组病理改变总体轻于MWT组。经过图像处理分析可见,胶原纤维面积MS3-KO组面积小于MWT组,组间比较均有显著性差异(P<0.05)。指标方面,SWT与SS3-KO组比较,α-SMA、CollageI、FN免疫荧光面积,Scr、Bun、U-Pro/24h水平,TGF-β1、α-SMA、CollageI、FN蛋白和mRNA相对表达水平,中性粒细胞阳性表达个数均无显著性差异(P>0.05),M组与其S组比较上述指标含量明显增高(P<0.01或P<0.05),而MS3-KO与MWT比较,MWT组上述指标上升更为明显,组间差异有统计学意义(P<0.01或P<0.05)。结论Smad3-KO能够减少模型组小鼠肾组织TGF-β1、α-SMA、FN、CollageI蛋白和mRNA的表达,改善肾功能,减少中性粒细胞的表达,能够减轻模型所致的肾脏纤维化病理损害,对肾脏损害有一定的保护作用。 展开更多

CMIP5/6模式对PDO调制ENSO爆发频率不对称的模拟评估

利用第五次和第六次国际间耦合模式比较计划(CMIP)中piControl情景下的模拟结果,结合观测资料,对比评估了19个CMIP5模式和23个CMIP6模式对太平洋年代际振荡(PDO)调制厄尔尼诺-南方涛动事件爆发频率不对称的模拟能力,并进一步揭示了PDO... 利用第五次和第六次国际间耦合模式比较计划(CMIP)中piControl情景下的模拟结果,结合观测资料,对比评估了19个CMIP5模式和23个CMIP6模式对太平洋年代际振荡(PDO)调制厄尔尼诺-南方涛动事件爆发频率不对称的模拟能力,并进一步揭示了PDO的调制过程。结果表明:在观测中,PDO正(负)位相下厄尔尼诺(El Niño)的爆发频率比拉尼娜(La Niña)多300%(少73%),53%(78%)的CMIP5(6)模式模拟出这一特征;尽管两个模式整体都低(高)估了PDO正(负)位相的调制能力,但CMIP6模式对PDO调制能力的模拟有所改进。进一步研究发现,在PDO正(负)位相下,赤道太平洋中西部会产生较强的西(东)风异常,风场通过平流的作用使得暖水向东流动,从而在太平洋中东部的海表面温度背景场中出现正(负)异常变化,而这有利于PDO正(负)位相下El Niño(La Niña)事件的发生。 展开更多

目的:探讨白藜芦醇(resveratrol,RSV)对5/6肾切除大鼠肌肉萎缩的保护作用及对肌营养不良蛋白(dystrophin)、肌肉环状脂蛋白-1(musclering finger-1,MuRF-1)、肌肉萎缩蛋白-1(MAF-bx,又名Atrogin-1)及肌肉生长抑制素(Myostatin)的影响。... 目的:探讨白藜芦醇(resveratrol,RSV)对5/6肾切除大鼠肌肉萎缩的保护作用及对肌营养不良蛋白(dystrophin)、肌肉环状脂蛋白-1(musclering finger-1,MuRF-1)、肌肉萎缩蛋白-1(MAF-bx,又名Atrogin-1)及肌肉生长抑制素(Myostatin)的影响。方法:采用两步法建立雄性Sprague-Dawley大鼠5/6肾切除模型。术后1周开始予20 mg/(kg·d)RSV灌胃,至术后12周处死大鼠,随机分为3组:对照组(Sham组,n=10)、5/6肾切除组(Nx组,n=10)、5/6肾切除+RSV干预组(Nx+RSV组,n=10),收集血液检测血清肌酐(serum creatinine,SCr)、血尿素氮(blood urea nitrogen,BUN)含量,取肾脏组织行过碘酸-雪夫(periodic acid-schiff,PAS)染色、Masson染色观察肾脏病理变化,取胫骨前肌用于HE染色观察肌纤维的形态和大小,免疫组织化学法观察肌肉中dystrophin表达及分布情况。Western Blot分析检测大鼠肌肉组织中肌萎缩相关蛋白MuRF-1、Atrogin-1、Myostatin的表达。结果:(1)与Sham组相比,Nx组大鼠在术后12周开始出现SCr、BUN水平升高,RSV干预可改善肾功能。(2)Nx组大鼠肾小球硬化指数(glomerular sclerosis index,GSI)及肾小管间质纤维化指数(tubulointerstitial fibrosis index,TFI)升高,RSV干预后得到改善。(3)Nx组大鼠胫骨前肌出现肌纤维横截面积缩小,dystrophin表达减少,Atrogin-1、MuRF-1、Myostatin表达增加,RSV干预后改善了肌纤维萎缩,上调了dystrophin表达,降低Atrogin-1、MuRF-1、Myostatin表达。结论:RSV可改善5/6肾切除大鼠的肾功能及肌肉萎缩,其机制可能与上调dystrophin表达,降低Atrogin-1、MuRF-1、Myostatin表达有关,提示RSV可能是5/6肾切除肌肉萎缩的一个潜在治疗药物。 展开更多

Using the historical simulations from 27 models in phase 5 of the Coupled Model Intercomparison Project(CMIP5)and 27 models in phase 6(CMIP6),the authors evaluated the differences between CMIP5 and CMIP6 models in simulating the climate mean of extreme temperature over China through comparison with observations during 1979–2005.The CMIP6 models reproduce well the spatial distribution of annual maxima of daily maximum temperature(TXx),annual minima of daily minimum temperature(TNn),and frost days(FD).The model spread in CMIP6 is reduced relative to CMIP5 for some temperature indices,such as TXx,warm spell duration index(WSDI),and warm days(TX90 p).The multimodel median ensembles also capture the observed trend of extreme temperature.However,the CMIP6 models still have low skill in capturing TX90 p and cold nights(TN10 p)and have obvious cold biases or warm biases over the Tibetan Plateau.The ability of individual models varies for different indices,although some models outperform the others in terms of the average of all indices considered for different models.By comparing different version models from the same organization,the updated CMIP6 models show no significant difference from their counterparts from CMIP5 for some models.Compared with individual models,the median ensembles show better agreement with the observations for temperature indices and their means. 展开更多

The Role of Angiopoietin-1 and Thrombospondin-1 in the Kidney of Rats Subject to 5/6 Nephrectomy 被引量:2

The expression of angiopoietin- 1 (Ang- 1) and thrombospondin- 1 (TSP- 1) in 5/6 subtotal nephrectomy (STN) rats model, and its correlation to the renal microvasculature injury were investigated. Rat 5/6 STN model was established in adult male SD rats, and the sham-operated group and 5/6 STN group were set up. The renal function and histopathological changes were examined at the 1st, 2nd, 4th, 8th and 12th week after operation. The expression orAng-1, TSP-1 and CD31 in renal tissues was detected by using immunohistochemistry. From 2nd to 8th week after operation, Ang-1 was significantly expressed in glomeruli of rats with STN. Ang-1 staining in glomeruli of STN group was increased significantly as compared with that in sham-operated group at 4th and 8th week after operation, and subsequently decreased after the 12th week. The expression of TSP-1 was increased significantly in STN group. As compared with sham-operated group, the CD31 expression was significantly down-regulated from the 2nd week. The expression of Ang-1 mRNA was detected by using RT-PCR at the same time points. The expression of Ang-1 mRNA in renal tissue of rats with STN was significantly up-regulated at the 2nd, 4th and 8th week after operation as compared with that in STN group at other time points or in sham-operated group at the same time points, while decreased evidently at the 12th week as compared with that in sham-operated group. It is concluded that there are changes in the mRNA expression of Ang-1, and the significant up-regulation of the expression of TSP-1 in renal tissue of rats with STN, which may be involved in the remnant renal microvasculature injury. 展开更多

Summary: This study investigated the effects of benazepril administered in the morning or evening on the diurnal variation of renin-angiotensin-aldosterone system (RAAS) and clock genes in the kidney. The male Wistar rat models of 5/6 subtotal nephrectomy (STNx) were established. Animals were ran- domly divided into 4 groups: sham STNx group (control), STNx group, morning benazepril group (MB) and evening benazepril group (EB). Benazepril was intragastfically administered at a dose of 10 mg/kg/day at 07:00 and 19:00 in the MB group and EB group respectively for 12 weeks. All the animals were synchronized to the light:dark cycle of 12:12 for 12 weeks. Systolic blood pressure (SBP), 24-h urinary protein excretion and renal function were measured at 11 weeks. Blood samples and kidneys were collected every 4 h throughout a day to detect the expression pattern of renin activity (RA), angio- tensin Ⅱ (Ang Ⅱ ) and aldosterone (Aid) by radioimmunoassay (RIA) and the mRNA expression profile of clock genes (bmall, dbp and per2) by real-time PCR at 12 weeks. Our results showed that no signifi- cant differences were noted in the SBP, 24-h urine protein excretion and renal function between the MB and EB groups. There were no significant differences in average Aid and RA content of a day between the MB group and EB group. The expression peak of bmall mRNA was phase-delayed by 4 to 8 h, and the diurnal variation of per2 and dbp mRNA diminished in the MB and EB groups compared with the control and STNx groups. It was concluded when the similar SBP reduction, RAAS inhibition and clock gene profile were achieved with optimal dose of benazepril, morning versus evening dosing of benazepril has the same renoprotection effects. 展开更多

Terutroban, a TP receptor antagonist, restores renal artery tone, but not renal function in mouse with 5/6 nephrectomy

Aim Thromboxane A2(TXA2) is assumed to contribute to the process of renal dysfunction. The pres- ent study was designed to investigate whether terutroban, a specific antagonist of thromboxane/prostaglandin? (TP) receptor, protects against renal damage in 5/6 nephrectomy. Methods C57/BL6 mice were randomly grouped into sham-operated (2K), 5/6 nephroectomy groups (5/6K-off) and 5/6 nephroectomy treated with ter- utroban (10 mg · kg^-1 · d^-l) groups (Yerutroban). Renal artery and kidney were collected for vascular function study, Western blot, immunohistochemistry (IHC) assay and enzyme-linked immunosorbent assay (ELISA), re- spectively. Results Four weeks after the surgery, arterial blood pressure was comparable among the three groups. However mice in terutroban group had higher levels of serum creatinine and lower survival. Compared with 2K groups, 5/6K-off mice had significantly higher levels of renal blood flow as well as a blunted relaxation to acetyl- choline. Production of prostacyclin (PGI2) and thromboxane B2 ( TXB2), but no prostaglandin E2 ( PGE2), were significantly increased in the renal artery of 5/6K-off group. Terutroban restored the renal blood flow, but not the acetylcholine-induced relaxation in the renal artery. It is probably due to the blockade effect of terutroban on the smooth muscle since terutroban treatment significantly reduced U46619-induced vasconstriction in renal arteries. Interestingly, terutroban increased the production of TXB2, but not PGI2 or PGE2, in the renal artery. This proba-bly is a compensatory effect on prostaglandins production. In kidney cortex, 5/6K-off group had significantly lower levels of PGE2 and TXB2 when compared with 2 K group. Terutroban markedly increased all three prostaglandins levels. Conclusion Terutroban restores renal artery function, but not renal function in mouse with 5/6 nephrecto- my. It suggests that kidney has more complicated regulations than renal artery. High levels of prostanoids in kid- neys may contribute to renal damage in terutroban group. Further experiments will focus on examining the underly- ing mechanisms. 展开更多

The South Pacific Quadrupole(SPQ) is the extratropical South Pacific’s second principal sea surface temperature mode.Previous observational studies have shown that the SPQ promotes the onset of the El Nino-Southern Oscillation(ENSO).The present study evaluates and compares simulations of the SPQ-ENSO relationship by 20 climate models from CMIP6 and their corresponding 20 previous models from CMIP5.It is found that 16 of the20 pairs of models are able to consistently reproduce the spatial pattern of the SPQ.In terms of simulating the SPQ-ENSO relationship,9 of the 16 CMIP6 models show significant improvement over their previous CMIP5 models.The multi-model ensemble(MME) of these 16 CMIP6 models simulates the SPQ-ENSO connection more realistically than the CMIP5 MME.Further analysis shows that the performance of the model simulations in reproducing the SPQ-ENSO relationship is strongly dependent on their ability to simulate the SPQ-related surface air-sea coupling processes over the southwestern and southeastern South Pacific,as well as the response of the SPQ-related equatorial subsurface ocean temperature anomalies.The improvement of the CMIP6 models in simulating these two processes is responsible for the improved performance of the CMIP6 models over their CMIP5 counterparts in simulating the SPQ-ENSO relationship. 展开更多

The nearest neighbor spacing distribution (NSD) and the spectral rigidity (△3)are investigated, and the factors that affect the properties of level statistics are also discussed. The results show that the boson number N is a dominant factor. If N is small, both the interaction strengths of subgroups SOB(5) and SOBF(5)and the spin play important roles in the energy level statistics, however, along with the increase of N, the statistics distribution would tend to be in Poisson form. 展开更多

目的:探讨左归丸对5/6肾大部切除模型并肾性骨病大鼠甲状旁腺功能亢进的影响。方法:SPF级Wistar大鼠进行5/6肾大部切除并给予高磷饮食诱导肾性骨病模型。造模大鼠分为模型组、左归丸组、骨化三醇组,并设正常组、假手术组。药物干预12周... 目的:探讨左归丸对5/6肾大部切除模型并肾性骨病大鼠甲状旁腺功能亢进的影响。方法:SPF级Wistar大鼠进行5/6肾大部切除并给予高磷饮食诱导肾性骨病模型。造模大鼠分为模型组、左归丸组、骨化三醇组,并设正常组、假手术组。药物干预12周处死大鼠,检测血清全段甲状旁腺激素(iPTH);双能X线测量大鼠股骨骨密度(BMD);分别采用Real-time PCR、western blotting检测甲状旁腺PTH mRNA与蛋白的表达;免疫组化的方法检测甲状旁腺PCNA的表达。结果:12周时,模型组大鼠出现了血iPTH升高,甲状旁腺增生,骨密度下降。左归丸组大鼠iPTH水平与模型组比较(P<0.01)明显下降,左归丸组大鼠骨密度与模型组比较明显改善(P<0.01),明显抑制甲状旁腺增生。结论:左归丸能降低5/6肾大部切除模型并肾性骨病大鼠甲状旁腺素合成以及血iPTH水平,抑制甲状旁腺增生,改善肾性骨病。 展开更多

目的观察复方黄甘对5/6肾切除慢性肾衰模型大鼠的疗效,并探讨其可能的作用机制。方法用HPLC检测复方黄甘提取物组分并定量;构建5/6肾切除大鼠慢性肾衰模型,将模型大鼠随机分为尿毒清组、科素亚组、复方黄甘低、中、高剂量组和模型组,另... 目的观察复方黄甘对5/6肾切除慢性肾衰模型大鼠的疗效,并探讨其可能的作用机制。方法用HPLC检测复方黄甘提取物组分并定量;构建5/6肾切除大鼠慢性肾衰模型,将模型大鼠随机分为尿毒清组、科素亚组、复方黄甘低、中、高剂量组和模型组,另加假手术组共7组,每组10只大鼠,经灌胃给药12周后用全自动生化分析仪检测Scr、BUN、Ucr及24 h UPr;对残余肾组织进行HE和Masson染色观察病理变化;用RT-PCR和免疫组化检测肾组织FN、MCP-1、ICAM-1的表达。结果复方黄甘提取物中主要成分及其含量为:甘草苷(0.06%)、丹皮酚(1.06%)、芦荟大黄素(0.1%)、大黄酸(0.47%)、大黄素(0.19%)、大黄酚(0.41%)和大黄素甲醚(0.13%);复方黄甘低、中、高剂量组Scr、BUN、Ucr、Ccr及24h UPr都显著低于模型组(P<0.05);复方黄甘给药组肾小球硬化和肾间质纤维化改善较为明显,且肾组织中FN、ICAM-1 mRNA表达和蛋白表达均显著低于模型组(P<0.05)。结论复方黄甘可改善慢性肾衰模型大鼠肾功能,减轻肾小球硬化和肾间质纤维化,在一定程度上延缓慢性肾功能衰竭的进展。 展开更多

目的观察血管紧张素Ⅱ受体拮抗剂厄贝沙坦对5/6肾切除及腹主动脉缩窄大鼠左心室肥厚及心肌尿素转运蛋白B(UT-B)表达的影响。方法 SD大鼠随机分为假手术组、5/6肾切除组(肾衰组)、腹主动脉缩窄组(心衰组)、肾衰+心衰组及相应的厄贝沙坦... 目的观察血管紧张素Ⅱ受体拮抗剂厄贝沙坦对5/6肾切除及腹主动脉缩窄大鼠左心室肥厚及心肌尿素转运蛋白B(UT-B)表达的影响。方法 SD大鼠随机分为假手术组、5/6肾切除组(肾衰组)、腹主动脉缩窄组(心衰组)、肾衰+心衰组及相应的厄贝沙坦干预组,每组8只。分组处理后,测定各组大鼠血清肌酐(SCr)、血尿素氮(BUN)、血管紧张素Ⅱ(AngⅡ)、B型利钠肽(BNP);取大鼠心脏,测定左心室质量与全心质量的比值(LVW/HW)、全心质量与体质量的比值(HW/BW)、左心室质量与体质量的比值(LVW/BW);RT-PCR和Western blotting检测心肌UT-B表达。结果与假手术组比较,肾衰组、心衰组及肾衰+心衰组大鼠出现心肌肥厚,LVW/HW、HW/BW、LVW/BW增大,血清AngⅡ和BNP水平增高,心肌组织UT-B mRNA和UT-B蛋白表达上调;肾衰组和肾衰+心衰组SCr、BUN水平升高。与肾衰组、心衰组及肾衰+心衰组比较,相应的厄贝沙坦干预组大鼠的心肌肥厚明显减轻,LVW/HW、HW/BW、LVW/BW降低,血清AngⅡ和BNP下降,心肌UT-B mRNA和UT-B蛋白表达下调。与肾衰组和肾衰+心衰组比较,相应的厄贝沙坦干预组大鼠SCr、BUN水平降低。结论 5/6肾切除及腹主动脉缩窄均可引起大鼠心功能明显下降,出现左心室肥厚,同时心肌UT-B表达增加;厄贝沙坦在改善心、肾功能的同时抑制心肌UT-B表达,UT-B可能在肾功能不全合并心力衰竭的发生中具有重要作用。 展开更多

[目的]查阅已有的心肾综合征大鼠模型文献报道,将“5/6肾切除术制备心肾综合征大鼠模型”方法适当改进,评价其方案造模效果,探讨简捷高效的心肾综合征造模方法。[方法]将20只雄性SD大鼠随机分为两组,即空白组与手术组,空白组大鼠不做处理,手术组大鼠均按“两次手术法”行5/6肾切除,手术1周后连续2 d(间隔24 h)皮下注射异丙肾上腺素(2.5、1.5 mg/kg)制备心肾综合征模型。分别检测两组大鼠造模后的血肌酐(Cr)、血尿素氮(BUN)、脑钠肽(BNP)水平,比较造模前、造模后、造模6周大鼠体质量,并在造模6周后处杀两组大鼠,称取心脏质量,计算心脏质量指数(心脏质量/大鼠体质量),观察两组大鼠心、肾病理组织变化。[结果]大鼠经5/6肾切除联合异丙肾皮下注射方法造模后,空白组与手术组BNP、Cr水平差异有统计学意义(P<0.05),手术组大鼠活动后喘息明显、耳廓苍白、被毛稀疏干枯且生长缓慢,体质量较空白组明显降低(P<0.05),心、肾组织病理均较空白组有明显改变。[结论]5/6肾切除联合ISO皮下注射制备心肾综合征大鼠模型方法有效,是一种简捷高效的心肾综合征造模方法,可以为心肾综合征动物实验提供模型基础。但该法由于不完全模拟慢性肾功能衰竭导致心力衰竭的自然病程,可能不适用于研究该病早期发展变化规律,而对于心肾综合征的药物治疗评价及机制研究较为合适。 展开更多

目的观察高磷饮食对5/6肾切除大鼠继发性甲状旁腺功能亢进、肾脏纤维化的影响及金雀异黄素的干预作用。方法 8~10周龄雄性Sprague-Dawley大鼠24只,体重200 250 g,随机分为4组,每组6只:假手术正常磷饮食组(Sham十NP组),5/6肾切除正常磷... 目的观察高磷饮食对5/6肾切除大鼠继发性甲状旁腺功能亢进、肾脏纤维化的影响及金雀异黄素的干预作用。方法 8~10周龄雄性Sprague-Dawley大鼠24只,体重200 250 g,随机分为4组,每组6只:假手术正常磷饮食组(Sham十NP组),5/6肾切除正常磷饮食组(STNx+NP组),5/6肾切除高磷饮食组(STNx+HP组),5/6肾切除高磷饮食+金雀异黄素灌胃组(STNx+HP+Gen组)。高磷饮食配方:磷1.2%,钙1.6%,维生素D 1 U/kg。正常磷饮食配方:磷0.6%,钙0.6%,维生素D 1 U/kg。5/6肾脏切除分两步进行。造模术后第4、8、12、16周,检测血清肌酐(SCr)、磷、钙、1,25(OH)_2D_3、全段甲状旁腺激素(iPTH)水平。特定饮食结束时,处死大鼠,取肾脏组织,石蜡切片行苏木精-伊红(H-E)染色,进行病理积分。结果 Sham+NP组各项指标在实验过程中均无明显变化(P值均>0.05)。STNx+NP组、STNx+HP组、STNx+HP+Gen组SCr、磷、iPTH的水平均随着特定饮食喂养时间的延长逐渐升高。16周时与4周时的差异均有统计学意义(P值均<0.01)。16周时,STNx+HP组、STNx+HP+Gen组的SCr、磷、iPTH与STNx+NP组的差异均有统计学意义(P值均<0.01)。而4组不同时间钙、1,25(OH)_2D_3的水平均无明显改变(P值均>0.05)。Sham+NP组、STNx+NP组、STNx+HP组和STNx+HP+Gen组的肾小球硬化指数分别为0、1.01±0.02、1.83±0.03、1.51±0.03,STNx+HP组和STNx+HP+Gen组显著高于STNx+NP组(P值均<0.01),STNx+HP组显著高于STNx+HP+Gen组(P<0.01)。结论高磷饮食可促进5/6肾大部切除大鼠甲状旁腺功能亢进的发生,并加剧其肾功能恶化,金雀异黄素可部分减轻其肾脏毒性作用,改善甲状旁腺功能亢进。 展开更多
