Objectives Endotheliallipase(EL) is a major determinantof high-density lipoprotein-cholesterol(HDL-C) metabolism,but the association of endothelial lipase gene (LIPG) polymorphism and serum HDL-C levels is scarce and conflicting in diverse populations.Bai Ku Yao is an isolated subgroup of the Yao minority in China.This study was designed to detect the association of LIPG 584C】T(rs2000813) polymorphism and several environmental factors with serum lipid levels in the Guangxi Bai Ku Yao and Han populations. Methods A total of 645 subjects of Bai Ku Yao and 638 participants of Han Chinese were randomly selected from our previous stratified randomized cluster samples.Genotyping of the LIPG584C】T was performed by polymerse chain reaction and restriction fragment length polymorphism combined with gel electrophoresis,and then confirmed by direct sequencing. Results The levels of serum total cholesterol(TC),HDL-C, low-density lipoprotein cholesterol(LDL-C) and apolipoprotein (Apo) AI and ApoB were lower in Bai Ku Yao than in Han(P【0.05-0.001).The frequency of C and T alleles was 73.5%and 26.5%in Bai Ku Yao,and 67.9%and 32.1%in Han(P【0.01).The frequency of CC,CT and TT genotypes was 50.4%,46.2%and 3.4%in Bai Ku Yao,and 41.4%, 53.1%and 5.5%in Han(P【0.01);respectively.Serum HDL-C levels in both ethnic groups were different among the three genotypes(P【0.05 for each).Serum TC levels in both ethnic groups were also different between CC and CT/TT types(P【 0.05 for each).The T allele carriers had higher serum HDL-C and TC levels than the T allele noncarriers.Multivariate logistic regression analysis showed that the levels of HDL-C and ApoB were correlated with genotype in Bai Ku Yao(P【 0.05 for each),whereas the levels of TC and HDL-C were associated with genotype in Han Chinese(P【0.05 and P【 0.01).Serum lipid parameters were also correlated with gender, age,weight,body massindex,alcohol consumption, cigarette smoking,and blood pressure in the both ethnic groups.Conclusions The frequency of LIPG 584Tallele is lower in the Bai Ku Yao than in the Han populations.The LIPG 584Tallele is associated with serum HDL-C and TC levels.The differences in serum lipid profiles between the two ethnic groups might partly result from different genotypic and allelic of LIPG 584C 】T or different LIPG-enviromental interactions. 展开更多

目的探讨载脂蛋白E(Apo E)基因526 C>T多态性与氟伐他汀调脂作用的相关性。方法纳入血脂异常患者152例,抽取其静脉血并提取基因组DNA后采用聚合酶链反应(PCR)-荧光探针法检测ApoE基因,并分析ApoE基因多态性与低密度脂蛋白受体(LDLR)... 目的探讨载脂蛋白E(Apo E)基因526 C>T多态性与氟伐他汀调脂作用的相关性。方法纳入血脂异常患者152例,抽取其静脉血并提取基因组DNA后采用聚合酶链反应(PCR)-荧光探针法检测ApoE基因,并分析ApoE基因多态性与低密度脂蛋白受体(LDLR)结合的相关性,比较ApoE等位基因出现的频率和各基因型出现的频率。根据基因检测结果将152例患者分为E2组(E2/E2和E2/E3基因型)37例、E3组(E3/E3和E3/E4基因型)110例、E4组(E2/E4和E4/E4基因型)5例,比较3组患者的上述检测结果。结果E2组、E3组、E4组的LDLR最大置换量分别为(3.89±0.37)μg/mg、(6.87±0.22)μg/mg和(6.12±0.34)μg/mg,其中E3组明显高于E2组和E4组,E4组明显高于E2组(P<0.05)。152例血脂异常患者中,ApoE等位基因E3出现的频率(63.82%)高于E2(15.46%)和E4(20.72%),差异有统计学意义(P<0.05)。E2组、E3组、E4组疗效优良率分别为19.74%(30/152)、55.92%(85/152)和9.21%(14/152),E2组明显高于E3组、E4组(P<0.001)。结论对于血脂异常而应用氟伐他汀治疗患者而言,ApoE基因526 C>T(E2)多态性对氟伐他汀有较好的降脂效能。 展开更多

目的探讨载脂蛋白E(ApoE)基因526C>T多态性与瑞舒伐他汀调脂疗效及超敏C反应蛋白(hs-CRP)的关系。方法选取本院2018年1月—2020年3月收治的血脂异常病人155例进行前瞻性研究,利用等位基因特异性多重聚酶链式反应(PCR)对高脂血症病人... 目的探讨载脂蛋白E(ApoE)基因526C>T多态性与瑞舒伐他汀调脂疗效及超敏C反应蛋白(hs-CRP)的关系。方法选取本院2018年1月—2020年3月收治的血脂异常病人155例进行前瞻性研究,利用等位基因特异性多重聚酶链式反应(PCR)对高脂血症病人进行ApoE基因分型,并分为E3/3组、E3/2组、E4/3组,分析不同的基因型和血脂水平及其对瑞舒伐他汀调脂疗效影响,并探究其与hs-CRP的关系。提取3组的基因组DNA后利用PCR-荧光探针法对其ApoE基因进行测定,并分析其与hs-CRP的关系。结果治疗前,E3/2组的总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平低于E3/3和E4/3组,且E3/3组TC、LDL-C水平低于E4/3组,差异有统计学意义(P<0.05);瑞舒伐他汀治疗12周后,E3/2基因组血清TC、LDL-C水平下降幅度大于E3/3组,差异有统计学意义(P<0.05)。瑞舒伐他汀治疗前,E4/3组病人血清hs-CRP水平低于E3/2组和E3/3组,差异有统计学意义(P<0.05);瑞舒伐他汀治疗后,E4/3组病人血清hs-CRP水平和E3/2组、E3/3组比较,差异无统计学意义(P>0.05)。结论ApoE基因多态性会影响高脂血症病人血清TC、LDL-C水平,从而影响瑞舒伐他汀的调脂疗效,临床可根据病人ApoE基因多态性指导他汀类药物治疗方案。 展开更多
