The leaves are elongated round and hollowed inside. This plant had been using for the treatment of breast cancer. Extraction with percolation method has been done in ekor naga’s leaves with ethanol, and fractionated by nhexane, chloroform and ethyl acetate using liquid-liquid extraction (LLE). Cytotoxicity assay of ethanol extract, n-hexane fraction, chloroform fraction, ethyl acetate fraction and water fraction against MCF-7 cells were done using MTT method (3-(4,5-dimetiltiazol-2-il)-2,5-diphenyl tetrazolium bromide). Phytochemical screening results showed the presence of the compounds such as triterpenoida/steroid, alkaloid, flavonoid, tannin, and saponin. n-hexane fraction was positive for the presence of triterpenoida/steroid, chloroform fraction containing alkaloids, saponin and triterpenoid;ethyl acetate fraction contained, flavonoid, tannin, and the fraction of water indicated the presence of tannin and saponin. Secondary metabolite compounds in ethanol extract, chloroform fraction and ethyl acetate fraction gave positive results against MCF-7 cells. Cytotoxicity assay of MCF-7 cell line showed that crude ethanol extracts had 112.240 mcg/ml IC50 chloroform fraction IC50 was 59.082 mcg/ml, and ethyl acetate fraction IC50 was 812.663 mcg/ml.展开更多

Methods: Anticancer activity of fungal extracts was determined by observing its...Objective: To analyze anticancer activity of an ethyl acetate extract of endophytic fungi isolated from soursop leaf(Annona muricata L.). Methods: Anticancer activity of fungal extracts was determined by observing its toxicity against MCF-7(Michigan Cancer Foundation-7) cells in vitro by using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay method. At an extract concentration of 100 μg/m L, 4 isolates out of 12 showed high activity against the cancer cell growth. The four isolates were then selected for further IC50 determination, by measuring the inhibition of cancer cell proliferation at extract concentration of 25 μg/m L, 50 μg/m L, 100 μg/m L, 200 μg/m L and 400 μg/m L. Results: Results showed that isolate Sir-G5 had the highest anticancer activity with an IC50 of 19.20 μg/m L. The best isolates were screened again using a normal cell(Chang cells) to determine its toxicity against normal cells. Results indicated that the extracts do not affect the proliferation of normal cells. Molecular identification showed that the fungal isolate Sir-G5 has a close relationship with Phomopsis sp. Conclusions: The endophytic fungi isolated from soursop leaf has the potential to be used as a source of anticancer agents.展开更多

The response of insulin and insulin resistance to artichoke leaf extract (ALE) may be affected by TCF7L2-rs7903146 polymorphism. Objective: This study examined the effects of ALE supplementation on metabolic parameters of the TCF7L2-rs7903146 polymorphism in patients with metabolic syndrome (MetS). Design, setting, participants and interventions: This double-blind clinical trial was conducted on 80 patients with MetS in Sina Clinic, Khoy, Iran. The patients were randomized into ALE or placebo groups to receive either ALE (1800 mg/d as four tablets) or matching placebo for 12 weeks. Main outcome rneasures: Anthropometric indices, blood pressure, glucose and lipid profile levels were measured before and after the study. Moreover, patients were genotyped for TCF7L2 polymorphism. Results: ALE supplementation decreased insulin level and the homeostasis model assessment of insulin resistance (HOMA-IR) in patients with the "IT genotype of TCF7L2-rs7903146 polymorphism (P 〈 0.05). There was no significant interaction between blood pressure, glucose and lipid profile response to ALE supplementation. Conclusion: The responses of insulin and HOMA-lR to ALE supplementation have shown an interaction with single-nucleotide polymorphism rs7903146 in TCF7L2.
