1-油酸-2-棕榈酸-3-亚油酸甘油三酯(OPL)是人乳中主要的脂质组成,为满足婴幼儿配方食品母乳化需求,该试验探究了新型固定化脂肪酶ANL-MARE催化制备OPL的方法。试验成功制备和表征了固定化酶ANLMARE,并以ANL-MARE为生物催化剂,建立了一... 1-油酸-2-棕榈酸-3-亚油酸甘油三酯(OPL)是人乳中主要的脂质组成,为满足婴幼儿配方食品母乳化需求,该试验探究了新型固定化脂肪酶ANL-MARE催化制备OPL的方法。试验成功制备和表征了固定化酶ANLMARE,并以ANL-MARE为生物催化剂,建立了一种用三棕榈酸甘油三酯(PPP)、油酸(OA)和亚油酸(LA)高效酶催化制备富含OPL结构脂的方法。经单因素和响应面试验优化,获得OPL最优合成工艺为:PPP与总脂肪酸的摩尔比1:14.27,OA与LA摩尔比为1:0.76,脂肪酶添加量12.70%,反应温度50℃,反应4 h,此条件下产物中OPL相对含量为47.93%,2位棕榈酸(sn-2 PA)占总棕榈酸(PA)的质量分数(sn-2 PA相对含量)为71.69%。此外,固定化酶ANL-MARE与商业脂肪酶相比,表现出较好的催化合成OPL的活性。综上所述,固定化酶ANL-MARE具有催化制备OPL结构脂的重大潜力,为人乳脂替代脂的高效制备提供了新策略和理论基础。 展开更多

Improved LS+MAR hybrid method to UT1-UTC ultra-short-term prediction by using first-order-difference UT1-UTC

Accurate ultra-short-term prediction of the Earth rotation parameters(ERP)holds paramount impor-tance for real-time applications,particularly in reference frame conversion.Among them,diurnal rota-tion(UT1-UTC)which cannot be directly estimated through Global Navigation Satellite System(GNSS)techniques,significantly affects the rapid and ultra-rapid orbit determination of GNsS satellites.Pres-ently,the traditional LS(least squares)+AR(autoregressive)and LS+MAR(multivariate autoregressive)hybrid methods stand as primary approaches for UT1-UTC ultra-short-term predictions(1-10 days).The LS+MAR hybrid method relies on the UT1-UTC and LOD(length of day)series.However,the correlation between LOD and first-order-difference UT1-UTC is stronger than that between LOD and UT1-UTC.In light of this,and with the aid of the first-order-difference UT1-UTC,we propose an enhanced LS+MAR hybrid method to UT1-UTC ultra-short-term prediction.By using the UT1-UTC and LOD data series of the IERS(International Earth Rotation and Reference Systems Service)EOP 14 C04 product,we conducted a thorough analysis and evaluation of the improved method's prediction performance compared to the traditional LS+AR and LS+MAR hybrid methods.According to the numerical results over more than 210 days,they demonstrate that,when considering the correlation information between the LoD and the first-order-difference UT1-UTC,the mean absolute errors(MAEs)of the improved LS+MAR hybrid method range from 21 to 934μs in 1-10 days predictions.In comparison to the traditional LS+AR hybrid method,the MAEs show a reduction of 7-53μs in 1-10 days predictions,with corresponding improvement percentages ranging from 1 to 28%.Similarly,when compared to the traditional LS+MAR hybrid method,the MAEs have a reduction of 5-42μs in 1-10 days predictions,with corresponding improvement percentages ranging from 4-20%.Additionally,when aided by GNSS-derived LOD data series,the MAEs of improved LS+MAR hybrid method experience further reduction. 展开更多

SIL1 improves cognitive impairment in APP23/PS45 mice by regulating amyloid precursor protein processing and Aβ generation

SIL1,an endoplasmic reticulum(ER)-resident protein,is reported to play a protective role in Alzheimer’s disease(AD).However,the effect of SIL1 on amyloid precursor protein(APP)processing remains unclear.In this study,the role of SIL1 in APP processing was explored both in vitro and in vivo.In the in vitro experiment,SIL1 was either overexpressed or knocked down in cells stably expressing the human Swedish mutant APP695.In the in vivo experiment,AAV-SIL1-EGFP or AAV-EGFP was microinjected into APP23/PS45 mice and their wild-type littermates.Western blotting(WB),immunohistochemistry,RNA sequencing(RNA-seq),and behavioral experiments were performed to evaluate the relevant parameters.Results indicated that SIL1 expression decreased in APP23/PS45 mice.Overexpression of SIL1 significantly decreased the protein levels of APP,presenilin-1(PS1),and C-terminal fragments(CTFs)of APP in vivo and in vitro.Conversely,knockdown of SIL1 increased the protein levels of APP,β-site APP cleavage enzyme 1(BACE1),PS1,and CTFs,as well as APP mRNA expression in 2EB2 cells.Furthermore,SIL1 overexpression reduced the number of senile plaques in APP23/PS45 mice.Importantly,Y-maze and Morris Water maze tests demonstrated that SIL1 overexpression improved cognitive impairment in APP23/PS45 mice.These findings indicate that SIL1 improves cognitive impairment in APP23/PS45 mice by inhibiting APP amyloidogenic processing and suggest that SIL1 is a potential therapeutic target for AD by modulating APP processing. 展开更多

Cancer poses a significant threat due to its aggressive nature,potential for widespread metastasis,and inherent heterogeneity,which often leads to resistance to chemotherapy.Lung cancer ranks among the most prevalent forms of cancer worldwide,affecting individuals of all genders.Timely and accurate lung cancer detection is critical for improving cancer patients’treatment outcomes and survival rates.Screening examinations for lung cancer detection,however,frequently fall short of detecting small polyps and cancers.To address these limitations,computer-aided techniques for lung cancer detection prove to be invaluable resources for both healthcare practitioners and patients alike.This research implements an enhanced EfficientNetB1 deep learning model for accurate detection and classification using histopathological images.The proposed technique accurately classifies the histopathological images into three distinct classes:(1)no cancer(benign),(2)adenocarcinomas,and(3)squamous cell carcinomas.We evaluated the performance of the proposed technique using the histopathological(LC25000)lung dataset.The preprocessing steps,such as image resizing and augmentation,are followed by loading a pretrained model and applying transfer learning.The dataset is then split into training and validation sets,with fine-tuning and retraining performed on the training dataset.The model’s performance is evaluated on the validation dataset,and the results of lung cancer detection and classification into three classes are obtained.The study’s findings show that an enhanced model achieves exceptional classification accuracy of 99.8%. 展开更多

[Objectives]To establish a thin-layer chromatography(TLC)method for the determination of rubiadin-1-methyl ether in Yao Medicine Chuanlianzhu(Damnacanthus giganteus).[Methods]A silica gel G thin-layer plate was adopted for TLC.Petroleum ether(60-90℃)-chloroform-methanol-water(7:15:3:1)was used as the developing solvent and inspected under ultraviolet lamp(365 nm).The content was determined by Inertsil ODS-3 C 18 column(4.60 mm×250 mm,5μm),mobile phase:acetonitrile-0.2%phosphoric acid gradient elution,detection wavelength 277 nm,flow rate 1.0 mL/min,column temperature 30℃,injection volume 10μL.[Results]The spots of 10 Chuanlianzhu samples from different origins showed the same color at the same position as the control,and the spots were clear and specific.The injection volume of rubiadin-1-methyl ether showed a good linear relationship in the range of 2.90-145μg(R=0.9996).The average recovery rate of rubiadin-1-methyl ether in the low,medium and high dose groups of Yao Medicine Chuanlianzhu was 98.72%,and RSD=1.78%.[Conclusions]This method can effectively identify Yao Medicine Chuanlianzhu medicinal materials and accurately determine the content of rubiadin-1-methyl ether in the medicinal materials.It provides a scientific basis for the development and utilization of Yao Medicine Chuanlianzhu medicinal resources. 展开更多

Angiotensin II (Ang II) is the main mediator of the Renin-Angiotensin-System acting on AT<sub>1</sub> and other AT receptors. It is regarded as a pleiotropic agent that induces many actions, including functioning as a growth factor, and as a contractile hormone, among others. The aim of this work was to examine the impact of Ang II on the expression and function of α<sub>1</sub>-adrenergic receptors (α<sub>1</sub>-ARs) in cultured rat aorta, and aorta-derived smooth muscle cells. Isolated Wistar rat aorta was incubated for 24 h in DMEM at 37˚C, then subjected to isometric tension and to the action of added norepinephrine, in concentration-response curves. Ang II was added (1 × 10<sup>−5</sup> M), and in some experiments, 5-Methylurapidil (α<sub>1A</sub>-AR antagonist), AH11110A (α<sub>1B</sub>-AR antagonist), or BMY-7378 (α<sub>1D</sub>-AR antagonist), were used to identify the α<sub>1</sub>-AR involved in the response. Desensitization of the contractile response to norepinephrine was observed due to incubation time, and by the Ang II action. α<sub>1D</sub>-AR was protected from desensitization by BMY-7378;while RS-100329 and prazosin partially mitigated desensitization. In another set of experiments, isolated aorta-derived smooth muscle cells were exposed to Ang II and α<sub>1</sub>-ARs proteins were evaluated. α<sub>1D</sub>-AR increased at 30 and 60 min post Ang II exposure, the α<sub>1A</sub>-AR diminished from 1 to 4 h, while α<sub>1B</sub>-AR remained unchanged over 24 h of Ang II exposure. Ang II induced an increase of α<sub>1D</sub>-AR at short times, and BMY-7378 protected α<sub>1D</sub>-AR from desensitization. 展开更多

血清c5ar1、ALP及25(OH)D3的表达规律对股骨颈骨折术后骨折愈合情况的预测价值

目的:探讨血清补体成分5a受体1(Complement component 5a receptor,c5ar1)、碱性磷酸酶(Alkaline phosphatase,ALP)及25羟基维生素D(Vitamin D325-hydroxy monohydrate,25(OH)D3)的表达规律对股骨颈骨折术后骨折愈合情况的预测价值.方法... 目的:探讨血清补体成分5a受体1(Complement component 5a receptor,c5ar1)、碱性磷酸酶(Alkaline phosphatase,ALP)及25羟基维生素D(Vitamin D325-hydroxy monohydrate,25(OH)D3)的表达规律对股骨颈骨折术后骨折愈合情况的预测价值.方法:选取2019年1月至2022年12月本院治疗的股骨颈发生骨折且实施全髋关节置换术的98例患者作为研究对象.依据其术后6 m的影像学方面检查结果评估患者的骨折愈合情况,并将患者分成骨折愈合组(77例)和未愈合组(21例).于入院及术后42 d分别对两组的血清c5ar1、ALP、25(OH)D3水平进行测定,采用Pearson相关性分析c5ar1、ALP、25(OH)D3的相关性.采用受试者工作(Receiver operating characteristic,ROC)曲线分析血清c5ar1、ALP及25(OH)D3对股骨颈骨折术后骨折的愈合情况的预测价值.结果:骨折愈合组术后血清c5ar1、ALP及25(OH)D3的表达水平均高于术前(P<0.05),未愈合组术后血清c5ar1、ALP及25(OH)D3的表达水平均低于术前(P<0.05);Pearson相关性分析结果显示,c5ar1与25(OH)D3呈正相关关系(P<0.05);c5ar1、ALP、25(OH)D3诊断股骨颈骨折术后骨折愈合情况的敏感度分别为92.20%、62.30%、76.60%,特异度分别为90.50%、47.60%、52.40%,ROC曲线分析显示,c5ar1、ALP、25(OH)D3诊断股骨颈骨折术后骨折愈合情况的曲线下面积(area under curve,AUC)分别为0.953、0.515、0.765.结论:血清c5ar1、ALP及25(OH)D3的表达水平升高可促进股骨颈骨折术后骨折愈合,c5ar1与25(OH)D3呈正相关关系,c5ar1、25(OH)D3诊断股骨颈骨折术后骨折愈合情况具有较高的准确性,而ALP预测股骨颈骨折术后骨折愈合情况的准确性较低. 展开更多

目的:探讨三阴性乳腺癌中信号转导及转录激活因子1 (STAT1)、趋化因子CXC配体9 (CXCL9)及雄激素受体(AR)的表达水平、相关性及其临床病理特征。方法:收集已经确诊并经过改良根治手术切除的三阴性乳腺癌(TNBC)病例114例,通过免疫组织化... 目的:探讨三阴性乳腺癌中信号转导及转录激活因子1 (STAT1)、趋化因子CXC配体9 (CXCL9)及雄激素受体(AR)的表达水平、相关性及其临床病理特征。方法:收集已经确诊并经过改良根治手术切除的三阴性乳腺癌(TNBC)病例114例,通过免疫组织化学染色(SP)的方法,测定STAT1、CXCL9及AR它们三者在TNBC中的表达情况、相关性分析及其与临床相关资料的关系。结果:STAT1、CXCL9及AR在三阴性乳腺癌中的阳性率分别为90.4% (103/114)、3.5% (4/114)及11.4% (13/114);STAT1、CXCL9及AR它们三者表达不相关(P > 0.05);STAT1与年龄相关(P 0.05)。结论:STAT1在三阴性乳腺癌(TNBC)中显著表达,并且与年龄相关,有望成为TNBC治疗及预后评价的重要靶标。 展开更多

奥灰岩溶裂隙含水层是影响华北型煤矿深部开采的重要水害,在水-岩相互作用下奥灰含水层易导致煤层底板突水。为进一步认识奥灰岩溶突水问题,文章以新集矿区深部1煤层开采为例,利用矿区近些年最新积累的奥灰钻孔资料,选取断层强度指数、... 奥灰岩溶裂隙含水层是影响华北型煤矿深部开采的重要水害,在水-岩相互作用下奥灰含水层易导致煤层底板突水。为进一步认识奥灰岩溶突水问题,文章以新集矿区深部1煤层开采为例,利用矿区近些年最新积累的奥灰钻孔资料,选取断层强度指数、断层交叉点与尖灭点、含水层水压、富水性、隔水层等效厚度、脆塑比7个因素作为奥灰岩溶突水的主控因素,并结合层次分析法(analytic hierarchy process,AHP)确定各主控因素影响权重。运用地理信息系统(geographic information system,GIS)空间分析功能建立各主控因素专题图,通过对专题栅格图归一化处理,将各主控因素按照权重进行空间复合叠加,最终获得1煤层底板奥灰岩溶突水危险性评价分区结果。将评价结果与突水系数法计算结果对比分析可知,基于GIS的煤层底板突水危险性评价方法更符合矿区实际地质情况,可以为矿区深部煤层开采与水害防治工作提供参考依据。 展开更多

为利用干酪乳杆菌制备发酵菠萝果汁饮品,本研究对干酪乳杆菌LK-1(Lactobacillus casei LK-1,L. casei LK-1)的生长曲线、产酸、耐酸和耐糖等特性进行研究分析,并以活菌数和总酸为指标,通过单因素实验及响应面法考察L. casei LK-1发酵菠... 为利用干酪乳杆菌制备发酵菠萝果汁饮品,本研究对干酪乳杆菌LK-1(Lactobacillus casei LK-1,L. casei LK-1)的生长曲线、产酸、耐酸和耐糖等特性进行研究分析,并以活菌数和总酸为指标,通过单因素实验及响应面法考察L. casei LK-1发酵菠萝果汁的最佳条件。结果表明,L. casei LK-1在pH5~7和0%~10%质量分数葡萄糖环境下,具备良好的生长繁殖能力;在酸性(pH3)和高糖(40%质量分数葡萄糖)环境中培养3 h,其存活率分别为76%和71%;在MRS培养基中培养48 h,产酸量为5.35 g/kg,产酸能力良好,适用于果汁的发酵。L. casei LK-1发酵菠萝果汁的最佳条件为:初始pH6.8,发酵温度37℃,接种量1%(v/v),发酵时间30 h,此条件下制备的发酵菠萝果汁饮品活菌数为8.99±0.04 lg CFU/mL,总酸含量为7.16±0.26 g/kg,且色泽鲜亮,酸甜适口,风味较好,为L. casei LK-1在发酵食品中的进一步应用提供了理论和技术依据。 展开更多

目的通过活化β3AR-PKA通路,探讨成纤维细胞生长因子21(FGF21)对白色脂肪组织(WAT)棕色化的影响。方法C57BL/6小鼠分别采用标准小鼠饲料(16只)及45%千卡脂肪高脂饲料(16只)喂养。12周后,标准饲料喂养小鼠随机分为2组:对照组(NC组)注射0... 目的通过活化β3AR-PKA通路,探讨成纤维细胞生长因子21(FGF21)对白色脂肪组织(WAT)棕色化的影响。方法C57BL/6小鼠分别采用标准小鼠饲料(16只)及45%千卡脂肪高脂饲料(16只)喂养。12周后,标准饲料喂养小鼠随机分为2组:对照组(NC组)注射0.9%氯化钠溶液,注射CL316,243组(CL组)注射CL316,243(1 mg/kg);高脂饲料喂养小鼠随机分为2组:高脂饮食组(HFD组)注射0.9%氯化钠溶液,高脂饮食注射CL316,243组(HFD+CL组)注射CL316,243(1 mg/kg)。14周后,Western blot检测WAT蛋白激酶A(PKA)及棕色化相关蛋白(FGF21、PGC-1α及UCP1)表达;观察肝脏及脂肪组织病理学变化。培养3T3-L1前脂肪细胞,诱导成熟后分为3组:NC组,DMEM培养液;CL组,培养液+10μmol/L CL316,243;SU+CL组,培养液+50μmol/L SU5402+10μmol/L CL316,243。48 h后,检测蛋白激酶A(PKA)及棕色化相关蛋白表达,免疫荧光检测线粒体表达。结果与NC组比较,CL组PKA水平及棕色化相关蛋白升高;与CL组比较,HFD+CL组PKA水平及棕色化相关蛋白降低;而HFD+CL组相关蛋白表达高于HFD组。体外实验,与NC组比较,CL组及CL+SU组PKA水平显著升高;CL+SU组较CL组棕色化相关蛋白表达降低;免疫荧光结果显示,与NC组比较,CL组线粒体表达增多;而SU+CL组线粒体表达较CL组减少。结论活化β3AR-PKA通路可增加WAT中FGF21分泌,进而上调PGC-1α及UCP1表达,促进WAT棕色化。 展开更多

Exploring novel high molecular weight glutenin subunits(HMW-GSs)from wild related species is a strategy to improve wheat processing quality.The objective of the present investigation was to identify the chromosomes of the wheatalien introgression line N124,derived from the hybridization between Triticum aestivum with Aegilops kotschyi,and characterize the effects on quality-related traits.Fluorescence in situ hybridization karyotypes showed that N124 is a disomic 1U^(k)(1A)substitution line.Sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE)and reversedphase high-performance liquid chromatography verified N124 expressed two HMW-GSs of the Ae.kotschyi parent.PacBio RNA sequencing and phylogenetic analysis confirmed that the two HMW-GSs were U^(k)x and U^(k)y.Compared to the wheat parent,the substitution line had no obvious agronomic defects except fewer grains per spike but improved several major quality parameters.It can be served as a donor or bridge material for wheat quality improvement. 展开更多

Al-3Ti-1B master alloys were prepared at different processing parameters by the reaction of halide salts,and the grain refining response of Al-7Si alloy was investigated with Al-3Ti-B master alloy.The microstructure of master alloy and its grain refining effect on Al-7Si alloy were investigated by means of OM,XRD and SEM.Experimental results show that,the size of Al3Ti particles presented in Al-3Ti-1B master alloys increases with the increase of reaction temperature and decreases with the increase of cooling rate.The grain refining efficiency of Al-3Ti-1B master alloy on Al-7Si alloy is mainly attributed to heterogeneous nucleation of Al3Ti particles,and the morphology ofα(Al)changes from coarse dendritic to fine equiaxed.As a result,Al-3Ti-1B master alloy is prepared by permanent mold,and holding at 800 ℃for 30min,which has better grain refining performance on Al-7Si alloy. 展开更多
