The (His)6 fusion protein was identified by SDS-PAGE and Western blot. The recombinant protein was purified by affinity chromatography, and the calculated molecular mass(Mr) was 88000. The results of the sequence analysis indicate that the cloned gene(GeneBank accession No. EU124675) was a supplement and revision of the reported ACCase CT partial cDNA from Chinese Spring wheat plastid. The recombinant protein will be significant for us to investigate the recognizing mechanism between ACCase and herbicides, and further to screen new herbicides.展开更多

目的通过观察石榴皮多酚乳膏对金黄地鼠皮脂腺斑腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)、乙酰辅酶A羧化酶-1(acetyl-COA carboxylase-1,ACC-1)的影响,研究外用石榴皮多酚乳膏治疗痤疮的作用机制。方法将24只雄性金黄...目的通过观察石榴皮多酚乳膏对金黄地鼠皮脂腺斑腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)、乙酰辅酶A羧化酶-1(acetyl-COA carboxylase-1,ACC-1)的影响,研究外用石榴皮多酚乳膏治疗痤疮的作用机制。方法将24只雄性金黄地鼠随机分成4组,分别是空白组、基质组、石榴皮多酚乳膏组、阳性对照组,每组6只,分别于双侧皮脂腺斑处涂抹蒸馏水、基质、石榴皮多酚乳膏、0.025%维A酸乳膏,连续给药4周,2次/d。观察各组金黄地鼠皮脂腺斑面积变化,并取材观察组织病理学变化,免疫组化法检测各组AMPK、ACC-1的表达水平。结果与空白组和基质组相比,各治疗组皮脂腺体积变小,数量减少,排列疏松;AMPK水平均明显升高,差异有统计学意义(P<0.05);ACC-1水平明显降低,差异有统计学意义(P<0.05)。与阳性对照组相比,石榴皮多酚乳膏组ACC-1水平降低,差异有统计学意义(P<0.05)。结论石榴皮多酚乳膏治疗痤疮的作用机制可能与激活AMPK/ACC-1信号通路表达,从而改善皮脂稳态失衡有关。展开更多
