Involvement of ERK1/2 and p38 MAPK in up-regulation of 14-3-3 protein induced by hydrogen peroxide preconditioning in PC12 cells

Objective To investigate the protective effects of hydrogen peroxide preconditioning (HPP) on the pheochromocytoma (PC12) cells treated with 1-methyl-4-phenylpyridinium (MPP^+) and to explore the potential mechanisms. Methods The viability and apoptosis of PC 12 cells were determinded by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and 4′,6′-diamidino-2-phenylindole (DAPI) staining, respectively. The expressions of 14-3-3 protein and phospholylated p38 mitogen-activated protein kinase (MAPK) were determined by Western blot. Enzyme-linked immunosorbent assay (ELISA) was used to measure the activity of extracellular signal-regulated protein kinase 1/2 (ERK1/2). Results The cell viability decreased and the number of apoptotic cells increased dramatically in MPP^+ group compared with that in Control group. HPP induced a significant increase in cell viability and a marked decrease in population of apoptotic cells of the MPP^+- treated PC 12 cells, accompanied with up-regulation of 14-3-3 protein and increase of ERK 1/2 and p38 MAPK activities. The 14-3-3 protein expression was positively correlated with the phosphorylation of ERK1/2. Furthermore, inhibition of the ERK1/2 with PD98059 abolished the 14-3-3 protein up-regulation in PC 12 cells induced by HPP. Conclusion HPP protects PC 12 cells against MPP+ toxicity by up-regulating 14-3-3 protein expression through the ERK1/2 and p38 MAPK signaling pathways. 展开更多

As an important agronomic trait, inclination of leaves is crucial Ior crop architecture and grain yields. 10 understand the molecular mechanism controlling rice leaf angles, one rice leaf inclination2 (1c2, three alleles) mutant was identified and functionally characterized. Compared to wild-type plants, lc2 mutants have enlarged leaf angles due to increased cell division in the adaxial epidermis of lamina joint. The LC2 gene was isolated through positional cloning, and encodes a vernalization insensitive 3-like protein. Complementary expression of LC2 reversed the enlarged leaf angles of lc2 plants, confirming its role in controlling leaf inclination. LC2 is mainly expressed in the lamina joint during leaf development, and particularly, is induced by the phytohormones abscisic acid, gibberellic acid, auxin, and brassinosteroids. LC2 is localized in the nucleus and defects of LC2 result in altered expression of cell division and hormone-responsive genes, indicating an important role of LC2 in regulating leaf inclination and mediating hormone effects. 展开更多

Complexes 1~5 all feature one-dimensional chain structures, and complex 6 exhibits a zero-dimensional structure. Using 2'-Hydroxyacetophenone and as the raw material, 1-( 2-Hydroxyphenyl )-3-Phenyl-1, 3- Propanedione (HPPPD) was synthesized by esterification reaction and Fries rearrangement reaction. And the complexes of HPPPD with europium were also synthesized. The compounds were characterized by IR, thermo-gravimetric analysis and ^1H-NMR. The effects of phenolic hydroxyl on the luminescence properties of complexes were studied. The results show that the complexes emit strong red fluorescence and the fluorescence intensity of the ternary complex is better than that of the duality. But the complexes of HPPPD with Sm( Ⅲ ), Tb ( Ⅲ ), Dy ( Ⅲ ) emit weak fluorescence. The phenomenon was interpreted by electron-effect and energy-matching mechanism. Energy transfer mechanism of the luminescence in the complex was discussed. 展开更多

The title compound, [Ni(C8N3H7)3]4[Mo8O26]2·6H2O 1, has been synthesized from the reaction of 2-(1H-pyrazol-3-yl)-pyridine (L) with (NH4)2MoO4·2H2O and NiCl2·6H2O. Elemental analysis, IR, UV spectra and X-ray single-crystal diffraction were carried out to determine the composition and crystal structure of the compound. Crystal data: C96H96Mo16N36Ni4O58, Mr = 4451.97, monoclinic system, space group P21/n, a = 20.846(5), b = 14.825(5), c = 23.122(5) A ,β= 91.594(5)°, V = 7143(3)A^3, Z = 2, F(000) = 4344, Dc = 2.070 g/cm^3,μ = 1.968 mm^-1, R = 0.0452 and wR = 0.1056 for 17102 independent reflections (Rint = 0.0442) and 11074 observed reflections (I 〉 2σ(I)). Structural analysis indicates that two kinds of octamolybdates ([α-Mo8026]^4- and [β-Mo8026]^4-) co-exist in the compound. The fluorescence emission mechanism of the complex was discussed by fluorimetric spectra. The single-crystal structure was determined by X-ray diffraction analysis. The crystal belongs to the triclinic system,space group P1 with a = 0.9918(4),b = 1.1297(4),c = 1.1331(4) nm,α = 76.860(7),β = 75.105(6),γ = 89.022(7)°,V = 1.1936(8) nm^3,Z = 1,μ = 0.472 mm^-1,Dc = 1.319 g/cm^3,F(000) = 502 and Rint = 0.0913. In the title compound,the nickel(II) atom is four-coordinated with two nitrogen atoms from amide and two oxygen atoms from keto group. The structure consisted of anion part and a 2,3,5-(triphenyl) tetrazolium cation as counter ions. The copper complex has a distorted tetrahedral geometry, the CI(1)-Cu-CI(2) [124.33 (2°)] and Cl(3)-Cu-Cl(4) [131.03 (2°)] angles are significantly larger than the ideal tetrahedral. 背景与目的:肌动蛋白相关蛋白2/3复合体5(actin related protein 2/3 complex subunit5,ARPC5)参与肌动蛋白的组装,影响细胞的运动能力,在形成癌细胞侵袭性伪足的过程中起到重要作用。本研究通过基因沉默手段,探讨下调ARPC5基因表达对... 背景与目的:肌动蛋白相关蛋白2/3复合体5(actin related protein 2/3 complex subunit5,ARPC5)参与肌动蛋白的组装,影响细胞的运动能力,在形成癌细胞侵袭性伪足的过程中起到重要作用。本研究通过基因沉默手段,探讨下调ARPC5基因表达对肺鳞癌细胞株SK-MES-1增殖、侵袭和转移能力的影响。方法:应用阳离子脂质体转染法构建ARPC5基因沉默(si-ARPC5)的SK-MES-1肺鳞癌细胞株,qRT-PCR和蛋白质印迹法(Western blot)鉴定转染效率和表达情况。实验设置si-ARPC5实验组、空白对照组(mock组)及阴性对照组(NC组)。采用细胞技术试剂盒(cell couiting kit-8,CCK-8)法测定si-ARPC5对SK-MES-1细胞的增殖的影响;划痕修复实验测定si-ARPC5对SK-MES-1细胞迁移能力的影响;Transwell小室体外侵袭实验测定siARPC5对SK-MES-1细胞的侵袭能力的影响。结果:转染si-ARPC5后的SK-MES-1细胞在mRNA和蛋白水平上均下调ARPC5的表达。与mock组及NC组比较,si-ARPC5实验组SK-MES-1细胞增殖能力明显下降(t=7.993,t=8.681,P<0.05)。si-ARPC5实验组划痕修复率(43.32±0.23)%,而mock组和NC组划痕修复率分别为(73.11±0.43)%和(76.58±0.88)%,差异有统计学意义(t=7.348,t=10.614,P<0.05)。si-ARPC5组的SK-MES-1细胞穿膜数为(27±6)个,mock组为(101±11)个,NC组为(92±9)个。si-ARPC5组穿膜细胞数明显少于其他2组,差异有统计学意义(t=10.229,t=8.391,P<0.05)。结论:下调ARPC5基因的表达能够显著抑制肺鳞癌细胞株SK-MES-1的增殖、侵袭和转移能力。 展开更多

目的探讨缺氧诱导因子-1α(hypoxia inducible factor-1,HIF-1α)和BCL-2/腺病毒E1B19KDa相关蛋白3(Bcl2/adenovirus E1B 19 kD interacting protein 3,BNIP3)在中耳胆脂瘤中的表达及胆脂瘤上皮的凋亡情况。方法采用免疫组织化学方法检... 目的探讨缺氧诱导因子-1α(hypoxia inducible factor-1,HIF-1α)和BCL-2/腺病毒E1B19KDa相关蛋白3(Bcl2/adenovirus E1B 19 kD interacting protein 3,BNIP3)在中耳胆脂瘤中的表达及胆脂瘤上皮的凋亡情况。方法采用免疫组织化学方法检测30例中耳胆脂瘤标本与18例外耳道皮肤标本中HIF-1α和BNIP3蛋白的表达情况,使用末端脱氧核苷酸转移酶介导的dUTP缺口末端标记(terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling,Tunel)检测20例中耳胆脂瘤标本和18例外耳道皮肤标本的凋亡情况。使用Pearson相关分析检验HIF-1α和BNIP3蛋白之间的相关性。结果 HIF-1α在胆脂瘤组和对照组的平均光密度分别为0.16±0.07和0.08±0.03,两组比较差异有统计学意义(t=4.279,P<0.01);BNIP3在胆脂瘤组和对照组的平均光密度分别为0.16±0.08和0.11±0.06,两组比较差异有统计学意义(t=2.463,P=0.0185);经pearson相关分析,在胆脂瘤上皮中,HIF-1α和BNIP3之间呈正相关(r=0.418,P=0.003);Tunel染色中,凋亡指数在胆脂瘤组和对照组分别为(52.8±12.5)%和(9.99±2.97)%,两组比较差异有统计学意义(t=14.166,P<0.01)。结论 HIF-1α和BNIP3在中耳胆脂瘤中的异常表达可能与胆脂瘤的高凋亡特性有关。 展开更多

The title complex [Cu3L3(H2O)]DMFH2O (H2L = 4-(3-hydroxy-2-ethyl-4- pyridinone-1-yl)-aniline condensation salicylaldehyde) was obtained. The single-crystal X-ray study shows that it is a trinuclear compound [Cu3(C20H15N2O3)3(H2O)]DMFH2O. The coordi- nation sphere about each copper ion in the complex consists of two oxygen atoms from hydroxylpyridinone moiety of one ligand and one oxygen and one nitrogen atoms from salicyladehyde Schiff-base moiety of another ligand arranged in a slightly distorted square planar geometry. Among the three copper ions, one (Cu(2)) is coordinated by the other oxygen atom of water molecule on the fifth coordinate position to form a distorted square pyramid geometry. The crystal is of monoclinic, space group P21/c with a = 12.9202(5), b = 27.197(1), c = 17.0116(7) ? b = 100.588(1), V = 5875.9(4) 3, Z = 4, C63H57N7O12Cu3, Mr = 1294.78, Dc = 1.464 g/cm3, m = 1.146 mm-1, F(000) = 2668, R = 0.0784 and wR = 0.1546 for 6926 observed reflections with I > 2s(I). The differences of coordinate bond lengths are observed between anhydrous and hydrous units: in the former unit, the average bond lengths are 1.978 ?for CuN (azomethine), 1.883 ?for CuO (phenolic) in Schiff-base moiety, 1.959 ?for CuO (keto), and 1.919 ?for CuO (hydroxy) in hydroxypyridinone moiety; while those in the latter are longer with the following corresponding values: 1.985(5), 1.908(5), 1.993(5) and 1.919(4) ? respectively. Background: We investigated the immunohistochemical expression of Arp2/3 complex in 88 oral squamous cell carcinomas (OSCCs) and correlated it with the tumor type and grade. Materials and methods: This is a retrospective study of 88 cases of OSCCs (9 verrucous carcinomas, 4 carcinomas in situ, 37 well differentiated OSCCs, 32 moderately differentiated OSCCs, and 6 poorly differentiated OSCCs), stained with mouse monoclonal antibodies for Arp2 and Arp3. Staining was graded according to the extent of positivity by two observers. Results: Arp2 positivity was seen in 79 (89.77%) cases and Arp3 positivity in 78 (88.63%) cases. In positive cases, Arp2 expression was graded as (+) in 12.5%, (++) in 28.4% and (+++) in 48.86% of the cases;Arp3 was graded as (+) in 36.36%, (++) in 30.68% and (+++) in 21.59% of the cases. Conclusion: Expression of Arp2 and Arp3, indicative of the formation of the Arp2/3 complex, was seen in most OSCCs examined. Although no statistically significant correlation was found between the immunohistochemical expression of Arp2/Arp3 and tumor type or grade, the more common expression in invasive OSCCs than in non-invasive verrucous carcinomas and carcinomas in situ is indicative of its involvement in the biological behaviour of the lesions. The availability of Arp2/3 inhibitors that could be used in clinical practice warrants further study of the expression of Arp2/3 in OSCC. 展开更多

The complex [Fe(C14H14NO2)3]2H2O has been prepared by reaction of N-p-methylphenyl-3-hydroxy-2-ethyl-4-pyridinone with FeCl36H2O. A single-crystal X-ray study shows that the iron atoms lie in a trigonally distorted octahedral environment coordinated to the hydroxy and ketone oxygen atoms of three ligands in the mer configuration Mr=773.57(C42H46N3O8Fe). The crystal is hexagonal with space group P31c; a=15.943(2), c=17.612(4)? V=3877.0(12)?, Z=4, Dc=1.325g/cm3, m=0.445mm-1, F(000)=1634, R=0.0446, wR= 0.1154 for 3085 reflections with I >2s(I). The bond lengths from iron to oxygens are 1.980(1)?for the ketone oxygens and 2.071(1)?for the hydroxy oxygens. The molecule exhibits the expected propeller shape, and the angle of the trigonal twist is 48.37. The dihedral angles are 0.5(2)?between chelate ring plane and pyridine ring plane and 71.31(7)?between pyridine ring plane and benzene ring plane. X-ray single-crystal analysis revealed that the complex crystallizes in a monoclinic system, space group P21/c, a = 13.9707(10), b = 10.0435(7), c = 10.3910(6) ?, β = 91.181(3)o, V = 1457.70(17) ?3, Z = 4, C8H12Cl2N4S2Co, Mr = 358.17, Dc = 1.632 g/cm3, μ = 1.813 mm–1, F(000) = 724, the final R = 0.0710 and wR = 0.1224 for 1549 observed reflections with I > 2σ(I). The Co atom is coordinated by two S atoms from two 1-methylimidazoline-2(3H)-thione ligands and two chloride ions in a slightly distorted tetrahedral geometry. The intramolecular classical hydrogen-bonding interactions involving chloride ions and N–H groups of the heterocyclic thione ligands are observed. AP3M2在胃癌中的作用及临床意义分析

目的探讨接头相关复合体蛋白3亚基2(adaptor related protein complex 3 subunit mu 2,AP3M2)在胃癌中的表达情况及其临床意义。方法通过TCGA数据库、实时荧光定量PCR及免疫印迹实验检测AP3M2基因的mRNA及蛋白在胃癌组织中的表达情况。... 目的探讨接头相关复合体蛋白3亚基2(adaptor related protein complex 3 subunit mu 2,AP3M2)在胃癌中的表达情况及其临床意义。方法通过TCGA数据库、实时荧光定量PCR及免疫印迹实验检测AP3M2基因的mRNA及蛋白在胃癌组织中的表达情况。通过检测KM Plotter网站探索AP3M2的表达与胃癌患者预后的关系。通过平板克隆形成实验检测AP3M2对胃癌细胞的作用。结果TCGA数据库数据及本研究收集的胃癌患者组织显示AP3M2相比于癌旁组织,其在胃癌组织中的mRNA表达水平升高,同时AP3M2的蛋白在胃癌组织中的表达也有上调的现象。同时本研究通过KM Plotter网站分析得到AP3M2的mRNA表达水平与胃癌患者的预后有关,高表达AP3M2的胃癌患者其预后较低表达AP3M2的胃癌患者差。通过在AGS细胞(人胃腺癌细胞)中过表达AP3M2并检测其平板克隆形成能力的变化,发现过表达AP3M2可以增强胃癌细胞的克隆形成能力。结论AP3M2在胃癌中高表达,高表达AP3M2与胃癌患者的不良预后有关,且能增强胃癌细胞的克隆形成能力。 展开更多

In the present study, we constructed a lentivirus, FIV-CMV-GFP-miR-7-3, containing the microRNA-7-3 gene and the green fluorescent protein gene, and used it to transfect human glioma U251 cells. Fluorescence microscopy showed that 80% of U251 cells expressed green fluorescence. Real-time reverse transcription PCR showed that microRNA-7-3 RNA expression in U251 cells was significantly increased. Proliferation was slowed in transfected U251 cells, and most cells were in the G1 phase of the cell cycle. In addition, the expression of the serine/threonine protein kinase 2 was decreased. Results suggested that transfection with a lentivirus carrying microRNA-7-3 can effectively suppress epidermal growth factor receptor pathway activity in U251 cells, arrest cell cycle transition from GI phase to S phase and inhibit glioma cell growth. 展开更多

目的 探讨肌动蛋白相关蛋白2/3复合体亚基4(ARPC4)、环氧合酶2(COX2)在结直肠癌组织中的表达及与患者临床特征和预后的关系。方法 收集96例结直肠癌患者的病历资料,所有患者均采用免疫组织化学染色法检测ARPC4、COX2表达情况,比较不同... 目的 探讨肌动蛋白相关蛋白2/3复合体亚基4(ARPC4)、环氧合酶2(COX2)在结直肠癌组织中的表达及与患者临床特征和预后的关系。方法 收集96例结直肠癌患者的病历资料,所有患者均采用免疫组织化学染色法检测ARPC4、COX2表达情况,比较不同临床特征结直肠癌患者结直肠癌组织中ARPC4、COX2阳性表达率,采用回归分析法分析结直肠癌组织中ARPC4与COX2表达的相关性,比较不同ARPC4、COX2表达情况结直肠癌患者的3年生存率。结果 结直肠癌患者中ARPC4阳性表达率为61.46%(59/96),COX2阳性表达率为59.38%(57/96)。有淋巴结转移、低分化、Ⅲ~Ⅳ期结直肠癌患者结直肠癌组织中ARPC4、COX2阳性表达率均高于无淋巴结转移、中高分化、Ⅰ~Ⅱ期患者,差异均有统计学意义(P﹤0.05)。结直肠癌组织中ARPC4与COX2的表达呈正相关(r=0.618,P﹤0.05)。ARPC4阳性表达结直肠癌患者的3年生存率低于ARPC4阴性表达患者,COX2阳性表达结直肠癌患者的3年生存率低于COX2阴性表达患者,差异均有统计学意义(P﹤0.05)。结论结直肠癌组织中ARPC4与COX2的表达呈正相关,其表达与结直肠癌患者的临床特征及预后有关。 展开更多
