Aeromonas hydrophila is a Gram-negative pathogen that can infect various fish,including largemouth bass(Micropterus salmoides),which have caused huge economic losses.In present study,largemouth bass anti-A.hydrophila ...Aeromonas hydrophila is a Gram-negative pathogen that can infect various fish,including largemouth bass(Micropterus salmoides),which have caused huge economic losses.In present study,largemouth bass anti-A.hydrophila antibodies were produced,then a highly immunogenic outer membrane proteins,AHA_3793,was identified by combined western blotting and mass spectrometry analysis.Moreover,AHA_3793 was expressed,and its immunogenicity was further verified by western blotting.Subsequently,the protective efficacy of AHA_3793 were evaluated in largemouth bass.The results showed that rAHA_3793 could produce a relative percentage survival(RPS)of 61.76% for largemouth bass against A.hydrophila challenge.ELISA analysis showed the specific serum antibodies of largemouth bass against rAHA_3793 and A.hydrophila in vaccinated group in weeks 4 and 5 after immunization were significantly higher than those in control group,which suggested that rAHA_3793 induced production of specific serum antibodies against rAHA_3793 and A.hydrophila.The qRT-PCR analysis showed that expressions of CD4-2 and MHC Ⅱα were also significantly up-regulated after immunization.These results collectively demonstrated that rAHA_3793 could induce a strong humoral immune response of largemouth bass,and then produce high immune protection ef fects against A.hydrophila infection.展开更多

The fishes were injected either intraperitoneally or intra...The effects of injectable killed Aeromonas hydrophila on lymphocyte populations and on serum proteins of juvenile common carp, Cyprinus carpio, were examined. The fishes were injected either intraperitoneally or intramuscularly with killed A. hydrophila bacterin isolated from a diseased fish. 15 days after injection the lymphocyte count was significantly higher in the kidney of intraperitoneally bacteria injected fishes than in the normal saline injected fishes (p 0.05). In the intramuscular bacteria injected fishes in comparison with the normal saline group, the lymphocyte count was significantly higher only in the blood (p 0.05). In comparison between the serum protein parameters in different groups, in intraperitoneal injection of killed-bacteria, total albumin as well as the ratio of albumin/globulin was greater than the control group (p 0.05). In intramuscular injection, the normal saline-injected group, amount of α?-globulin was significantly higher than bacteria-injected group (p 0.05).展开更多
The aim of the study was to investigate the antibacterial potential of garlic(Allium sativum),pinus(Pinus elliottii),eucalyptus(Eucalyptus sp.),yerba mate(Ilex paraguariensis)and guava tree(Psidium guajava Linn.)in vi...The aim of the study was to investigate the antibacterial potential of garlic(Allium sativum),pinus(Pinus elliottii),eucalyptus(Eucalyptus sp.),yerba mate(Ilex paraguariensis)and guava tree(Psidium guajava Linn.)in vitro and the effect of yerba mate supplementation on Oreochromis niloticus physiology and resistance to Aeromonas hydrophila.The in vitro antibacterial potential of the plants was verified by the minimum inhibitory concentration(MIC)method and by the antimicrobial sensitivity test(TSA).The in vivo assay was performed using diet supplementation with yerba mate and challenge with A.hydrophila.After the supplementation and challenge periods biochemical,immune and physiological parameters were evaluated.The in vitro assays showed that only yerba mate presented antibacterial potential against A.hydrophila and no one of the plant extracts were effective against Streptococcus agalactiae.The use of yerba mate in the diet did not influence the immune and metabolic status of O.niloticus.Also,the supplementation did not act as a growth promoter for the fishes.Considering the challenge of O.niloticus with A.hydrophila,the yerba mate did not improve the resistance of the fishes to the bacteria.The yerba mate extract presented potential to be used as an antibacterial agent for fish diseases.Also,more detailed studies are necessary to stablish effective doses and times required for fish treatments.展开更多
